Abstract
本发明提供了人β3肾上腺素受体(ADRB3)及其抑制剂在制备防治脱发、白发产品中的新用途,具体用途包括:诱导毛囊干细胞(HFSCs)和黑素细胞干细胞(MeSCs)的分化,促进含黑素头发再生。根据本发明研究,ADRB3分布在增殖期的HFSCs和MeSCs,调控HFSCs和/或MeSCs增殖、分化和迁移而影响含黑素头发的再生,通过减少ADRB3基因表达,减少毛囊组织中ADRB3蛋白水平,抑制ADRB3活性,对防治脱发、白发具有显著的效果。因此,ADRB3抑制剂在制备开发防治脱发、白发产品方面具有很好的应用价值。本发明提供了ADRB3抑制剂在防治脱发、白发方面的新用途,也为脱发与白发的防治提供了新的策略和技术手段。

背景技术
人类毛囊中有两种类型的干细胞:毛囊干细胞(HFSCs)和黑素细胞干细胞(MeSCs),二者通常处于休眠期,只有在毛发早期生长期,HFSCs和MeSCs同时被激活而进入细胞周期,经过有丝***而分别产生2个后代细胞,后代细胞将全部退出细胞周期。其中一个后代细胞进入休眠期,另一个分化为成熟细胞,如HFSCs分化为毛囊细胞,MeSCs分化为黑色素细胞,毛囊细胞决定头发的形成,而头发的颜色由黑色素细胞决定。因此,后代HFSCs和MeSCs能够退出细胞周期并分化成熟,是决定色素头发再生的关键因素。如果后代细胞不能退出细胞周期,将会干扰休眠和分化,引起脱发和头发变白。
现有技术《Hyperactivation of sympathetic nerves drives depletion ofmelanocyte stem cells》中,张等人在2020年公开交感神经的活动可以刺激人黑素细胞干细胞(MeSCs)表达的人β2肾上腺素受体(ADRB2),使得MeSCs从毛囊中迁移,最终导致白发。现有技术揭示了ADRB2与人黑素细胞干细胞的迁移有关,但ADRB2对人黑素细胞干细胞的细胞增殖分化和染色质去凝聚是否有影响、有什么样的影响还不得而知,ADRB2在防治人类白发上有无作用暂时还不清楚。另外ADRB2与毛囊干细胞有无关系未有任何报道,ADRB2在防治脱发上有无作用也未有任何报道。
发明内容
本发明要解决的技术问题是探索能够防治脱发、白发的新技术手段,为防治脱发、白发提供了全新的制剂来源。
本发明的目的提供ADRB3抑制剂在制备防治脱发和/或白发的制剂中的应用。
本发明上述目的通过以下技术方案实现:
我们的研究基础为我们经过大量的探索研究发现,与休眠期相比,增殖期HFSCs和MeSCs的ADRB3表达水平均明显上调,ADRB3促进染色质去凝聚,阻止增殖期细胞退出细胞周期,抑制新生的HFSCs和MeSCs进入休眠期和分化期,一方面导致休眠期HFSCs和MeSCs的数量均明显降低,无法维持毛囊再生和黑色素细胞再生;另一方面干扰HFSCs和MeSCs的终末分化,无法发育为成熟的毛囊细胞和黑色素细胞,最终引起脱发和白发。
HFSCs和MeSCs所在的干细胞生态位受到交感神经的调控,我们研究发现,交感神经释放出去甲肾上腺素,激活处于增殖阶段的HFSCs和MeSCs的ADRB3,借助于ADRB3介导的染色质去凝聚功能,使有丝***后新生的细胞无法退出细胞周期,从而干扰分化和休眠,阻止产生毛囊细胞和黑色素细胞。通过使用ADRB3阻滞剂SR59230A,可以阻滞ADRB3的活化,促使新生HFSCs和MeSCs退出细胞周期,诱导它们分化成为毛囊细胞和黑色素细胞,保障色素性头发的再生,为制备防治脱发和白发的制剂提供了线索。
