Description
The present invention relates to a process for the production of formamidine acetate.
Amidines are strongly basic compounds that are used, for example, in pharmacy, biochemistry, organometallic science or synthetic organic chemistry. The pKa values for amidines are typically in the range of 11 to 12. The basic property is based on the protonation of the imine nitrogen and the resulting mesomeric stabilization of the charge. The simplest amidine is formamidine, which is derived from formic acid. Due to the high instability of the unsubstituted formamidine to hydrolysis and condensation with amines or cyclocondensation to form the triazine, the product can only be isolated as a salt.
In the context of the development of sustainable processes, electroorganic synthesis has gained importance in recent decades. Electro-organic syntheses are reactions in which the oxidation or reduction of a substrate is carried out using an electric current as a reagent. The electron transfer can take place directly on the electrode surface by a single electron transfer SET to the substrate, by a redox mediator or by secondary processes on an electrically activated surface of the electrode.
Soc 1957, 104, 160-162). Mit Hilfe dieser Ergebnisse entwickelten sie eine elektrochemische Synthese für Formamidinsalze ausgehend von Cyanamid in schwefelsaurer und salzsaurer Lösung unter Verwendung einer Pd/schwarz-Elektrode (japanische Patentanmeldung JPS36-2657 A1).Electrochemical investigations into the reduction of nitriles in acidic aqueous solution were carried out as early as the 1950s. Thus, by K.Odo et al. described the electrochemical reduction of cyanamide to methylamine (K.Odo et al., J. Electrochem. Soc 1957, 104, 160-162). Using these results, they developed an electrochemical synthesis for formamidine salts starting from cyanamide in sulfuric acid and hydrochloric acid solution using a Pd/black electrode (Japanese patent application JPS36-2657 A1).
In addition, Trümpler et al. on the electrochemical reduction of cyanamide on a nickel sponge electrode (G.Trümpier et al., Helv. Chim. Acta 1959, 42 (2), 407-416). The studies have shown that the actual reduction takes place via formamidine as an intermediate and not via hydrocyanic acid. However, neither complete conversion of the cyanamide nor isolation of a formamidine salt could be achieved.
The present invention is therefore based on the object of providing an electrochemical process for the preparation of formamidine acetate which can be used in a wide variety of forms and enables a high space-time yield, with complex work-up of the resulting product being dispensed with entirely or carried out very easily can be.
- a) an aqueous solution comprising cyanamide and acetic acid is provided in a first process step,
- b) in a second process step, the aqueous solution is passed through a flow electrolysis cell comprising at least one cathode and at least one anode without the addition of a conductive salt, the cyanamide being reduced by means of an electric current at the cathode, and
- c) in a third process step, the formamidine acetate formed in the second process step is isolated.
A method in which direct current is used as the electrical current, in particular smoothed direct current, is preferred. The cyanamide is thus reduced at the cathode by means of direct current, in particular smoothed direct current.
The investigations on which the invention is based have shown that formamidine acetate can be produced by means of electrochemical reduction in a continuous electrolysis process. What is surprising here is that formamidine acetate, as a product from the electrochemical reduction of cyanamide in aqueous acetic acid, is accessible without the addition of a conducting salt, despite its low conductivity. The desired product can be prepared in high yields directly from the starting compounds. High losses due to side reactions can be suppressed to an acceptable level. The isolation of the target substance can therefore be carried out without any effort and leads to a high purity of the desired product because no conductive salt has to be separated off. Furthermore, it has been found that the reaction can be carried out continuously by using a flow electrolytic cell. In addition, the use of a flow electrolysis cell means that the contact time with the electrodes can be limited to a necessary level in relation to the amount of cyanamide to be reduced. With the results of the underlying investigations and the present invention, implementation in large-scale processes has become feasible. Taken together, these facts are totally unpredictable and surprising.
