Abstract
本发明提供一种盐生杜氏藻提取物的制备方法及其应用,制备的盐生杜氏藻提取物含有丰富的多糖、β‑胡萝卜素等活性成分,具有优异的滋润保湿、抗衰抗热老化的效果,有助于皮肤再生和提高皮肤光泽度。本发明同时提供一种含盐生杜氏藻提取物的组合物,制备原料包括:盐生杜氏藻提取物0.1‑3%、珍珠提取物0.01‑2%、金黄洋甘菊提取物0.01‑1%、三七提取物0.01‑1%、小分子醇30‑40%、角鲨烷1‑5%、抗氧化剂1‑8%、螯合剂0.01‑0.1%、余量水。其以天然植物提取成分为主,温和不刺激;配合多种抗氧化、保湿等或活性成分共同使用,从抗氧化、抗糖化和抗热老化等方面防止皮肤老化,具有优异的滋润保湿、抗皱、抗热老化和保湿提亮的效果。

技术领域
本发明涉及化妆品技术领域,具体涉及一种盐生杜氏藻提取物的制备方法及其应用。
背景技术
人体的皮肤,一方面能够防止水分、营养物质等的流失,另一方面能防止外部有害物质的侵入,形成皮肤屏障,同时还参与了人体的代谢过程。但是皮肤的衰老是不可避免、且不可逆的,光照、糖分的摄取以及氧化等问题都容易导致皮肤老化,特别是强烈的紫外线对表皮层会带来直接的损伤,如果长期在室外就会导致皮肤松弛,长纹;而长期在空调房里强劲的冷风又会带走肌肤的水分,导致皮肤粗糙、长纹、老化。
目前脸部抗衰除皱的方式和产品种类繁多,但由于导致皮肤老化的原因较多,当下如何延缓皮肤衰老,有效抗衰、抗皱、抗热老化、保持皮肤紧致滋润,仍然是目前化妆品领域研究的一大课题。
盐生杜氏藻是一种绿色微藻,其含有大量的天然类胡萝卜素、维生素A、盐藻多糖等成分,用于护肤产品使用,具有优异的滋润保湿、抗衰修护的效果。因此,如何有效进行盐生杜氏藻的活性成分提取,并将其进行应用是目前的一大研究方向。
发明内容
针对上述问题,本发明提供一种盐生杜氏藻提取物的制备方法,具体步骤如下:
步骤S1:将盐生杜氏藻进行粉碎处理,烘干后过筛;
步骤S2:在藻粉中加入80%乙醇,在50-60℃进行浸提2-4h,离心得上清液A和滤渣;
步骤S3:在滤渣中加入碳酸钠水溶液,在60-70℃进行浸提2-4h,离心取上清液B,将上清液A与上清液B混合,旋蒸浓缩,100-120℃灭菌即得所述盐生杜氏藻提取物。
在一种优选的实施方式中,所述盐生杜氏藻粉碎后过100目筛。
在一种实施方式中,所述藻粉和乙醇的质量体积比为1:25-30g/ml。优选的,所述藻粉和乙醇的质量体积比为1:28g/ml。
在一种实施方式中,所述滤渣和碳酸钠水溶液的质量体积比为1:20-25g/ml。可列举的有:1:21g/ml、1:22g/ml、1:23g/ml、1:24g/ml、1:25g/ml。优选的,所述滤渣和碳酸钠溶液的质量体积比为1:23g/ml。
为了更好的进行活性成分的提取,在一种优选的实施方式中,所述碳酸钠水溶液的浓度为0.2-0.4%。更优选的,所述碳酸钠水溶液的浓度为0.3%。
优选的,所述离心速率为3000-5000r/min,离心时间为20-30min。
通过本发明的提取工艺制得的盐生杜氏藻提取物,用于化妆品产品中,具有滋润保湿、抗衰提亮的效果,其通过提高ATP水平来保护和刺激线粒体,有助于皮肤再生和提高皮肤光泽度。在进行提取时,提取溶剂、提取温度、提取时间、料液比等都有可能影响提取效率和提取物中的活性成分含量,本发明中通过使用乙醇和碳酸钠水溶液作为提取溶剂,设定特定的提取温度和时间,获得的提取物用于化妆品中具有最好的抗衰抗老化的效果。发明人分析,由于衍生杜氏藻提取物中含有大量的胡萝卜素、多糖、维生素和多种氨基酸等活性成分,随着提取时间增长、温度升高,有利于活性成分的溶出,但是若时间过长或温度过高,会使得部分活性成分如多糖出现降解等现象,反而影响使用效果。
