Abstract

Description
技术领域
本发明涉及活化内皮生长因子的方法,更具体地讲,本发明涉及用纤溶酶(plasmin)活化血管内皮生长因子D和血管内皮生长因子C的方法。另外,本发明也涉及识别上述内皮生长因子的活化和/或抑制因子的方法和实验,这些内皮生长因子包括VEGF-D和VEGF-C。
背景技术
血管生成(angiogenesis)是组织正常生长和发育必需的基本过程,其涉及从已有的血管增生出新的毛细血管。血管生成不仅仅包含在胚胎发育以及正常组织生长、修复和再生中,还包含在女性生殖周期中如怀孕的发生和维持以及创伤和骨折的修复中。除在正常个体发生血管生成外,许多病理过程也涉及血管生成的增加,如肿瘤生长和转移及其它的血管增殖状态,尤其是微血管***中,如糖尿病性视网膜病、牛皮癣及关节病。抑制血管生成有助于抑制或减轻这些病理过程。
另一方面,某些情况下我们需要促进血管生成,此时要建立或扩大血管化的过程,例如在组织或器官移植后,或者要促进组织梗死或动脉狭窄中侧支循环的建立,如在冠心病以及血管闭塞性脉管炎中。
因为在许多生理性和病理性过程中血管生成发挥重要作用,因此,我们对控制血管生成的相关因素进行了深入研究。许多生长因子被发现与调节血管生成有关,其中包括成纤维细胞生长因子(FGFs)、血小板源性生长因子(PDGF)、转化生长因子α(TGFα)以及肝细胞生长因子(HGF),如参阅:Folkman等,″Angiogenesis″,J.Biol.Chem.,267:10931-10934,1992。
有证据表明一个独特的内皮细胞特异性生长因子及其相应受体的家族起着如下主要作用:刺激内皮细胞生长和分化以及分化细胞的某些功能。这些因子是PDGF家族成员,并且通过内皮细胞受体酪氨酸激酶(RTKs)发挥作用。至少四种血管内皮细胞生长因子亚型已经得以识别。
已经从几个来源中分离出血管内皮细胞生长因子(VEGF),现在也叫VEGF-A,。VEGF-A呈现出对内皮细胞高度的特异性促有丝***活性,并且可以刺激导致血管生成的整个事件的发生。此外,它还对单核细胞具有强烈的趋化作用,能够诱导内皮细胞中的纤溶酶原活化因子和纤溶酶原活化因子抑制剂,并且还可影响微血管的通透性。后一种活性,有时被称作血管通透性因子(VPF)。关于VEGF的分离和特性的相关综述,可参见Ferrara等,″The Vascular EndothelialGrowth Factor Family of Polypeptides″,J.Cell.Biochem.,47:211-218,1991以及Connolly,″Vascular Permeability Factor:A Unique Regulatorof Blood Vessel Function″,J.Cellular Biochem.,47:219-223,1991。
最近,VEGF家族的另外三个成员得以识别,分别被指定为VEGF-B(在Ludwig Institute for Cancer Research and The University ofHelsinki的国际申请中有描述:PCT/US96/02957(WO 96/26736))、VEGF-C(在以下文献中有描述:Joukov等,EMBO J.,1996,15:290-298)以及VEGF2(在Human Genome Sciences,Inc的国际申请中有描述:PCT/US94/05291(WO 95/24473))。在血管生成和其它特性方面,VEGF-B与VEGF极为相似,但在组织中的分布和表达方面存在差异。尤其是,VEGF-B在心脏强烈表达,在肺部表达较弱,而VEGF与之相反。这表明尽管VEGF和VEGF-B在许多组织中共同表达,但是在功能上可能存在差异。
利用酵母共杂交作用搜集筛选技术来分离VEGF-B,对可能同I型细胞视黄酸结合蛋白(CRABP-I)作用的细胞蛋白进行筛选。其分离方法以及特性的详细描述参见:PCT/US96/02597以及Olofsson等,Proc.Natl.Acad.Sci.,1996 93:2576-2581。
VEGF-C从PC-3***癌细胞系(CRL1435)的条件培养基中分离出来,通过对能够产生内皮细胞特异性受体酪氨酸激酶Flt-4的酪氨酸磷酸化的培养基进行筛选而得到,其中利用细胞转染来表达Flt-4。VEGF-C利用重组Flt-4的亲和色谱法进行纯化,并且根据PC-3 cDNA文库进行克隆。其分离方法和特性的详细描述见:Joukov等,EMBOJ.,15:290-298,1996。
VEGF-C作为一种前蛋白进行合成,其中受体结合VEGF同源性结构域(VHD)通过肽的氨基末端和羧基末端进行侧面相连。成熟VHD的生物合成包括前肽蛋白水解的去除,以及VHD对VEGFR-2和VEGFR-3亲和力的大幅增长,其中VEGFR-2和VEGFR-3是未被加工过的全长形态(Joukov等(1997),EMBO 16:3898-3911)。故蛋白水解的处理活化了VEGF-C。这说明VEGF-C在***内皮中发挥原发性功能,在血管生成和通透性调节方面发挥继发性功能,其中血管通透性的调节是与VEGF共同承担(Joukov等,EMBO J.,1996 15:290-298)。