本发明公开了ADRB3在调控HFSCs和MeSCs的表观遗传学、增殖、分化、迁移和形成色素毛发中的生理功能和在脱发、白发病理机制中的作用。公开了ADRB3作为一种表观遗传印记而传递到HFSCs和MeSCs的后代细胞,ADRB3通过促进染色质去凝聚而阻止新生HFSCs和MeSCs退出细胞周期,既能阻止新生细胞的分化命运,使其无法分化成色素性头发生长所必须的毛囊细胞和黑色素细胞;还能阻止新生细胞休眠,无法补充因HFSCs和MeSCs活化而损耗的休眠干细胞储备库;另外,ADRB3通过改造细胞骨架而促进增殖期HFSCs和MeSCs迁移出毛囊,造成HFSCs和MeSCs在毛囊部位的永久性消耗,引起脱发和头发变白。
本发明公开了ADRB3抑制剂通过降低ADRB3的活性,诱导HFSCs和MeSCs退出增殖期,而分化为毛囊细胞和黑色素细胞,并且维持正常的休眠状态而补充减少的干细胞,还能抑制HFSCs和MeSCs迁移出毛囊,防止毛囊内HFSCs和MeSCs的耗竭,具有刺激毛囊和黑色素细胞再生、防治脱发白发的作用。
因此,基于上述研究成果,本发明要求保护:
ADRB3抑制剂在制备诱导毛囊干细胞和/或黑素细胞干细胞分化的制剂中的应用。
ADRB3抑制剂在制备抑制毛囊干细胞和/或黑素细胞干细胞迁移出毛囊的制剂中的应用。
ADRB3抑制剂在制备促进毛囊干细胞和/或黑素细胞干细胞再生的制剂中的应用。
ADRB3抑制剂在制备抑制毛囊干细胞和/或黑素细胞干细胞增殖的制剂中的应用。
ADRB3抑制剂在制备促进毛囊干细胞和/或黑素细胞干细胞退出细胞周期的制剂中的应用。
ADRB3抑制剂在制备防治脱发和/或白发的制剂中的应用。
一种用于防治脱发和/或白发的制剂,包含ADRB3抑制剂。
可用于本发明的ADRB3抑制剂没有特别的限制,所有能够抑制或阻滞ADRB3活性、功能和/或降低ADRB3蛋白水平、浓度及数量的化合物、抗体、多肽、短肽、针对ADRB3基因的CRISPR-Cas9基因编辑技术等都适用于本发明。
作为可选择的优选方案,上述ADRB3抑制剂是指ADRB3阻滞剂或人ADRB3单克隆抗体。
所述ADRB3阻滞剂可的例子是(但不限于):SR59230A(分子式:C23H29NO6;CAS号:174689-39-5),L748337(分子式:C26H31N3O5S;CAS号:244192-94-7),以及以SR59230A、L748337为母核的化合物均适用于本发明。
本发明的ADRB3抑制剂的剂型和制备方法没有特别限制,可用本领域常规通用的制法制成片剂、胶囊、缓释剂、注射剂、外用剂型等各种剂型。
本发明创新性地发现了ADRB3调控毛囊中两种干细胞的机制,并且基于此机制提供了对防治脱发、白发有显著作用的化合物,为防治脱发、白发持续开发更多的制剂具有重要的临床意义。
具体在实验方面:
1、成功地分离具有干细胞特性的人毛囊干细胞和人黑素细胞干细胞。分离细胞通过流式细胞仪鉴定毛囊干细胞表面标记物Lgr5阳性率为91%,Lgr6阳性率为92%。黑素细胞干细胞多呈两极状或者多角状,L-DOPA染色呈棕黑色阳性反应,流式细胞术以及Westernblot分析结果显示获得的细胞表达MITF、TYR、TRP1蛋白。结果证明分离细胞均具有较强的增殖潜能,可由单细胞生成明显的克隆集落,符合干细胞特性。
2、通过免疫荧光法测定两种干细胞在不同的细胞***期中ADRB3的含量,证明ADRB3在染色质去凝聚中的作用。ADRB3蛋白在G1早期(Ki67阴性)和增殖期(Ki67阳性)人毛囊干细胞(图1)和人黑素细胞干细胞表达(图2)。与ADRB3弱阳性的细胞相比,ADRB3强阳性细胞的细胞核更大,染色质更松散。分别给予SR59230A(10uM)或人ADRB3单克隆抗体(5ug/ml)处理24小时,毛囊干细胞和黑素细胞干细胞的细胞核均明显固缩,染色质致密(图3),表明ADRB3促进染色质去凝聚。