本发明另一方面提供一种盐生杜氏藻提取物在化妆品中的应用。
在一种实施方式中,所述盐生杜氏藻提取物可用于制备一种含有盐生杜氏藻提取物的组合物,按重量百分比计,制备原料包括:
盐生杜氏藻提取物0.1-3%、珍珠提取物0.01-2%、金黄洋甘菊(CHRYSANTHELLUMINDICUM)提取物0.01-1%、三七(PANAX NOTOGINSENG)提取物0.01-1%、小分子醇30-40%、角鲨烷1-5%、抗氧化剂1-8%、螯合剂0.01-0.1%、余量水。
盐生杜氏藻提取物中含有胡萝卜素、多糖、维生素和多种氨基酸等活性成分,能够有效滋润保湿、抗衰提亮,保持皮肤弹性。珍珠提取物含有大量碳酸钙和硬蛋白,含有多种氨基酸成分和各种微量金属元素,能对皮肤起到优异的美白、抗老、保湿效果。金黄洋甘菊(CHRYSANTHELLUM INDICUM)提取物富含黄酮类活性成分,该成分具有抗氧化,消炎等功效。三七(PANAX NOTOGINSENG)提取物含皂甙,主要为人参皂甙Rb1、Rg1、Rg2和少量人参皂甙Ra、Rb2、Rb和Re,此外,尚含黄酮甙、淀粉、蛋白质、油脂等。本发明中通过使用多种植物提取物成分共同使用,具有优异的抗衰抗皱、抗热老化、消炎舒缓等效果。
在一种实施方式中,所述小分子醇为甘油、丁二醇、1,2-己二醇、中的一种或多种组合。
为了进一步提高活性成分在体系中的稳定性,在一种实施方式中,所述小分子醇为甘油、丁二醇、1,2-己二醇。在该条件下,本发明的植物提取物成分等能更好的溶解,并均匀分散,有效提高了组合物体系的稳定性。优选的,所述甘油、丁二醇、1,2-己二醇的质量比为30:4-6:2-4。更优选的,所述甘油、丁二醇、1,2-己二醇的质量比为30:5:3。
在一种实施方式中,所述抗氧化剂为维生素A、维生素C、维生素E、生育酚乙酸酯、泛醌中的一种或多种组合。
优选的,所述抗氧化剂为生育酚乙酸酯(维生素E醋酸酯)和泛醌。更优选的,所述生育酚乙酸酯和泛醌的质量比为1:(3-5)。
当泛醌和生育酚乙酸酯共同使用,会产生更强的抗氧化作用。意外的时,当配合本发明中的植物提取物等成分共同使用时,其抗氧化效果、稳定性和渗透性都有进一步的增强。发明人分析,可能是由于本发明所使用的原料中含有大量的维生素C、维生素E等成分,多种成分共同作用时有极好抗氧化等效果,但是在较高含量时,维生素C等的稳定性和渗透性都较差,本发明中通过多种植物提取物配合使用,其中含有大量的多糖、多酚、蛋白质、氨基酸等成分,分子间作用力能够使体系更稳定,如多糖与蛋白质等可能会以非共价键形态结合在一起,增强体系的稳定性,同时配合角鲨烷等成分,有效促进了活性成分的渗透。
在一种实施方式中,所述螯合剂为EDTA-2Na。
在一种优选的实施方式中,所述含有盐生杜氏藻提取物的组合物的制备方法为:
(1)将盐生杜氏藻提取物、珍珠提取物、金黄洋甘菊(CHRYSANTHELLUM INDICUM)提取物、三七(PANAX NOTOGINSENG)提取物、小分子醇混合均匀,得到混合物A;
(2)将混合物A、角鲨烷、抗氧化剂、螯合剂依次加入水中,在35-45℃条件下混合均匀,即得所述组合物。
有益效果:
本发明制备的盐生杜氏藻提取物含有丰富的多糖、β-胡萝卜素等活性成分,具有优异的滋润保湿、抗衰抗老化的效果,有助于皮肤再生和提高皮肤光泽度。本发明同时提供一种含有盐生杜氏藻提取物的抗衰抗热老化组合物,其以天然植物提取成分为主,温和不刺激;配合多种抗氧化、保湿等或活性成分共同使用,易于被皮肤吸收,能够有效清除自由基,从抗氧化、抗糖化和抗热老化等方面防止皮肤老化,具有优异的滋润保湿、抗皱、抗老化和保湿提亮的效果。

