人体的皮肤屏障主要由皮肤表皮的角质层构成,其中“砖—墙”结构起到保护机体内各组织器官免受外界有害侵袭以及保持机体内环境的稳定的作用。但是当个体存在自身基因缺陷、免疫紊乱或者受到外界刺激的时候,“砖—墙”结构被破坏,引起表皮细胞分化异常、脂质和蛋白合成异常和微生物群紊乱等,最后导致皮肤疾病的发生。
创伤、炎症和自然退化等造成组织缺损后,人的自身修复能力有限,仿生角质膜能够保护角质层抵御外界侵害,但目前制备方法得到仿生角质膜透气性差、生物相容性不佳。为了克服现有仿生角质膜的缺点,本发明涉及一种仿生角质膜的制备方法。
发明内容
本发明的目的就在于解决上述背景技术的问题,而提出一种仿生角质膜的制备方法。
本发明的目的可以通过以下技术方案实现:
一种仿生角质膜的制备方法,包括以下步骤:
步骤1:将乙醇先加入水相锅,然后将VP/VA共聚物缓慢加入水相锅中,搅拌直至溶解均匀;
步骤2:将聚乙二醇-8、辛酸/癸酸甘油三酯依次投入水相锅,搅拌分散均匀;
步骤3:将贻贝提取物和微囊添加剂投入水相锅,搅拌分散均匀,即得到仿生角质膜;
其中,仿生角质膜包括以下重量份原料:乙醇40-90份、VP/VA共聚物1-10份、聚乙二醇-81-20份、辛酸/癸酸甘油三酯1-20份、贻贝提取物0.01-0.2份、微囊添加剂1-6份。
作为本发明进一步的方案:微囊添加剂的制备方法包括以下步骤:
步骤1:首先将壳聚糖和复合材料溶于2wt%的醋酸溶液配成的质量浓度为2wt%的壳聚糖溶液作为分散相,溶液在磁力搅拌下搅拌过夜,以保证壳聚糖和复合材料的完全溶解;
步骤2:在30℃下,将分散相逐滴加入质量分数为2%的含有乳化剂(span-80)的液体石蜡中;搅拌乳化15min,使得体系形成稳定的油包水(w/o)的乳液之后,此时整个体系呈现出乳白色的乳液状,再向乳液中加入相应质量的戊二醛溶液,保持温度为30℃的条件下,搅拌固化2小时;
步骤3:待固化完全后,将所得到的样品,以10000rpm离心5min,倒出上层的液体石蜡,随后将离心管底下的固体;进行通过孔径为220nm的滤膜进行抽滤,并用石油醚,丙酮和去离子水先后洗涤几次获得微胶囊,即得到微囊添加剂。
作为本发明进一步的方案:壳聚糖、复合材料、2wt%的醋酸溶液、质量分数为2%的含有乳化剂(span-80)的液体石蜡和戊二醛溶液的质量比为2-5:0.2-0.8:50-80:500:10-20。
作为本发明进一步的方案:复合材料包括以下重量份原料:银耳多糖10-30份、海藻提取液10-30份、菊花蒸馏液10-30份、薄荷提取液10-30份和保鲜剂1-5份。
作为本发明进一步的方案:保鲜剂的制备工艺包括以下步骤:
步骤1:将乙基纤维素加入到无水乙醇中,开动电动搅拌器搅拌;
步骤2:再加入焦亚硫酸钠,继续搅拌,然后采用喷雾干燥即得保鲜剂。
作为本发明进一步的方案:乙基纤维素、焦亚硫酸钠和无水乙醇的质量比为10-15:10—15:100。
本发明的有益效果:
本发明通过乙基纤维素、焦亚硫酸钠复配制备得到安全无毒可以吸收的保鲜剂,其可以大大提高仿生角质膜的使用期;
然后,与银耳多糖、海藻提取液、菊花蒸馏液和薄荷提取液,制备得到复合材料,然后使得复合材料与壳聚糖之间生成含有多种有效物质的微胶囊,其中不仅可以保证有效物质的保质期,还可以进行很好的贮藏;并且在使用过程中,可以有效地进行释放,对皮肤进行很好的保养;
本发明模拟天然皮肤表皮层-角质层,具有防水透气的保护层的效果,另外负载有修复物质贻贝提取物,具有生物粘合剂、促进愈合及形成保护膜的功能,且生物亲和性良好,无毒害作用,不会引起人体产生免疫反应,可以形成透明、透气、防水及有弹性的物理屏障保护膜,保护表皮不受感染和损伤。既可以为细胞生长提供环境,有利于缺损的皮肤修复;以及通过筛选合适的组份原料搭配工艺,得到薄如蝉翼、柔软、富有弹性的纤维仿生角质膜,该制备方法步骤简易,易于制备。

