- Technisches Gebiet
- Die Erfindung betrifft ein Verfahren zum Herstellen einer optisch aktiven Allylverbindung, welche als Zwischenprodukt für Pharmazeutika, Agrarchemikalien oder physiologisch aktive Substanzen wichtig ist, welche zum Beispiel sehr nützlich als Zwischenprodukt zum Synthetisieren für Antibiotika ist.
- Stand der Technik
- Verfahren, die bekannt sind, oder von denen angenommen wird, dass sie zum Herstellen einer optisch aktiven Allylverbindung geeignet sind, schließen (1) einmal Synthetisieren einer racemischen Form einer gewünschten Allylverbindung ein, gefolgt von Enantiomerentrennung unter Verwenden eines optisch aktiven Spaltungsreagenz oder eines Enzyms, (2) Beginnen mit einer asymmetrischen Verbindung oder (3) Verwenden eines asymmetrischen Katalysators.
- Das Verfahren (1), welches ein optisch aktives Spaltungsreagenz verwendet, erfordert eine Äquivalentmenge oder mehr eines Spaltungsreagenz bezogen auf die Allylverbindung. Darüber hinaus sind komplizierte Prozeduren wie Kristallinisierung, Trennung und Aufreinigung vor dem Erhalt der optisch aktiven Allylverbindung einbezogen. In dem Verfahren (1), welches ein Enzym verwendet, ist, während es imstande ist, eine Allylverbindung mit relativ hoher optischer Reinheit zu ergeben, der Typ des Reaktionssubstrats und die absolute Konfiguration der resultierenden Allylverbindung beschränkt.
- Das Verfahren (2) ist vorstellbar, aber dahingehend problematisch, dass eine optisch aktive Ausgangsverbindung nicht nur teuer ist, sondern in stöchiometrischem Überschuss verwendet werden muss.
- Bezüglich des Verfahrens (3) wurden kürzlich ausführliche Untersuchungen in der katalytischen asymmetrischen Synthese einer optisch aktiven Allylverbindung durchgeführt, welche eine hohe Wirksamkeit und asymmetrische Ausbeute ergeben kann, wie es in Trost, B. M. & Van Vranken, D. L. Chem. Rev., 96, 395–422 (1996) und Trost, B. M. & Crawley, M. L., Chem. Rev., 103, 2921–2944 (2003) berichtet wurde. Die Techniken in diesem Fach sind immer noch in Untersuchung.
- Im Stand der Technik werden Phosphor enthaltende Verbindungen offenbart, die bei der Herstellung von optisch aktiven Verbindungen verwendet werden. So offenbart die

- Angesichts der obigen Gegebenheiten ist ein Gegenstand der Erfindung das Bereitstellen eines Verfahrens zum Herstellen einer optisch aktiven Allylverbindung mit hoher Effizienz und hoher asymmetrischer Ausbeute.
- Als Ergebnis umfangreicher Studien der katalytischen asymmetrischen Synthese einer optisch aktiven Allylverbindung haben die Erfinder herausgefunden, dass die asymmetrische Kupplungsreaktion einer Allylverbindung mit einer organischen nucleophilen Verbindung in Anwesenheit eines Katalysators die gewünschte optisch aktive Allylverbindung mit einer verringerten Anzahl von Schritten bei hoher Wirksamkeit in guter asymmetrischer Ausbeute ergibt. Die vorliegende Erfindung wurde basierend auf diesen Befunden erreicht.
