The pH value of the larch woodland is 6.8-7.2, and the humidity is not lower than 60%.
In the step S3, the mother strains of morchella esculenta are cultured in the culture medium of the cultivar at the environment temperature of 10-20 ℃ and are cultured for 10-20 days in a dark place.
The invention solves the defects in the background technology and has the following beneficial effects:
1. the cultivation method can improve the yield of the morchella in high-altitude areas, the yield per mu reaches more than 310kg, the yield per mu can be improved by more than 20% compared with the traditional cultivation method, and the growth period is shortened by 5-8 days.
2. s2), inoculating the selected morchella to a test tube with a culture medium under an aseptic condition for culture; when hyphae grow in the test tube and sclerotium grows out, selecting the sclerotium with the diameter larger than 1cm, and inoculating the sclerotium or bottom hyphae into a mother culture medium; the mother culture medium comprises the following components in parts by weight: 35 parts of peeled potato, 20 parts of rice bran, 3 parts of glucose powder, 10 parts of corn flour, 12 parts of wheat flour, 5 parts of calcium superphosphate, 2 parts of magnesium sulfate, 3 parts of agar, 1.2 parts of gibberellin, 0.3 part of cytokinin and 15 parts of water.
S3), inoculating the morchella mother strain obtained in the step S2 into a cultivar culture medium for culture, and obtaining a cultivar after hypha on the cultivar culture medium overgrows; the culture medium comprises the following components in parts by weight: 25 parts of organic fertilizer, 15 parts of sawdust, 8 parts of wheat bran, 7 parts of decomposed soil, 2 parts of glucose, 1.3 parts of vinasse, 3 parts of bifidobacterium and 3 parts of bacillus. In addition, the influence of the planting environment on crops is not considered too much in the traditional cultivation method, the morchella belongs to the biological strains which like yin and moisture, and the conventional method is only to build a sun-proof net in a planting place, and the sun-proof net is blown and dried by the sun for a long time, so that the conditions of abnormal use and the like caused by air drying can occur. The method selects 8-15 years old larch forest land as the cultivation land, and utilizes a natural shielding screen formed by leaves and branches of pine trees for shielding. In winter, fallen leaves have the effects of heat preservation and moisture preservation, so that the humidity is ensured, and the damage of direct sunlight to strains is avoided; the nutrients required in the planting process are all from soil under the pine forest, and the growth mode of wild morchella is simulated. s3), inoculating the morchella mother strain obtained in the step S2 into a culture medium of a cultivated species for culture, and obtaining the cultivated species after the hypha on the culture medium of the cultivated species overgrows;
s4), selecting 8-15 years old larch forest land as a cultivation land, turning over the land, making furrows, and arranging and flattening the furrows; mixing the growth-promoting matrix with the soil on the surface layer of the ridge bed, and then scattering a layer of thin lime powder on the surface of the ridge bed for soil sterilization;