下面将结合本发明实施例,对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有作出创造性劳动前提下所获得的所有其它实施例,都属于本发明保护的范围。
实施例1
本发明为一种仿生角质膜,包括以下重量份原料:乙醇40份、VP/VA共聚物1份、聚乙二醇-81份、辛酸/癸酸甘油三酯1份、贻贝提取物0.01份、微囊添加剂1份。
实施例2
本发明为一种仿生角质膜,包括以下重量份原料:乙醇70份、VP/VA共聚物6份、聚乙二醇-8 10份、辛酸/癸酸甘油三酯10份、贻贝提取物0.1份、微囊添加剂3份。
实施例3
本发明为一种仿生角质膜,包括以下重量份原料:乙醇90份、VP/VA共聚物10份、聚乙二醇-820份、辛酸/癸酸甘油三酯20份、贻贝提取物0.2份、微囊添加剂6份。
实施例4
基于上述实施例1-3,一种仿生角质膜的制备方法,包括以下步骤:
步骤1:将乙醇先加入水相锅,然后将VP/VA共聚物缓慢加入水相锅中,搅拌直至溶解均匀;
步骤2:将聚乙二醇-8、辛酸/癸酸甘油三酯依次投入水相锅,搅拌分散均匀;
步骤3:将贻贝提取物和微囊添加剂投入水相锅,搅拌分散均匀,即得到仿生角质膜;
其中,仿生角质膜的制备工艺过程中,温度控制在23-27℃。
实施例5
基于上述实施例1-4,微囊添加剂的制备方法包括以下步骤:
步骤1:首先将壳聚糖和复合材料溶于2wt%的醋酸溶液配成的质量浓度为2wt%的壳聚糖溶液作为分散相(水相),溶液在磁力搅拌下搅拌过夜,以保证壳聚糖和复合材料的完全溶解;
步骤2:在30℃下,将分散相逐滴加入质量分数为2%的含有乳化剂(span-80)的液体石蜡中(油相);搅拌乳化15min,使得体系形成稳定的油包水(w/o)的乳液之后,此时整个体系呈现出乳白色的乳液状,再向乳液中加入相应质量的戊二醛溶液,保持温度为30℃的条件下,搅拌固化2小时;

步骤2:再加入焦亚硫酸钠,在转速2000—3000r/min的条件下继续搅拌50-80min,然后采用喷雾干燥,喷雾进口温度为105-110℃,出口温度为50-58℃,喷雾干燥后收集粉末即得保鲜剂;
其中,乙基纤维素、焦亚硫酸钠和无水乙醇的质量比为10:10:100。
实施例6
微囊添加剂的制备方法包括以下步骤:
步骤1:首先将壳聚糖和复合材料溶于2wt%的醋酸溶液配成的质量浓度为2wt%的壳聚糖溶液作为分散相(水相),溶液在磁力搅拌下搅拌过夜,以保证壳聚糖和复合材料的完全溶解;
步骤2:在30℃下,将分散相逐滴加入质量分数为2%的含有乳化剂(span-80)的液体石蜡中(油相);搅拌乳化15min,使得体系形成稳定的油包水(w/o)的乳液之后,此时整个体系呈现出乳白色的乳液状,再向乳液中加入相应质量的戊二醛溶液,保持温度为30℃的条件下,搅拌固化2小时;
步骤3:待固化完全后,将所得到的样品,以10000rpm离心5min,倒出上层的液体石蜡,随后将离心管底下的固体;进行通过孔径为220nm的滤膜进行抽滤,并用石油醚,丙酮和去离子水先后洗涤几次获得微胶囊,即得到微囊添加剂。
其中,壳聚糖、复合材料、2wt%的醋酸溶液、质量分数为2%的含有乳化剂(span-80)的液体石蜡和戊二醛溶液的质量比为3:0.5:67:500:15;
其中,复合材料包括以下重量份原料:银耳多糖10-30份、海藻提取液10-30份、菊花蒸馏液10-30份、薄荷提取液10-30份和保鲜剂1-5份;
其中,保鲜剂的制备工艺包括以下步骤:
步骤1:将乙基纤维素加入到无水乙醇中,开动电动搅拌器搅拌,在转速1200-1500r/min的条件下搅拌15-30min;