实施例1
本实施例提供一种盐生杜氏藻提取物的制备方法,具体步骤如下:
步骤S1:将盐生杜氏藻进行粉碎处理,烘干后100目过筛;
步骤S2:在藻粉中加入80%乙醇,藻粉和乙醇的质量体积比为1:28g/ml;在55℃进行浸提3h,4000r/min离心20min得上清液A和滤渣;
步骤S3:在滤渣中加入浓度为0.3%的碳酸钠水溶液,滤渣和碳酸钠水溶液的质量体积比为1:23g/ml;在65℃进行浸提3h,4000r/min离心20min取上清液B,将上清液A与上清液B混合,旋蒸浓缩,110℃灭菌30min即得所述盐生杜氏藻提取物。
实施例2
本实施例提供一种含有盐生杜氏藻提取物的组合物,按重量百分比计,其制备原料包括:
盐生杜氏藻提取物1.5%、珍珠提取物1%、金黄洋甘菊(CHRYSANTHELLUMINDICUM)提取物0.5%、三七(PANAX NOTOGINSENG)提取物0.5%、小分子醇35%、角鲨烷3%、抗氧化剂4%、螯合剂0.05%、余量水。
盐生杜氏藻提取物的制备方法同实施例1。珍珠提取物购于西安瑞尔丽生物科技有限公司;金黄洋甘菊(CHRYSANTHELLUM INDICUM)提取物购于西安维珍生物科技有限公司;三七(PANAX NOTOGINSENG)提取物购于陕西新天域生物科技有限公司。
小分子醇为质量比为30:5:3的甘油、丁二醇、1,2-己二醇;抗氧化剂为质量比为1:4的生育酚乙酸酯和泛醌,生育酚乙酸酯购于陕西晨明生物科技有公司;泛醌购于广州科进生物科技有限公司;螯合剂为EDTA-2Na。
本实施例另一方面提供一种含有盐生杜氏藻提取物的组合物的制备方法,具体步骤如下:
(1)将盐生杜氏藻提取物、珍珠提取物、金黄洋甘菊(CHRYSANTHELLUM INDICUM)提取物、三七(PANAX NOTOGINSENG)提取物、小分子醇混合均匀,得到混合物A;
(2)将混合物A、角鲨烷、抗氧化剂、螯合剂依次加入水中,在40℃条件下搅拌50min混合均匀,即得所述组合物。
实施例3
本实施例提供一种含有盐生杜氏藻提取物的组合物,按重量百分比计,其制备原料包括:
盐生杜氏藻提取物0.1%、珍珠提取物0.01%、金黄洋甘菊(CHRYSANTHELLUMINDICUM)提取物0.01%、三七(PANAX NOTOGINSENG)提取物0.01%、小分子醇30%、角鲨烷1%、抗氧化剂1%、螯合剂0.01%、余量水。

盐生杜氏藻提取物3%、珍珠提取物1%、金黄洋甘菊(CHRYSANTHELLUM INDICUM)提取物2%、三七(PANAX NOTOGINSENG)提取物1%、小分子醇40%、角鲨烷5%、抗氧化剂8%、螯合剂0.1%、余量水。

性能测试
1、多糖含量测定
使用苯酚-硫酸法进行多糖含量测定,以葡糖糖为标准品。
制作标准曲线:准确称取标准葡聚糖(或葡萄糖)20mg于500ml容量瓶中,加水至刻度,分别吸取0.4、0.6、0.8、1.0、1.2、1.4、1.6及1.8ml,各以蒸馏水补至2.0ml,然后加入6%苯酚1.0ml及浓硫酸5.0ml,摇匀静置30分钟以后于490nm测吸光度,以2.0ml水按同样显色操作为空白,横坐标为多糖微克数,纵坐标为光密度值,得标准曲线。
取样品1.0ml,加1.0ml蒸馏水,然后加入6%苯酚1.0ml,迅速加入浓硫酸5.0ml,在涡旋仪上震荡,充分混匀,静置30分钟,于490nm测定吸光度,并将测得的吸光度带入标准曲线中,可获得多糖浓度。
将实施例1、对比例1-2的盐生杜氏藻提取物进行多糖含量测试,测试结果如下表1。

2、弹性蛋白酶抑制率测试