3、实时定量PCR检测毛囊细胞标志物角蛋白Krt5、Krt10、Krt14和Krt19的mRNA表达情况,与对照组相比,ADRB3阻滞剂和抗体组的毛囊干细胞角蛋白Krt5、Krt10、Krt14和Krt19的mRNA表达均明显升高2~4倍(图4),说明毛囊干细胞分化为毛囊细胞。实时定量PCR检测黑色素细胞分化蛋白酪氨酸酶(TYR)和小眼畸形相关转录因子(MITF)的mRNA表达情况,与对照组相比,ADRB3阻滞剂和抗体组的毛囊干细胞角蛋白TYR和MITF的mRNA表达均明显升高(图5)。倒置显微镜下观察,对照组细胞无黑素产生,而ADRB3阻滞剂和抗体组的黑色素细胞发灰或灰黑,说明诱导人黑素细胞干细胞分化为黑色素细胞。
4、体外MTT实验中,使用针对ADRB3的阻滞剂SR59230A、L748337和人ADRB3单克隆抗体,结果证明采用小分子化合物阻滞剂或大分子的抑制性抗体,阻滞ADRB3,降低ADRB3的活性,将会抑制人毛囊干细胞和人黑素细胞干细胞增殖(图6、7)。
5、体外实验Transwell细胞迁移实验,与对照组相比,SR59230A处理组和人ADRB3单克隆抗体处理组的两种干细胞迁移数量都明显减少(P<0.01)。表明ADRB3促进HFSCs和MeSCs的迁移。抑制ADRB3活性,将会使HFSCs和MeSCs保留在毛囊,防止数量减少或耗竭。
6、动物实验中,通过为多只已经发生脱发、白发的老年小鼠腹腔注射SR59230A和人ADRB3单克隆抗体组与对照组进行对比,显示实验组老年小鼠乌黑油亮,毛发茂密,脱发和白发均明显减轻(图8),毛囊细胞ADRB3蛋白阳性率分别为20%和10%,而对照组老年小鼠毛发稀疏花白,ADRB3在毛囊隆突区组织中的阳性率为100%(图9)。与对照组ADRB3阳性的毛囊细胞相比,SR59230A和人ADRB3单克隆抗体组ADRB3阳性的毛囊细胞的细胞核变小,染色质更加致密,表明ADRB3促进染色质去凝聚。
以上技术方案展现了ADRB3抑制剂在诱导毛囊干细胞和/或黑素细胞干细胞分化的制剂中的应用;本发明的技术方案能够支持发明的首要目的的实现。
本发明也提供了ADRB3抑制剂在制备抑制毛囊干细胞和/或黑素细胞干细胞迁移出毛囊的制剂中的应用,说明本发明的技术方案能够支持发明的另一目的的实现。
本发明也提供了ADRB3抑制剂在促进毛囊干细胞和/或黑素细胞干细胞再生中的应用,说明本发明的技术方案能够支持发明的另一目的的实现。
本发明也提供了ADRB3抑制剂在抑制毛囊干细胞和/或黑素细胞干细胞增殖中的应用,说明本发明的技术方案能够支持发明的另一目的的实现。
本发明也提供了ADRB3抑制剂在促进毛囊干细胞和/或黑素细胞干细胞退出细胞周期中的应用,说明本发明的技术方案能够支持发明的另一目的的实现。
同时,本发明提供ADRB3抑制剂在制备防治脱发和/或白发的制剂中的应用也可以为技术方案所支持。具体实验包括三种物质,SR59230A、L748337、人ADRB3单克隆抗体,实时定量PCR实验证明,分别采用小分子化合物阻滞剂或大分子的抑制性抗体,阻滞ADRB3,降低ADRB3的活性,将会促进人毛囊干细胞和人黑素细胞干细胞的分化(图4、5)。体外MTT实验中证明,分别采用小分子化合物阻滞剂或大分子的抑制性抗体,阻滞ADRB3,降低ADRB3的活性,将会抑制人毛囊干细胞和人黑素细胞干细胞增殖(图6、7)。在动物实验中,注射了SR59230A和人ADRB3单克隆抗体的实验组老年小鼠乌黑油亮,毛发茂密,脱发和白发均明显减轻(图8),毛囊细胞ADRB3蛋白阳性率,阳性率分别为20%和10%,而对照组老年小鼠毛发稀疏花白,并且其ADRB3在毛囊隆突区组织中的阳性率为100%(图9)。