Description
Es ist bekannt, daß man 5-Cyan-uracile ausÄthoxymethylencyanacetylurethanen
mit Ammoniak oder einem primären Amin oder aus /?-(S-Äthyl-isothioureido)
- α - cyan - acrylsäure - (S - äthylisothioureid) in Gegenwart von Salzsäure oder aus 2-Äthylthio-4-hydroxy-5-cyan-pyrimidin
durch Verseifen der Äthylthiogruppe erhält.
Diese Verfahren sind aber technisch von geringem Interesse, da sie von Verbindungen ausgehen, die
teils mit unbefriedigenden Ausbeuten, teils über mehrere oft uneinheitlich verlaufende Reaktionsstufen
hergestellt werden müssen.
Es wurde nun gefunden, daß man 5-Cyan-uracile der allgemeinen Formel I

in der R1 ein Wasserstoffatom oder eine niedere
Alkylgruppe bedeutet, leicht und in sehr guten Ausbeuten herstellen kann, wenn man eine Verbindung
der allgemeinen Formel II

in der R2 und R3 je einen niederen Alkylrest bedeuten
oder zusammen mit dem Stickstoffatom einen nicht aromatischen, gegebenenfalls weitere Heteroatome
aufweisenden Ring bilden, R4 einen niederen Alkoxyrest oder eine Dichlorphosphinylgruppe und Xe das
Anion einer niederen Alkylschwefelsäure oder das Chlorion bedeutet, in Gegenwart einer Base in einem
Lösungsmittel oder im Überschuß der Verbindung der allgemeinen Formel II bei Temperaturen von
etwa —10 bis 1200C mit Cyanacetylharnstoff umsetzt
und die erhaltene Verbindung der allgemeinen Formel III

bei 30 bis 1500C in einem polaren Lösungsmittel
mit einer alkalisch reagierenden Verbindung behandelt.
Vorzugsweise stellt R1 ein Wasserstoffatom dar.
R2 und R3 können zusammen mit dem Stickstoffatom
ζ. B. einen 5gliedrigen Ring, wie den Pyrrolidinring, oder einen 6gliedrigen Ring, wie den Piperidin-
oder den Morpholinring, bilden, bevorzugt ist R2 und R3 je eine Methyl- oder Äthylgruppe.
Für R4 ist die Methoxy- oder Äthoxygruppe und
für X das Methyl- oder Äthylsulfation bevorzugt.
Die als Ausgangsstoffe verwendbaren Verbindungen der allgemeinen Formel II sind zum größten
Teil bekannt, oder sie können auf an sich bekannte Art und Weise aus Amiden der allgemeinen Formel

