Abstract
本发明涉及含有细菌染色体DNA片段和无毒脂多糖的免疫刺激和控制组合物。在工业上,本发明的组合物可被应用于治疗癌症的有效材料和佐剂。
Description
技术领域
本发明涉及含有细菌染色体DNA片段和无毒脂多糖的免疫刺激和控制组合物。
背景技术
自19世纪60年代以来发展的癌症疗法主要包括使用手术、放射疗法和化学疗法。这些治疗方法已经显示出了疗效,即一直剧增到1973年的美国癌症死亡率上升曲线开始变得缓慢。然而,手术和放射疗法是局部治疗,因此它们存在局限性,即只有当癌症在早期被阻断为局部癌症时患者才会顺利地康复。只有当所有癌症细胞被完全消灭时化学疗法才成功,所以化学疗法可能会损害宿主、正常组织,如患者的免疫***,并会威胁老年患者和体弱患者的生命。免疫疗法的主要目的是通过增强免疫监视来抵抗癌化。以下是几个试验。
1)免疫预防:给同类动物接种癌组织来预防同源的癌症。例如,动物的病毒性白血病可以通过使用其致病病毒来预防(Morton等。1991年,proc.am.Assoc.cancer res.2:492:494)。然而,该方法从未被应用于人类,并且很难诱导细胞免疫。
2)免疫疗法:
■主动而特异的免疫作用
该免疫作用是通过给患者接种自体癌细胞或同源癌细胞或受X-射线照射或丝裂霉素C调控的失活的自体或异体癌细胞来避免癌细胞激活特异的免疫癌症监控细胞。然而,该方法只在动物实验中而没有在人类中成功。最近,为提高癌组织中特异抗原的表达,已经使用了多种方法,如用神经氨酸酶处理以结合伴刀豆球蛋白A(Con-A)或使隐藏的抗原暴露出来,或者与异源细胞形成杂交瘤。然而,树突细胞的使用(Sprinzl等,Cancer Treat Rev.2001年8月;27(4):247-55)或其它多种DNA疫苗药物的开发(Pantuck等,Int J Urol.2001年7月;8(7):S1-4)在其安全性和功效上仍然有局限。
■非特异性免疫疗法
目前这种免疫作用备受瞩目,其可单独使用或与化疗试剂联合使用来治疗几乎所有类型的肿瘤。非特异性免疫疗法意味着它并不受肿瘤的类型的限制。虽然已经提出了关于其机理的多种理论,然而这些理论仍处于研究之中,只是指出了非特异性免疫疗法可特异地刺激淋巴细胞的网状内皮***。在临床试验中,棒状杆菌实际上作为首选材料来使用。已在韩国用于患者的溶链菌(Picibanil)(OK-432),已经被研制并主要在日本制药公司中生产。其已经在日本、韩国或东南亚销售。很早以前,由溶链菌形成的材料已经在癌症治疗中使用。1968年,德国的Buch Fehleison等人发现癌症停止发展或原先存在的癌症消失。1891年,美国芝加哥的外科医生Coley用从链球菌培养基中提取的物质制成了混合毒素,并将其用于许多患者。
■BCG(或结核菌)及其相关材料
活的BCG生物体:在20世纪60年代,美国的Old和法国的Mathe报道了动物癌症可以通过接种BCG而治愈。1970年,美国的Morton报道了黑素瘤也可以通过接种BCG而治愈。因此,BCG及其相关材料作为非特异性免疫药物被广泛使用。要增加免疫反应需要接种大量BCG。BCG能够在皮肤下接种、或直接在癌组织区域接种或口服。然而,口服BCG对于在新生期已经接种BCG的人来说没有效果,但对之后接触过结核菌的人有效(结核菌素为阳性的人不吸收BCG或结核菌)。用活的BCG生物体治疗存在副作用,如需要大量活的BCG生物体并且注射区周围会产生溃疡,***症状如发冷、发热或肝功能紊乱。然而,如果使用小剂量来减少副作用,那么功效会降低或减弱。
■未甲基化的CpG DNA
