Description

Plant extract composite bacteriostatic agent and application thereof
Technical Field
The invention belongs to the field of cosmetics, and particularly relates to a plant extract composite bacteriostatic agent and application thereof.
Background
The preservative is an indispensable component in the cosmetics, and the preservative can be added to inhibit the breeding of microorganisms in the cosmetics and protect the products from being polluted by the microorganisms in the production or use process, so that the selection of the efficient and safe preservative is one of the keys in the formula development of the cosmetics. According to technical Specification for safety of cosmetics (2015 edition), there are 51 kinds of preservatives for cosmetic preparations at present, including p-hydroxybenzoate esters, phenoxyethanol, isothiazolinone and the like.
In recent years, along with the development of society and the improvement of living standard, people have made higher demands for the diversification of cosmetic functions and product quality. As consumers are deeply fixed on the concept of nature and health, the cosmetic has strong psychological repulsion on chemical additives, and the concepts of 'zero addition' and 'no corrosion' are vigorously developed in the cosmetic industry. Therefore, in order to meet the increasing demand of consumers, the development of cosmetics without adding chemical preservatives has been a hot spot.

Detailed Description
The invention is further illustrated by the following examples. These examples are intended to illustrate the invention and are not intended to limit the scope of the invention. Experimental procedures without specific conditions noted in the examples below, generally according to conditions conventional in the art or as suggested by the manufacturer; the raw materials, reagents and the like used are, unless otherwise specified, those commercially available from the conventional markets and the like. Any insubstantial changes and substitutions made by those skilled in the art based on the present invention are intended to be covered by the claims.
Example 1
The extraction process of the cinnamon bark comprises the following steps:
taking 55kg of cinnamon bark, crushing by using a traditional Chinese medicine crusher, and sieving by using a 50-mesh sieve to obtain cinnamon bark powder. Taking 50kg of sieved cortex Cinnamomi powder, adding 5-15 times of distilled water, soaking for 18-24h, putting into a steam distillation device, rapidly heating at high power, reducing heating power after steam is generated, and continuously extracting for 4 h. Extracting the distillate with 50L diethyl ether twice, mixing the two extracted diethyl ether layers, adding small amount of anhydrous sodium sulfate, drying, heating for 20min, and steaming to remove diethyl ether to obtain oily extract of cortex Cinnamomi.
Example 2
The extraction process of root bark of the peony tree comprises the following steps:
50kg of cortex moutan radicis is taken, crushed by a traditional Chinese medicine crusher and sieved by a sieve of 80 meshes for standby. Adding the fine powder of cortex moutan into Soxhlet extractor, extracting with n-hexane as solvent at 70-80 deg.C under reflux for 24 hr. After extraction, the extract is decompressed and concentrated by a rotary evaporator to obtain the oily extract of the cortex moutan which is preserved in dark.
Preparation of plant-derived composite bacteriostatic agents of examples 3-13 and comparative examples 1-6 in each ratio:
according to the mass percentage composition of the table 1, the bacteriostat, the synergistic agent and the solvent are uniformly stirred and mixed and dissolved, and the plant extract composite bacteriostat with each proportion can be obtained.

4. And (4) preparing a bacterial suspension.
TABLE 2 proportion of McLeod turbiditubes
Figure GDA0003769030270000071
The turbidimetric vials prepared according to the proportions in table 2 were used as standard bacterial suspension controls. Respectively adding sterile 0.85% of normal saline (0.85% of NaCl) into each strain slant, respectively washing out a certain amount of bacteria, then adding the washed bacteria into a sterile test tube with the same diameter (size) as that of the McLeod's turbidimetric standard tube, respectively adding sterile 0.85% of normal saline (0.85% of NaCl) for dilution until the concentration is the same as that of the McLeod's turbidimetric standard tube, taking the diluted bacteria as bacterial suspension, and finally diluting each bacterial suspension by 100 times for later use.
5. And (4) determining the minimum inhibitory concentration.
Taking 12 sterile test tubes, sequentially marking, transferring 3000 μ L of liquid culture medium (nutrient broth for bacteria, and wort extract for fungi) into the sterile test tubes with pipette gun, and sequentially adding 3000 μ L of prepared liquid culture medium C into the 12 test tubes 1 ,C 2 ,C 3 ,C 4 … … sample (C) 1 ,C 2 ,C 3 ,C 4 … … concentrations were set according to each example test range and gradient decreased), 3 replicates per concentration; in addition, with 1 control sample, 3000. mu.L of liquid medium and 3000. mu.L of 0.85% physiological saline were added, and finally, 300. mu.L of each bacterial suspension was added to the test tube; plugging the inoculated test tube with a plug, and placing the test tube inoculated with the microorganism in a shaking table at 30 ℃ for culturing for 48 hours; taking out the culture medium at 24h and 48h, shaking the test tube, and if sediment is produced at the bottom or the solution is turbid, the strain cannot be inhibited from growing at the concentration; on the other hand, the concentration is such that the growth of the species is inhibited (for example, in E.coli inoculated tubes, the concentration is C 1 ,C 2 ,C 3 The test tube of (A) produced no precipitate and the solution was relatively clear at a concentration of C 4 The bottom of the tube is precipitated or the solution is cloudy, and the minimum concentration of the sample for inhibiting Escherichia coli is C 3 )。
As shown in table 3, it can be seen from the comparison of examples 3, 6 and 9 with comparative example 1 that the schisandra chinensis extract, levulinic acid and saccharomyces cerevisiae fermentation product extract all enhance the bacteriostatic performance of the cinnamon bark extract; comparing examples 4, 7 and 10 with comparative examples 2, 3, 4 and 5, it can be seen that the peony root-bark extract has slightly poor bacteriostatic performance, but the compound of the peony root-bark extract and the synergist shows synergistic bacteriostatic effect; in addition, by respectively comparing examples 3-5, 6-8 and 9-11, the peony root-bark extract and the cinnamon extract can be synergized to improve the bacteriostatic effect; in addition, in comparative examples 5, 8, 11, 12 and 13, it can be found that the schisandra chinensis extract, levulinic acid and the saccharomyces cerevisiae fermentation product extract have a synergistic effect, and the antibacterial effect of the compound of the cinnamon bark extract and the peony root bark extract can be further improved.