und niedrigen Dialkylester der Schwefelsäure, z. B. Dimethylsulfat oder Diäthylsulfat, oder mit Phosphoroxychlorid
hergestellt werden.
Der Cyanacetylharnstoff ist bekannt oder kann nach an sich bekannten Methoden hergestellt werden,
beispielsweise durch Umsetzung von Cyanessigsäure mit Harnstoff in Essigsäureanhydrid.
Die Umsetzung der Verbindungen der allgemeinen Formel II mit dem Cyanacetylharnstoff zu Verbindungen
der allgemeinen Formel III erfolgt in einem polaren, wasserarmen bis wasserfreien organischen
Lösungsmittel, in Gegenwart von Verbindungen, die in Wasser alkalisch reagieren, und bei Temperaturen
von etwa —10 bis 1200C. Als polare organische
Lösungsmittel eignen sich z. B. Amide niederer Fettsäuren, insbesondere Dimethylformamid, oder niedere
Alkanole, wie Methanol, Äthanol, Glykol, Methylcellosolve oder Äthylcellosolve, Dialkylsulfoxyde, wie
Methylsulfoxyd, oder auch Acetonitril. Die Umsetzung kann auch in apolaren organischen Lösungsmitteln
oder im Überschuß der Verbindung der allgemeinen Formel II durchgeführt werden. Als geeignete
apolare organische Lösungsmittel seien beispielsweise genannt: Ketone, wie Aceton, Äther, wie Dioxan
oder Tetrahydrofuran, oder Kohlenwasserstoffe, wie Toluol, die gegebenenfalls halogeniert sein können,
wie Chlorbenzol. Geeignete, in Wasser alkalisch reagierende Verbindungen sind beispielsweise sekundäre
und insbesondere tertiäre aliphatische Amine, wie Trimethylamin oder Triäthylamin, Alkalicarbonate,
z. B. Natrium- oder Kaliumcarbonat, Alkalihydroxyde, wie Natrium-, Kalium- oder Lithiumhydroxyd,
Natriumamid oder Alkalialkoholate, insbesondere -alkylate, wie Natrium- oder Kalium-methylat oder;
-äthylat.
Das Zwischenprodukt der allgemeinen Formel III kann man entweder isolieren oder direkt zur ringförmigen
Verbindung der Formel I umsetzen. Der Ringschluß erfolgt mit Vorteil in polaren Lösungsmitteln,
z. B. in den vorstehend genannten oder in Wasser, und in Gegenwart von in Wasser alkalisch reagierenden
Verbindungen, wie von Alkalicarbonaten, Alkalialkoholaten oder wäßrigen Alkalihydroxyden,
z. B. von Natrium- oder Kalium-hydroxyd, und bei einer Temperatur von 30 bis 1500C, vorzugsweise
50 bis 1000C. Die Cyclisierung erfolgt unter Abspaltung
der Gruppe
—N
Die nach dem neuen Verfahren erhaltenen 5-Cyanuracile der allgemeinen Formel I sind wertvolle Zwischenprodukte
für die Synthese von Farbstoffen. So erhält man aus dem 5-Cyanuracil der allgemeinen
Formell, in der R1 ein Wasserstoffatom bedeutet,
durch Verseifung der Cyangruppe die Uracil-5-carbonsäure und aus letzterer durch Ersetzen der Hydroxylgruppen
durch Halogen mittels Phosphorhalogenid
das für die Herstellung wertvoller Reaktivfarbstoffe geeignete Farbstoffzwischenprodukt 2,4-Dichlor-pyrimidin-5-carbonsäurechlorid.
In den folgenden Beispielen sind die Temperaturen in Celsiusgraden angegeben.
12,7 g Cyanacetylharnstoff werden in 50 ml Dimethylformamid
gelöst und mit 24 g der Verbindung aus Dimethylformamid und Dimethylsulfat (hergestellt nach B. 96, 1354 [1963]) vereinigt. Man
kühlt dieses Gemisch auf 0° ab und läßt dazu bei 0 bis 10° 12 g Triäthylamin zutropfen. Dann rührt
man das Reaktionsgemisch noch 2 Stunden bei 20 bis 30°, saugt den während der Reaktion ausgefallenen
N,N - Dimethylaminoniethylen - cyanacetylharnstoff ab, wäscht ihn mit Methanol und trocknet ihn. Sein
Schmelzpunkt beträgt 204° und die Ausbeute 14,7 g (80,8% der Theorie, bezogen auf den Cyanacetylharnstoff).
18,2 g Ν,Ν-Dimethylaminomethylen-cyanacetylharnstoff
werden in 50 ml Wasser angerührt. Zu der entstehenden Suspension läßt man eine Lösung von
10 g Natriumhydroxyd in 50 ml Wasser innerhalb einer Stunde zufließen und steigert in derselben Zeit
die Reaktionstemperatur von 20 bis 30 auf 60°. Man rührt die Mischung noch 1 Stunde bei dieser Temperatur
und säuert dann die entstandene Lösung mit Salzsäure an, kühlt auf 5 bis 10° ab und saugt das
ausgefallene 5-Cyan-uracil der Formel abgekühlt, das ausgefallene 6-Methyl-5-cyan-uracil
der Formel

