Acne is a disease caused by multiple factors, local androgen metabolism is one of the most main causes, and excessive androgen can cause the volume of sebaceous glands to be increased and sebum secretion to be increased, the duct of the pilosebaceous glands is abnormal in keratinization, sebum excretion is not smooth, lipid embolism is formed, and acne is caused. In sebaceous gland cells, androgen can be converted into more active Dihydrotestosterone (DHT) under the action of 5 a-reductase, and further high expression of inflammatory factor IL-6 is stimulated, so that the capacity of inducing acne is enhanced. Therefore, the inhibition of the inflammatory factor IL-6 can inhibit acne, and achieve the effect of treating the acne.
This example compares the inhibition of the inflammatory factor IL-6 with a single component and a composition, respectively.
The main experimental process is as follows: raw264.7 mouse macrophage cells were cultured normally in DMEM medium containing 10% FBS at 37 ℃ in 5% CO 2. When the cells grow to 90% density, inoculating the cells into a 96-well plate according to the density of 8x105/ml, after adhering the cells to the wall for 8 hours, adding samples with different concentrations for respectively treating for 24 hours, and adding corresponding test samples into blank control groups (normal groups), LPS (induction groups), LPS + positive control groups (80 mu M dexamethasone) and LPS + sample groups, wherein the sample groups are respectively 1% of sweet wormwood herb volatile oil; and sweet wormwood volatile oil, o-cymene-5-ol and extract of Spanish sage (SALVIA HISPANICA) seeds, wherein the composition content is 1%, and the composition is prepared from sweet wormwood oil: 0.8%, cymene: 0.1 percent. Spanish sage (SALVIA HISPANICA) seed extract: 0.1 percent of the total saponin is directly mixed, cultured for 24 hours at 37 ℃ and saturated humidity with 5 percent of CO2, and supernatant is taken for detection, and each group is repeated for 3 times. The test results are shown in fig. 1.
As can be seen from FIG. 1, the secretion of inflammatory factor IL-6 by Raw264.7 cells is obviously up-regulated after LPS treatment, and the amount of IL-6 is inhibited after the positive control dexamethasone treatment. The herba Artemisiae Annuae volatile oil has inhibitory effect on inflammatory factor IL-6, but is lower than positive control dexamethasone; to date, there is no report in the literature that o-cymene-5-ol and extract of Salvia Spanish (SALVIA HISPANICA) seed have inhibitory effects on IL-6, an inflammatory factor when used alone; the inhibition rate of the composition compounded by the three components is obviously higher than that of positive control dexamethasone (P is less than 0.01), which shows that the three components of the sweet wormwood herb volatile oil, the o-cymene-5-ol and the spanish sage (SALVIA HISPANICA) seed extract have obvious synergistic effect on the inhibition effect of the inflammatory factor IL-6.
Example 3 Effect of amount of volatile oil of Artemisia annua on the inhibition of IL-6 Effect of inflammatory factor
The preparation method of the acne-removing skin care product containing the bacteriostatic composition provided by the embodiment comprises the following steps: 0.1 part of o-cymene-5-alcohol, 3 parts of dipropylene glycol and 0.3 part of sodium polyacryl dimethyl taurate are premixed, 81.9 parts of deionized water, 3 parts of 1, 2-pentanediol, 0.1 part of salvia officinalis (SALVIA HISPANICA) seed extract, 0.2 part of allantoin and 0.2 part of dipotassium glycyrrhizinate are added, the mixture is stirred uniformly and homogenized for 5 minutes at the temperature of below 35 ℃, the mixture is heated to 80-85 ℃, the mixture is cooled to below 45 ℃ after being stirred for 30 minutes at the temperature, the mixture is continuously stirred and added with sweet wormwood volatile oil, and the mixture is cooled, stood and discharged after being stirred uniformly. Wherein the addition amount of the sweet wormwood volatile oil is 0.05, 0.1, 1 and 2 parts respectively, and the rest is complemented by deionized water.