哺乳动物DNA与细菌DNA不同之处在于它们具有许多CpG抑制作用并且CpG二核苷酸的胞嘧啶被选择性地甲基化。最近,已经认识到细菌DNA中的CpG基元(motifs)迅速刺激多克隆的B细胞,因而增加了IgM的分泌,然后通过抗IgM抗体来阻止细胞周期进程,有效地抑制细胞程序死亡(apoptosis)的诱导以抑制c-myc的表达,并使得myn、blc2和bcl-XL mRNA表达增加以此来防止细胞遭受细胞程序死亡。在其它研究中,已报道CpG基元可直接激活B细胞使得在短时间内增加IL-6和IL-12的分泌。美国的CPG公司正在进行关于免疫佐剂以及使用合成的含CpG序列的寡核苷酸治疗哮喘的临床试验。
如上所述,虽然已经开发了使用不同免疫调控物质的药物,但是在这些药物中只有BCG和CpG应用于人类。尽管BCG具有广谱效应,但是由于其稳定性而很难使用大量的BCG或通过血液注射。至于CpG,合成的寡核苷酸过于昂贵。
发明内容
因此,本发明的目的是提供用于诱导免疫反应的比传统材料更稳定、经济、有效和特异的材料。
提供了免疫刺激和控制组合物,其含有:细菌染色体DNA片段;和无毒的细菌脂多糖。
优选细菌染色体DNA片段的大小在2.0到0.5kb之间,脂多糖在5,000到10,000道尔顿之间。
优选混合最小数量的细菌染色体DNA片段和脂多糖以此来显示本发明的效果。特别地是,本发明在质量比为500∶1到1∶500之间时显示出功效随剂量的增加而增加。在上述质量比中本发明是无毒且经济的。
优选细菌染色体DNA片段和脂多糖通过振荡而混合。
本发明的组合物可用于免疫佐剂或抗癌药物。可通过诱导第1型T辅助细胞的免疫激活作用来表现这些效果。
本发明中的细菌优选大肠杆菌或分支杆菌。更优选地,细菌是大肠杆菌,特别是
E.coli EG0021(KCCM-10374)。
在本发明的组合物中,CIA02在稳定性、细胞免疫诱导上被认为有协同效应,CIA05在免疫增强尤其是癌症药物上被认为有协同效应。
将简要描述含有细菌染色体DNA片段和无毒脂多糖的已公开的免疫刺激和控制组合物。
本发明者成功地生产出作为抗癌佐剂的细菌寡核苷酸的有效产品,并且开发出作为抗癌药物并用于适当激活作用的改良脂多糖。通过将细菌寡核苷酸和脂多糖混合最终获得一种新的免疫佐剂CIA07。
通常,脂多糖和DNA的结合可显示出协同效应。脂多糖显示出多种反应,如作为T细胞的独立抗原。在此,协同效应可能会引起严重后果,如脓毒病。
本发明者获得一个株系E.coli EG0021,其具有来自健康者肠内的大肠杆菌的短糖链脂多糖。2002年5月2日他们把该株系保存在位于韩国汉城西大门区洪界洞361-221(361-221 Hongje-dong,Seodaemun-gu,Seoul,Korea)的韩国微生物保藏中心KCCM中,其编号为No.KCCM 10374。他们建立了从该菌株中纯化脂多糖的方法。
从E.coli EG0021的基因组DNA中分离出表现免疫激活作用的E.coliDNA,CIA02。CIA02是在对该分离的DNA进行片段化以及常规处理后获得的。
通过CIA02和CIA05的结合最终获得CIA07。
附图说明

具体实施方式
参照下列实施例将更具体地描述含有细菌染色体DNA片段和无毒脂多糖的已公开的免疫刺激和控制组合物,但本发明并不限制于此。
实施例1:获得无毒株系
筛选并分离具有短糖链脂多糖的突变的E.coli
从健康者肠中分离出具有短糖链脂多糖的E.coli EG0021,并建立了从该株系中纯化脂多糖的方法。
将健康成年男性肠中分离的E.coli的液体培养的单菌落注射到实验动物Balb/c小鼠中,该步骤重复5次。
从其中选出50个株系,从平板上得到所选50个株系中的一个菌落。把该菌落溶解在4ml0.9%的生理盐水中之后,取1ml溶液转移到eppendorf管中。用2μl DNase I处理溶液并在37℃培养箱中反应1小时。DNase I处理后,用50μlRnase(10mg/ml)处理溶液并在37℃培养箱中反应1小时。然后,在其中加入100μl蛋白酶K(20mg/ml),并在37℃下反应过夜。用由此获得的各株系的LPS处理由GM-CSF区分出的人体淋巴细胞系。检测TNF-α的分泌,选出数值最小的株系(见表1)并通过电泳证实脂多糖的分子量。毒性减弱的株系在形态上或其性状上没有显示出变化。在电泳中显示分子量为5,000到10,000的脂多糖而没有分子量为50,000到100,000的脂多糖梯(见图1)。该株系被称为EG0021。