A continuous process according to the present invention is a process in which electrolysis can be carried out constantly under the same conditions and in flow. And in which it is possible, without interrupting the process, to remove product from the reaction solution and add new starting material to the reaction.
A flow electrolysis cell is a preparative structure in which an electrolyte is pumped through a cell through which an electric current, in particular direct current, in particular smoothed direct current, is passed. The direction of flow of the electrolyte is predominantly orthogonal to the applied electric field. The electrical current causes chemical reactions at the electrodes. Both cathodic (reductive) and corresponding anodic (oxidative) processes take place.
The reaction volume in the cell can be designed in such a way that the reaction spaces for the cathodic reaction and the anodic reaction are locally separated; this is referred to as a divided structure. The separation takes place through semi-permeable diaphragms or membranes. Alternatively, both partial reactions can also be carried out in a common reaction volume, in a so-called undivided structure.
A flow electrolytic cell according to the present invention is thus an electrolytic cell which comprises at least one reaction space and within the reaction space a cathode and/or an anode, the reaction space having an inflow for at least one reactant and an outflow for has at least one formed product, so that a continuous process is made possible. Thus, a flow electrolytic cell differs from an electrolytic cell that is operated using the pot process (prior art) by having an inlet and an outlet that enable a continuous process.
According to the present invention, the cell volume of the flow electrolytic cell is defined as the total volume of all spaces within the flow electrolytic cell, both cathodes and anode spaces, through which the electrolyte flows and in which active chemical reactions take place.
This flow electrolytic cell also differs from an electrolytic cell that is operated using the pot process in that it has an increased surface-to-volume ratio of electrode surface to reaction chamber volume compared to a pot-type electrolytic cell. If the flow electrolytic cell has a divided structure, it also differs from an electrolytic cell that is operated using the pot process in that it has an increased surface-to-volume ratio of membrane surface to reaction chamber volume compared to a pot-type electrolytic cell having. A flow electrolytic cell also differs from an electrolytic cell that is operated using the pot process in that it has a reduced electrode spacing.
Thus, the method according to the invention can preferably be carried out in a flow electrolysis cell, which comprises a reaction space and within the reaction space at least one cathode and at least one anode, the cyanamide being reduced by means of an electric current at the cathode, and the reaction space having an inflow for the cyanamide and an outlet for the product formed, namely formamidine acetate.
According to the present invention, a large number of different flow electrolytic cells can be used as flow electrolytic cells, which differ in terms of their geometry and their apparatus structure. However, the underlying investigations have shown that particularly good results can be achieved using a divided flow electrolysis cell. Thus, according to a further development, the present invention also relates to a method in which a divided flow electrolysis cell is used as the flow electrolysis cell. A flow electrolysis cell which comprises a first half-cell, to which the anode is assigned, and a second half-cell, to which the cathode is assigned, can further preferably be used. Even more preferably, this flow electrolysis cell can have a membrane, in particular a semi-permeable membrane, which separates the first and second half-cells from one another.
More preferably membranes, in particular semipermeable membranes, which have a thickness in the range from 0.08 mm to 0.50 mm, in particular from 0.08 mm to 0.30 mm and particularly preferably from 0.08 mm to 0.18 mm .
Furthermore, a flow electrolysis cell according to the present invention preferably has a surface-to-volume ratio of geometric electrode surface to reaction volume in the range of 0.1 cm -1 to 100 cm -1 , in particular from 4 cm -1 to 80 cm -1 and particularly preferably from 6 cm -1 to 60 cm -1 .
A flow electrolysis cell according to the present invention also preferably has a surface-to-volume ratio of membrane surface area to reaction chamber volume in the range from 0.1 cm -1 to 100 cm -1 , in particular from 4 cm -1 to 80 cm -1 and particularly preferably from 6 cm -1 to 60 cm -1 .
The reaction volume is to be understood as the volume of the entire cell, with the volumes of any half-cells or cells divided by membranes being added. In particular, the internal cellular resistance of the electrolytic cell can be significantly reduced by these design features. This means that a higher yield can be achieved in relation to the electricity used.