The main experimental process is as follows: raw264.7 mouse macrophage cells were cultured normally in DMEM medium containing 5% CO2 at 37 ℃ using 10% FBS. When the cells grow to 90% density, inoculating the cells into a 96-well plate according to the density of 8x105/ml, adhering the cells to the wall for 8h, adding samples with different concentrations, respectively treating for 24h, culturing at 37 ℃ and containing 5% CO2 for 24h under saturated humidity, and taking supernatant for detection. Dividing into blank control group (normal group), LPS (induced group), LPS + positive control group (80 μ M dexamethasone), and LPS + sample group, adding corresponding test sample, wherein in the positive control group, 80 μ M dexamethasone is used as positive control, judging whether inflammatory factor IL-6 is inhibited, examining the influence of different addition of composition herba Artemisiae Annuae volatile oil on inhibiting inflammatory factor IL-6 effect, and the detection result is shown in figure 3.
As can be seen from fig. 3, different amounts of the volatile artemisia apiacea oil have a large influence on the expression of the inflammatory factor IL-6, the effect of inhibiting the inflammatory factor IL-6 is gradually increased with the gradual increase of the amount of the volatile artemisia apiacea oil, but when the amount of the volatile artemisia apiacea oil is more than 1 part, the effect of inhibiting the inflammatory factor IL-6 is decreased, and therefore, the amount of the volatile artemisia apiacea oil is preferably 1 part.
Example 4 Artemisia annua volatile oil 1 part + o-cymene-5-ol 0.1 part + Salvia Spanish (SALVIA HISPANICA) seed extract 0.1 part + allantoin 0.2 part + dipotassium glycyrrhizinate 0.2 part
The preparation method of the acne-removing skin care product containing the bacteriostatic composition provided by the embodiment comprises the following steps: 0.1 part of o-cymene-5-alcohol, 3 parts of dipropylene glycol and 0.3 part of sodium polyacryl dimethyl taurate are premixed, 92.1 parts of deionized water, 3 parts of 1, 2-pentanediol, 0.1 part of salvia officinalis (SALVIA HISPANICA) seed extract, 0.2 part of allantoin and 0.2 part of dipotassium glycyrrhizinate are added, the mixture is stirred uniformly and homogenized for 5 minutes at the temperature of below 35 ℃, the mixture is heated to 80-85 ℃, the mixture is cooled to below 45 ℃ after being stirred for 30 minutes at the temperature, the mixture is continuously stirred and added with 1 part of sweet wormwood volatile oil, and the mixture is cooled, stood and discharged after being stirred uniformly.
Example 5 contains no Artemisia annua volatile oil
The difference between the present example and example 4 is that, the acne-removing skin care product containing the bacteriostatic composition is prepared without the volatile oil of artemisia apiacea as follows: 0.1 part of o-cymene-5-alcohol, 3 parts of dipropylene glycol and 0.3 part of sodium polyacryl dimethyl taurate are premixed, 93.1 parts of deionized water, 3 parts of 1, 2-pentanediol, 0.1 part of salvia sclarea (SALVIA HISPANICA) seed extract, 0.2 part of allantoin and 0.2 part of dipotassium glycyrrhizinate are added, the mixture is stirred uniformly and homogenized for 5 minutes at the temperature of below 35 ℃, the mixture is heated to 80-85 ℃, the mixture is kept warm and stirred for 30 minutes, cooled and kept stand, and the material is discharged.
Example 6 No o-cymene-5-ol
The difference between the embodiment and the embodiment 4 is that o-cymene-5-alcohol is not contained, and the acne-removing skin care product containing the antibacterial composition is prepared by the following method: 0.1 part of Spanish sage (SALVIA HISPANICA) seed extract, 0.2 part of allantoin, 0.2 part of dipotassium glycyrrhizinate and 98.5 parts of deionized water are added, the mixture is stirred uniformly at the temperature of below 35 ℃ and homogenized for 5 minutes, then the mixture is heated to 80-85 ℃, the mixture is kept warm and stirred for 30 minutes, then the temperature is reduced to below 45 ℃, the mixture is continuously stirred, 1 part of sweet wormwood herb volatile oil is added, and the mixture is cooled and kept stand after being stirred uniformly, and then the mixture is discharged.