VEGF2从高度肿瘤发生性、非***依赖的人乳腺癌细胞系中分离而来。而这种分子据称与PDGF大约有22%的同源性,与VEGF有30%的同源性,编码VEGF2的基因的分离方法还不清楚,并且还未揭示其生物活性特征。
血管内皮生长因子通过结合到PDGF受体家族的受体酪氨酸激酶发挥作用。已经有五种内皮细胞特异性受体酪氨酸激酶得以识别,分别命名为FLT-1(VEGFR-1)、KDR/FLK-1(VEGFR-2)、FLT-4(VEGFR-3)、Tie以及Tek/Tie-2。所有这些受体均具有信号转导所必需的内在酪氨酸激酶活性。
在小鼠胚胎中利用靶向变异使这些受体失活,证明Flt-1、Flk-1、Tie以及Tek/Tie-2在血管生成和血管生成中能发挥特异性的作用。
VEGFR-1和VEGFR-2与VEGF以高亲和力相结合,并且VEGFR-1也同VEGF-B以及胎盘生长因子(P1GF)相结合。VEGF-C作为Flt-4(VEGFR-3)的配体,并且还可以活化VEGFR-2(Joukov等,EMBO J.,15:290-298,1996)。Tek/Tie-2的配体已经得以阐明(Regeneron Pharmaceutcals,Inc.的PCT/US95/12935(WO 96/11269)),而Tie的配体尚未得以识别。
受体Flt-4在胎儿的静脉和***内皮中表达,并且在成人的***内皮中显著表达(Kaipainen等,Cancer Res,1994 54:6571-6577;Proc.Natl.ACAD.SCI.USA,92:3566-3570,1995)。
血管内皮生长因子D(VEGF-D)是一种分泌的糖蛋白,可以结合以及活化VEGF受体-2(VEGFR-2)和VEGFR-3(Achen等,Proc.Natl.Acad.Sci.USA 95:548-553,1998),细胞表面受体酪氨酸激酶在血管和***内皮中分别显著表达(Stacker等,FASEB J.16:922-934,2002)。VEGFR-3为***生成(lymphangiogenesis)的信号(Veikkola等,EMBO J.20:1223-1231,2001),而VEGFR-2被认为是血管生成的信号。如所期望的,鉴于人VEGF-D的受体特异性,这种生长因子可以刺激血管生成和***生成(Byzova等,Blood 99:4434-4442,2002;Veikkola等,EMBO J.20:1223-1231,2001;Marconcini等,Proc.Natl.Acad.Sci.USA 96:9671-9676,1999)。
重要的是,VEGF-D刺激肿瘤血管生成,从而增强实体瘤的生长,以及诱导***生成,促进肿瘤细胞转移扩散到***和***中(Stacker等,Nature Med.7:186-191,2001)。最近有报道说VEGF-D的表达是结直肠癌全生存率和无瘤生存率的一个独立预后因素(White等,Cancer Res.62:1669-1675,2002)。

因此,VEGF-D和VEGF-C的蛋白水解过程是一种活化生长因子的机制。然而,这种活化过程中所涉及的蛋白酶类还是未知的。许多生物学过程中也涉及这种活化,如调整蛋白定位、生物利用度及受体特异性的过程。而这些过程与不同的疾病相关。根据患者所需对这些过程及其中的活化因子进行抑制或者活化,从而可提供治疗选择。
已知某种临时基质在血管生成中发挥重要作用。该临时基质充当内皮细胞粘附、迁移和侵入的底物,对于内皮细胞的生存是必需的。如同重塑过程,临时基质不断的生成和分解,直到新血管完全形成。
这种临时基质的重塑通过大量分子的平衡活动进行高度调节。纤溶酶,一种通过激活其酶原纤溶酶原而形成的丝氨酸蛋白酶,在介导该临时基质的降解方面发挥重要作用。因为纤溶酶在临时基质的重塑过程中的重要作用,所以通过对纤溶酶原活化因子、纤溶酶原活化因子抑制剂以及纤溶酶抑制剂之间进行复杂的调协,来严密控制纤溶酶水平,可参见国际申请WO 01/62799 A2。
发明内容
本发明首次确定出一种能够活化***生成的生长因子的蛋白酶。在一个实施方式中,本发明提供了一种方法来活化至少一种血管内皮生长因子,包括用丝氨酸蛋白酶来处理该至少一种血管内皮生长因子,该至少一种血管内皮生长因子选自VEGF-C和VEGF-D中。丝氨酸蛋白酶优选为纤溶酶(plasmin,胞质素)。至少一种的血管内皮生长因子可以是未加工或部分加工过的形态。
在另一个实施方式中,本发明提供了一种方法来筛选可以活化VEGF-C和VEGF-D中至少一种的蛋白酶,其中,VEGF-D具有C端或N端的至少一端,这种方法包括用候选的蛋白酶来处理VEGF-C或VEGF-D中至少一种,以及检测VHD,而VHD的检测表明候选的蛋白酶是否能够活化VEGF-C或VEGF-D。
在更进一步的实施方式中,本发明提供了一种方法来确定纤溶酶活化VEGF-C或VEGF-D中至少一种的抑制剂。此方法包括将VEGF-C或VEGF-D中至少一种同候选物质和纤溶酶相混合,以及测量从该VEGF-C或VEGF-D中至少一种释放的VHD的抑制情况。优选的是,此方法进一步包括如下内容:检验所谓的候选物质是否除了抑制VEGF-C或VEGF-D之外,还抑制其它纤溶酶底物的降解,如果某种物质抑制纤溶酶对VHD的释放,但是并不抑制其它物质的降解,那么就表明这种物质为VEGF-C或VEGF-D活性的抑制剂。如此确定的抑制剂仅仅抑制纤溶酶对VEGF-C或VEGF-D的活性,但并不另外影响纤溶酶对其他底物的活性。虽然不希望作任何理论方面的限制,但是可以相信这种抑制剂结合到VEGF-C或VEGF-D上,从而抑制纤溶酶对其的活性。