According to the present invention, the flow electrolytic cell comprises at least one cathode and at least one anode. Flow electrolysis cells can also be used in the method according to the invention, which more preferably comprise two, three, four or five cathodes, in particular cathodes connected in series, and/or flow electrolysis cells which more preferably have two, three, four or include five anodes, in particular anodes connected in series.
A large number of different electrodes can be used in the method according to the invention. However, it has been shown that a selective reduction of the cyanamide can be realized when using a metal cathode. Thus, according to a preferred embodiment, it is provided that a flow electrolysis cell comprises a metal cathode as the cathode. A metal cathode which comprises a metal from the group consisting of iron, nickel, cobalt and palladium, in particular nickel, is also preferred. A flow electrolysis cell whose cathode is a nickel foam electrode is particularly preferred.
Furthermore, it has been shown that metal electrodes, in particular nickel electrodes and more preferably nickel foam electrodes and very particularly preferably nickel foam electrodes with a metal content of 2 vol .-% to 10 vol .-% and / or with a pore number of 4 cm -1 to 40 cm -1 and / or with a nickel content of 93% to 100% deliver particularly good results.
The proportion of metal describes the proportion by volume that the metal occupies in relation to the entire volume of the metal foam. The void count indicates the average number of voids found per unit length in the foam. Nickel content refers to the mass content of elemental nickel in the metal.
It has been shown that when using a nickel foam electrode in the flow cell, a non-linear flow behavior of the electrolyte is achieved, which leads to an increased mass transfer at the electrode. In this process, this leads to an increased yield of target substance. In addition, by massively increasing the active surface, the current density could be reduced without increasing the process time, which led to an improved yield in this process. Information on current densities relates to a geometric area (top view of the foam).
Derartige Nickel-Schaumelektroden können kommerziell erworben werden bei beispielsweise Goodfellow GmbH, Hamburg, Deutschland oder Aqua Titan GmbH, Dortmund, Deutschland oder IKA-Werke GmbH & Co. KG, Staufen, Deutschland.Such nickel foam electrodes can be purchased commercially from, for example, Goodfellow GmbH, Hamburg, Germany or Aqua Titan GmbH, Dortmund, Germany or IKA-Werke GmbH & Co. KG, Staufen, Germany.
Regardless of the type of cathode, it has been shown that a metal electrode or a graphite electrode can preferably be used as the anode in the method according to the invention. A metal anode is particularly preferred as the anode. These metal anodes enable the anodic counter-reaction through a far-reaching, trouble-free current flow. An anode which is a metal electrode which comprises an electrocatalytic coating which in turn comprises a mixed metal oxide, in particular from the group of iridium dioxide, ruthenium dioxide and/or platinum dioxide with titanium dioxide on titanium or tantalum dioxide on tantalum, is very particularly preferred.
Furthermore, it has been shown that such metal electrodes, in particular coated metal electrodes whose coating comprises a mixed metal oxide from the group of iridium dioxide, ruthenium dioxide and platinum dioxide with titanium dioxide on titanium or tantalum dioxide on tantalum, deliver particularly good results.
It has been shown that when using a metal electrode whose coating comprises a mixed metal oxide from the group of iridium dioxide, ruthenium dioxide and platinum dioxide with titanium dioxide on titanium or tantalum dioxide on tantalum, there is a low overvoltage for the generation of oxygen. As a result, the voltage required for the process can be lowered and the development of by-products can be further suppressed.
Derartige dimensionsstabile Anoden (DSA) können kommerziell erworben werden bei beispielsweise De Nora, Mailand, Italien oder METAKEM GmbH, Usingen, Deutschland.Such dimensionally stable anodes (DSA) can be purchased commercially from, for example, De Nora, Milan, Italy or METAKEM GmbH, Usingen, Germany.
