Abstract
本发明涉及美容护肤产品领域,具体是具有抗衰修护功效的蓝铜胜肽及其制备方法和应用。搅拌下,向反应容器内依次加入纯化水、GHK三肽,用盐酸调pH=7±0.5,加入一水醋酸铜或硫酸铜或二水氯化铜搅拌一定时间,缓慢向料液滴加乙醇,加完后缓慢降温至10℃,保持料温110℃搅拌一定时间,过滤,滤饼真空干燥得到产品蓝铜胜肽晶体。本发明提供可以在不伤肌肤、不刺激肌肤的情况下,增加细胞的活力,逐渐修复体内流失的胶原蛋白,使皮下组织坚强,伤口迅速愈合,进而达到除皱抗老化的目的具有抗衰修护功效的蓝铜胜肽及其制备方法和应用。

背景技术
铜肽是胜肽的始祖,胜肽其实就是小分子的蛋白质,是由氨基酸所组成,这些小分子的蛋白质更容易被皮肤所吸收,胜肽是由一些具有特定序列的氨基酸通过酰胺键排列相连而成,2个氨基酸组成的叫做二胜肽,3个氨基酸组成的就叫做三胜肽,依次类推。即使完全相同的氨基酸,如果排列相连的方式不同,将会形成结构完全不同的胜肽,蓝铜胜肽可以刺激胶原蛋白与弹力蛋白形成,紧实肌肤及减少细纹,恢复肌肤修复能力,增加皮肤细胞间黏质产生,减少皮肤受损,刺激葡萄糖聚胺形成,增加皮肤厚度,减少皮肤松垮紧实肌肤促进血管增生,增加皮肤供氧量等。
发明内容
本发明提供可以在不伤肌肤、不刺激肌肤的情况下,增加细胞的活力,逐渐修复体内流失的胶原蛋白,使皮下组织坚强,伤口迅速愈合,进而达到除皱抗老化的目的具有抗衰修护功效的蓝铜胜肽及其制备方法和应用。
本发明所采用的技术方案为:具有抗衰修护功效的蓝铜胜肽及其制备方法和应用,其特征在于:包括三肽化合物合成步骤如下:
步骤一:合成H-Lys(Ac)-OH,将赖氨酸完全溶解于蒸馏水中,室温搅拌,用质量分数为10%的氢氧化钠水溶液调节pH值至10.5,加入乙酸对硝基苯酯与1,4-二氧六环或四氢呋喃的质量-体积比为1g:5~10mL比为1:2~5的混合液,赖氨酸与乙酸对硝基苯酯的摩尔比为1:1.5,室温搅拌反应4小时,分离纯化产物,得到H-Lys(Ac)-OH;
步骤二:将Boc-Gly-OH与1,4-二氧六环或四氢呋喃按质量-体积比为1g:10mL混合,室温搅拌,加入N-羟基丁二酰亚胺,搅拌至溶解,滴加N,N'-二环己基碳二亚胺与1,4-二氧六环或四氢呋喃的质量-体积比为1g:3mL的混合溶液,Boc-Gly-OH与N-羟基丁二酰亚胺、N,N'-二环己基碳二亚胺的摩尔比为1:1.3:1.5,室温反应2小时,减压过滤除去固体不溶物,得到Boc-Gly-COOSu的1,4-二氧六环或四氢呋喃溶液;
步骤三:将组氨酸和碳酸氢钠完全溶解于蒸馏水中,所得溶液滴加到步骤2得到的Boc-Gly-COOSu的1,4-二氧六环或四氢呋喃溶液中,室温搅拌反应20小时;滴加二碳酸二叔丁酯与1,4-二氧六环或四氢呋喃的质量-体积比为1g:5mL的混合液,加入氢氧化钠,Boc-Gly-OH与组氨酸、碳酸氢钠、二碳酸二叔丁酯、氢氧化钠的摩尔比为1:1.5:1.5:1.5:3,室温反应3~5小时,分离纯化产物,得到Boc-Gly-His(Boc)-OH;
步骤四:将Boc-Gly-His(Boc)-OH与1,4-二氧六环或四氢呋喃按质量-体积比为1g:8mL混合,24℃下搅拌,加入N-羟基丁二酰亚胺,搅拌至溶解,滴加N,N'-二环己基碳二亚胺与1,4-二氧六环或四氢呋喃的质量-体积比为1g:4mL的混合溶液,Boc-Gly-His(Boc)-OH与N-羟基丁二酰亚胺、N,N'-二环己基碳二亚胺的摩尔比为1:1.2:1.5,室温反应3小时,减压过滤除去固体不溶物,得到Boc-Gly-His(Boc)-COOSu的1,4-二氧六环或四氢呋喃溶液;
步骤五:将H-Lys(Ac)-OH和碳酸氢钠完全溶解于蒸馏水中,所得溶液滴加到步骤4得到的Boc-Gly-His(Boc)-COOSu的1,4-二氧六环或四氢呋喃溶液中,H-Lys(Ac)-OH与碳酸氢钠、Boc-Gly-His(Boc)-OH的摩尔比为1.5:15:1,室温搅拌反应20小时,分离纯化产物,得到Boc-Gly-His(Boc)-Lys(Ac)-OH;
步骤六:将Boc-Gly-His(Boc)-Lys(Ac)-OH加入三氟乙酸与蒸馏水的体积比为1:1的混合液中,Boc-Gly-His(Boc)-Lys(Ac)-OH与混合液的质量-体积比为1g:10mL,室温搅拌反应2~3小时,用乙酸乙酯萃取,水相减压浓缩至干,得到GHK三肽。

