Die Erfindung betrifft den in den Ansprüchen senkende Wirkung des I -[N-Methyl-N-(3,4-dimethoxy-
gekennzeichneien Gegenstand. benzyl)-/?-aminoäthyl]-theobromin beschrieben.
Es ist bekannt, daß bestimmten, basisch substituierten Es wurde nun gefunden, daß I -[N-Methyl-N-iJJ-phe·

bedeuten sowie deren Salze mit physiologisch verträglichen
Säuren starke antiallergische sowie bronchodilatorische Eigenschaften aufweisen.
Als physiologisch verträgliche Säuren kommen u. a. in
Betracht: Salzsäure, Schwefelsäure, Phosphorsäure, Essigsäure, Malonsäure, Bernsteinsäure, Fumarsäure,
Zitronensäure.
Bei der Verfahrensweise a) arbeitet man zweckmäßig in alkoholischer Lösung mit Alkalialkoholaten als
halogenwasserstoffbindende Mittel oder in alkoholischwäßriger Lösung mit Natronlauge, wobei man die
Reaktionspartner einige Zeit unter Rühren am Rückflußkühler erhitzt Als besonders vorteilhaft hat es sich
erwiesen, eine Suspension von Benzimidazol, trocknem Kaliumcarbonat und der Verbindung II in einem inerten
Lösungsmittel — vorzugsweise Aceton oder Butanon — bei Siedetemperatur zu rühren.
Bei dem Verfahren b) hat es sich bewährt, als halogenwasserstoffbindendes Mittel ein zweites Moi
der Base III zuzufügen und die Mischung der beiden Reaktionspartner einige Stunden im ölbad auf etwa
1400C zu erhitzen. Fügt man darauf ein geeignetes Lösungsmittel (z. B. Toluol oder Xylol) in der Wärme zu,
so scheidet sich ein Mol Halogenv asserstoffsalz der Base Ml kristallin aus und kann abgetrennt werden. Das
Filtrat enthält dann das basische Verfahrensprodukt Es ist auch möglich, die Umsetzung in siedendem Xylol,
oder bei reaktionsfähigem Halogen in Toluol durchzuführen. Man ka...i in diesem Falle statt eines
zusätzlichen Mols der Bise II' auch trockenes Kaliumcarbonat oder tertiäre Basen, wie Pyridin oder
Triethylamin, als säurebindendes Mitte' einsetzen. .
Die gleichen Arbeitsbedingungen gelten für das Verfahren c).
Die Einführung der Methylgruppe in die sekundären Basen der allgemeinen Formel V zur Überprüfung in die
erfindungsgemäßen Verbindungen I kann gemäß Verfahrensweg d) durch Behandeln mit Methylhalogeniden
oder Dimethylsulfat erfolgen, ferner durch Kochen mit Formalin bzw. Paraformaldehyd und
Ameisensäure. Die Methylierung kann auch in der Weise vorgenommen werden, daß man die mit
Formaldehyd versetzte alkoholische Lösung der sekundären Base V in Gegenwart von Palladium- oder
Nickelkatalysator mit Wasserstoff schüttelt, oder mit amalgamiertem Aluminium oder Natriumboranat oder
einem anderen geeigneten Reduktionsmittel behandelt.
Die neuen Verbindungen besitzen ein breites histaminolytisches Wirkungsspektrum, das sich gegen
endogene (anaphylaktische) und exogene Histaminreaktionen richtet. Diese Eigenschaften lassen sie als
wertvolle Heilmittel bei der Behandlung einer ganzen' Reihe von Krankheiten, z. B. Asthma bronchiale.
Urticaria. Heufieber. Hauterscheinungen und Allergosen, erscheinen.
Die histaminolytische Wirksamkeit wurde nach der
Methode von Konzett H.. Rossler R.. Arch. cxp. Path.
Pharm, 195. 71 (1940) am Bronchospasrrms des
Meerschweinchens geprüft. In der Tabelle unter (A) ist die Dosis aufgeführt, die den Bronchospasmus des
Ausgangswertes nach Substanzgabe um die Hälfte herabsetzt (KD50).
Am isolierten Meerschweinchenileum zeigen die neuen Verbindungen gleichfalls eine hohe histaminolytisehe
Wirksamkeit, wie nach der Methode von Magnus [R, Pflügers Arch. ges. Physiol. 102, 123 (1904)] gezeigt
werden kann. In der Spalte (B) der Tabelle sind die Konzentrationen in μg/ml Badflüssigkeit aufgeführt, die
eine vollständige Hemmung des Ausgangsspasmus bewirken.
Für die therapeutische Anwendung können die neuen Stoffe als Ampullen, Tabletten, Dragees und Inhalat
eingesetzt werden.