与对照组ADRB3阳性的毛囊细胞相比,SR59230A和人ADRB3单克隆抗体组ADRB3阳性的毛囊细胞的细胞核变小,染色质更加致密,以此证明ADRB3促进染色质去凝聚。借助于抑制ADRB3介导的染色质去凝聚功能,有丝***后新生的细胞可以退出细胞周期,HFSCs和MeSCs得以顺利分化和休眠,从而产生毛囊细胞和黑色素细胞。通过使用ADRB3抑制剂SR59230A、L748337、人ADRB3单克隆抗体,抑制ADRB3的活化,抑制ADRB3介导的染色质去凝聚功能,促使新生HFSCs和MeSCs退出细胞周期,诱导它们分化成为毛囊细胞和黑色素细胞,保障色素性头发的再生。说明此本发 明提供的ADRB3抑制剂,在制备防治脱发和/或白发的制剂中的应用,可以为技术方案所支 持。
ADRB3是毛囊干细胞和黑素细胞干细胞的关键受体,ADRB3介导的信号通路调控毛囊干细胞和黑素细胞干细胞的增殖、分化、迁移、毛囊形成和黑色素细胞再生等色素性头发形成过程。ADRB3的过度活化将导致脱发和白发。ADRB3阻滞剂、中和抗体等抑制剂能够特异结合并阻滞ADRB3的活性,可用于防治脱发和白发。
本发明具有以下有益效果:
本发明公开了HFSCs和MeSCs中ADRB3在调控细胞的表观遗传学、增殖、分化、迁移和形成色素毛发中的生理功能和ADRB3抑制剂在脱发、白发病理机制中的应用,公开了ADRB3及其抑制剂在调控关于HFSCs和MeSCs上的作用,并且为防治脱发、白发提供了全新的制剂来源。

以下结合说明书附图和具体实施例来进一步说明本发明,但实施例并不对本发明做任何形式的限定。除非特别说明,本发明采用的试剂、方法和设备为本技术领域常规试剂、方法和设备。

以下实施例中所用的人ADRB3单克隆抗体为专利ZL201710183354.8中披露的5D9杂交瘤细胞产生的抗体。
实施例1人毛囊干细胞和人黑素细胞干细胞的分离培养与鉴定
显微分离人头部皮肤标本毛囊隆突区组织,0.1%I型胶原酶37℃下消化真皮组织1-2小时,反复吹打使毛囊从真皮中游离出来。1500rpm离心5分钟,获得毛囊,0.25%胰酶37℃下消化毛囊1-5分钟,获得毛囊来源干细胞,首先消化脱落下来的细胞类型为人黑素细胞干细胞,后续脱落的是毛囊干细胞,分别收集备用。将两种干细胞分别接种于Matrigel包被的培养板中培养。细胞迁出后,待细胞融合达到70%-80%时,传代培养。取传代培养2周的干细胞进行L-DOPA染色、流式细胞学分析及克隆形成能力测定,鉴定毛囊干细胞和黑素细胞干细胞的表型及增殖潜能。
结果:分离培养所得毛囊干细胞呈铺路石样生长,细胞核大,具有较高的核质比。流式细胞仪鉴定毛囊干细胞表面标记物Lgr5阳性率为91%,Lgr6阳性率为92%。黑素细胞干细胞多呈两极状或者多角状,L-DOPA染色呈棕黑色阳性反应,流式细胞术以及Westernblot分析结果显示获得的细胞表达MITF、TYR、TRP1蛋白。细胞增殖能力测定结果显示,毛囊干细胞和黑素细胞干细胞均具有较强的增殖潜能,可由单细胞生成明显的克隆集落,符合干细胞特性。
以上表明成功在体外分离、培养出人毛囊干细胞和人黑素细胞干细胞。
实施例2免疫荧光检测人毛囊干细胞和人黑素细胞干细胞中ADRB3
人毛囊干细胞和人黑素细胞干细胞分别以105/孔接种于6孔板,内置无菌盖玻片,孵育至细胞贴片后,4%多聚甲醛固定10min,PBS洗3遍。0.1%TritonX-100通透10分钟,3%BSA封闭1h,滴加Ki67抗体和人ADRB3单克隆抗体(1∶100),4℃孵育过夜。滴加FITC标记的二抗(anti-rabbit,1∶800)和PE标记的二抗(anti-mouse,1∶800),室温孵育1h,DAPI 0.5μg/ml,3min,PBS漂洗5min×3,50%甘油/PBS封片,激光共聚焦显微镜下观察,随机观察5~7个视野并拍片,Fluorchem8900软件测量细胞平均红色荧光强度和平均绿色荧光强度。