- Die Erfindung stellt ein Verfahren zum Herstellen einer durch die Formel (III) wiedergegebenen optisch aktiven Allylverbindung bereit: worin R1, R2, R3, R4 und R5 jeweils ein Wasserstoffatom und eine Arylgruppe wiedergeben;
Nu eine nucleophile Gruppe wiedergibt, ausgewählt aus einer α-Ketocarbaniongruppe, einer Aminogruppe und einer substituierten Aminogruppe; und der Stern * ein asymmetrisches Zentrum angibt,
oder einer optisch aktiven Allylverbindung, wiedergegeben durch Formel (IV): worin R1, R2, R3, R4 und R5, Nu und * wie oben definiert sind,
oder einer Mischung der Verbindung der Formel (III) und der Verbindung der Formel (IV),
wobei das Verfahren das asymmetrische Kuppeln einer durch die Formel (I) wiedergegebenen Allylverbindung: worin R1, R2, R3, R4 und R5 wie oben definiert sind; und E eine Abgangsgruppe wiedergibt, ausgewählt aus einer Gruppe umfassend ein Sauerstoffatom und die Gruppe, die durch R1, R2, R3, R4 und R5 wiedergegeben wird und an das Sauerstoffatom gebunden ist; und eine Halogengruppe;
mit einer organischen nucleophilen durch Formel (II) wiedergegebenen Verbindung:Nu-H (II)
in Gegenwart eines Katalysators, welcher eine Pd-Komplexverbindung ist, die ein durch Formel (a) wiedergegebenes Chinoxalinderivat als Ligand aufweist, worin * wie oben definiert ist, umfasst. - Ausführliche Beschreibung der Erfindung
- Die Allylverbindung, die als Ausgangsmaterial in dem Verfahren der Erfindung verwendet werden kann, wird durch Formel (I) wiedergegeben. Das durch das Verfahren der Erfindung erhaltene Produkt ist die durch Formel (III) wiedergegebene optisch aktive Allylverbindung, die durch die Formel (IV) wiedergegebene optisch aktive Allylverbindung oder durch eine Mischung derselben. Die von R1, R2 R3, R4 und R5 in den Formeln (I), (III) und (IV) wiedergegebenen Gruppen werden beschrieben.
- Der Ausdruck „Arylgruppe” schließt Arylgruppen mit sechs bis 14 Kohlenstoff atomen ein, wie Phenyl, Naphtyl und Anthryl.
- In den Formeln (I), (III) und (IV) können R1, R2 R3, R4 und R5 gleich oder verschieden sein. Sie können voneinander unabhängig sein oder zwei oder mehr von ihnen können miteinander vernetzt sein.
- In der Formel (I) gibt E eine Abgangsgruppe wieder. Insbesondere gibt E eine Gruppe wieder, umfassend (a) die durch R1, R2 R3, R4 und R5 wiedergegebene Gruppe, und (b) ein Sauerstoffatom. In diesem Fall ist E über das Sauerstoffatom an die Carbonylgruppe gebunden. E gibt ebenso eine Halogengruppe wieder.
- Beispiele der Gruppe, die durch R1, R2 R3, R4 und R5 wiedergegeben wird und ein Sauerstoffatom umfasst, schließen eine Hydroxylgruppe, eine Phenyloxygruppe, eine Benzyloxygruppe und eine p-Methoxybenzyloxygruppe ein. Beispiele der Halogengruppe sind Fluor, Chlor, Brom und Iod.
- Die durch die Formel (II) wiedergegebene organische nucleophile Verbindung, die in der Kupplungsreaktion der Allylverbindung der Formel (I) verwendet wird, wird unten beschrieben. Die durch Nu in Formel (II) wiedergegebene Gruppe ist eine nucleophile Gruppe, ausgewählt aus einer α-Ketocarbaniongruppe, einer Aminogruppe und einer substituierten Aminogruppe.
- Beispiele des α-Ketocarbanions schließen organische Verbindungen mit einer wie unten beschriebenen Strukturformel ein. Beispiele solcher organischer Verbindungen schließen Aceton-α-Dehydroanion, Ethylacetat-α-Dehydroanion, Diethylmalonat-α-Dehydroanion ein.
- Der Ausdruck „substituierte Aminogruppe” bezieht sich auf eine Aminogruppe, bei der wenigstens ein Wasserstoff mit einem Substituenten ersetzt ist, wie eine Alkylgruppe, eine Cycloalkylgruppe, eine halogenierte Alkylgruppe, eine Alkoxygruppe oder ein Halogenatom. Der Ausdruck „substituierte Aminogruppe” schließt eine cyclische Aminogruppe, wie eine Morpholingruppe, eine Pyrrolidingruppe und eine 3-Oxazolin-2-on-1-yl-Gruppe ein.