实施例2:E.coli DNA制备方法
纯化E.coli染色体DNA
将E.coli EG0021在TSB(胰蛋白酶大豆肉汤;Difco)培养基(30g/L)中37℃振荡培养10小时。
10小时培养之后,将通过8,000G离心获得的150g细胞在TE(10mM Tris,pH 8.0,25mM EDTA)缓冲液(300ml)中清洗并离心。将离心获得的细胞(150g)溶解在750ml的裂解液中(10mM Tris(pH 8.0),25mM EDTA,100ug/ml溶菌酶),并在37℃下处理1小时。
此后,在溶液中加入蛋白酶K(Sigma)至终浓度为100μg/ml,并在50℃下处理12小时。
将溶液与苯酚/氯仿/异戊醇(25∶24∶1)以1∶1的比例混合,重复三次混合以此来获得水层。
用乙醇沉淀法获得E.coli染色体DNA。
用无菌蒸馏水稀释纯化的E.coli DNA之后,使用紫外分光光度计在260nm和280nm下测定E.coli DNA的浓度。
根据下列方法计算该浓度:
双链DNA浓度(μg/ml)=OD260nm×稀释比例×50
单链DNA浓度(μg/ml)=OD260nm×稀释比例×40
OD260nm/OD280nm=1.7~1.8
E.coli DNA片段化
将纯化的E.coli染色体DNA溶解于TE缓冲液中至0.5mg/ml,并使用超声仪在玻璃烧杯中进行超声处理。-使用500watt超声仪VCX500(SonicsCo.)作为超声仪并使用630-0220(探头直径:1/2″(13mm))作为探头来一次破碎20ml该溶液。
-在此,为鉴定代表最佳效果的E.coli DNA的大小,使用超声仪,根据时间阶段,在20,000J将全部E.coli染色体DNA分开,然后通过大小进行分离(见图1)。根据大小,E.coli DNA被分成超声处理前的完整DNA(完整的,大于10kb),2.0~0.5kb,0.5~0.1kb以及低于0.1kb。
-为鉴定依大小分离的E.coli DNA的免疫增加效果,在小鼠中检测作为免疫佐剂的效果(见图2)。在ICR小鼠(4周大,雄性,20g)中注射(i.p腹腔注射)作为抗原的50μg HEL(Sigma)和作为佐剂的50μg各种E.coliDNA,一周两次。最终注射final injection后7天,收集全血并分离血清。使用ELISA方法,用HEL作为抗原来检测血清中的抗体校价(见图2a)。
分析结果表明2.0~0.5kb大小显示最高的抗体校价。此后,在重复试验中显示出大约1kb显示出了最佳效果。
使用相同的ELISA方法鉴定出血清中抗体亚类中体液免疫和细胞免疫的效果(见图2b和2c)。
根据上述结果确定的用于获得1kb E.coli DNA的超声处理的条件是20,000J,7分钟。
实施例3:从E.coli DNA中除去内毒素并检测DNA纯度
除去内毒素
超声处理后,DNA与氯仿在4℃下反应12小时,在其中用三倍体积乙醇进行处理来获得沉淀。
用终浓度至0.5%的Triton X-114(Sigma)处理该沉淀。获得的沉淀在4℃下反应4小时,在37℃下温育5分钟,然后与苯酚/氯仿/异戊醇(25∶24∶1)以1∶1的比例混合,以此获得水层。
获得的E.coli DNA用乙醇沉淀,并溶解于无热原的水中。
使用鲎变形细胞溶解物(LAL)试剂盒(BioWhittaker QCL-1000)分析已除去内毒素的DNA来检测残留的内毒素。