步骤2:再加入焦亚硫酸钠,在转速2000-3000r/min的条件下继续搅拌50-80min,然后采用喷雾干燥,喷雾进口温度为105-110℃,出口温度为50-58℃,喷雾干燥后收集粉末即得保鲜剂;
其中,乙基纤维素、焦亚硫酸钠和无水乙醇的质量比为12:12:100。
实施例7
微囊添加剂的制备方法包括以下步骤:
步骤1:首先将壳聚糖和复合材料溶于2wt%的醋酸溶液配成的质量浓度为2wt%的壳聚糖溶液作为分散相(水相),溶液在磁力搅拌下搅拌过夜,以保证壳聚糖和复合材料的完全溶解;
步骤2:在30℃下,将分散相逐滴加入质量分数为2%的含有乳化剂(span-80)的液体石蜡中(油相);搅拌乳化15min,使得体系形成稳定的油包水(w/o)的乳液之后,此时整个体系呈现出乳白色的乳液状,再向乳液中加入相应质量的戊二醛溶液,保持温度为30℃的条件下,搅拌固化2小时;

步骤1:将乙基纤维素加入到无水乙醇中,开动电动搅拌器搅拌,在转速1200-1500r/min的条件下搅拌15-30min;
步骤2:再加入焦亚硫酸钠,在转速2000-3000r/min的条件下继续搅拌50-80min,然后采用喷雾干燥,喷雾进口温度为105-110℃,出口温度为50-58℃,喷雾干燥后收集粉末即得保鲜剂;
其中,乙基纤维素、焦亚硫酸钠和无水乙醇的质量比为15:15:100。
本发明的工作原理:本发明通过乙基纤维素、焦亚硫酸钠复配制备得到安全无毒可以吸收的保鲜剂,其可以大大提高仿生角质膜的使用期;
然后,与银耳多糖、海藻提取液、菊花蒸馏液和薄荷提取液,制备得到复合材料,然后使得复合材料与壳聚糖之间生成含有多种有效物质的微胶囊,其中不仅可以保证有效物质的保质期,还可以进行很好的贮藏;并且在使用过程中,可以有效地进行释放,对皮肤进行很好的保养;

Claims (6)
1.一种仿生角质膜的制备方法,其特征在于,包括以下步骤:
步骤1:将乙醇先加入水相锅,然后将VP/VA共聚物缓慢加入水相锅中,搅拌直至溶解均匀;
步骤2:将聚乙二醇-8、辛酸/癸酸甘油三酯依次投入水相锅,搅拌分散均匀;
步骤3:将贻贝提取物和微囊添加剂投入水相锅,搅拌分散均匀,即得到仿生角质膜;
其中,仿生角质膜包括以下重量份原料:乙醇40-90份、VP/VA共聚物1-10份、聚乙二醇-81-20份、辛酸/癸酸甘油三酯1-20份、贻贝提取物0.01-0.2份、微囊添加剂1-6份。
2.根据权利要求1所述的一种仿生角质膜的制备方法,其特征在于,微囊添加剂的制备方法包括以下步骤:
步骤1:首先将壳聚糖和复合材料溶于2wt%的醋酸溶液配成的质量浓度为2wt%的壳聚糖溶液作为分散相;
步骤2:在30℃下,将分散相逐滴加入质量分数为2%的含有乳化剂的液体石蜡中;搅拌乳化15min,再向乳液中加入戊二醛溶液,保持温度为30℃的条件下,搅拌固化2小时;
步骤3:待固化完全后,离心,抽滤,洗涤,即得到微囊添加剂。
3.根据权利要求2所述的一种仿生角质膜的制备方法,其特征在于,壳聚糖、复合材料、2wt%的醋酸溶液、质量分数为2%的含有乳化剂(span-80)的液体石蜡和戊二醛溶液的质量比为2-5:0.2-0.8:50-80:500:10-20。
4.根据权利要求3所述的一种仿生角质膜的制备方法,其特征在于,复合材料包括以下重量份原料:银耳多糖10-30份、海藻提取液10-30份、菊花蒸馏液10-30份、薄荷提取液10-30份和保鲜剂1-5份。
5.根据权利要求4所述的一种仿生角质膜的制备方法,其特征在于,保鲜剂的制备工艺包括以下步骤:
步骤1:将乙基纤维素加入到无水乙醇中,开动电动搅拌器搅拌;
步骤2:再加入焦亚硫酸钠,继续搅拌,然后采用喷雾干燥即得保鲜剂。
6.根据权利要求5所述的一种仿生角质膜的制备方法,其特征在于,乙基纤维素、焦亚硫酸钠和无水乙醇的质量比为10-15:10—15:100。
