Abstract

Description
技术领域

背景技术

实施例8
体外切片电生理学
[0122]水平海马切片(400μm)从3-7月龄的PDAPPJ20、PDAPP(D664A)B21和非转基因同窝对照小鼠(J20小鼠被选择用来与B21比较,因为它们表达相似水平的APP转基因)制得。用标准方法制备切片(参见,例如,Contractor,A.et al.J Neurosci23:422-9(2003))。简言之,用异氟烷麻醉动物并断头。移走脑,在冰冷的蔗糖人造CSF(ACSF)下切片,所述人造脑脊液含有85mM NaCl、2.5mM KCl、1.25mMNaH2PO4、25mM NaHCO3、25mM葡萄糖、75mM蔗糖、0.5mM CaCl2和4mMMgCl2,并用还含有10μM D,L-APV和100μM犬尿喹啉酸的95%O2和5%CO2进行平衡。在记录之前,在室温下温育切片至少1小时,在标准ACSF中进行,所述ACSF含有:125mM NaCl、2.4mM KCl、1.2mM NaH2PO4、25mM NaHCO3、25mM葡萄糖、1mM CaCl2和2mM MgCl2。在记录期间,用含有2mM CaCl2和1mM MgCl2的ACSF持续灌注切片。
[0123]用注满细胞外液的玻璃记录管(尖端电阻是2-3MOhm),在海马CA1区的辐射层记录细胞外场电位(fEPSPs)。使用通有恒定电流的同心双极电极记录响应于Schaffer侧支/联合通路刺激的诱发fEPSPs。基础突触传递通过比较记录的fEPSPs的输入和输出关联而被测定(Hsia et al);输入为纤维群峰的峰幅度,输出为fEPSP的起始斜率。对于每一动物,测定纤维群峰(FV)幅度和fEPSP反应的起始斜率,刺激范围为5-800μA,对这两种值绘制反应曲线。随后通过用fEPSP的斜率除以FV 隔度(从沿着反应曲线上的线性部分的各点)并取得平均值,计算输入输出关联。用40ms的刺激间间隔引发双脉冲易化,比值测定为fEPSP(2)的峰值/fEPSP(1)的峰值。用单因素方差分析(ANOVA)并接着用Tukey post-hoc检验来分析结果。
实施例9
神经发生
[0124]将BrdU(50mg/kg)溶解在盐水中并以8小时间隔每日两次腹膜内给予,持续3日,1周后处死小鼠。用小鼠单克隆抗BrdU(Roche,2μg/ml)和生物素化的山羊抗小鼠IgG(Vector,1∶200)对脑切片染色,用二氨基联苯胺和H2O2对染色显色。在每一小鼠的间隔200μm的5-7、50-μm冠状切片中,在不知内情的情况下计数齿状回中的BrdU阳性细胞。在带有Magnifire数字照相机的NikonE800显微镜上,在高倍率下对细胞进行计数,图像显示在计算机监视器上。结果表示为每一切片的BrdU阳性细胞的平均数。

Claims (21)
1.非人转基因动物,其包含编码突变型人β淀粉样前体蛋白(APP)或APP样蛋白的核苷酸序列,其中所述突变型人APP或APP样蛋白包括使得所述突变型人APP或APP样蛋白对Asp664处的切割具有抵抗性的突变,并且其中编码所述突变体APP或APP样蛋白的所述核苷酸序列可操纵地与适当的启动子连接。
2.权利要求1所述的转基因动物,其中在Asp664处的切割是胱天蛋白酶诱导的。
3.权利要求1所述的转基因动物,其中所述突变型人APP或APP样蛋白包括在残基664处的突变。
4.权利要求3所述的转基因动物,其中所述在残基664处的突变将Asp改变为Ala、Glu或Gln。

11.权利要求1所述的转基因动物,其中所述动物是啮齿动物。
12.权利要求1所述的转基因动物,其中所述动物是小鼠。
13.治疗神经变性疾病的方法,所述方法包括阻断蛋白酶切割β淀粉样前体蛋白(APP)和/或β淀粉样蛋白(Aβ)产生羧基端肽APP-C31的能力。
14.权利要求13的方法,其中所述蛋白酶切割APP和/或Aβ的能力是被抗蛋白酶抗体、基于蛋白酶编码序列的反义核苷酸、肽、肽模拟物、核酶、干扰RNAs、蛋白酶拮抗剂、或阻断蛋白酶切割APP和/或Aβ的胞质内区域的能力的小分子阻断的。
15.防止海马突触丢失的方法,所述方法包括阻断胱天蛋白酶切割β淀粉样前体蛋白(APP)的能力。
16.防止齿状回萎缩的方法,所述方法包括阻断胱天蛋白酶切割β淀粉样前体蛋白(APP)的能力。
17.鉴定用于治疗神经变性疾病的化合物的方法,所述方法包括鉴定阻断蛋白酶切割β淀粉样前体蛋白(APP)产生羧基端肽APP-C31的能力的化合物。
18.权利要求17所述的方法,其中阻断胱天蛋白酶切割β淀粉样前体蛋白(APP)产尘羧基端肽APP-C31的能力的化合物是通过在胱天蛋白酶存在的情况下使APP或APP样蛋白与测试化合物接触,和监测APP-P31的形成而被鉴定的,其中胱天蛋白酶不能诱导APP-P31产生提示了用于治疗神经变性疾病的化合物。
19.权利要求17所述的方法,其中阻断胱天蛋白酶切割β淀粉样前体蛋白(APP)产生羧基端肽APP-C31的能力的化合物是通过在胱天蛋白酶存在的情况下使APP或APP样蛋白与测试化合物接触,和筛选切割证据而被鉴定的,所述证据选自切割产物的易位、细胞死亡的诱导、萎缩的诱导和突触丢失的诱导。
20.用于治疗神经变性疾病的方法,所述方法包括向需要的对象施用有效量的通过权利要求17所述的方法鉴定出的化合物。
21.用于治疗神经变性疾病的基因治疗方法,所述方法包括将突变导入β淀粉样前体蛋白(APP),以至于防止胱天蛋白酶介导的对APP的切割。