表2显示根据上述方法除去内毒素后,纯化的E.coli DNA(CIA002)的内毒素值和产量。

-使用GC-MSD(气相色谱/质谱法)HP-5890A/HP-5670B检测残留的有机溶剂的量。使用带50m.ultra-1柱的SIM(选择性离子监测)检测乙醇、丙酮、氯仿和苯酚(见表3)。

实施例4:从突变E.coli中纯化脂多糖(CIA04)
从突变E.coli中纯化脂多糖
使用与如上所述DNA分离方法相同的方法制备E.coli。
制备的E.coli与2倍体积的乙醇混合,4,000g离心获得沉淀。在该沉淀中加入1.5倍体积的丙酮并混合,然后4,000g离心。
在得到的沉淀中加入相同量的***并混合,然后4,000g离心。用铝箔把由此获得的细胞小球用铝箔包好,刺破铝箔,并使其干燥来测量细胞质量。此后,在每1g干重细胞中加入7.5ml提取混合物(90%的苯酚∶氯仿∶石油醚=2∶5∶8)。
把得到的溶液分到玻璃离心管中,在25℃下3,000rpm(1,200g)离心20分钟以获得上清液。把上清液置于通风橱中12小时。然后,把溶液分到玻璃离心管中,在25℃下3,000rpm(1,200g)离心20分钟,脂多糖溶解于***中,然后将其转移到eppendorf管中。在通风橱中使溶液干燥,并用化学天平测量干重。然后,在其中加入乙醇并保存至使用前。
完全除去储存于乙醇的纯化的E.coli脂多糖中的乙醇之后,使用脂多糖标准品检测脂多糖中KDO(2-酮基-3-脱氧辛酸2-Keto-3-deoxyoctonate)的含量(Lsit Biological Lab.)。根据标准品计算浓度后,使用SDS-PAGE根据大小对脂多糖进行分析,并用银染方法进行鉴定(见图3)。脂多糖分子量在5,000到10,000之间,与普通的E.coli脂多糖相比,其非常小。
实施例5:从纯化的来自突变E.coli的脂多糖中除去毒素
通过脂质A降解除去纯化的脂多糖中的毒素
将纯化的E.coli脂多糖稀释到3mg/ml的浓度,然后与0.2N NaOH以1∶1的比例混合。将得到的溶液在60℃下每10分钟振荡一次,并将其脱酰化140分钟。
-在上述溶液中加入大约占最初0.2N NaOH体积的1/5的乙酸以滴定到pH 7.0。
-pH滴定后,获得乙醇沉淀的无毒脂多糖。
-用KDO方法检测无毒脂多糖的浓度之后,用SDS-PAGE和银染方法鉴定其大小变化并与处理前的脂多糖进行比较,。
-染色结果显示出脂多糖的脂质A通过碱处理而降解,而且脂多糖变得更小(见图4)。
验证无毒脂多糖中毒素的去除
为检测无毒脂多糖的稳定性,进行了有关炎症蛋白的分泌、热原性和异常毒性的实验。
-关于炎症蛋白分泌的实验
用从高浓度到低浓度的无毒脂多糖处理THP-1(急性单核细胞白血病),检测分泌出的TNF-α的量,并与纯化的脂多糖对照组比较。
当对照组中1μg脂多糖中分泌出5pg TNF-α时,1μg无毒脂多糖中分泌出0.1pg TNF-α。在此,由毒性诱导的炎症反应显示出降低了50倍。另外,E.coli DNA中分泌的TNF-α的量也显示出低于100fg。因此,证明了无毒脂多糖是非常安全的材料(见图5)。
-关于常规安全性检测的实验
在两种以上的啮齿动物中注射高剂量的样品以观察异常的重量变化。
A、在豚鼠中的实验
使用前,对大约350g的豚鼠持续观察5天以上,其没有显示异常并且体重逐渐增加。
每只豚鼠使用5ml样品。
将样品一次注射(i.p)到两只以上的豚鼠中,并观察5天以上。
B、在小鼠中的实验
使用前,对大约5周大的小鼠持续观察5天以上,其没有显示异常并且体重逐渐增加。
将样品一次注射(i.p)到两只以上的小鼠中,并观察7天以上。
在观察期间动物没有显示异常,证明样品在本次实验中是适用的。
实验结果表明在注射样品之后没有观察到异常的重量变化(见图6a)。
-热原性试验