本发明进一步提供了一种治疗方法,这种方法包括给予患者所需要的、至少一种纤溶酶抑制剂的有效剂量。优选的是,这种纤溶酶活化VEGF-C或VEGF-D的抑制剂并不另外影响纤溶酶对其他底物的活性。优选的是,这种抑制剂是VEGF-C或VEGF-D的某种抗体或其免疫活性片断。
本发明还提供了一种治疗方法,该方法包括给予患者所需要的、有效剂量的纤溶酶,来活化VEGF-C或VEGF-D或二者一起。同时,本发明也提供了某种药物组合物来活化VEGF-C或VEGF-D或二者一起,这种组合物含有有效剂量的纤溶酶以及药物学上适宜的赋形剂。优选地,此治疗方法包括给予患者所需要的、有效剂量的上述药物组合物。优选地,该方法使用某种抗体或其片断,其中,这种抗体或其片断可以结合到VEGF-C或VEGF-D中至少一种,从而抑制纤溶酶对VEGF-C或VEGF-D中至少一种的活化。

图2所示为纤溶酶对VEGF-D的蛋白酶解——蛋白质印迹分析(Western blotting)。图2a为用10、1、0.1或0U/ml纤溶酶消化后,人VEGF-D-FULL-N-FLAG(100ng/lane)与抗-VHD抗体的分析。图2b为纤溶酶的α2-抗纤溶酶抑制。纤溶酶(1U/ml;130nM)同不同浓度的α2-抗纤溶酶在加入VEGF-D-FULL-N-FLAG之前共同在37℃孵育1小时。α2-抗纤溶酶:纤溶酶的摩尔比率显示在印迹上方。图2c为小鼠VEGF-D同型异构体(100ng/labe)的分析。小鼠VEGF-D358(358)和VEGF-D326(326)用1U/ml纤溶酶处理(+)或不消化(-)。分子量标记的大小(kDa)显示在每一面板的左边。
图3所示为纤溶酶生成的成熟生长因子结合以及交联受体。图3a所示为结合到可溶性受体。含有人VEGFR-2或VEGFR-3的细胞外结构域的受体-Ig融合蛋白与蛋白-A琼脂糖接合,并与成熟的重组人VEGF-D共同孵育作为阳性对照(成熟的),PBS作为阴性对照,未消化和纤溶酶消化的全长VEGF-D(未消化和消化)。上部为VEGFR-2结合,下部为VEGFR-3结合。结合到受体-Ig融合蛋白的物质利用抗-VHD抗体进行蛋白印迹分析。纤溶酶产生成熟VEGF-D(-21kDa)比较明显。分子量(kDa)大小显示在左边。图3b和图3c所示为Ba/F3生物检测中受体结合和交联的分析。用纤溶酶消化的或未消化的全长VEGF-D(b)或VEGF-C(c)进行处理Ba/F3细胞,该Ba/F3细胞表达嵌合受体,其包含VEGFR-2或VEGFR-3的细胞外结构域以及EpoR的细胞质结构域。上部面板:VEGFR-2/EpoR生物检测。下部面板:VEGFR-3/EpoR生物检测。对照是缺乏生长因子的介质(介质)或缺乏生长因子的纤溶酶消化(纤溶酶)。数值为两次重复平均值±标准误,并且代表三次实验。同未消化物质比较的纤溶酶消化结果的P值用Students′t试验进行计算。

图5显示了同VEGF-C的VEGFR-2生物检测(左边)和VEGFR-3生物检测(右边)。结果显示了未消化的全长VEGF-C和纤溶酶消化的物质。无VEGF-C的为阴性对照。数值为三次重复平均值±标准误,消化同未消化样本比较的P值用Students’t试验进行计算。
具体实施方式
本发明提供了一种方法,即利用蛋白酶纤溶酶活化VEGF-C或VEGF-D或二者一起,并公开了确定VEGF-C或VEGF-D蛋白水解过程中抑制剂或活化剂的实验。
以前,用来处理VEGF-D和VEGF-C的蛋白酶还未为人所知。本发明首次发现丝氨酸蛋白酶纤溶酶可以剪切未加工和部分加工过的VEGF-D以及VEGF-C形态,从而产生具有生物学活性的成熟形态。在下面的实施例2中,发明者在利用VEGFR-2和VEGFR-3的生物测定中证明VEGF-C同样由纤溶酶活化。

对来自于小鼠肺部的VEGF-D的分析表明,仅仅一部分、而不是全部的VEGF-D属于短链约21kDa全激活状态(Stacker等,J.Biol.Chem.274:32127-32136,1999)。因而,局部给予纤溶酶可以增高体内VEGF-C或VEGF-D或二者(下文用“VEGF-C/D”表示)的活性,因为额外的纤溶酶可以将剩余的全长或部分加工的生长因子转换为全活性状态,从而提升了组织中总的VEGF-C或VEGF-D的活性。
在一个优选的实施方式中,活化VEGF-C/VEGF-D的方法用来处理淋巴水肿。在淋巴水肿中,因为不适当的淋巴引流而致的组织膨胀可以导致***或***损伤。已知VEGF-C活性的升高(通过VEGF-C蛋白的局部释放)可以引起淋巴水肿组织中***的生长,从而帮助消除淋巴水肿(Szuba等,FASEB J.16:1985-1987,2002)。