Test example 2 Induction of inhibition of histamine Release amount from macrophage tumor cells in P815 mice.
Histamine is often used as one of the early diagnostic indicators of acute allergic reactions. The histamine is directly involved in hypersensitivity, the content of the histamine is related to the occurrence and the severity of the hypersensitivity, and the antiallergic capability of the sample can be evaluated by detecting the content of the histamine.
1. Experimental materials: DMEM medium, FBS, MTT, dansyl chloride, NaHCO 3 Ammonia, acetonitrile, ammonium acetate.
2. Preparing a sample to be tested: the samples of examples 3-13 and comparative examples 1-6 were diluted to a concentration of 10% with DMEM medium, respectively, for use.
3. The experimental method comprises the following steps: taking P815 cells (mouse mastocytoma cells, GDC099) in logarithmic growth phase, and diluting the cells to 6 × 10 concentration by DMEM medium 4 one/mL. The diluted cell solutions were inoculated into 96-well plates with an inoculum size of 1mL per well at 37 ℃ with 5% CO 2 The culture box is used for culturing for 24 hours. Then dividing the test sample into a blank control group, a model control group and a sample group, wherein each group is provided with 6 complex holes, 50 mu L of corresponding samples to be tested are respectively added into the test sample group, 50 mu L of DMEM culture medium is respectively added into the blank control group and the model control group, the test sample group and the model control group are placed in an incubator to be pre-incubated, and after 1h, except the blank control group, cells of each group are placed under a 15W ultraviolet lamp and irradiated for 30s to induce P815 cells to generate anaphylactic reaction and release histamine. Then put 5% CO 2 The culture box is used for culturing for 24 hours. After 24h, 200. mu.L of the culture medium was removed for derivatization of histamine, and saturated NaHCO was added in sequence 3 The solution (300. mu.L) and dansyl chloride (300. mu.L) at 20mg/mL were reacted at room temperature for 1 hour, then 20% ammonia-acetonitrile solution (100. mu.L) was added to terminate the reaction, and after 1 hour, 100. mu.L of acetonitrile solution was added, and HPLC analysis was performed after filtration through a membrane.
4. Result judgment
Preparing a solution with the concentration of 1,5, 10, 20 and 50ppm by using a histamine standard sample, filtering the solution by using a filter membrane, performing HPLC analysis, drawing a standard curve by using the concentration of the histamine standard sample as a horizontal coordinate and using a corresponding chromatographic peak area as a vertical coordinate, and calculating the concentration value of each sample according to the standard curve.
Table 4. test results of the inhibition effect experiment of the plant extract composite bacteriostatic agent on histamine release amount.
Figure GDA0003769030270000101
As shown in Table 4, it can be seen from comparative examples 3 to 11 and comparative examples 1 to 6 that the peony root bark extract and the Schisandra chinensis extract have a significant inhibitory effect on histamine release amount; due to the addition of the schisandra chinensis extract and the saccharomyces cerevisiae fermentation product extract, the inhibition effect of the peony root bark extract on the histamine release amount can be effectively enhanced; in addition, the results of comparative examples 5, 8, 11, 12 and 13 show that the Schisandra chinensis extract and the Zygosaccharomyces bifidus fermentation product extract have synergistic effect.
Test example 3 human body patch test.

adding 70 parts of deionized water, and then heating to the system temperature of about 85-90 ℃;
standing, keeping the temperature for 20min for sterilization, slowly cooling to below 60 deg.C under stirring, adding sufficient deionized water, and adding collagen powder, vitamin B3, and K2 into the system;
continuously stirring and cooling to about 40 ℃, adding the composition into the system, continuously stirring for about 10min, stopping stirring, cooling, and then inspecting to discharge materials;
and sealing the storage barrel after discharging, and moving to a standing room for later use.
3. The test method comprises the following steps: a48 h closed patch test was used. Respectively weighing 0.02mL of diluted aqueous solution of different mask liquid products into a spot tester, and simultaneously setting a blank control group, wherein the control group is the diluent of the cosmetic. 50 healthy volunteers are selected, and the healthy volunteers are 18-24 years old and have no history of allergy. After cleaning the inner forearm of the subject, 0.02mL of sample was applied to a piece of filter paper attached to the plaque tester. The patch tester is applied to the inner side of forearm of the subject with non-irritating adhesive tape, and then is applied to skin by pressing with fingers for 48 h. The examinee is ordered to keep the spot-pasting part dry within 48h, and violent exercise, scratching the spot-pasting part, long-time sunlight irradiation and the like are avoided. After 48h, the tester is removed and marked, and after 30min, the pressure marks disappear under sufficient light for judgment and return visit observation is carried out within 72h and 96h respectively.