所述蓝铜胜肽在面部护理方面的应用为蓝铜胜肽修护锁水抗氧化精华液,其制备方法为玫瑰花提取液8wt%、甘油单硬脂酸酯8wt%、金花茶提取液4wt%、红花提取液2wt%、桃仁提取液0.5wt%、谷物酵素液2wt%、薏苡仁油2wt%、保湿因子2wt%、蓝铜胜肽0.8wt%、藻类活性肽0.8wt%、余量为水在室温下搅拌20小时。
所述蓝铜胜肽在面部护理方面的应用为蓝铜胜肽修复喷雾,其制备方法为水73.1%,1,3-丁二醇8%,甘油5%,二丙二醇4%,尿囊素0.2%混合,在室温下搅拌10分钟,燕麦仁提取物5%,库拉索芦荟叶提取物2%,蓝铜胜肽1%,1,2-戊二醇1%,对羟基苯乙酮0.5%,苯氧乙醇0.2%在室温下搅拌10分钟,然后将上述两者混合,室温下搅拌十分钟。
所述燕麦提取物为禾本科植物燕麦的全草提取物,主要含有β-葡聚糖和黄酮类化合物及多种抗氧化的物质,拥有优异的抗衰老功效,能够抚平细小皱纹,改善皮肤纹理度。
所述玫瑰花提取液主要成分为玫瑰多酚,可消除疲劳、改善体质,玫瑰花茶的味道清香幽雅,能令人缓和情绪、纾解抑郁,能改善内分泌失调,解除腰酸背痛,对消除疲劳和伤口愈合,滋润养颜,护肤美容。
所述金花茶提取液主要成分为多酚、多糖、黄酮以及皂苷,具有抗氧化、抗菌、抗过敏等功效。
所述蓝铜胜肽在面部护理方面的应用为蓝铜胜肽抗衰修护精华霜,所述蓝铜胜肽抗衰修护精华霜包括A相和B相,所述A相制备方法为poly matrix7 0.5%,丁二醇3%,EDTA-二钠0.02%,小核菌胶0.05%,尿囊素0.1%,对羟基苯乙酮0.5%,EMT-10(丙烯酸羟乙酯/丙烯酰二甲基牛磺酸钠共聚物)2.3%,余量为水,在烧杯中加热至85℃以上,搅拌均匀至无颗粒物后静置一分钟,开始搅拌降温,B相制备方法为每千克1,2己二醇0.5%,蓝铜胜肽(粉末)0.06%,依克多因1%,类肉毒杆菌3%,类蛇毒肽3%,血清白蛋白(毕赤酵母源)1%,所述A相与B相总量为一千克,所述A相搅拌至45℃以下时加入B相,搅拌均匀即可出料。
本发明的有益效果:
本发明借助氨基酸复合物作为载体,使具有生化效果的二价铜离子成分进入细胞,发挥生理功能,这种蓝铜胜肽可以有效促进胶原蛋白(Collagen)和弹性(Elastin)蛋白的制造,增加血管生长和抗氧化能力,并刺激葡萄糖聚胺(GAGs)产生,帮助皮肤恢复自我修补的天赋能力。