已知VEGF-C和VEGF-D可以诱导血管生成(Byzova等,Blood 99:4434-4442,2002;Cao等,Proc.Natl.Acad.Sci.USA 95:14389-14394,1998)。因此,局部给予纤溶酶可以用于治疗某些疾病,例如缺血症,其中纤溶酶能够激活这些分子并促使血管生长。
在周围血管或者冠状动脉疾病的血管成形术后,VEGF-C/D活性的升高有利于引起血管修复,阻止血管再狭窄。VEGF-C/D活性的升高也有利于引起血管修复来阻止血管移植后的狭窄,血管移植可用于心脏旁路治疗或治疗外周缺血。VEGF-C/D活性的升高进一步还有利于引起治疗性血管生成或动脉血管形成,用来治疗缺血性心脏疾病和其它缺血疾病,包括外周组织缺血。
因此,根据本发明的另外一个实施方式,VEGF-C/D的纤溶酶活化可以通过应用配体结合分子来阻止其同处理蛋白酶相互作用而加以抑制。根据本发明,能够结合VEGF-C/D的物质可以用来阻止这些生长因子的活化。这些抑制活化作用的物质可以用来治疗某些疾病,例如癌症。首选的可以用来治疗的癌症属于以下情况:癌瘤中的VEGF-C或VEGF-D诱导肿瘤***生成,从而促使肿瘤细胞沿***转移扩散(Stacker等,Nature Med.7:186-191,2001;Skobe等,NatureMed.7:192-198,2001;Mandriota等,EMBO J.20:672-682,2001)。因此这些生长因子的抑制成为癌症治疗中的抗转移手段。VEGF-C/D也可以引起肿瘤血管生成,刺激实体瘤的生长(Stacker等,Nature Med.7:186-191,2001),因此这种抑制也有利于实体瘤的治疗。
可以用VEGF-C/D抑制剂治疗的其它疾病包括***瘤、***肉瘤以及非控制性血管增生性疾病,例如糖尿病性视网膜病以及关节炎。VEGF-C/D的抑制也可以阻止眼睛新生血管的形成,眼睛新生血管的形成的特征是新生血管侵入到眼睛结构中,例如视网膜或角膜。这是失明最常见的原因,并且涉及大约二十种眼部疾病,例如糖尿病性视网膜病,以及严重的年龄相关的黄斑变性。在严重的年龄相关的黄斑变性中,相关的视觉问题由以下因素引起:脉络毛细管经由正常人眼玻璃膜(Bruch′s membrane)缺损处向内生长,伴有视网膜色素上皮下的纤维管组织的增殖。
某种抗VEGF-D抗体可以优先选择给予需要VEGF-C/D抑制剂的患者,这种抗体可以阻止VEGF-D结合到VEGFR-2和VEGFR-3。在另一个具体实施方式中,可溶解的细胞外VEGFR-3结构域可用来隔离VEGF-C和VEGF-D,从而达到抑制效用。在动物模型中的研究表明,VEGFR-3信号旁路的抑制阻止了肿瘤***的生成以及沿***进行转移(Stacker,S.A.,C.Caesar,M.E.Baldwin,G.E.Thornton,R.A.Williams,R.Prevo,D.G.Jackson,S.-I.Nishikawa,H.Kubo和M.G.Achen.2001.VEGF-D promotes the metastatic spread of tumor cellsvia the lymphatics.Nat.Med.7:186-191;He,Y.,K.Kozaki,T.Karpanen,K.Koshikawa,S.Yla-Herttuala,T.Takahashi和K.Alitalo.2002.Suppression of tumor lymphangiogenesis and lymph nodemetastasis by blocking vascular endothelial growth factor receptor 3signaling.J;Natl.Cancer Inst.94:819-825)。

在一个优选的实施方式中,活化的封闭物质是一种对抗VEGF-C/D的抗体。
在又一实施方式中,本发明提供了一种方法来筛选特异性封闭纤溶酶对VEGF-C以及VEGF-D活性的抑制剂。VEGF-C/D活性的抑制可以通过纤溶酶或其他丝氨酸蛋白酶抑制剂来完成。已知纤溶酶可以剪切许多其它蛋白质(包括VEGF-A和血纤维蛋白凝块)。由于潜在的较大范围的副作用,通常的纤溶酶抑制剂也许并不是非常有效的。相反,仅封闭纤溶酶对VEGF-C和VEGF-D活化能力的抑制剂反而比较有效,可特异性的阻止肿瘤***生成以及血管生成。
本发明中的筛选方法就是用来鉴别这种抑制剂。在一个具体实施方式中,这样的抑制剂可以与VEGF-C/D结合,抑制其同纤溶酶相互作用,导致纤溶酶对VEGF-C/D活化的抑制,但是并不干扰其剪切与VEGF-C/D不相关的蛋白质能力,例如临时基质的重新塑造。
这里所使用的“VEGF-D”总体上是指血管内皮生长因子D,及其具有VEGF-D生物活性的片断或类似物,如同本领域内所熟知的。例如这种生物活性实例包括受体结合以及内皮细胞增殖。
这里所使用的“VEGF-C”总体上是指血管内皮生长因子C,及其具有VEGF-C生物活性的片断或类似物,如同本领域内所熟知的。例如这种生物活性实例包括受体结合以及内皮细胞增殖。
这里所使用的“抗体及其片断”包括任何以及所有生物活性部分,或者完全抗体,包括但不局限于Fab、Fab2、Fscv、Fab′等。抗体类型及其片断可在任何免疫学教科书中找到,例如Paul的FundamentalImmunoloy.

