Description
The Invention relates to a composite material (biocomposite material, Biocer), an inorganic gel and distributed biological materials includes. The invention further relates to a process for its preparation. The composite material is used as biofilter in environmental technology Removal of heavy metal ions from aqueous solutions suitable.
- High mechanical, thermal and photochemical stability,
- - high transparency,
- - biological inertness (ie no food source for microorganisms), and
- Controllable porosity and variable degree of immobilization. Such biocomposite materials offer numerous new potential advantageous applications, e.g. B. for the production of biocompatible surfaces in medical technology, biocatalysis, biogenesis and novel drug delivery systems.
C. J. Brinker, G. Scherer, ”Sol-Gel Science: The Physics and Che mistry of Sol-Gel-Processing”, Academic Press Inc., Boston 1990).Next The possibility, biomolecules or bacterial cells on inorganic carriers such as silica gel, bentonite u. a. adsorptive on the surface to fix, as described for example in IN 171047 exists the possibility a direct embedding of biomolecules in an inorganic matrix by using the modern sol-gel technique (see C. J. Brinker, G. Scherer, "Sol-Gel Science: The Physics and Chemistry of Sol-Gel Processing, "Academic Press Inc., Boston 1990).
The procedure is evident from the following scheme (method for forming a biocomposite material (BM = biomolecule)):

- (A) by the drying process in the inorganic gel, the microorganisms are practically crushed and used as coatings after cracking z. T. torn directly,
- (b) the surviving microorganisms starve after a short time due to lack of food and are thereby deformed so that their metal-binding capacity is greatly reduced. As a result, the biofilter effect of living microorganisms in a pure SiO 2 matrix drops drastically after a short time, and such biocomposites decrease due to their low storage stability and high moisture dependency are not suitable for practical applications.

It The object of the invention is an improved composite material with biomolecules Specifically, as a filter material for the removal of Heavy metal ions such as uranium from aqueous Solutions suitable is. The composite material should be stable on storage and dry and have a high biofilter effect. It is also the job of Invention, a method for producing such a composite material specify.
These Task is by a composite material and a procedural ren to its Production with the features according to claims 1 or 8 solved. Advantageous embodiments The invention will become apparent from the dependent claims.
The The basic idea of the invention is in particular the use of biocomponents, although a similar high metal binding capacity as living microorganisms have, but much more stable against mechanical and thermal influences as well as drying effects are. Surprisingly the objects could be achieved according to the invention, that for the production of a biocomposite material that is used for biosorption heavy metals such as uranium, dry-stable cell products, namely Spores, cell wall proteins (so-called S-layers) or killed cells into an inorganic Silicon or metal oxide gel embedded according to the sol-gel method. The cell products or killed cells come from living microorganisms that consist of a heavy metal-containing Environment were isolated.
One The invention particularly relates to a biocomposite material (Biocer) containing an inorganic gel and distributed homogeneously therein, Dry-stable cell products (spores, cell wall proteins, or killed Cells). Among the dry-stable cell products are generally substances or understood materials derived from biological cells have been. It is for example by deprivation of nutrients or mechanical Influence of living cells Separates or forms cell parts, which are stable for a long time. The cell products consist of biomolecules.
The Inventors have succeeded in drying stable cell products derived from metal-binding microorganisms from a heavy metal-containing environment are derived through a porous sol-gel matrix immobilize effectively so that both biocomposites Coating of carriers or as bulk products (granulates) new industrial applications z. B. as a filler for biofilter columns.
As inorganic gel metal oxides of elements of II.-V. Main group and subgroup of the periodic table such as SiO 2 , Al 2 O 3 , ZrO 2 , TiO 2 , B 2 O 3 , ZnO, CaO, P 2 O 5 or mixtures thereof, which is obtained by a sol-gel process , By acidic or alkaline hydrolysis of the corresponding Me metal halides corresponding to the scheme shown above are formed inorganic nanosols, which can be gelled by neutralization, heating, fluoride addition or solvent evaporation to solvent-containing lyogels and converted after drying to xerogels in the form of bulk products. By coating the nanosols on any support (eg glass, paper, films, metal or textiles) and subsequent drying, it is also possible to produce thin transparent xerogel films.
For the modification of the sol-gel layer properties, the hydrolysis process of the metal alkoxides can be carried out in the presence of mixed alkyltrialkoxysilanes R-Si (OR ') 3 and / or dialkoxysilanes R 2 -Si (OR') 2 , whereby modified metal oxide gels are formed, which, based on 1 part by weight of metal oxide gel, contains 0 to 2 parts by weight of R-SiO 3/2 and / or R 2 = SiO. R is an organic alkyl or aryl radical which may contain amino, hydroxy, epoxy or alkoxy groups or is substituted by halogens. R 'is an alkyl radical, preferably with 1-4 carbon atoms. By this modification z. B. the mechanical properties of the composites are specifically improved.
For embedding the biocomponent in the inorganic gel matrix it is advantageous to remove the alcohol from the nanosols, on the one hand as a solvent for homogenization of the hydrolysis mixture (alkoxysilanes are water-insoluble), on the other hand is present as a hydrolysis product. You can do that through Evaporation of the alcohol with adequate addition of water at 20 ° C by passing reach an inert gas stream.
- (1) in spores due to reduced food supply and addition of manganese salts,
- (2) in S-layer, ie in isolated cell wall proteins, which are responsible in the cell envelopes for the metal binding ability, by mechanical destruction of the cell walls and enzymatic purification according to Engelhardt et al., J. Bacteriol. 168 (1): 309 (1986),
- (3) in killed cells by the addition of cytotoxins such as sodium azide.
The mentioned different biocomponents are called aqueous suspension with aqueous nanosols mixed and either on carriers coated or neutralized by gelation, dried and ground. The proportion of dry-stable cell products is 1-50 wt .-% based on the dry biocomposite.
Zur Erhöhung der Porosität und damit verbesserten Reaktivität gegenüber Schwermetallionen können dem Nanosol wasserlösliche Zusätze wie organische Salze, Polyole, Polycarbonsäuren, Harnstoff-Derivate oder Kohlenhydrate zugesetzt werden.to increase the porosity and thus improved reactivity across from Heavy metal ions can do that Nanosol water-soluble additions such as organic salts, polyols, polycarboxylic acids, urea derivatives or Carbohydrates are added.
The additives mentioned are gradually washed out and increased in the application of biocomposites in aqueous solutions which in addition created pores accessibility the heavy metal ions to the immobilized biocomponent. The amount of additives can be 5-50 Wt .-% based on the dry biocomposite amount.
- (1) preparation of alcoholic SiO 2 nanosols by acidic or alkaline hydrolysis of tetraalkoxysilanes, optionally modified by cohydrolysis with metal alcoholates or halides, tri- or dialkoxysilanes,
- (2) optionally substitution of the volatile alcohol by water in the nanosol by passing an inert gas stream at temperatures below 20 ° C,
- (3) homogeneously mixing the purely aqueous inorganic nanosol with an aqueous dispersion of the drying stable cell products,
- (4) Gelation by neutralization, heating or fluoride addition and subsequent comminution and freeze-drying or air drying of the gel.
To produce a layered biocomposite material is carried out after homogeneous mixing of the purely aqueous inorganic nanosol with an aqueous dispersion of the drying-stable cell products, the coating of a material surface, for. B. by dipping, spraying, watering. Subsequently, the formed composite film is dried in air or at moderate temperatures.
- High biosorbability against heavy metal ions such as uranium, copper (see examples),
- - storage of biocomposites of any length for different drying conditions and humidities without loss of bioactivity,
- Ensuring high reversibility of metal adsorption, since the metal ions can be completely removed from the Biocer by washing with 0.5 M citric acid.
- - Various application options in the form of bulk product with adjustable grain size and as a well-adhering coating with adjustable layer thickness.
The Biocomposite material according to the invention can therefore z. B. advantageous as a biofilter in environmental technology be used to remove heavy metal ions from aqueous solutions.
The In particular, the invention has the following advantages. It is possible the Sol-gel immobilization of cell products for the production of biofilters to use, the z. B. the removal of heavy metal ions from natural Water or industrial wastewater serve. In contrast to the embedding of living cells is the composite formation with cell products technologically easier to handle. The cell products can targeted and reproducibly provided and as a chemical Substance can be used. The filtering effect of the composite material is thereby reproducibly adjustable. A special advantage results from the use of spores in the composite material, since spores, which are formed for example from bacteria, on the one hand in the dry Condition extremely durable and on the other hand by moisture supply again are reactivatable. A filter with the composite material according to the invention allows thus an independent one Activation of the filter function when starting to use.

