Abstract
文冠果果壳提取物在制备提高脑功能食品和药品中的应用,文冠果果壳醇提取物包括粗脂肪2-10%,粗蛋白15-20%、文冠果总皂甙15-25%、糖20-30%、水7-12%,还含有二种甾醇。对治疗智力低下、尿频和提高人肌体抗糖尿活性具有明显疗效,还可以制成提高脑功能的功能性食品。
Description
(一)技术领域
本发明属于一种由文冠果果壳中提取的功能食品原料和药物原料。
(二)技术背景
文冠果(Xanthoceras sorbifolia)系无患子科文冠果属的木本油料植物,为我国特有种。原产我国北方黄土高原,野生,具有抗旱、耐寒、耐瘠薄、耐盐碱等特点,在我国北方多数省区有人工种植,主要用于防风固沙。
我国古书《救荒本草》记载:文冠果种仁味甘可食。
文冠果种仁含油达60%,油中富含14种不饱和脂肪酸,是质量上乘的食用油。用文冠果籽榨油食用,榨油后的油渣含丰富的蛋白质,是优质饲料。文冠果种仁油也可作工业用油和药用,如高级润滑油、增塑剂、烫伤膏、治癫痫药等,此外,民间也一直用文冠果种仁治疗遗尿症。
中国科学院沈阳应用生态研究所,从文冠果仁中提取出文冠果总皂甙、粗脂肪、粗蛋白,并证明文冠果总皂甙可提高脑功能,制备治疗智力低下的药。已向国家专利局申报了发明专利,专利号为ZL93111010.6。
文冠果果壳虽然有文献报导,可以作为生产活性炭、糠醛、木糖、酒精产品等的化工原料,这些都是根据文冠果果壳成分推断的。事实上至今无任何单位或个人把文冠果果壳作为化工原料应用,只有少量果壳被农民用作烧火柴或引火柴。总体看仍然是废弃物。经检索国内外文献,更没有文冠果果壳提取物用于制备功能食品和药品的报导。为此我们对文冠果果壳的成份进行了分析研究,提取和开发。
(三)发明内容
本发明要解决的技术问题是确定文冠果果壳的有效成份和对有效成份的提取方法及文冠果果壳提取物在制备提高脑功能食品和药品中的应用。
文冠果果壳醇提取物,包含以下成分:粗脂肪2-10%,粗蛋白15-20%、文冠果总皂甙15-25%、糖20-30%、水7-12%。
文冠果果壳用正己烷回流热提取,所得脂肪酸经甲酯化后进行气-质联用分析,测得的文冠果果壳中含有21种脂肪酸,其中己酸、庚酸、辛酸、壬酸、癸酸、10-甲基-十一烷酸、12-甲基-十四烷酸、十七烷酸等8种脂肪酸是文冠果种仁油脂中所没有的。
文冠果果壳中还含2种甾醇:(3β,5α,20R,24S)-豆甾-7,反-22二烯-3-醇和(3β,5α,20R,24R)-豆甾-7-烯-3-醇,这2种甾醇在文冠果籽及文冠果植株其它部位中均未见有相关记载,而7-烯-3-甾醇类化合物具有抗糖尿活性和抗肿瘤活性。
文冠果果壳有效物质提取工艺过程如下:
文冠果果壳经粉碎,用乙醇、甲醇、正丁醇等醇液浸提,将浸出液回收除去溶剂的固体物质,干燥后得棕色粒状粉末,精制则为黄白色粒状粉末。
本发明的核心是发现和实施了从文冠果果壳中提取文冠果总皂甙、粗脂肪、粗蛋白、甾醇等具有保健和药用价值的物质。
文冠果果壳提取物在制备提高脑功能食品和药物的应用,是将文冠果果壳提取物包括粗脂肪2-10%,粗蛋白15-20%、文冠果总皂甙15-25%、糖20-30%、水7-12%,及甾醇等物粉碎成粒状粉末,制成胶囊或片剂、冲剂等剂型。可治疗智力低下、痴呆症、遗尿病等疾病,还可使人体获得不饱合脂肪酸以及有利于抗糖尿和抗肿瘤的甾醇物质,提高人体的免疫功能。

(五)具体实施方式
实施例一
取2000g文冠果果壳,粉碎加3倍乙醇热回流提取4次,取浸提液回收乙醇,干燥粉碎,得110g棕色粒状粉末,其成份百分含量如下:

将该粉末装入胶囊中,200mg/粒,1粒/次,3次/日,可治疗小儿遗尿症;治疗中老年尿频尿急、小便失禁症、治疗儿童智力低下、治疗多发性栓塞引发的记忆障碍症。
也可以制成冲剂、颗粒剂、片剂等。
也可以制成功能食品。
实施例二
取2000g文冠果果壳,粉碎加3倍乙醇热回流提取4次,取浸出液回收乙醇,脱色、干燥粉碎,得106g黄白色粒状粉末,其成份百分含量如下:

实施例五
取实施例4之产品40g,经柱层析,高压液相制备,得白色结晶40g。该品干品含文冠果总皂甙97%,为纯品,用于作标准品。

取2000g文冠果果壳,粉碎,用3倍乙醇热回流浸提4次,取浸出液回收乙醇,用乙酸乙脂萃取,将萃取液回收乙酸乙酯,干燥粉碎,得28g棕色粒状粉末,其成份百分含量如下:

将该粒状粉末制成胶囊,片剂,颗粒剂等,可治疗糖尿病和癌症。制成功能食品,可提高人肌体抗糖原活性和抗肿瘤活性。

将实施例六所得28g产品,经柱层析和重结晶,得200mg白色结晶,为二种甾醇之纯品(干品含量99%),作标准品用。

1、文冠果果壳提取物在制备治疗脑功能低下、尿频症、提高人肌体抗糖尿活性等药物及功能性食品中的应用。
2、根据权利要求1所述的文冠果果壳提取物在制备提高脑功能食品和药品中的应用,其特征在于所述的提取物,包括粗脂肪2-10%,粗蛋白15-20%、文冠果总皂甙15-25%、糖20-30%、水7-12%。
3、根据权利要求1所述的文冠果果壳提取物在制备提高脑功能食品和药品中的应用,其特征在于所述的提取物,还含有两种甾醇:(3β,5α,20R,24S)-豆甾-7,反-22二烯-3-醇和(3β,5α,20R,24R)-豆甾-7-烯-3-醇。