肽的标记:将pH6.0、200μl 200mM的2-吗啉代乙磺酸(MES)中的1毫克生物素化的肽(Auspep,Parkville,Australia)在戊烷中(PerkinElmer Lifesciences,Boston,MA)同455μCi的[3H]N-乙基马来酰亚胺(NEM)相混合,在N2下移除戊烷。在室温下孵育10分钟后,添加含有50μg NEM的pH6.0的200mM MES并在室温下孵育1小时。进一步添加含有500μg NEM的pH6.0的200mM MES并在室温下孵育1小时。在150mM NaCl中利用Seph adex G-10的色谱法将标记的多肽从未整合的标记物分离开。
VEGF-C和VEGF-D的纯化:VEGF-C-FULL-N-FLAG或VEGF-D-FULL-N-FLAG(在N端用FLAG八肽标记的全长人VEGF-C或VEGF-D)、VEGF-DΔNΔCFLAG(在N端用标记物标记的人VEGF-D的VHD)(Stacker,S.A.,K.Stenvers,C.Caesar,A.Vitali,T.Domagala,E.Nice,S.Roufail,R.J.Simpson,R.Moritz,T.Karpanen,K.Alitalo和M.G.Achen.1999.Biosynthesis of vascular endothelial growth factor-Dinvolves proteolytic processing which generates non-covalent homo-dimers.J.Biol.Chem.274:32127-32136)和小鼠VEGF-D326-FLAG以及VEGF-D358-FLAG(在C端用标记物标记的全长异构体)(Baldwin,M.E.,S.Roufail,M.M.Halford,K.Alitalo,S.A.Stacker和M.G.Achen.2001.Multiple forms of mouse vascular endothelial growth factor-D aregenerated by RNA splicing and proteolysis.J Biol.Chem.276:44307-44314)从转染293EBNA细胞的条件培养基中纯化。
蛋白酶水解:蛋白酶在pH7.5、150mM NaCl和10mM磷酸钾缓冲液中,37℃下消化一个小时。消化液包含来自于人血清的10-102U/ml的纤溶酶(Calbiochem,San Diego,CA)。在加入VEGF-D-FULL-N-FLAG之前,α2-抗纤溶酶(Calbiochem)与纤溶酶在PBS中室温条件下孵育30分钟,在37℃消化1小时。然后用含有0.5-50kU/ml酶的组织纤溶酶原活化因子(Calbiochem)进行消化。近似闪烁检测包含纤溶酶(0.1U/ml)、凝血酶(0.1U/ml)、AMP-2(5mU/ml)或MMP-9(9mU/ml)(Calbiochem)。
近似闪烁检测(scintillation proximity assay):用蛋白酶孵育后,104cpm标记的生物素化的肽在pH6.0、10mM磷酸钾缓冲液中同200mg链霉抗生物素闪烁珠(Amersham Biosciences,Uppsala,Sweden)相混合,在β-计数之前室温下反应20分钟。
蛋白印迹分析:SDS-PAGE和转膜之后,用生物素化的小鼠VEGF-D的VHD抗体以及HRP接合的链霉抗生物素抗体探查蛋白质,并用化学荧光(Pierce Biotechnology,Rockford,IL)进行显影。
氨基酸测序:氨基酸N端序列用双相NH2-端蛋白质测序仪(ModelG1005A,Agilent Technologies,Palo Alto,CA)进行分析。
质谱分析:将肽用μC18 ZipTips(Millipore,Bedford,MA)除去盐分,然后与含有2,5-二羟苯甲酸基质(Agilent Technologies)的0.1%TFA/60%氰化甲烷共同结晶于10×10MALDI不锈钢盘(AppliedBiosystems,Foster City,CA)上,并干燥10min。在o-MALDI QStarTMPulsar质谱计(Applied Biosystems)上分析样本,每分钟收集700-3000Da的阳性TOF MS。
受体结合和交叉连接的测定:同VEGF-D-FULL-N-FLAG以及可溶性受体-Ig融合蛋白的结合测定如先前所描述的进行,即用Ba/F3和750ng/ml的配体进行生物测定(Achen等,1998,Proc.Natl.Acad.Sci.USA 95:548-553),可溶性受体-Ig融合蛋白包含人VEGFR-2或VEGFR-3的细胞外结构域以及人IgGI的Fc部分(分别来自Y.Gunji,Haartman Institute和K.Pajusola,Biotechnology Institute,Helsinki)。
实施例1:VEGF-D处理的分析
为了确定活化VEGF-D的蛋白酶,用近似闪烁检测(SPA)来监测C端前肽从VHD的剪切。此测定基于C端剪切是因为这种剪切只出现在VEGF-D的一个部位,而N端前肽的剪切较为复杂,有两个截然不同的部位(Stacker等,1999,Biosynthesis of vascular endothelialgrowth factor-D involves proteolytic processing which generatesnon-covalent homodimers,J.Biol.Chem.274:32127-32136)。为了这一测定,将跨VEGF-D C-端剪切部位的一种17肽(包含人VEGF-D的198-213残基)的N端进行生物素化,C端用放射性同位素进行标记(图1a)。SPA的原理在图1b进行了概述。将多肽用蛋白酶处理后,其结合到闪烁素饱和的链霉抗生物素接合的珠子上。当多肽是完整的时候,其C端放射性标记对闪烁物质的亲近可以充分的产生可见的光子。相反,当多肽出现剪切的时候,计数检测显著减少,这是因为放射性标记物不能有效地接近闪烁物质从而产生光子。