- Die Kupplungsreaktion zwischen der Allylverbindung der Formel (I) und der organischen nucleophilen Verbindung der Formel (II) wird in Gegenwart eines Katalysators: durchgeführt. In der vorliegenden Erfindung wird eine Pd-Komplexverbindung mit einem Chinoxalinderivat als Ligand als Katalysator verwendet.

- Die Chinoxalinderivate der Formel (a) können in Übereinstimmung mit dem Verfahren, das in der allgemein gültig zugeteilten US-Patentanmeldung 2007/0021610 A1 offenbart ist, deren Offenbarung hierin unter Bezugnahme aufgenommen ist, hergestellt werden.
- Das das Chinoxalinderivat der Formel (a) reagiert mit dem Übergangsmetall zum Bilden einer Komplexverbindung, welche als Katalysator füx die asymmetrische Synthese verwendet werden kann. Ein Palladiumkomplex, der das Chinoxalinderivat der Formel (a) als Ligand aufweist, kann zum Beispiel durch Mischen eines 2,3-bis(Dialkylphosphin)pyrazinderivats und einer Palladiumverbindung, die eine Allylverbindung an einem Palladiumatom koordiniert hat, zum Beispiel [PdCl(η3-C3H5)]2, hergestellt werden.
- Der die Übergangs-Metallkomplexverbindung umfassende Katalysator wird bevorzugt in einer Menge von 0,0001 bis 100 Mol-% verwendet, stärker bevorzugt 0,001 bis 10 Mol-%, basierend auf dem Reaktionssubstrat. Um die Reaktion moderat zu fördern, während die Menge des Katalysators gering gehalten wird, ist eine noch stärker bevorzugte Menge von zu verwendendem Katalysator 0,02 bis 5 Mol-%.
- Um den gleichmäßigen Fortgang der asymmetrischen Kupplungsreaktion zu beschleunigen kann eine Base verwendet werden. Beispiele für geeignete Basen schließen N,O-bis(Trimethylsilyl)acetamid (BSA) Kaliumacetat und Mischungen derselben ein, sind jedoch nicht darauf beschränkt.
- Die asymmetrische Kupplungsreaktion wird gewöhnlich in einem Lösungsmittel, das üblicherweise in allgemeinen organischen chemischen Reaktionen eingesetzt wird, wie Toluol, Hexan, Tetrahydrofuran (THF), Diethylether, Dioxan, Aceton, Ethylacetat, Chlorbenzol, Dimethylformamid (DMF), Essigsäure und Wasser, durchgeführt. Bevorzugte Läsungsmittel sind Methanol, Ethanol und Dichlormethan.
- Die Menge des zu verwendenden Lösungsmittels wird entsprechend unter Berücksichtigung der Fluidität der Reaktionsmischung während der Umsetzung und der Wirkungen, die das Lösungsmittel auf die Reaktion ausübt, bestimmt. Wenn die Reaktion ohne Lösungsmittel gut voranschreitet, zum Beispiel wenn die umzusetzende Reaktantenmischung ohne Hilfe eines Lösungsmittels eine homogene Flüssigkeit mit geringer Viskosität ist, ist es nicht notwendig, ein Lösungsmittel zu verwenden.
- Die Temperatur der asymmetrischen Kupplungsreaktion beträgt bevorzugt –80°C bis 150°C, stärker bevorzugt 0°C bis 120°C, ein Bereich, in dem die Reaktion begünstigt wird, während Nebenreaktionen und Racemisierung unterdrückt werden.
- Die asymmetrische Kupplungsreaktion wird bevorzugt für eine Zeitdauer von einer Minute bis zu einem Monat, stärker bevorzugt für eine Zeitdauer von drei Stunden bis drei Tagen durchgeführt, eine Zeitdauer, die zur Vollendung der Reaktion angemessen ist.