将疫苗注射到三只兔子之后,观察直肠温度的变化。在兔子的耳静脉中注射样品,每1kg兔子注射0.2μg/ml的样品。然后,将温度计***直肠来测量异常温度的变化。
在此,兔子的重量超过1.5kg。在被用于实验后超过三天之后,兔子可被重新使用。使用至少可检测到0.1℃温度的仪器对体温进行检测。注射器和针头在250℃加热灭菌30分钟以上。从开始使用前16小时到实验结束,只给动物供给水。尽可能使动物不被紧紧地固定着。
将温度计***到直肠60mm到90mm范围内的一个固定的深度并持续固定时间来测量体温。将注射前测量的温度定义为对照温度。对照测量后大约15分钟内将已加热到37℃的样品注射到耳静脉中。每3小时测量一次体温,并至少在注射后1小时测量。把对照温度和样品温度之间的差额定义为温度上的差异。把温度差异的最大值定义为实验动物的热原反应。在此,使用了三个动物的样品。
当三个动物的总量低于1.3℃时热原材料实验为阴性,而当超过2.5℃时为阳性。这些实验进行三次,阴性反应适用于热原材料实验。
结果在表4中显示。

实施例6:使E.coli DNA片段(CIA02)和无毒脂多糖(CIA05)结合并鉴定其活性
使E.coli DNA片段(CIA02)和无毒脂多糖(CIA05)结合
把根据各标准方法制备的E.coli DNA(CIA02)和无毒脂多糖(CIA05)混合起来以确定最佳混合方法和剂量。
为了鉴定根据混合方法而产生的效果上的变化,用两种不同的用于结合的化学材料(MPBH和SPB)处理抗原。CIA02和CIA05与改良的抗原结合。在另一种方法中,把CIA02、CIA05和抗原混合起来并振荡。另外,对各剂量的免疫增加效果进行分析。为鉴定免疫佐剂在小鼠中的效果,在ICR小鼠(4周大,雄性,20g)中注射(i.p)作为抗原的0.1ml 50ug的HEL(Sigma),且一周注射两次。最终注射的七天之后,收集全血并从中分离血清。用ELISA方法并使用HEL作为抗原而分析血清中的抗体校价(见图7a)。
对照组为0.5μg和1μg的LPS(CIA04),而实验组是抗原、0.5μgCIA05和50μg CIA,振荡混合组含有抗原、1μg CIA05和50μg CIA,结合组含有改良的抗原、1μg CIA05和50μg CIA。
实验结果证实了LPS(CIA04)显示出最高的抗体活性,但是由于它有毒性副作用而并不适用抗癌佐剂(见图7a)。虽然结合方法没什么区别,但是振荡混合方法比其他方法更好些。特别地,振荡混合方法显示出是有效的,因为在免疫球蛋白亚类中,与细胞免疫有关的IgG2a显示出差异(见图7b)。总的来说,振荡混合方法简单并且在处理过程中没有损耗。因此,选择了振荡混合方法。根据CIA05的量确定了剂量。1μg的剂量显示出的功效比0.5μg的好。因此,确定了使用1μg的剂量。
用ELISA方法分析血清中抗体亚类的体液免疫和细胞免疫的效果。
与传统免疫佐剂的比较实验
为了检测已确定的制造方法和剂量的免疫增加效果,进行与传统免疫佐剂的免疫增加效果对比的实验(见图8a)。
通过动物实验对CIA07作为免疫佐剂的适用性进行了分析。在ICR小鼠(4周人,雄性,20g)中注射(i.p)作为抗原的0.1ml 50μg的HEL(SigmaL-6876),一周注射两次。最终注射的七天之后,收集全血并从中分离血清。用ELISA方法并使用HEL作为抗原以分析血清中的抗体活性。
CIA07的抗体活性显示出与CFA和Alum相同的效果(见图8a),其中CFA(弗氏完全佐剂)是用于动物实验的传统的免疫佐剂;Alum(氢氧化铝凝胶)只被认可应用于人体。然而,分析同型转换的结果显示传统的免疫佐剂如CFA和Alum激活Th(T-辅助细胞)-2型的免疫作用,其中主要产生IgG1的抗体,而CIA07激活Th(T-辅助细胞)-1型的免疫作用,其中特异地产生IgG2a的抗体而不是IgG1的抗体(见图8b和8c)。