蛋白酶类的范围在此测定中进行了检验,包括纤溶酶\凝血酶以及基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)和MMP-9。这些蛋白酶被选定是因为其涉及血管生成或肿瘤形成。MMP-2和MMP-9对SPA检测中的计数无作用,而纤溶酶引起超过90%的信号减少,指示了肽的实质性剪切(图1c)。凝血酶引起少量的信号减少。为了确定蛋白酶水解肽的部位,样本用质谱测定进行分析。如所预期的,未经消化的肽包含一个分子质量2282.15的单峰(图1d,上部面板)。随后纤溶酶处理可观察到一个主要的分子质量1267.68的峰,与Biotin-HPYSIIRR相符合(图1d,下部面板)。如在293EBNA细胞中表达的VEGF-D所观察到的,这一分子种类是在肽的相同部位剪切的预期产物,例如R205和S206之间(图1a)(Stacker等,1999,J.Biol.Chem.274:32127-32136)。某种生成Biotin-HPYSIIR(分子质量1111.59)的可选择的剪切因子也可以检测到。
实施例2:纤溶酶处理VEGF-D。
为了确定VEGF-D是否是纤溶酶作用的底物,将这种蛋白酶同全长人VEGF-D(VEGF-D-FULL-N-FLAG)共同孵育,其中,全长人VEGF-D从转染293EBNA细胞的培养基中纯化。一定程度的蛋白水解出现在这些细胞的培养基中,产生含有全长材料(约50kDa)的VEGF-D标本,以及包含N端前肽和VHD的部分处理过的形态(~31kDa)(图2a)(StackerR,S.A.,K.Stenvers,C.Caesear,A.Vitali,T.Domagala,E.Nice,S.Roufall,R.J.Simpson,R.Moritz,T.Karpanen,K.Alitalo和M.G Achen.1999.Biosynthesis of vascular endothelial growthfactor-D involves proteolytic processing which generates non-covalenthomodimers.J.Biol.Chem.274:32127-32136)。纤溶酶消化后,用蛋白印迹可检测到一条约21kDa的条带(图2a)。这一种类与先前观察到的成熟的全部处理过的VEGF-D形态相符合,表明纤溶酶可以将N端和C端前肽从VHD分离。THIS-21kDa的N端氨基酸序列分析显示了两种序列:FAATFYDIE和VIDEE,表明N端前肽的剪切发生在两个部位。FAATF(VEGF-D的89-93残基)代表完全处理过的、成熟VEGF-D主要形态的N端序列,这种VEGF-D从293EBNA细胞条件培养基中纯化而来。VIDEE(VEGF-D的101-105残基)代表N端,此N端定位于朝向C端的一个残基,与293EBNA细胞培养基中检测到的成熟VEGF-D(KVIDEE)其它形态相比较而言(Stacker,S.A.,K.Stenvers,C.Caesar,A.Vitali,T.Domagala,E.Nice,S.Roufail,R.J.Simpson,R.Moritz,T.Karpanen,K.Alitalo和M.G.Achen.1999.Biosynthesis of vascular endothelial growth factor-D involves proteolyticprocessing which generates non-covalent homodimers.J.Biol.Chem.274:32127-32136)。因此,纤溶酶将N端前肽从VHD的基本相同的部位剪切下来,这些部位在先前已得以描述。与纤溶酶相反,丝氨酸蛋白酶凝血因子和组织纤溶酶原活化因子不能将人VEGF-D前肽从VHD剪切下来(未给出数据)。
本研究中使用的纤溶酶是从人血浆中纯化而来。为了去除因纤溶酶标本污染而导致的VEGF-D处理的可能性,α2-抗纤溶酶,一种能够同纤溶酶1∶1复合产生灭活的特异性抑制剂(Collen等,1991,Blood78:3114-3124),在VEGF-D消化之前与纤溶酶样本共同孵育。当纤溶酶的量超过5倍摩尔量时,对消化产物的分析证明消化受α2-抗纤溶酶的完全抑制(图2b)。因此,观察到的纤溶酶样本对VEGF-D的蛋白水解作用归因于纤溶酶。
全长小鼠VEGF-D有两个亚型(同型异构体):VEGF-D326和VEGF-D358,它们在蛋白的C端存在差异(Baldwin等,2001,J:Biol.Chem.276:44307-44314)。为了分析不同C端对蛋白水解的作用,用纤溶酶对小鼠VEGF-D亚型进行消化。两种亚型的纤溶酶处理产生含VHD的约21kDa的成分,表明该酶可完全处理这两种亚型(图2c)。
实施例3:纤溶酶产生生物活性的VEGF-D和VEGF-C。
为了确定是否由纤溶酶产生的成熟VEGF-D结合VEGFR-2和VEGFR-3,我们利用可溶性受体-Ig融合蛋白进行免疫沉淀研究,此融合蛋白含有VEGFR-2或VEGFR-3的细胞外结构域(图3a)。这一研究揭示了由纤溶酶产生的成熟形态与VEGFR-2和VEGFR-3的细胞外结构域相结合。
为了比较全长和由纤溶酶产生的成熟VEGF-D在细胞表面结合以及交联受体的能力,利用Ba/F3和前B(pre-B)细胞进行生物测定,这些细胞表达嵌合受体,包括人VEGFR-2或VEGFR-3的细胞外结构域,以及***受体(EpoR)的跨膜和细胞质内结构域(Stacker等,1999,A Mutant form of vascular endothelial growth factor(VEGF)that lacks VEGF receptor-2 activation retains the ability toinduce vascular permeability,J.Biol.Chem.274:34884-34892;Achen等,2000,Monoclonal antibodies to vascular endothelial growth factor-Dblock interactions with both VEGF receptor-2 and VEGF receptor-3,Eur.J.Biochem.267:2505-2515)。这些细胞系是IL-3依赖性的,然而,从EpoR细胞质结构域发出的信号可以在缺乏IL-3的情况下使细胞生存和增殖,此信号在嵌合受体的细胞外结构域由配体进行交联时出现。