In the method according to the invention it is provided that an aqueous solution comprising acetic acid and cyanamide is provided in the first method step and this aqueous solution is passed through the flow electrolysis cell in the second method step. It has been found that cyanamide and acetic acid can be used in a wide concentration range, with a cyanamide concentration in the range from 5 to 500 g/l and an acetic acid concentration in the range from 5 to 800 g/l showing particularly good yields. Thus, an aqueous solution can also preferably be used which has a cyanamide concentration in the range from 5 to 500 g/l, in particular in the range from 5 to 400 g/l, more preferably in the range from 5 to 300 g/l in the range from 5 to 200 g/l and most preferably in the range from 5 to 100 g/l.
At the same time or independently of this, a method is preferred in which the aqueous solution has an acetic acid concentration in the range from 5 to 800 g/l, in particular in the range from 5 to 700 g/l, in particular in the range from 5 to 600 g/l, more preferably in the range from 5 to 500 g/l, more preferably in the range from 5 to 400 g/l and very particularly preferably in the range from 5 to 300 g/l.
However, it has also been found that an excess of acetic acid (relative to cyanamide) in the aqueous solution allows particularly good yields to be achieved. Thus, in a preferred method, an aqueous solution comprising acetic acid and cyanamide is provided in the first method step, which solution comprises acetic acid (with respect to cyanamide) in excess. According to a further advantageous embodiment of the method, it is therefore also provided that the aqueous solution contains cyanamide and acetic acid in a molar ratio of cyanamide to acetic acid in the range from 1: 1 to 1: 100, in particular in the range from 1: 1 to 1: 50, in particular in range from 1:1 to 1:30, more preferably in the range from 1:1 to 1:20, more preferably in the range from 1:1 to 1:10 and very particularly preferably in the range from 1:1 to 1:2 .
A process for the preparation of formamidine acetate is therefore particularly preferred by providing an aqueous solution comprising cyanamide and acetic acid in the first process step, the cyanamide concentrations being in the range from 5 to 500 g/l and an acetic acid concentration in the range from 5 to 800 g/l and wherein the molar ratio of cyanamide to acetic acid is in the range of 1:1 to 1:100.
In addition, it was surprisingly found that the process according to the invention can be carried out without the addition of conducting salts, such as are usually used in electrolysis. This is all the more surprising since acetic acid itself has a much lower conductivity than common strong acids. According to a preferred embodiment, the subject of the invention is a method in which the aqueous solution contains no conductive salt or the aqueous solution is free of conductive salt.
Conductive salts, such as are commonly used in electrochemical reactions, are salts that cannot be oxidized or reduced themselves. Conductive salts of this type are usually used in electrolysis processes in high concentrations up to the saturation limit in order to increase the conductivity of the reaction medium or reaction solvent used in the reaction.
Conductive salts used in methods not according to the invention, which should be explicitly excluded here, are for example salts of sodium, potassium, magnesium, calcium or ammonium with the anions chloride, bromide, sulfate, sulfonate, acetate, phosphate or nitrate.
According to the present invention, a conductive salt-free solution should be understood to mean a solution that contains less than 1% by weight, in particular less than 0.1% by weight and particularly preferably less than 0.01% by weight, of any one salt that is different of formamidine acetate. Traces of salts can occur due to the salts contained in process water due to impurities in the outlet starting materials or due to side reactions and should not be regarded as conductive salts.
Since no conductive salt is used in the process according to the invention, the desired product can be isolated from the resulting reaction mixture in a largely unproblematic manner. In addition, it is surprising that the product can be isolated in high yields and purity. Thus, a process can be provided that enables large-scale application and economical production of formamidine acetate.
Particular preference is therefore also given to a process for the production of formamidine acetate in which an aqueous solution comprising cyanamide and acetic acid is provided in the first process step, the cyanamide concentrations being in the range from 5 to 500 g/l and an acetic acid concentration in the range from 5 to 800 g/l, the molar ratio of cyanamide to acetic acid being in the range from 1:1 to 1:100, and the aqueous solution containing no conductive salt or the aqueous solution being free of conductive salt.