结果:ADRB3蛋白在G1早期(Ki67阴性)和增殖期(Ki67阳性)人毛囊干细胞(图1)和人黑素细胞干细胞表达(图2)。与ADRB3弱阳性的细胞相比,ADRB3强阳性细胞的细胞核更大,染色质更松散。分别给予SR59230A(10uM)或人ADRB3单克隆抗体(5ug/ml)处理24小时,毛囊干细胞和黑素细胞干细胞的细胞核均明显固缩,染色质致密(图3),表明ADRB3促进染色质去凝聚。
实施例3实时定量PCR检测毛囊细胞、黑色素细胞分化标志物基因。
(1)对数生长期的贴壁人毛囊干细胞和人黑素细胞干细胞,分别用胰酶消化,含10%小牛血清的RPMI l640培养基配成5000个/ml的细胞悬液,接种在6孔培养板中,37℃,5%CO2培养24h。
(2)实验组换新的含不同浓度SR59230A、L748337和人ADRB3单克隆抗体的培养基,对照组则换含等体积溶剂或鼠源IgG的培养基,每组设3~5平行孔,培养4~5d。
(3)TRlzol法提取细胞RNA,采用随机引物利用逆转录酶反转录成cDNA,实时荧光定量PCR检测毛囊细胞标志物角蛋白Krt5、Krt10、Krt14、Krt19的表达量和黑色素细胞分化相关基因酪氨酸酶(TYR)和小眼畸形相关转录因子(MITF)的表达。
结果:与对照组相比,ADRB3阻滞剂和抗体组的毛囊干细胞角蛋白Krt5、Krt10、Krt14和Krt19的mRNA表达均明显升高2~4倍(*P<0.01,图4)。ADRB3阻滞剂和抗体组的毛囊干细胞角蛋白TYR和MITF的mRNA表达均明显升高(*P<0.01,图5)。
实施例4MTT实验检测SR59230A、L748337和人ADRB3单克隆抗体抑制人毛囊干细胞和人黑素细胞干细胞增殖
(1)对数生长期的贴壁人毛囊干细胞和人黑素细胞干细胞,分别用胰酶消化,含10%小牛血清的RPMI l640培养基配成5000个/ml的细胞悬液,接种在96孔培养板中,每孔接种200ul,37℃,5%CO2培养24h。
(2)实验组换新的含不同浓度SR59230A、L748337和人ADRB3单克隆抗体的培养基,对照组则换含等体积溶剂或鼠源IgG的培养基,每组设3~5平行孔,37℃,5%CO2培养4~5d。
(3)弃去上清液,每孔加入200μl新鲜配制的含0.5mg/ml MTT的无血清培养基。37℃继续培养4h。加入200μl DMSO,用超声振荡器混匀后,酶标仪以试验波长为570nm,参比波长为450nm测定光密度值。
结果:SR59230A、L748337剂量依赖性抑制人毛囊干细胞和人黑素细胞干细胞增殖,与对照组比,*P<0.05,#P<0.01(图6,纵坐标为细胞增殖抑制率,阻滞剂浓度单位为umol/L)。人ADRB3单克隆抗体剂量依赖性抑制人毛囊干细胞和人黑素细胞干细胞增殖,与对照组比,*P<0.05,#P<0.01(图7)。
实施例3、4的结果表明,ADRB3与人毛囊干细胞和人黑素细胞干细胞的增殖和分化高度相关,采用小分子化合物阻滞剂或大分子的抑制性抗体,阻滞ADRB3,降低ADRB3的活性,将会抑制人毛囊干细胞和人黑素细胞干细胞增殖,促进分化。
实施例5Transwell细胞迁移实验检测SR59230A和人ADRB3单克隆抗体对人毛囊干细胞和人黑素细胞干细胞迁移的影响