Die Herstellungsverfahren werden durch nachstehende Ausführungsbeispiele erläutert
Die Temperaturen sind in "Celsius angegeben. Die Schmelzpunkte sind korrigierte Werte.
1-[N-MethyI-N-(P-3.4-dichlorphenyläthyl)-3-aminopropyl]-benzimid,"zol
A) Herstellung des Ausgangsmaterials
I. N-Methyl-/?-3,4-dichlorphenyläthylamin
I. N-Methyl-/?-3,4-dichlorphenyläthylamin
Eine Lösung von 41.2 g (0.217 MoI) 0-3,4- Dichlorphenyläthylamin
[F. Benington et al. J. Org. Chem. 25 (1960), 2066] wird mit 11,8g (1,2x0,217 Mol) Ameisensäure
versetzt und 4 Stunden am Wasserabscheider unter Rückfluß gekocht. Dabei werden 8,4 ml Wasser entfernt.
Man verdampft das Toluol, destilliert den Rückstand im Vakuum und erhält 41.0 g (90% der Theorie) der
N-Formyl-Verbindung vom Kp= 168- 17Γ/0.02 Torr. 41.0 g (0,188 Mol) des so erhaltenen Formamids werden
in 200 ml Tetrahydrofuran gelöst und zu einer im Dreihalskoiben siedenden Suspension von 11 g
(2x0,75x0.188 Mol) Lithiumalanat in 200 ml Tetrahydrofuran
unter Rühren getropft. Man hält weitere 4 Stunden am Sieden und zersetzt durch Eintropfen von
Wasser und Natronlauge. Man saugt vom Aluminiumhydroxid ab. dampft das Filtrat ein. löst den Rückstand in
2 N-Salzsäure und schüttelt mit Toluol. Die wäßrige Lösung wird mit konzentrierter Natronlauge versetzt,
die Base in Toluol aufgenommen, das Lösungsmittel verdampft und der Rückstand im Vakuum destilliert. Bei
96-97°/0,3 Torr gehen 32,3g (86% der Theorie) N-Methyl-/3-3,4-dichlorphenyläthylamin vom n" 1.5489
über. Das Hydrochlorid (aus Alkohol und Äther) schmilzt bei 197-200°.
11. N-Methyl-N-(j3-3,4-dichlorphenyläthyl)-3-aminochlorpropan
133,5 g (2 χ 0328 MoI) N-Methyl-p-3,4-dichlorphenyläthylamin
werden mit 31 g (0,328 MoI) 3-Chlorpropanol 5
in Toluol 24 Stunden unter Rückfluß und Rühren gekocht. Nach Abtrennen des ausgeschiedenen N-Methyl-ß-S/t-dichiorphenyläthylamin-Hydrochlorids
wird dasN-Methyl-N-p-hydroxypropylJ-jii-S^dichlorphenyläthylamip
das nach Abdampfen des Lösungsmittels als öl zurückbleibt, in Chloroform aufgenommen, tropfenweise
mit 43,0 g (1,1x0,328 MoI) Thionylchlorid versetzt und 2 Stunden unter Rückfluß gekocht Man
nimmt das Reaktionsprodukt nach dem Abdampfen des Chloroforms in heißem Aceton auf. Beim Abkühlen fällt
das Hydrochlorid des N-Methyl-N-(/?-3,4-dichIorphenyläthyl)-3-aminochlorprupan
vom Fp= 145-146° aus. Ausbeute 73,9 g = 76,2% der Theorie. Die freie Base
siedet bei 112 — 116°/0,2 Torr. Der Brechungsindex
beträgt n'i 1,5392.
B) Herstellung des Endproduktes
1-[N-Mnthyl-N-G3-2,4-dichlorphenyläthyl)-3-aminopropyl]-benzimidazol
A) Herstellung des Ausgangsmaterials
1. N-Methyl-^/l-dichlorphenyläthylamin

168 g (0,53 Mol) N-Methyl-N-(JJ-3,4 dichiorphenyl-8thyl)-3-aminochlorpropan-Hydrochlorid
werden in die freie Base übergeführt, die in 100 ml Toluol gelöst
innerhalb 30 Minuten zu einer unter Rückfluß siedenden Mischung von 250 ml Isopropanol, 21,4g (033 MoI)
Natriumhydroxid. 30 ml Wasser und 63 g (033 MoI)
Benzimidazol getropft wird. Die Reaktionslösung wird weitere 3 Stunden unter Rückfluß gekocht. Man saugt
vom ausgeschiedenen Natriumchlorid ab und dampft das Filtrat ein. Der ölige Rückstand wird in Toluol
gelöst. Chlorwasserstoffgas wird bis zur stark sauren Reaktion eingeleitet. Es scheidet sich das
1 -[N-Methyl-N-^-S/t-dichlorphenyläthyl^-aminopropyl]-benzimidazol-Hydrochlorid
als weißes, kristallines Pulver aus. Nach Umlösen aus Isopropanol erhält man 187 g(81%derTheorie)desSalzes, Fp =142-144°.
Die Kondensation von Benzimidazol mit N-Methyl-N-(/3-3,4-dichlorphenyläthyl)-3-aminochlorpropan
läßt sich au.h mit trockenem Kaliumcarbonat als halogenwasserstoffbindendem
Mittel in Aceton oder Butanon durchführen.