The subject matter of the present invention is therefore also preferably a process in which the reduction of the cyanamide is carried out without the addition of a conductive salt. A process is very particularly preferred in which b) in a second process step, the aqueous solution is passed through a flow electrolysis cell comprising at least one cathode and at least one anode, with the cyanamide being reduced by means of an electric current at the cathode, and with the reduction of the cyanamide is carried out without the addition of a conductive salt.
Furthermore, it has been shown that the method according to the invention can be carried out particularly advantageously if aqueous acetic acid is used both as the anolyte and as the catholyte. Aqueous acetic acid can thus more preferably be used both as the anolyte and as the catholyte in the process according to the invention. A method can thus be provided that can be classified as being particularly simple to implement with regard to the variability of the substances to be used.
Also preferred is a method in which the reduction is carried out at a pH in the range from
The subject of the present invention is therefore also a process in which aqueous acetic acid is used both as the anolyte and as the catholyte and/or in which the reduction is carried out at a pH in the range from
According to a further advantageous embodiment, the process, in particular the reduction, can be carried out at a temperature in the range from 5 to 50° C., in particular in the range from 10 to 50° C., more preferably in the range from 10 to 40° C., more preferably in the range from 10 to 30° C. and very particularly preferably at room temperature.
According to a further advantageous embodiment of the invention, the method, in particular the reduction, can be carried out at normal pressure or elevated pressure, in particular at up to 50 bar, in particular up to 3 bar overpressure. Higher local concentrations and thus better productivity can be achieved at the electrode, particularly when carried out at elevated pressure.
The subject of the present invention is therefore also a method in which the reduction is carried out at a temperature in the range from 5 to 50° C. and/or at a pressure in the range from 0.3 to 50 bar, in particular at 0.3 to 10 bar. in particular at 0.3 to 4 bar, in particular at 0.3 to 3 bar.
According to a preferred embodiment of the method, the surface of the anode is aligned parallel to the direction of flow of the anolyte and/or the surface of the cathode is aligned parallel to the direction of flow of the aqueous solution. The electrolyte, in particular the catholyte or the anolyte, can thus be guided orthogonally to the applied electric field and a good yield can be achieved.
The charge or current is applied either a) completely when the electrolyte is passed through the flow electrolytic cell once, or b) gradually when the same electrolyte is repeatedly pumped through the same cell or a different cell.
In the method according to at least one of the aforementioned characteristics, the aqueous solution is conducted through the flow electrolysis cell in such a way that the flow rate of the aqueous solution is in a range from 0.002 to 16.667 cm 3 /s, in particular from 0.017 to 0.333 cm 3 /s.
In the method according to at least one of the aforementioned characteristics, the anolyte is conducted through the flow electrolysis cell in such a way that the flow rate of the anolyte is in a range from 0.002 to 16.667 cm 3 /s, in particular from 0.008 to 0.025 cm 3 /s.
In the method according to at least one of the aforementioned characteristics, the geometric current density for reduction is in the range from 0.1 to 300 mA/cm 2 , in particular in a range from 2 to 5 mA/cm 2 (in each case based on the electrode surface of the cathode) .
The applied current intensity is defined as the geometric current density in relation to the cathode or anode surface, if this is viewed as a flat surface.

A screening cell for electrochemical reactions in flow was used. A planar nickel electrode with a nickel foam spacer was used as the cathode and a DSA (IrOx on tantalum) as the anode. Two half-cell spaces were created with the help of EPDM spacers, which were separated by a Nafion membrane. Both half-cell spaces were connected to separate reservoirs and the electrolytes were pumped from the reservoirs through the half-cell spaces and back into the corresponding reservoirs using peristalsis or diaphragm pumps.