The difference between the present example and example 4 is that, without the extract of the seeds of salvia spanish (SALVIA HISPANICA), the acne-removing skin care product containing the bacteriostatic composition is provided by the following preparation method: 0.1 part of o-cymene-5-alcohol, 3 parts of dipropylene glycol and 0.3 part of sodium polyacryloyldimethyl taurate are premixed, 92.2 parts of deionized water, 3 parts of 1, 2-pentanediol, 0.2 part of allantoin and 0.2 part of dipotassium glycyrrhizinate are added, the mixture is stirred uniformly and homogenized for 5 minutes at the temperature of below 35 ℃, the mixture is heated to 80-85 ℃, the mixture is stirred for 30 minutes under heat preservation, the temperature is reduced to below 45 ℃, the mixture is continuously stirred, 1 part of sweet wormwood volatile oil is added, and the mixture is cooled, stood and discharged after being stirred uniformly.
Example 8 No allantoin

Example 14 stability test
In this embodiment, the stability test of the acne-removing skin care products prepared in examples 4 to 13 specifically includes: the stability test of high temperature 40 ℃, low temperature-4 ℃, centrifugation 2000-4000r/min x 30min is carried out, whether the appearance shapes such as thinning, color change, layering, hardness change and the like exist or not is observed by naked eyes, whether the odor change exists or not is judged by olfaction, and the stability test result is shown in table 2.

As can be seen from Table 2, the acne-removing skin care products prepared in examples 4-13 are stable at the conditions of high temperature of 40 ℃, low temperature of-4 ℃, and centrifugation of 2000-4000r/min x 30min, and the stability completely meets the requirements.
Example 15 safety test
One, multiple (acute) skin irritation test
In this example, the acne-removing skin care products prepared in examples 4 to 13 were subjected to multiple (acute) skin irritation tests, and animal experiments were performed according to the "technical specification for cosmetics" 2015 edition, wherein at least 4 adult, healthy, and skin-intact white rabbits were used as test animals. Before the test, the hair on both sides of the spine of the test animal is cut off, the hair removing range is respectively 3cm multiplied by 3cm, and the smearing area is 2.5cm multiplied by 2.5 cm. About 0.5g of the test substance was applied to one side of the skin 1 time per day for 14 days. From the next day, shearing hair before each application, removing residual test substance with water or non-irritant solvent, observing the result after one hour, and recording the result and intensity grading according to the skin irritation reaction standard in the current effective technical specification. The results of multiple (acute) skin irritation tests are shown in table 5.
Second, acute eye irritation test
In this example, the acne-removing skin care products prepared in examples 4 to 13 were subjected to acute eye irritation test by animal experiments according to 2015, which is the "technical specification for cosmetics", and at least 3 adult and healthy white rabbits were used as test animals. 0.1g of the test substance was instilled (or applied) into the conjunctival sac, and the upper and lower eyelids were passively closed for 1s, while the other eye was left untreated as a self-control. The eyes of the animals were examined 1/24/48/72h and 4d and 7d after instillation of the test substance. If no stimulus response occurs for 72 hours, the test is terminated. If the patient does not recover within 7 days after discovering involvement of the cornea or other eye irritation, the observation time needs to be prolonged to determine the reversibility or irreversibility of the damage, generally not exceeding 21 days, and the result and response grading are recorded according to the eye damage standard in the current effective technical specification. The results of the acute eye irritation test are shown in table 3.
Three, human body skin patch test
In this example, the skin patch test of human body was performed on the acne-removing skin care products prepared in examples 4 to 13, referring to "technical safety standards for cosmetics" 2015 edition, 34 persons in total were selected from the skin care products of 19 to 60 years old and the average age was 41.09 ± 2.15 years old, and then qualified spot test devices were selected, and by the closed spot patch test method, about 0.020 to 0.025g of the test object was placed in a spot tester, and then the test object was externally applied to the back of the subject with hypoallergenic tape, and after 24 hours, the test object was removed, and then the skin reactions were observed at 0.5, 24, and 48 hours after removal, and the results were recorded according to the skin reaction grading standards in the current effective standards. The results of the human skin patch test are shown in table 4.