用BD公司的Matrigel1:8稀释,包被Transwell小室底部膜的上室面,置37℃30min使Matrigel聚合成凝胶。消化细胞,调整细胞密度至5×105/ml。取细胞悬液100μl加入Transwell小室。24孔板下室加入600μl含SR59230A或人ADRB3单克隆抗体的培养基。常规培养24-48h。取出Transwell小室,用无钙的PBS洗2遍,甲醇固定30分钟。0.1%结晶紫染色20min,用棉签轻轻擦掉上层未迁移细胞,用PBS洗3遍。400倍显微镜下随即五个视野观察细胞,记数。
结果:相比对照组人毛囊干细胞和人黑素细胞干细胞的迁移数量[(21.3±3.4)×104,(24.1±3.6)×104],SR59230A处理组的两种干细胞迁移数量[(9.2±1.4)×104,(7.5±1.2)×104]均明显减少(P<0.01),人ADRB3单克隆抗体处理组的两种干细胞迁移数量[(8.6±1.7)×104,(8.3±1.3)×104]均明显减少(P<0.01)。表明ADRB3与HFSCs和MeSCs的迁移高度相关,抑制ADRB3活性,将会使HFSCs和MeSCs保留在毛囊,防止数量减少或耗竭。
实施例6SR59230A和人ADRB3单克隆抗体治疗老年小鼠脱发、白发
1、实验分组
30只雄性15月龄老年C57BL/6J小鼠,全部出现脱发、白发。随机分为3组:
1○.对照组;对照组给予相同体积溶剂。
2○.SR59230A组,每3天腹腔注射SR59230A100nmol/只,连用5周;
3○.人ADRB3单克隆抗体组,每3天腹腔注射人ADRB3单克隆抗体,5mg/kg,连用5周。
2、实验观察脱发、白发发生情况。
3、并收集小鼠头部皮肤,免疫组化检测毛囊中ADRB3的表达情况。
实验步骤:石蜡切片:厚度4μm。65℃,3h。脱蜡至水。PH8.0TRIS-EDTA修复液高压修复,从排气阀开始排气算起3min,自然冷却。3%过氧化氢孵育10min。10%山羊血清封闭10min。一抗(1:100)4℃过夜。二抗室温孵育30min。DAB显色。(A液与B液比例为1:50),在显微镜下观察,表达适当,背景清晰,即可终止。苏木素复染1min,温水返蓝15min。过梯度酒精脱水,中性树胶封片。染色强度判定:每个组织点随机观察6~8个高倍视野,弱阳性为淡黄色、中阳性为棕黄色、强阳性为棕褐色。染色阳性率判定:选取3个染色强度不同的视野在高倍镜下进行判读,每个视野中随机记100个细胞,记100个细胞中阳性细胞占的百分率X1%,同样原理看另外2个视野的阳性细胞占的百分率后X2%、X3%,最终该组织点染色阳性率取X1%、X2%及X3%的平均数。
3、实验结果:
与对照组相比,SR59230A和人ADRB3单克隆抗体组小鼠毛发乌黑油亮,毛发茂密,脱发和白发均明显减轻,表明SR59230A和人ADRB3单克隆抗体均能促进老年C57BL/6J小鼠色素性毛发的再生(图8)。对照组小鼠ADRB3蛋白阳性的细胞主要位于毛囊隆突区,该区域是HFSCs和MeSCs富集的位置。ADRB3在毛囊隆突区组织中的阳性率为100%(10/10)(图9)。SR59230A或人ADRB3单克隆抗体均明显减少毛囊细胞ADRB3蛋白阳性率,阳性率分别为20%和10%,明显低于对照组(P<0.05)。与对照组ADRB3阳性的毛囊细胞相比,SR59230A和人ADRB3单克隆抗体组ADRB3阳性的毛囊细胞的细胞核变小,染色质更加致密,表明ADRB3很可能作为表观遗传因子来促进染色质去凝聚。
上述实施例为本发明较佳的实施方式,但本发明的实施方式并不受上述实施例的限制,其他的任何未背离本发明的精神实质与原理下所作的改变、修饰、替代、组合、简化,均应为等效的置换方式,都包含在本发明的保护范围之内。