Man erhält diese Verbindung aus ß-2A- Dichlorphenylathylamin
[W. N. Cannon et al.. J. Org. Chem. 22 (1957)
1323] durch Reduktion ihrer N-Formylverbindung mit Lithiumalanat. Das Hydrochlorid schmilzt bei
183-185°.
II. N Mcthyl-N-(0-2.4-dichlorphenyläthyl)-3-aminochlorpropan
Diese Verbindung wird analog Beispiel IA) Il aus N-Methyl-^-2,4-dichlorphenyläthylamin durch Kondensation
mit 3-Chlorpropanol und anschließende Behandlung des Aminoalkohols mit Thionylchlorid erhalten. Ihr
Bioxalat schmilzt bei 152 - I 54°.
B) Herstellung des Endproduktes
66,6 g (0,21 Mol) N-Methyl-N-(0-2,4-dichlorpheuyläthyl)-3-aminochloiyropan-Hydrochlorid
und 25,0 g (0,21 Mol) Benzimidazol werden analog Beispiel I kondensiert und aufgearbeitet. Die Base wird zur
Reinigung in das Maleinat übergeführt, das nach Umlösen aus Isopropanol bei 112-114° schmilzt.
Ausbeute: 74 g (59% der Theorie),
Ausbeute: 74 g (59% der Theorie),
I-[N-MethyI-N-(j3-4-chlorphenyläthyl)-3-aminopropyl]-benzimidazol
148 g (0,525 Mol) N-Methyl-N-(j8-4-chIorphenyläthyl)-3-aminochIorpropan-Hydrochlorid
und 62 g (0325 Mol) Benzimidazol werden analog Beispiel ! kondensiert und aufgearbeitet. Die entstandene Base
wird in das Maleinat überführt und aus Aceton umkristallisiert.
Es werden 218,5 g = 75,5% der Theorie des Salzes erhalten, Fp =122-124°.
1 -[N-MethyI-N-0?-3.4-diir, i.hoxyphenyläthyl)-3-aminopropyi
j-beiizi· nidazol
78,6 g (0,255 MoI) N-Methyl-N-(j?-3,4-dimethoxyphenyläthyl)-3-aminochlorpropan-Hydrochloriu
werden mit 30 g (0,255 Mol) Benzimidazol analog Beispiel 1 kondensiert und aufgearbeitet. Die Base wird in Toluol
gelöst und in diese Lösung wird Chlorwasserstoffgas eingeleitet, wobei sich das Hydrochlorid als weißes
Kristallpulver ausscheidet Nach Umlösen aus Äthanol schmilzt es bei 120-122°.
Ausbeute: 67.7 g (623% der Theorie)
l-[N-Methyl-N-(0-3-chlor-4-methoxy-phenyläthyl)-3-aminopropyl]-benzimidazol
In einen Dreihalskolben werden 33 g (0,143 Mol) l-P-ChlorpropylJ-benzimidazol-Hydrochlo-id, 200 ml
Toluol und 28,6 g (0,284 Mol) Triethylamin gegeben. Die Mischung wird 1 Stunde lang bei Zimmertemperatur
gerührt und anschließend mit 28.6 g (0,143 Mol) N-Methyl-j9-3-chlor-4-methoxy-phenyläthylamin (M. Julia
et al. Bull. Soc. chim. I- rance 1966.1335) versetzt. Das
Reaktionsgemisch wird dann 25 Stunden lang unter Rückfluß am Sieden gehalten. Man saugt vom
ausgeschiedenen Triäthylamin-Hydrochlorid ab und destilliert das Toluol ab. Man erhält 29.4 g (57,6% der
Theorie) eines Öles.
Die Charakterisierung des Öles erfolgte durch die Überführung in das Bioxalat. welches bei 155-156°
(Methanol) schmilzt.
1-[N-Meth>i-N-(j3-3-methoxy-4-chlotphenyläihyl)-3-aminopropyl]-benzimidazol
A) Herstellung des Ausgangsmaterials

l.^-3-Methoxy-4chlorphenyläthylamin
150 g (0,83 Mol) 3-Methoxy-4-chlorbenzylcyanid (S. Munavalli et al.. Bull. Soc. Chim. Fr. !966 (!0) 3311-18)
werden in 600 ml Methanol, das 50 g Ammoniak enthält gelöst und mit Raney-Kobalt bei 5O°/7O at H? hydriert.
Die tiase wiro als Hydrochlorid isolier;. Man erhält
202 g (91% der Theorie) ,9-3-Mcthoxy-4-clilorphenyläthylamin-Hydrochlorid
vom Fp= 164 — 16b'.