- Die erfindungsgemäße asymmetrische Kupplungsreaktion resultiert im Allgemeinen in der Bildung einer optisch aktiven Allylverbindung der Formel (III). Wenn während der Umsetzung eine Umordnung der Abgangsgruppe stattfindet, wird die optisch aktive Allylverbindung der Formel (N) erhalten. Wenn es gewünscht ist selektiv eine von beiden zu erhalten, werden Reaktionsbedingungen, die in einer erhöhten Selektivität zu der gewünschten Verbindung resultieren, durch geeignete Auswahl des Katalysators, des Lösungsmittels und dergleichen, geschaffen. Ansonsten kann eine Mischung der beiden Verbindungen, wie sie erhalten wurde, der Kristallinisierung, Destillation, Säulenchromatographie, präparativen HPLC und so weiter unterzogen werden, um die gewünschte optisch aktive Allylverbindung zu isolieren.
- Die durch das Verfahren der Erfindung synthetisierte optisch aktive Allylverbindung kann in Form der Reaktionsmischung, wie sie erhalten wurde, verwendet werden. Wenn gewünscht, kann die Reaktionsmischung den üblichen Aufbereitungs- und Reinigungsprozeduren unterzogen werden, wie Lösungsmittelentzug, Flüssig-Flüssig-Trennung, Kristallinisierung, Destillation, Sublimation und Säulenchromatographie.
- Das Herstellungsverfahren der Erfindung kann entweder in Chargen oder kontinuierlich durchgeführt werden.
- Die durch das Verfahren der Erfindung erhaltene optisch aktive Allylverbindung wird als Zwischenprodukt für Pharmazeutika, Agrarchemikalien und physiologisch aktive Substanzen verwendet. Sie ist zum Beispiel als Zwischenprodukt für die Synthese von Antibiotika geeignet.
- Beispiele
- Die vorliegende Erfindung wird jetzt ausführlicher unter Bezugnahme auf die Beispiele erläutert, aber es soll verstanden sein, dass die Erfindung nicht so ausgelegt werden sollte, dass sie darauf beschränkt ist.
- Alle Synthesevorgänge in den Beispielen wurden unter Verwenden gründlich getrockneter Glaswaren durchgeführt. Die Umsetzung wurde in Argon- oder Stickstoffatmosphäre durchgeführt. Ein kommerziell erhältlicher Phosphinligand, (R,R)-2,3-(bis(tert-Butylmethylphosphin)chinoxalin ((R,R)-t-Bu-ChinoxP*), von Sigma-Aldrich, Japan, wurde als solcher verwendet. Das Lösungsmittel und die Metallverbindung, wie [PdCl(η3-C3H5)]2, waren allgemeine Reagenzien.
- Eine NMR-Spektrummessung wurde unter Verwenden eines NMR-Spektrometers von JEOL, Ltd. (1H: 300 MHz; 13C: 75,4 MHz; 31P: 121,4 MHz) durchgeführt. Tetramethylsilan (1H) wurde als interner Standard verwendet. Eine GC-Analyse wurde unter Verwendung eines GC-14B FID-Detektors von Shimadzu Corp. durchgeführt. Die Massenspektrometrie wurde unter Verwendung von GC-MS von Shimadzu Corp. durchgeführt.
- Beispiel 1
- Synthese von Dimethyl-(S)-α-(1,3-diphenyl-2-propen-1-yl)malonat
- In 0,5 ml Dichlormethan wurden 0,9 mg (2,5 μmol) [PdCl(η3-C3H5)]2 und 1,8 mg (5,5 μmol) (R,R)-t-Bu-ChinoxP*(1) suspendiert, gefolgt von Rühren bei Raumtemperatur für 10 bis 30 min. Zu der resultierenden Lösung wurde eine Lösung aus 126 mg (0,50 mmol) 1,3-Diphenyl-2-propenylacetat in 1,5 ml Dichlormethan, 171 μl (1,50 mmol) Dimethylmalonat, 367 μl (1,50 mmol) N,O-bis(trimethylsilyl)acetamid (BSA) und eine Spurenmenge Kaliumacetat gegeben. Die Reaktionsmischung wurde bei Raumtemperatur für eine Stunde gerührt. Die Reaktionsmischung wurde mit Diethylether verdünnt, und eine gesättigte wässrige Lösung von Ammoniumchlorid wurde zum Beenden der Reaktion zugegeben. Die resultierende Mischung wurde mit Diethylether extrahiert und die organische Schicht wurde mit einer wässrigen Natriumchloridlösung gewaschen. Die organische Schicht wurde über wasserfreiem Natriumsulfat getrocknet. Jegliche flüchtige Bestandteile wurden abgedampft und der Rückstand wurde durch Silicagel-Säulenchromatographie gereinigt.