为确定CIA02的剂量,把1μgCIA05分别混合在25μg和50μgCIA02中。分析结果显示抗体活性根据剂量而改变。因此,把50μgCIA02和1μgCIA05确定为最佳剂量。
用全血分析进行CIA活性的鉴定试验
在含有肝素作为抗凝血剂的真空管中无菌地收集健康成人男性的静脉血。把从其中得到的全血与RPMI 1640培养基(2mML-谷氨酰胺,1mM丙酮酸钠,80μg/ml庆大霉素)以1∶1的比例混合。在1ml混有培养基的全血中加入20μl CIA07(50μgCIA02+1μg或500ng、100ng CIA05)或20μl HBSS,然后在5%的CO2培养基中于37℃下诱导24小时。用ELISA试剂盒分析培养基上清液中IFN-γ(R&D***,210-TA-010)和IL-12 p40(R&D***,219-IL-005)。结果在图9中显示。
使用同样的方法,在1ml混有培养基的全血中加入20μl CIA07(50μgCIA02+1μgCIA05)、CIA02(50μg)、分支杆菌(50μg)或HBSS(20μl)。然后,分析细胞因子的分泌。
分析的结果显示混合加入CIA02和CIA05比只加入CIA02或只加入CIA05时具有更高的协同免疫增加效果。在剂量为50μgCIA02和1μg CIA05时表现出最佳结果。与分支杆菌DNA比较,CIA07和CIA02一样显示出极好的结果。
荧光素酶检测
把原始细胞以每个孔5×104个细胞(1ml DMEM/10%FBS)铺在12孔板上,并在CO2培养箱中37℃培养24小时。在混有Fugene 6(1.5μl/孔,RocheCat.No.1 814 443)转染试剂的无血清DMEM(50μl/孔)中加入混有PRL无效质粒(20ng/孔)的IL-12荧光素酶报告基因质粒(0.2μg/孔),然后静置5至10分钟。将得到的混合物以52ul/孔的量加到原始细胞中,并在CO2培养箱中37℃培养24小时。此后,用CIA07(每孔CIA02 20μg+CIA05 400ng)处理原始细胞,并在CO2培养箱中37℃培养12小时。通过荧光素酶检测试剂盒进行荧光素酶反应以用光度计测量荧光素酶活性。
通过检测荧光素酶活性来鉴定CIA07对IL-8和IL-12启动子活性的效果,其结果是在IL-8和IL-12启动子中都存在有NF-κB结合位点。可显示NF-κB在RAW264.7细胞系中被CIA07激活来增加启动子的活性(见图11)。
使用CIA的细胞裂解活性来检测抗癌药物的效果
使用51铬释放法检测CIA07的癌细胞致死活性。
在5~8周大的雄性C3H/HeN小鼠足底的皮肤下单独注射抗原或与CIA07一起注射。
使用RPMI-1640(10mM HEPES,100单位/ml青霉素,100μg/ml链霉素,300μg/ml谷氨酰胺;Gibco实验室,Grand Island,NY)用于培养细胞系的基础培养基。在基础培养基中加入于56℃下加热30分钟的失活的10%胎牛血清。为了检测介导致死活性的LAK细胞和癌细胞的活性,使用肉瘤180和小鼠膀胱癌细胞系(MBT-2)作为靶细胞。
为制备反应细胞系,使用颈椎折断法处死实验组的老鼠。无菌分离该老鼠的脾脏,并用剪刀在不锈钢金属网上将其切碎。通过使用玻璃棒加入由磷酸盐缓冲的盐水而研磨碎片并过滤。然后,组织碎片流过金属网而被除去。在显微镜下鉴定单细胞悬浮液之后,用基础培养基把细胞洗涤一次。将细胞在37℃0.84%氯酸铵溶液中悬浮5分钟以溶解红血球。再用基础培养基把细胞洗涤两次,然后将其悬浮于完全培养基中。细胞悬浮液被分到培养瓶中,并在CO2恒温恒湿培养箱中37℃培养1小时。从其中获得没有附着在瓶上的细胞,然后用锥虫蓝染色排除法检测存活细胞数。然后,用完全培养基获得5×106的细胞并使用锥虫蓝染色排除法检测其存活细胞数。然后,使用完全培养基制成5×106/ml的细胞悬浮液。