ab, wäscht es mit Wasser und trocknet es. Die Ausbeute beträgt 12,6 g (93,5% der Theorie).
Verwendet man an Stelle von 24 g der Verbindung aus Dimethylformamid und Dimethylsulfat die äquimolare
Menge der Verbindung aus Dimethylformamid und Diäthylsulfat oder Di-isopropylsulfat oder
aus Dimethylformamid und Phosphoroxychlorid und verfährt wie im Beispiel vermerkt, so erhält man
ebenfalls N,N - Dimethylaminomethylen - cyanacetylharnstoff und daraus 5-Cyan-uracil der vorstehenden
Formel.
12,7 g Cyanacetylharnstoff werden mit 50 ml Methanol angerührt, und die Suspension wird mit 25,5 g
der Verbindung aus Dimethylacetamid und Dimethylsulfat vereinigt. Zu diesem Gemisch tropft man bei
10 bis 20° eine Lösung von 6,5 g Natriummethylat in 40 ml Methanol zu. Man rührt das Reaktionsgemisch noch 5 Stunden bei 30 bis 40° und versetzt
es dann mit einer Lösung von 10 g Natriumhydroxyd in 50 ml Wasser. Die Temperatur des Gemisches
wird 1 Stunde bei ungefähr 60° gehalten; dann wird die Lösung mit Salzsäure angesäuert, auf 10 bis 15°
abgesaugt, mit etwas Wasser gewaschen und getrocknet. Die Ausbeute beträgt 10,7 g (70,8% der
Theorie, bezogen auf den Cyanacetylharnstoff).
Die gleiche Verbindung erhält man, wenn man an Stelle von 25,5 g der Verbindung aus Dimethylacetamid
und Dimethylsulfat äquimolare Mengen der Verbindung aus Dimethylsulfat und Diäthylacetamid
oder N-Acetylpiperidin oder N-Acetylmorpholin verwendet.
Die entsprechende 6-Äthylverbindung erhält man, wenn man an Stelle von 25,5 g der Verbindung aus
Dimethylacetamid und Dimethylsulfat äquimolare Mengen der Verbindung aus Dimethylpropionamid
und Dimethylsulfat verwendet.
127 g Cyanacetylharnstoff werden mit 400 ml Methanol
angerührt und auf 10° gekühlt. Man tropft gleichzeitig 240 g der Verbindung aus Dimethylformamid
und Dimethylsulfat und eine Lösung von 46 g Natriumhydroxyd in 230 ml Methanol innerhalb
2 Stunden bei 10 bis 15° zu. Die Mischung wird anschließend noch eine weitere Stunde bei dieser
Temperatur gerührt, dann mit 400 ml Wasser versetzt, das ausgeschiedene Kristallisat filtriert, mit
Wasser gewaschen und getrocknet. Die Ausbeute beträgt 170 g Ν,Ν-Dimethylaminomethylencyanacetylharnstoff
(93,5% der Theorie, bezogen auf Cyanacetylharnstoff)." . ·
Das Produkt wird wie im Beispiel 1 in 5-Cyanuracil übergeführt.
Zu 22 g der Verbindung aus Dimethylformamid und Dimethylsulfat gibt man 16,3 g Diäthylamin,
wobei die Temperatur auf 40 bis 50° steigt. Zu dem Reaktionsgemisch läßt man eine Lösung von 12,7 g
Cyanacetylharnstoff in 85 ml Dimethylformamid fließen und rührt 2 Stunden bei 80 bis 90°. Das beim
Abkühlen ausfallende Kristallisat wird abfiltriert, in 50 ml Wasser angeschlämmt, die Suspension mit
15 ml 10%iger wäßriger Natronlauge versetzt und 3 Stunden bei 80 bis 90° gerührt, wobei völlige Lösung
eintritt. Beim Ansäuern mit 10 ml 36%iger Salzsäure fällt das 5-Cyan-uracil kristallin aus. Die Ausbeute
beträgt 3 g (etwa 30%, bezogen auf Cyanacetylharnstoff).
Zu 25,5 g der Komplexverbindung aus Dimethylformamid und Dimethylsulfat tropft man bei 20 bis
30° eine Lösung von 6,5 g Natriummethylat in 80 ml Methanol. Man rührt das Gemisch 1 Stunde' bei
der angegebenen Temperatur und streut dann 12,5 g
Cyanacetylharnstoff ein. Nach etwa 5 Stunden wird der gebildete N.N-Dimethylaminomethylen-cyanacetylharnstoff
abgesaugt, mit Methanol gewaschen und getrocknet. Die Ausbeute beträgt 7 g (etwa 40%,
bezogen auf Cyanacetylharnstoff). Das Produkt wird gemäß Beispiel 1 in 5-Cyan-uracil übergeführt.

1. Verfahren zur Herstellung von 5-Cyanuracilen der allgemeinen Formel I,

Alkylgruppe bedeutet, dadurch gekennzeichnet, daß man eine Verbindung der allgemeinen Formel II

oder zusammen mit dem Stickstoffatom einen nicht aromatischen, gegebenenfalls weitere
Heteroatome aufweisenden Ring bilden, R4 einen niederen Alkoxyrest oder eine Dichlorphosphinylgruppe
und ΧΘ das Anion einer niederen Alkylschwefelsäure
oder das Chlorion bedeutet, in Gegenwart einer Base in einem Lösungsmittel oder im Überschuß an der Verbindung der allgemeinen
Formeln bei Temperaturen von etwa -10 bis 1200C mit Cyanacetylharnstoff umsetzt
und die erhaltene Verbindung der allgemeinen Formel III

2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß R1 ein Wasserstoffatom oder
eine Methyl- oder Äthylgruppe, R2 und R3 die
Methyl- oder Äthylgruppe, R4 eine niedere Alkoxygruppe undX® ein niederes Alkylsulfation bedeutet.