步骤四:将Boc-Gly-His(Boc)-OH与1,4-二氧六环或四氢呋喃按质量-体积比为1g:8mL混合,24℃下搅拌,加入N-羟基丁二酰亚胺,搅拌至溶解,滴加N,N'-二环己基碳二亚胺与1,4-二氧六环或四氢呋喃的质量-体积比为1g:4mL的混合溶液,Boc-Gly-His(Boc)-OH与N-羟基丁二酰亚胺、N,N'-二环己基碳二亚胺的摩尔比为1:1.2:1.5,室温反应3小时,减压过滤除去固体不溶物,得到Boc-Gly-His(Boc)-COOSu的1,4-二氧六环或四氢呋喃溶液;
步骤五:将H-Lys(Ac)-OH和碳酸氢钠完全溶解于蒸馏水中,所得溶液滴加到步骤4得到的Boc-Gly-His(Boc)-COOSu的1,4-二氧六环或四氢呋喃溶液中,H-Lys(Ac)-OH与碳酸氢钠、Boc-Gly-His(Boc)-OH的摩尔比为1.5:15:1,室温搅拌反应20小时,分离纯化产物,得到Boc-Gly-His(Boc)-Lys(Ac)-OH。
步骤六:将Boc-Gly-His(Boc)-Lys(Ac)-OH加入三氟乙酸与蒸馏水的体积比为1:1的混合液中,Boc-Gly-His(Boc)-Lys(Ac)-OH与混合液的质量-体积比为1g:10mL,室温搅拌反应2~3小时,用乙酸乙酯萃取,水相减压浓缩至干,得到GHK三肽。

步骤一:合成H-Lys(Ac)-OH,将赖氨酸完全溶解于蒸馏水中,室温搅拌,用质量分数为10%的氢氧化钠水溶液调节pH值至10.5,加入乙酸对硝基苯酯与1,4-二氧六环或四氢呋喃的质量-体积比为1g:5~10mL比为1:2~5的混合液,赖氨酸与乙酸对硝基苯酯的摩尔比为1:1.5,室温搅拌反应4小时,分离纯化产物,得到H-Lys(Ac)-OH;

步骤六:将Boc-Gly-His(Boc)-Lys(Ac)-OH加入三氟乙酸与蒸馏水的体积比为1:1的混合液中,Boc-Gly-His(Boc)-Lys(Ac)-OH与混合液的质量-体积比为1g:10mL,室温搅拌反应2~3小时,用乙酸乙酯萃取,水相减压浓缩至干,得到GHK三肽。

8.根据权利要求1所述的具有抗衰修护功效的蓝铜胜肽及其制备方法和应用,其特征在于:所述蓝铜胜肽在面部护理方面的应用为蓝铜胜肽抗衰修护精华霜,所述蓝铜胜肽抗衰修护精华霜包括A相和B相,所述A相制备方法为poly matrix7 0.5%,丁二醇3%,EDTA-二钠0.02%,小核菌胶0.05%,尿囊素0.1%,对羟基苯乙酮0.5%,EMT-10(丙烯酸羟乙酯/丙烯酰二甲基牛磺酸钠共聚物)2.3%,余量为水,在烧杯中加热至85℃以上,搅拌均匀至无颗粒物后静置一分钟,开始搅拌降温,B相制备方法为每千克1,2己二醇0.5%,蓝铜胜肽(粉末)0.06%,依克多因1%,类肉毒杆菌3%,类蛇毒肽3%,血清白蛋白(毕赤酵母源)1%,所述A相与B相总量为一千克,所述A相搅拌至45℃以下时加入B相,搅拌均匀即可出料。