As can be seen from tables 3 and 4, the acne-removing skin care products prepared in examples 4 to 13 have no irritation or different skin reactions in the multiple (acute) skin irritation tests, acute eye irritation tests and human skin patch tests, and therefore, the safety completely meets the requirements.
Example 16 Corrosion protection challenge test
This example performed a preservative challenge Test on the anti-acne skin care products prepared in examples 4-13, with reference to German Shumei corporation standard SM021 "KoKoKoKoKo Test" (28 days for single inoculation, the inoculation numbers of bacteria, yeasts and molds were 10 respectively 8 germs/ml), the preservative challenge test was performed on each example for a 4 week visit period. As can be seen from table 5, the acne-removing skin care product provided by the invention has a remarkable antibacterial effect due to the compound combination of the sweet wormwood herb volatile oil and cymene, does not need to be added with a preservative, and is more green, safe and healthy.
Example 17 acne treatment efficacy test
In this example, the skin care products prepared in examples 4 to 13 were tested for anti-acne efficacy, and 300 healthy and pox-affected volunteers were recruited and screened and randomly divided into 10 groups of 30 persons each, and each of examples 3 to 12 was tested. The age of the volunteers is mainly concentrated on 18-30 years, and 77% of the volunteers have more than 5 pox growing amount per month. Any cosmetics or external medicines can not be used for 2-3 days before the tested part. Before the experiment, the volunteers needed to clean their faces. In the experiment, each group of volunteers applied the corresponding sample 2 times in the morning and evening, each time according to the dosage of 1mg/cm 2. After 3d, 7d and 14d were used, questionnaires were investigated and photographs were taken using a facial imaging instrument ViSia, taking a set of photographs see fig. 3. The results of the acne treatment are shown in table 6.

As can be seen from table 6, the acne removing effects of examples 3 and 8 are the best, and the higher the concentration of the oil containing artemisia apiacea is, the better the effect is; however, the sweet wormwood volatile oil has strong pungent and cool characteristic smell and is pungent, so the best combination mode is selected in the embodiment 3.
Although the present invention is disclosed above, the present invention is not limited thereto. Various changes and modifications may be effected therein by one skilled in the art without departing from the spirit and scope of the invention as defined in the appended claims.

Claims (10)

1. An antibacterial composition, which is characterized by comprising sweet wormwood volatile oil and o-cymene-5-alcohol.
2. Bacteriostatic composition according to claim 1, further comprising a sage seed extract from Spanish.
3. The bacteriostatic composition according to claim 1 or 2, further comprising allantoin.
4. The bacteriostatic composition according to claim 3, further comprising dipotassium glycyrrhizinate.
5. Bacteriostatic composition according to claim 4, wherein the bacteriostatic composition comprises 0.1-5 parts by weight of Artemisia annua volatile oil, 0.01-0.1 part by weight of o-cymene-5-ol, 0.01-1 part by weight of Salvia Spanish seed extract, 0.01-0.3 part by weight of allantoin and 0.01-0.3 part by weight of dipotassium glycyrrhizinate.
6. An acne-removing skin care product containing the bacteriostatic composition as defined in any one of claims 1-5.
7. Use of the bacteriostatic composition according to any one of claims 1-5 in preparation of acne-removing skin care products.
8. The use of claim 7, wherein the acne-removing skin care product is prepared by a method comprising the following steps: premixing o-cymene-5-alcohol and dipropylene glycol, and sodium polyacryl dimethyl taurate, adding deionized water, 1, 2-pentanediol, Spanish sage seed extract, allantoin and dipotassium glycyrrhizinate, stirring uniformly at 35 deg.C, homogenizing for 5 min, heating to 80-85 deg.C, stirring at constant temperature for 30min, cooling to 45 deg.C, stirring, adding herba Artemisiae Annuae volatile oil, stirring uniformly, cooling, standing, and discharging.
9. Use of a bacteriostatic composition according to any one of claims 1-5 in the preparation of a propionibacterium acnes inhibitor.
10. Use of a bacteriostatic composition according to any one of claims 1-5 in the preparation of an inhibitor of the inflammatory factor IL-6.