Herstellung des sauren wässrigen Nanosols BPreparation of the acidic aqueous Nanosols B
Es entsteht das saure Nanosol A (ca. 4.2% SiO2 in 70% Ethanol).100 ml of tetraethoxysilane, 400 ml of ethanol and 200 ml of 0.01 M hydrochloric acid are stirred for 20 hours at room temperature. The result is acidic nanosol A (about 4.2% SiO 2 in 70% ethanol).
For the production of bulk products after gelation of the sol, the aqueous gel formed is stored at 4 ° C. for 3 days, subsequently crushed, freeze-dried or at room temperature to constant weight air dried and classified to a particle size of 355-500 microns.

- (a) 17.23 mg (2.8 × 1010) Sporen
- (b) 36.4 mg S-layers
- (c) 36.4 mg (2.6 × 1010) abgetötete B. sphaericus Zellen.
- (a) 17.23 mg (2.8 x 10 10 ) spores
- (b) 36.4 mg S-layers
- (c) 36.4 mg (2.6 x 10 10 ) of killed B. sphaericus cells.
Zur
Bestimmung der Metallbindungskapazität wurden die Biokomposite mit
35 ml einer Lösung
0.9% NaClO4, pH 4.5 mit 9 × 10–4 M
UO2(NO3)2 × 6H2O bzw. CuCl2 bei
30°C für 48 h.
geschüttelt.
Die verbleibende Metallionen-Konzentration in der Lösung wurde
durch ICP-MS (Inductive coupled plasma mass spectroscopy) bestimmt.
Die Ergebnisse sind in
Man erkennt:
z. B. durch Gefriertrocknung oder Zusatz von 20 Gew.-% Sorbitol
(bezogen auf Oxidmatrix) beträchtlich
beschleunigt, wie aus
2 ersichtlich ist.
- (1) Cell composites have the highest metal binding capacity.
- (2) The binding capacity for uranium is considerably larger compared to copper, since the natural origin of the biocomponents means that their specific protein structure is primarily oriented towards uranium biosorption.
- (3) Biosorption is enhanced by increasing the porosity of the matrix, e.g. B. by freeze-drying or addition of 20 wt .-% sorbitol (based on oxide matrix) considerably accelerated, such as from
2 is apparent.