Il N-Methyl-^-B-methoxy^-chlorphenyläthylamin
155,5 g (0,7 Mol) des vorstehend erhaltenen /?-3-Methoxy-4-chlorphenyläthylamin-Hydrochlorids
werden in die Base übergeführt und gemäß Vorschrift Beispiel I A) I durch Behandeln mit Ameisensäure und anschließende
Reduktion des erhaltenen Formamids mit Lithiumalanat am Stickstoff methyliert. Man erhält 147 g (89% der
Theorie) N-Methyl-jJO-methoxy^-chlorphenyläthylamin-Hydrochlorid
vom Fp = 147 — 148°.
B) Herstellung des Endproduktes
44,4 g (0,192 Mol) l-p-ChlorpropyO-benzimidazol
und 38,4 g (0,192 Mol) N-Methyl-^-niethoxy^-chlorphenyläthylamin
werden analog Beispiel 5 mittels Triäthylamin kondensiert und aufgearbeitet. Man erhält
47.3 g (69% der Theorie) eines Öles.
Die Charakterisierung des Öles erfolgte durch die Überführung in das Bioxala», welches bei
Fp= 178— 179" (Methanol)schmilzt.
l-[N-Methyl-N-(0-3,4-dichlorphenyläthyl)-3-aminopropyl]-benzimidazol
A) Herstellung des Ausgangsmaterials
l-[N-(/}-3,4-dichlorphenyIäthyl)-3-aminopropyl]-benzimidazol
In einen Dreihalskolben werden 30 g (0,129 Mol) l-(3-Chiorpropyl)-benzimidazol-Hydrochlorid, 200 ml
Toluol und 26,1 g (0.258 Mol) Triäthylamin gegeben. Die
Mischung wird I Stunde lang bei Zimmertemperatur gerührt. Anschließend werden 24.6 g (0.129 Mol)
^-3,4-Dichlorphenyläthylamin zugegeben. Es wird 25
Stunden unter Rückfluß gekocht. Man saugt vom ausgeschiedenen Triäthylamin-Hydrochlorid ab und
dampft das Filmt unter vermindertem Druck ein. Der ölige Rückstand wird in 200 ml Toluol aufgenommen
und Chlorwasserstoffgas bis zur sauren Reaktion eingeleitet. Es scheiden sich Kristalle ab.die aus Äthanol
umkristallisiert werden.
Man erhält 32,5 g (59,5% der Theorie) l-[N-(/9-3,4-Dichlorphenylethyl)-i-aminopropyl]-benzimidazol-Hydrochlorid
vom Fp = 236-239°.
B) Herstellung des Endproduktes
26.1 g (0.075 Mol) l-[N(/J-3,4-Dichlorphenyläthyl)-3-aminopropylj-benzimidazol
(;ius dem vorstehend beschriebenen Hydrochlorid freigesetzt) werden in 140 ml
Äthanol gelost und mit 5.5 g (0.12 Mol) Ameisensäure
versetzt. Zu dieser Mischung gibt man unter Eiskühlung innnerhalb von 20 Minuten 8,36 g 35%ige (0,098 Mol)
wäßrige Formalinlösung erhitzt anschließend 2 Stunden unter Rückfluß. Der Abdampfrückstand der Reaktionslösung wird in verdünnter Salzsäure aufgenommen und
mit Toluol geschüttelt. Aus der wäßrig-salzsauren Lösung scheidet man mit Natronlauge die Base ab, die in
Toluol aufgenommen und getrocknet wird. Beim Einleiten vnn Chlorwasserstoffgas in die Toluollösung
scheidet sich das 1-[N-Methyl-N-(/?-3.4-dichlorphenyläthyl)-3-aminopropyl]benzimidazol-Hydrochlorid
aus. Es wird aus Isopropanol umkristallisiert. Man erhält 25,5 g (78.2% der Theorie) des Salzes. Fp= 142-144°.

- Patentansprüche:
1. !-[N-Methyl-N-OS-phenyläthyO-a-aminopropyll-benzimidazol-Abkönimlinge der allgemeinen Fomiel IR2CH3N—CH2-CH2-CH2-N—CH2-CH2-<fR1 ein Chloratom oder eine Methoxygruppe und R2 ein Wasserstoff- oder Chloratom oder eine Methoxygruppe - 2. Verfahren zur Herstellung der 1-[N-Methyl-N-QJ-phenyläthyl)-3-aminopropyI]-benzimidazol-Abkömmünge gemäß Anspruch 1, dadurch gekennzeichnet, daß man in an sich bekannter Weise