这些生物测定对结合以及交联的受体进行比较,并且用来说明VEGFR-2和VEGFR-3配体的受体间相互作用(Achen等,1998,Vascular endothelial growth factor D(VEGF-D)is a ligand for thetyrosine kinases VEGF receptor 2(Flk-1)and VEGF receptor 3(Flt-4),Proc.Natl.Acad.Sci.USA 95:548-553;Stacker等,1999,A mutant formof vascular endothelial growth factor(VEGF)that lacks VEGF receptor-2activation retains the ability to induce vascular permeability,J.Biol.Chem.274:34884-34892;Achen等,2000,Monoclonal antibodies tovascular endothelial growth factor-D block interactions with both VEGFreceptor-2 and VEGF receptor-3,Eur.J.Biochem.267:2505-2515;Wise等,1999,Vascular endothelial growth factor(VEGF)-like protein fromorf virus NZ2 binds to VEGFR2 and neuropilin-1,Proc.Natl.Acad.Sci.USA 96:3071-3076)。在缺乏IL-3的情况下,细胞同未经消化的或者纤溶酶消化的VEGF-D进行孵育,通过整合[3H]胸腺嘧啶脱氧核苷入DNA来评估增生性应答。表达VEGFR-2或者VEGFR-3嵌合受体的细胞暴露于纤溶酶消化的VEGF-D,较那些未经消化的蛋白处理的细胞而言,呈现出更大的应答(图3b)。因此,对比全长的VEGF-D,纤溶酶处理而产生的成熟VEGF-D形态更能够在细胞表面结合和交联VEGFR-2以及VEGFR-3。在VEGF-C观察到的可比较性的结果(图3c),表明纤溶酶活化两种已知的***生成性生长因子。
实施例4:纤溶酶释放成熟VEGF-D
质粒pEFBOS-SS-Myc-VEGF-D358-FLAG编码全长小鼠VEGF-D358,在N端用两个Myc抗原决定簇进行标记,在C端用FLAG八肽(octapeptide)进行标记(Baldwin等,J.Biol.Chem.,47:44307-44314,2001),根据厂家说明书,该质粒利用Fugene来短暂转染293EBNA细胞。利用抗标记性单克隆抗体M2(Sigma),用亲和性色谱法从细胞条件培养基中纯化小鼠VEGF-D358。纯化的小鼠VEGF-D(100ng)在37℃、10mM磷酸钾、150mM NaCl、pH7.5条件下与0、1×10-4、1×10-5、1×10-6、1×10-7单位的纤溶酶(Calbiochem)孵育1小时。作为对照,小鼠VEGF-D在37℃与0、1×10-4、1×10-5、1×10-6、1×10-7单位的凝血酶(Calbiochem)孵育1小时。
利用抗VEGF-D中的VHD的生物素化抗体(R & D Systems)以及链霉抗生物素-辣根过氧化物酶接合物(Zymed),将因此而产生的物质进行蛋白印迹分析。图4显示了蛋白印迹分析的结果。对照样本中的大量条带(即未与蛋白酶孵育的样本;条带C)在先前已经得以识别(Stacker等,J.Biol.Chem.274:32127-32136,1999)并描述如下:56kDa条带是全长VEGF-D,48kDa条带包括结合到C端前肽的VHD,33kDa条带是结合到N端前肽的VHD,21kDa条带是成熟VEGF-D的亚单位。与纤溶酶孵育完全改变了这些种类的相关丰富程度。当同1×10-4单位纤溶酶孵育时,除了成熟形态的VEGF-D,其他所有均检测不到,而成熟形态(21kDa)则显著地提高(图4,纤溶酶条带10-4)。
这表明纤溶酶蛋白水解性将VHD从全长物质以及部分处理过的形态中释放出来。与1×10-4单位处理的结果相比较,这种效应是剂量依赖性的,即随着纤溶酶浓度的降低,VHD变的越来越少,而其它种类则不断增多(图1,纤溶酶条带10-5-10-7)。相反,凝血酶的效用充其量只是边缘性的,即使在其最高浓度时(图1,凝血酶条带10-4-10-7)。这些结果证明纤溶酶活化VEGF-D,可以将成熟VEGF-D从未经处理的和部分处理的形态中释放出来。
实施例5:VEGF-D作用的抑制剂的鉴别
为了确立一种方法来筛选VEGF-D作用的抑制剂,我们利用近似闪烁检测法(″SPA″)来检测VEGF-D的C端前肽从VHD的剪切。具有以下氨基酸序列的某种肽类用标准方法进行合成,包含在N端的生物素部分(bio):
bio-His-Pro-Tyr-Ser-Ile-Ile-Arg-Arg-Ser-Ile-Gln-Ile-Pro-Glu-Glu-Asp-Cys(SEQ ID NO:1)
这种肽的1-16残基与人VEGF-D的198-213残基相应(Achen等,Proc.Natl.Acad.Sci.USA 95:548-553,1998),并且VHD从C端前肽的分离在此肽序列的C端精氨酸后立即出现(Stacker等,J.Biol.Chem.274:32127-32136,1999)。通过同乙基马来酰亚胺(N-[ethyl-1,2-3H],Perkin Elmer)作用在C端放射性标记此肽(Brown等,Anal.Biochem.217:139-147,1994)。
放射性标记的肽同纤溶酶(0、0.001、0.005、0.01单位)(Calbiochem)或凝血酶(0.001、0.005、0.01单位)(Calbiochem)在10mM磷酸钾、150mM NaCl、pH7.5条件下,在全容量为10μl的eppendorf管于37℃孵育1小时。随后产物同链霉抗生物素接合的闪烁珠(AmershamPharmacia)在eppendorf管中孵育20分钟,转移到Unifilter 96孔板中进行完全反应,并用Topcount NXT微孔板闪烁计数器(Packard)计数。