- Beispiele 2 bis 13
- Verschiedene optisch aktive Allylverbindungen wurden auf dieselbe Weise wie in Beispiel 1 synthetisiert, außer dass die nucleophile Verbindung (Nu-H), das Additiv und so weiter, wie in Tabelle 1 unten gezeigt, verändert wurden. Die erhaltenen Ergebnisse sind in Tabelle 1 zusammen mit den Ergebnissen des Beispiels 1 gezeigt. Sofern nicht anders angegeben, war das Allylalkylierungs-Reaktionssystem 1,3-Diphenyl-2-propenylacetat (3)/Malonester oder Acetylaceton/BSA/t-Bu-ChinoxP*(1)/[PdCl(η3-C3H5)]2/CH2Cl2 = 0,50 mmol/1,5 mmol/1,5 mmol/0,0055 mmol/0,0025 mmol/2,0 ml; 1 Mol-% Pd; und das Allylaminierungs-Reaktionssystem war 1,3-Diphenyl-2-propenylacetat(3)/Amin/BSA/t-Bu-ChinoxP*(1)/[PdCl(η3-C3H5)]2/CH2Cl2 = 0,50 mmol/1,5 mmol/1,5 mmol/0,011 mmol/0,0050 mmol/2,0 ml; 2 Mol-% Pd.

- Gemäß dem Verfahren der vorliegenden Erfindung kann eine optisch aktive Allylverbindung mit hoher optischer Reinheit mit einer verringerten Anzahl von Schritten unter Verwenden einer kleinen Menge eines Katalysators hergestellt werden, und der in dem Verfahren verwendete Katalysator erreicht eine ausgezeichnete katalytische Aktivität und Enantio- und Diastereoselektivität. Die durch das Verfahren der Erfindung erhaltene optisch aktive Allylverbindung ist als Zwischenprodukt für Pharmazeutika, Agrarchemikalien und physiologisch aktive Substanzen wichtig. Sie ist zum Beispiel als Zwischenprodukt zum Synthetisieren von Antibiotika sehr geeignet. Daher besitzt die vorliegende Erfindung eine hohe industrielle Verwendbarkeit.
Claims (1)
- Verfahren zum Herstellen einer optisch aktiven Allylverbindung, wiedergegeben durch die Formel (III): worin R1, R2, R3, R4 und R5 jeweils ein Wasserstoffatom und eine Arylgruppe wiedergeben; Nu eine nucleophile Gruppe wiedergibt, ausgewählt aus einer α-Ketocarbaniongruppe, einer Aminogruppe und einer substituierten Aminogruppe; und der Stern * ein asymmetrisches Zentrum angibt, oder einer optisch aktiven Allylverbindung, wiedergegeben durch Formel (IV): worin R1, R2, R3, R4 und R5, Nu und * wie oben definiert sind, oder einer Mischung der Verbindung der Formel (III) und der Verbindung der Formel (IV), wobei das Verfahren das asymmetrische Kuppeln einer durch Formel (I) wiedergegebenen Allylverbindung: worin R1, R2, R3, R4 und R5 wie oben definiert sind; und E eine Abgangsgruppe wiedergibt, ausgewählt aus einer Gruppe, umfassend ein Sauerstoffatom und die Gruppe, die durch R1, R2, R3, R4 und R5 wiedergegeben wird und an das Sauerstoffatom gebunden ist; und eine Halogengruppe; mit einer organischen nucleophilen durch Formel (II) wiedergegebenen Verbindung:
