Experiment 1 effect verification experiment principle of soothing repair composition: the experiment is based on the barrier principle, utilizes a 3D skin model (EpiKutis, lot number: ES 210202), adopts a surface administration mode (a mode of simulating human body exposure), and evaluates the effect of a sample to be tested on improving the skin barrier by detecting the change of the tissue morphology of the skin model and the change of the content of FLG and LOR proteins.
Sample to be measured: the soothing repair composition of each example and comparative example.
The experimental method comprises the following steps: the experiment is divided into 16 groups, wherein 1 group is a blank control group, 1 group is a positive control group, and 14 groups are sample groups to be detected. The 3D model was placed in 6-well plates (0.9 mL of model broth added in advance) and labeled according to experimental groupings.
The experimental group takes 25 mu L of a sample to be detected and adds the sample to be detected on the surface of a model, shakes the model lightly to ensure that the sample is uniformly distributed on the surface of the model, incubate the model for 24 hours in a carbon dioxide incubator at 37 ℃ and 5% carbon dioxide concentration, then wash the residual test object on the surface of the model by using a wash bottle filled with sterile PBS solution, and wipe the residual liquid inside and outside the model lightly by using a sterile cotton swab.
The blank control group is different from the experimental group in that 25 mu L of model culture solution is added on the surface of the model; the positive control group was different from the experimental group in that 25. Mu.L of positive control solution was applied to the surface of the model, and the positive control solution was prepared as follows: 10mg of WY14643 powder was weighed and dissolved in 1mL of DMSO to prepare 30mM WY14643 mother liquor, and 5. Mu.L of WY14643 mother liquor was added to 3mL of the model culture broth to prepare 3mL of 50. Mu.M positive control solution.
Verification 1: fixing the model with 4% paraformaldehyde for 24 hours, performing H & E dyeing treatment on the model, photographing and observing by a microscope (Olympus, CKX 41), and evaluating the effect, wherein 5 points represent that the skin is thickened obviously, the cells at the muscle base are arranged very tightly, and the boundaries of the four layers of skin structures are very clear; 4 points represent that the skin is obviously thickened, the arrangement of cells at the muscle base is compact, and the skin structure boundary is clear; 3 points represent skin thickening, the arrangement of myobasal cells is compact, and the skin structure boundary is clear; 2 points represent that the skin thickening effect is not obvious, the close arrangement effect of the cells at the muscle base is poor, and the definition of the skin structure boundary is poor; 1 is the fact that the skin thickening phenomenon is difficult to observe, the phenomenon that the cells at the muscle base are closely arranged is difficult to observe, and the skin structure boundary is unclear; the 0's point indicates that the skin has no thickening phenomenon, the myoblasts have no tight arrangement phenomenon, the skin structure boundary is not clear, and the experimental results are shown in Table 1.
Verification 2: the paraffin slice is placed in a slice baking machine at 70 ℃ for 4 hours, then the slice is placed in xylene for 10 minutes, the xylene is replaced and then soaked for 10 minutes, the slice is soaked in absolute ethyl alcohol for 5 minutes, the slice is soaked in 95% ethanol for 5 minutes, and PBS buffer solution is washed for 3 times, each time for 5 minutes. Then Gu Ang paraffin sections are put into a 0.01M sodium citrate antigen retrieval solution, high-pressure retrieval is adopted, the sections are taken out after cooling, and the sections are washed for 3 times with PBS buffer solution for 5min each time. Then, 1 drop of 3% hydrogen peroxide solution was added to each slice, incubated at room temperature for 30min, washed 3 times with PBS buffer for 5min each, and blocked for 60min at 37℃with serum homologous to the secondary antibody. Then, primary antibody incubation, secondary antibody incubation, nuclear staining (host 33342 working solution) were performed, photographs were taken (20×) with a fluorescence microscope (Leica, DM2500 LED), and the Integral Optical Density (IOD) of the blank control group was taken as 1, and FLG relative IOD value and LOR relative IOD value were counted, respectively, and the experimental results are shown in table 1.
