2 ml (11.5 mmol) Hünig-Base.
Man lässt über Nacht
reagieren, wobei die Temperatur auf RT ansteigt. Laut DC-Kontrolle
(Dichlormethan/Methanol = 10/1) ist die Reaktion beendet. Man dampft
alles i.V. zur Trockne ein, nimmt in 25 ml Essigsäureethylester
auf und wäscht
mit gesättigter
Natriumhydrogencarbonat-Lösung.
Die organische Phase wird dreimal mit 1 N Salzsäure und mit gesättigter
Kochsalz-Lösung
gewaschen. Nach dem Trocknen mit Natriumsulfat wird alles zur Trockne
eingedampft.
Ausbeute: 3.31 g (89% d.Th.)

versetzt man mit 1 N Salzsäure.
Man trennt beide Phasen und extrahiert die wässrige Phase noch zweimal mit
wenig Essigsäureethylester.
Nach Trocknen der organischen Phase über Natriumsulfat dampft man
i. V. zur Trockne ein. Der Rückstand
wird in Diethylether aufgenommen und mit gesättigter Natriumhydrogencarbonat-Lösung geschüttelt. Die
wässrige
Phase stellt man mit 5 N Salzsäure
auf einen pH-Wert = 2.7 ein und extrahiert sie zweimal mit Essigsäureethylester.
Die vereinigten organischen Extrakte werden über Natriumsulfat getrocknet
und i.V. zur Trockne eingedampft.

wird in 900 ml Dichlormethan aufgenommen und innerhalb von drei

Vorschrift des Beispiels 1 aus 80 mg (0.12 mmol) der Verbindung aus
Beispiel 23A und 20.73 mg (0.16 mmol) trans-2-Heptensäure mit
61.49 mg (0.16 mmol) HATU und 18 mg/36 mg (0.07 ml) Hünig-Base
in. 4 ml abs. DMF.
Ausbeute: 27.1 mg (30% d.Th.)

10 mg (0.02 mmol) der Verbindung
aus Beispiel 31A und 9.62 mg (0.03 mol) N-[(2E)-3-Cyclohexyl-2-propenoyl]-3,5-difluorphenylalanin
werden unter Argonatmosphäre
in 0.3 ml abs. DMF gelöst
und bei 0°C mit
10.84 mg (0.03 mmol) HATU und 0.004 ml (2.6 mg, 0.03 mmol) Hünig-Base
versetzt. Man rührt
30 min und versetzt dann mit weiteren 0.007 ml (5.3 mg ≅ 0.06 mmol)
Hünig-Base.
Nach einstündigem
Rühren
bei 0°C entfernt
man das Kältebad
und lässt über Nacht
bei RT weiterrühren.
Anschließend
wird der Reaktionsansatz mittels RP-HPLC aufgereinigt. Ausbeute:

MS (ESI pos.): m/z = 739 [M+H]+ 10 mg (0.02 mmol) of the compound from Example 31A and 9.62 mg (0.03 mol) of N - [(2E) -3-cyclohexyl-2-propenoyl] -3,5-difluorophenylalanine are dissolved in 0.3 ml of abs under an argon atmosphere. DMF dissolved and at 0 ° C with 10.84 mg (0.03 mmol) HATU and 0.004 ml (2.6 mg, 0.03 mmol) Hünig base. 15 mg (0.03 mmol) der Verbindung
aus Beispiel 34A und 14.36 mg (0.04 mmol) N-[(2E)-3-Cyclohexyl-2-propenoyl]-3,5-difluorphenylalanin
werden unter Argonatmosphäre
in 0.5 ml abs. DMF gelöst
und bei 0°C mit
16.19 mg (0.04 mmol) HATU und 0.007 ml (0.04 mmol) Hünig-Base
versetzt. Nach 30 min Rühren
versetzt man mit weiteren 0.014 ml (0.08 mmol) Hünig-Base und lässt noch
1 h bei 0°C
rühren. Über Nacht
lässt man bei
RT weiterrühren.
Anschließend
arbeitet man den Reaktionsansatz mittels RP-HPLC auf.
Ausbeute:
1 mg (4% d.Th.)

MS (ESI pos.): m/z = 741 [M+H]+ 15 mg (0.03 mmol) of the compound from Example 34A and 14.36 mg (0.04 mmol) of N - [(2E) -3-cyclohexyl-2-propenoyl] -3,5-difluorophenylalanine are dissolved in 0.5 ml of abs under an argon atmosphere. DMF dissolved and 16.19 mg (0.04 mmol) of HATU and 0.007 ml (0.04 mmol) of Hünig base added at 0 ° C. After stirring for 30 min, a further 0.014 ml (0.08 mmol) of Hünig base is added and the mixture is left to stir at 0 ° C. for 1 h. Stir at RT overnight. The reaction mixture is then worked up using RP-HPLC.
Yield: 1 mg (4% of theory)