按照实验室方法(HMC-WI-028弹性蛋白酶抑制率),对样品的抗皱功效进行测试,样品弹性蛋白酶抑制率高于阴性对照且具有显著性差异(P<0.05=,则可认为样品具有一定的抗皱功效。
样品溶液:用纯水稀释至样品浓度为1%;阳性对照物(EGCG):用水稀释至阳性对照物浓度为0.1%;阴性对照物:纯水。
设立样品组、样品本底组、溶剂组和溶剂本底组,每个组别需设立3个平行,在96孔板中分别加入不同试剂溶液,轻轻摇匀,25℃育15min后,将其放在酶标仪中,在410nm处测定吸光值。
弹性蛋白抑制率(%)=[1-(C-D)/(A-B)]×100
其中:A为不含样品的反应溶液的吸光度;B为不含样品和酶的反应溶液的吸光度;C为含有样品和酶的反应溶液的吸光度;D为含有样品和不含酶的反应溶液的吸光度。
统计分析软件为SPSS,试验样品、阳性对照物和阴性对照物弹性蛋白酶抑制率之间的比较采用独立样本t检验。上述统计分析均为双尾检验,显著性水平为Q-0.05。P>0.05,表示两组之间无显著性差异;P<0.05,表示两组之间具有显著性差异。
将实施例1、对比例1-2的盐生杜氏藻提取物和实施例2-4、对比例3-6的含有盐生杜氏藻提取物的组合物进行弹性蛋白酶抑制率测试,测试结果如下表2。

注:弹性蛋白酶抑制率保留三位小数,P<0.05,表示样品和阳性对照与阴性对照相比有显著性差异。
Claims (10)
1.一种盐生杜氏藻提取物的制备方法,其特征在于,具体步骤如下:
步骤S1:将盐生杜氏藻进行粉碎处理,烘干后过筛;
步骤S2:在藻粉中加入80%乙醇,在50-60℃进行浸提2-4h,离心得上清液A和滤渣;
步骤S3:在滤渣中加入碳酸钠水溶液,在60-70℃进行浸提2-4h,离心取上清液B,将上清液A与上清液B混合,旋蒸浓缩,100-120℃灭菌即得所述盐生杜氏藻提取物。
2.根据权利要求1所述的一种盐生杜氏藻提取物的制备方法,其特征在于,所述藻粉和乙醇的质量体积比为1:25-30g/ml。
3.根据权利要求1所述的一种盐生杜氏藻提取物的制备方法,其特征在于,所述滤渣和碳酸钠水溶液的质量体积比为1:20-25g/ml。
4.根据权利要求1所述的一种盐生杜氏藻提取物的制备方法,其特征在于,所述碳酸钠水溶液的浓度为0.3%。
5.一种根据权利要求1-4任一项所述的制备方法制备的盐生杜氏藻提取物在化妆品中的应用。
6.一种含有权利要求1-4任一项所述的制备方法制备的盐生杜氏藻提取物的组合物,其特征在于,所述含有盐生杜氏藻提取物的组合物,按重量百分比计,制备原料包括:
盐生杜氏藻提取物0.1-3%、珍珠提取物0.01-2%、金黄洋甘菊提取物0.01-1%、三七提取物0.01-1%、小分子醇30-40%、角鲨烷1-5%、抗氧化剂1-8%、螯合剂0.01-0.1%、余量水。
7.根据权利要求6所述的含有盐生杜氏藻提取物的组合物,其特征在于,所述小分子醇为甘油、丁二醇、1,2-己二醇、中的一种或多种组合。
8.根据权利要求7所述的含有盐生杜氏藻提取物的组合物,其特征在于,所述小分子醇为甘油、丁二醇、1,2-己二醇。
9.根据权利要求6所述的含有盐生杜氏藻提取物的组合物,其特征在于,所述抗氧化剂为维生素A、维生素C、维生素E、生育酚乙酸酯、泛醌中的一种或多种组合。
10.根据权利要求6所述的含有盐生杜氏藻提取物的组合物,其特征在于,所述含有盐生杜氏藻提取物的组合物的制备方法为:
(1)将盐生杜氏藻提取物、珍珠提取物、金黄洋甘菊提取物、三七提取物、小分子醇混合均匀,得到混合物A;
(2)将混合物A、角鲨烷、抗氧化剂、螯合剂依次加入水中,在35-45℃条件下混合均匀,即得所述组合物。