SPA的理论基础是肽的C端的放射性标记仅仅能被β-计数检测到,如果其与闪烁珠中的闪烁物质有非常紧密的亲和力的话。反过来,这又只出现在如果肽在闪烁珠上结合到链霉抗生物素部分(经由肽N端生物素化部分),以及如果肽仍然是完整的(即如果C端放射性标记还未因蛋白水解而从N端生物素化部分上分离)。相反,肽的蛋白水解剪切使得C端放射性标记对闪烁珠中的闪烁物质并不足够亲密到能够被β-计数所检测到。
总之,肽的蛋白水解剪切使VEGF-D的VHD从C端前肽上分离,而导致计数检测的显著降低。在不同量的纤溶酶或凝血酶条件下进行的测试结果显示在表1。结果表明,在所有测试浓度下,纤溶酶剪切放射标记的肽,如同计数检测之减少所指示的,而凝血酶则不可以。这些结果与前面讨论过的蛋白印迹中所观察到相一致。
本文中描述的SPA将来可被用于进行大范围的抑制剂的筛选,这些抑制剂是指抑制VEGF-D的VHD从C端前肽分离。例如,一些与筛选方法相关的抑制剂可以是小分子、抗体或者肽模拟物。这种试验或者方法可以用于鉴别纤溶酶抑制剂或者任何其它蛋白酶的抑制剂,这些蛋白酶能够执行这种剪切。
尽管最初的SPA是利用C端前肽的剪切来完成,然而,这种方法和检测***可以稍微修改,并且也可以用来鉴别VHD从VEGF-DN端前肽剪切的抑制剂。此外,这些方法也可以用于VEGF家族的其它成员,如VEGF-C。
表1显示了近似闪烁检测的结果,此检测用于VHD和VEGF-D的C端前肽结合部位的蛋白水解。

实施例6:VEGF-C的纤溶酶活化
VEGFR-2和VEGFR-3的结合和交联的生物检测在白介素-3(IL-3)依赖的Ba/F3前B细胞系中进行(参见国际申请PCT/US95/16755和PCT/US97/14696;Achen等(2000),Eur.J.Biochem.267,2505-2515;Stacker等(1999),J.Biol.Chem.274:34884-34892)。在缺乏IL-3的情况下,Ba/F3细胞在48h内就会死亡。来源于Ba/F3的细胞系表达嵌合受体,包含VEGFR-2或VEGFR-3的细胞外结构域,以及***受体(EpoR)的跨膜和细胞质内结构域。嵌合受体的细胞外结构域通过嵌合配体来交联,从而经由细胞质EpoR结构域产生信号,导致细胞存活和扩增。因此,在没有IL-3的情况下,VEGFR-2和VEGFR-3配体的活化可以促进这些细胞系的存活。
N端用FLAG八肽标记的人重组全长VEGF-C,利用抗-FLAG亲和色谱法将其从293EBNA细胞的条件培养基中纯化出来,这发生在短暂的转染质粒pEFBOS-S-FLAG-hVEGF-C-Full之后,该质粒为pEFBOS-S-FLAG的衍生物(Stacker等(1999),J:Biol.Chem.274:32127-32136),其中FLAG标记的全长人VEGF-C的编码区域在延伸因子-1的控制之下,该因子是一种基因增强剂。VEGF-C在37℃、PBs中用来自于人血清(IXLO-3U,Calbiochem)的纤溶酶消化1小时。
表达VEGFR-2/EpoR或VEGFR-3/EpoR嵌合受体的BA/F3细胞在PBS中洗涤三次,在缺乏IL-3的介质中洗涤一次,并在缺乏IL-3的介质中以7.4×104cells/ml的浓度进行重悬。大约10,000个细胞(135μl)加到96孔细胞培养板的每孔中。每一孔再加入250ng未消化的全长VEGF-C或者250ng纤溶酶消化的VEGF-C,或者含有15μl无IL-3的介质的空白对照。在37℃,10%CO2环境中孵育48小时后,每孔中加入1μCi[3H]胸腺嘧啶脱氧核苷并孵育4小时。然后收集细胞,并用闪烁计数评估其生存能力。
结果显示在图5。纤溶酶消化的VEGF-C导致VEGFR-2/EpoR和VEGFR-3/EpoR系的扩增均显著增高,因此,纤溶酶活化VEGF-C。
实施例7:利用抗VEGF-D的抗体和/或抗VEGF-C的抗体来抑制纤
溶酶对VEGF-D和/或VEGF-C的活化
利用标准方法产生全长或部分VEGF-D蛋白的抗体。初步的检测表明这些抗体阻止纤溶酶和VEGF-D蛋白的相互作用。用全长或部分VEGF-C的抗体可以重复这一结果。
实施例8:药物组合物和诊断试剂盒
纤溶酶和/或其它活性蛋白酶的抑制剂同合适的辅助剂结合形成药物学上可接受的形态。“患者”包括人和其它哺乳动物。抑制剂包括但不限于单克隆抗体、多克隆抗体、前述抗体的片段以及其它可以抑制血管内皮生长因子途径的分子。
本发明也包括诊断试剂盒,其包括含至少一种可用于筛选检验的商业手段,该手段将纤溶酶或其它活性蛋白酶或其抑制剂整合其中。
上面的描述及实例只是阐述本发明,而不是加以限制。本领域的熟练人员,完全可以根据此处所公开的内容,对本发明进行相应的改变,因此,本发明的保护范围应理解为权利要求及其等同变化。
Claims (17)
1、一种活化至少一种血管内皮生长因子的方法,所述的至少一种血管内皮生长因子选自于VEGF-C和VEGF-D中,其中,该方法包括用丝氨酸蛋白酶处理所述的至少一种血管内皮生长因子。
2、如权利要求1所述的方法,其中,所述的丝氨酸蛋白酶为纤溶酶。
3、如权利要求1所述的方法,其中,所述的至少一种血管内皮生长因子为未经处理的形式或部分经过处理的形式。
4、如权利要求1所述的方法,其中,所述的血管内皮生长因子为VEGF-D。
5、如权利要求1所述的方法,其中,所述的血管内皮生长因子为VEGF-C。
6、一种筛选蛋白酶的方法,该蛋白酶可以活化VEGF-C和VEGF-D中的至少一种,其中,所述的VEGF-C或VEGF-D具有C-前肽和N-前肽中的至少一种,该方法包括用候选的蛋白酶处理VEGF-C和VEGF-D中的至少一种,然后检测VHD;其中,VHD的检测指示出所述的候选蛋白酶活化VEGF-C或VEGF-D的能力。

15、一种处理方法,包括给予患者所需的、如权利要求14所述的药物组合物的有效剂量。
16、一种抑制VEGF-C或VEGF-D或二者的药物组合物,该组合物包括VEGF-C和/或VEGF-D纤溶酶活化的抑制剂和药物学上可接受的赋形剂。
17、如权利要求15所述的方法,其中,所述的抑制剂是抗体或该抗体的片段,其中,所述的抗体或其片段与VEGF-C和VEGF-D中至少一种相结合,而且,所述的抗体或其片段阻止纤溶酶活化所述的VEGF-C和VEGF-D中至少一种。