培养靶细胞系并计算细胞数。获得106的细胞并将其300g离心3分钟。用Pasteur吸液器除去上清液,保留0.2~0.3ml的上清液,而不损伤沉淀的细胞。加入100 Ci Na2 51CrO4(1ml Ci/ml,NEZ 030S,NEN,美国),在振荡恒温器中37℃标记1小时。用基础培养基洗涤细胞,然后使用锥虫蓝染色排除法检测其存活细胞数。在完全培养基中把标记的靶细胞重悬到5×104/ml。
把标记的靶细胞按每孔0.1ml分装到圆底的96孔板中,则每孔放5×103细胞。以反应细胞∶靶细胞=100∶1的比例加入0.1ml的反应细胞。将细胞在5%CO2恒温恒湿培养箱中37℃培养4小时。每个试验进行3孔以上,在4小时培养结束后,将细胞在500g离心15分钟。用gamma计数器(Packard,美国)检测0.1ml上清液中的放射性。在此,为诱导最大放射,将0.1ml的5%Triton X-100(Sigma,美国)加入到对照孔组中。为了测定自然放射,在含相同剂量的完全培养基中培养标记的细胞。根据下列公式计算细胞毒性:
细胞毒性(%)=(ER-SR/MR-SR)×100
ER:实验组的平均数(cpm)
SR:培养基中培养的靶细胞的平均数(cpm)
MR:用5%Triton X-100处理的靶细胞的平均数(cpm)。
实验结果在表5中显示。LAK细胞与非免疫细胞相比,其细胞裂解增加了8倍,与BCG注射组相比增加了1.5倍。MBT-2细胞与非免疫细胞相比,其细胞裂解增加了5倍。这些结果表示CIA取代BCG作为抗癌药物可能会产生多种副效果。

鉴定小鼠中抗癌活性的实验
在5~8周大的雄性C3H/HeJ小鼠的两个部位(左背部,右腿部)皮下地注射5×105的MBT2细胞(C3H/He来源的膀胱癌细胞)。在此,为产生单个癌组织应避免注射出几个针孔。每个实验组使用6到10只小鼠。从注射癌细胞后第二天起,在细胞系注射位置注射100μl CIA07(50μg CIA02+500ngCIA05)或生理盐水溶液,每天注射一次并持续一周,然后在接下来的两周内每两天注射一次。用游标卡尺测量皮下生成的癌的大小,每周测量三次。结果在表12中表示。与生理盐水溶液注射组相比,CIA05注射组的癌组织生长受到抑制。
工业适用性
根据本发明将两种E.coli来源的材料CIA02和CIA05混合而得到的抗癌药物CIA07与传统药物相比具有更高的安全性,并且由于制造过程简单而将制造成本最小化。同样,由于混合了两种材料,CIA07可诱导更有效、更特异的免疫作用。另外,由于DNA的物理过程,本发明比CpG更便宜,并且比BCG更有效。
因此,根据本发明的E.coli来源的抗癌药物CIA07在抗癌药物和免疫佐剂的工业应用上更重要。
Claims (11)
1、免疫刺激和控制组合物,其包括:
a)细菌来源的DNA片段;和
b)细菌来源的无毒脂多糖。
2、如权利要求1所述的组合物,其中所述的细菌来源的DNA片段的大小在2.0到0.5kb之间。
3、如权利要求1所述的组合物,其中所述的脂多糖的道尔顿在5,000到10,000道尔顿之间。
4、如权利要求1所述的组合物,其中a)和b)的质量比在500∶1到1∶500之间。
5、如权利要求1所述的组合物,其中a)和b)是通过振荡而混合的。
6、如权利要求1所述的组合物,其中所述的组合物可被用作免疫佐剂。
7、如权利要求1所述的组合物,其中所述的组合物可被用作抗癌药物。
8、如权利要求1所述的组合物,其中所述的组合物诱导第1型T辅助细胞的免疫激活作用。
9、如权利要求1所述的组合物,其中所述的细菌是大肠杆菌或分支杆菌。
10、如权利要求1或9所述的组合物,其中所述的细菌是大肠杆菌。
11、如权利要求10所述的组合物,其中所述的大肠杆菌株系是E.coliEG0021(KCCM-10374)。