An in-pot electrochemical reaction screening cell was used. A flat nickel electrode or a nickel foam electrode was used as the cathode. A planar graphite electrode was used as the anode. A Nafion membrane was used as the membrane, made of the same material as in the flow cell. The electrolytes were stirred by a magnetic stirrer.
c) Durchführung der Versuchec) Carrying out the tests
The bare electrolyte was pumped through both half-cells. When the catholyte reached the exit of the cell, a current was applied. A preliminary flow of approximately half the cell volume of the electrolyte was pumped through the cell and then discarded. This served to flush out potentially dissolved Ni ions from the cell during the filling process.
Immediately after the preparation, the reservoirs were replaced with new ones. The same electrolyte was used for the catholyte and anolyte. The cyanamide was additionally introduced into the catholyte. The electrolysis was carried out at a constant flow rate with a constant current. Charge was applied until complete conversion of cyanamide could be verified by precipitation as silver cyanamide. After the reaction had ended, 1 mL of the catholyte was transferred, and the solvent was removed at 40° C. under reduced pressure. For the analytical content determination, maleic acid was added and the residue was dissolved in as little H 2 O/D 2 O (1:1) as possible. The yield was then determined by means of 1 H-NMR spectroscopy against an internal standard.

In a divided cell for electrochemical screening with a membrane as a separator, the corresponding, identical electrolyte was placed in each half-cell. The cell, when tempered, was stirred with the electrolyte for half an hour in order to reach the desired reaction temperature. Then cyanamide was dissolved in the catholyte. The electrodes were immersed in the solution (electrode area A=3.6 cm 2 ) and the electrolysis started.
After the reaction was complete, the electrodes were removed and flushed with H 2 O over a flask. The reaction solution was transferred to the same flask (in the case of a Nafion® membrane as separator, only the catholyte was used). The solvent was removed under reduced pressure at 40°C. For the analytical content determination, maleic acid was added and the residue was dissolved in as little H 2 O/D 2 O (1:1) as possible. The yield was then determined by means of 1 H-NMR spectroscopy against an internal standard.
d) Bewertung der Ergebnisse gemäß Tabelle 1 und Tabelle 2d) Evaluation of the results according to Table 1 and Table 2
It was also possible to show that the addition of conductive salt in this process has no effect on the yield of formamidine acetate and is therefore completely superfluous (Example 5). In addition, technical cyanamide could also be used in the process developed without the yield being significantly reduced (Example 6).
e) Isolierung von Formamidinacetate) isolation of formamidine acetate
ErfindungsgemässAccording to the invention
A flow electrolysis was carried out under the conditions according to example 4. After the electrolysis was complete, the entire aqueous solution (approx. 25 mL) was transferred to a flask and the flow cell was rinsed with deionized water (approx. 50 mL). The aqueous solutions containing the formamidinium acetate were combined and evaporated to dryness under reduced pressure at a maximum of 50°C. Diethyl ether (15 mL) was added to the crude product, causing it to crystallize. The ether was filtered and the crude formamidinium acetate obtained (892 mg, corresponds to 72% chemical yield (based on cyanamide), Mp = 120 °C) was then recrystallized from ethanol (4 mL) from the boiling point, where it was in very

Das so erhaltene Salzgemisch zeigte ein Molverhältnis von Formamidinium zu Natrium von 1,0 zu 1,5.A pot electrolysis was carried out under the conditions according to example 1. After the electrolysis was complete, the aqueous solution was transferred to a flask and the cathode chamber was rinsed out with deionized water. The aqueous solutions containing the formamidine acetate in addition to sodium acetate were combined and concentrated under reduced pressure at a maximum of 50°C. Diethyl ether was added to the crude product, causing it to crystallize. The salt mixture thus obtained showed a molar ratio of formamidinium to sodium of 1.0 to 1.5.
The energy parameter per kg of formamidine acetate is a measure of the energy efficiency of the process, since it relates the electrical energy supplied to the yield.
