Abstract
The invention discloses a preparation method of a poria cocos planting nutrition bag, and relates to the technical field of poria cocos. The method comprises the following steps: s1, selecting strains; s2, culturing hyphae of the stock; s3, cultivating hyphae of the cultivated species; s4, guiding wood to select; s5, preparing cultivars; s6, preparing a nutrition bag; s7, inoculating and culturing the nutrition bag; s8, soil preparation and land selection; s9, grafting; s10, planting; and S11, performing later management. According to the preparation method of the poria cocos planting nutrition bag, the cultivation effect of the nutrition bag is improved under the matching of the nutrition bag and the cultivation bag, sufficient nutrient substances are provided for the growth of poria cocos, the survival rate of the cultivated poria cocos and the quality of the grown poria cocos are improved, the prepared nutrition medium is pure natural, pollution-free and rich in various nutrients, and the cultivated poria cocos is high in growth speed, developed in root, good in disease resistance and strong in pest and disease resistance.
Description
Technical Field
The invention relates to the technical field of poria cocos nutrition packages, in particular to a preparation method of a poria cocos planting nutrition package.
Background
Poria, the name of Chinese medicine, is the dried sclerotium of Poria of Polyporaceae fungus, it contains pachymaran, glucose, protein, amino acid, organic acid, fat, lecithin, adenine, choline, ergosterol, many enzymes and many ingredients such as potassium salt, etc., it has effects of excreting dampness, diuresis, invigorating the spleen, tranquilization, etc., its medicinal value lies in: has effects in enhancing immunity, resisting tumor, promoting urination, increasing discharge of electrolyte such as potassium, sodium, and chlorine, tranquilizing mind, protecting liver, inhibiting ulcer, lowering blood sugar, and resisting radiation, and can be used for treating edema, fullness, phlegm and fluid retention, cough, vomiting, diarrhea, nocturnal emission, stranguria with turbid urine, palpitation, and amnesia.
In the prior art, in order to improve the yield of poria cocos in the planting process, namely, a fresh poria cocos is grafted at the lower end of a pine section full of hypha and used for guiding the growth of the poria cocos, the time required for grafting combination of the fresh poria cocos block and the pine section is long, but the poria cocos is cultivated in the open air, the probability of rainwater climate is increased in advance if the combination time is long, and the survival rate of the poria cocos block is low once the poria cocos meets rainwater weather, so that the production yield is improved by using a nutrition bag for culture in a matched mode in the planting process of the poria cocos, but the conventional nutrition bag provides common nutrients for the poria cocos in the culture process.
Disclosure of Invention
Aiming at the defects of the prior art, the invention provides a preparation method of a poria cocos planting nutrition bag, and aims to solve the problems in the background art.
In order to achieve the purpose, the invention provides the following technical scheme: a preparation method of a poria cocos planting nutrition bag comprises the following steps:
s1, selecting strains: selecting high-quality poria cocos blocks suitable for local temperature and climate, cleaning and disinfecting the surfaces of the selected poria cocos blocks for later use;
s2, cultivation of stock hyphae: taking out the poria cocos blocks selected in the step S1, inoculating the poria cocos flesh in the poria cocos blocks to a culture medium I in a triangular flask, transferring the poria cocos blocks to a thermostat with the temperature of 22-25 ℃ for culturing for 5-7 days until mycelium grows in a culture container, and obtaining required stock hypha;
s3, cultivation of hyphae of the cultivated species: inoculating the stock spawn hyphae prepared in the step S2 onto a culture medium II in a polypropylene cultivation bag with the diameter of 6cm, and performing cultivation operation until the cultivation bag is full of poria cocos hyphae, so as to obtain cultivation spawn hyphae;
s4, wood guiding operation: peeling pine tree branches with the diameter of 3-5cm and the length of 30-40cm, air-drying, soaking in saturated brown sugar water for a day and a night, and sterilizing for later use;
s5, preparation of cultivars: putting the peeled pine branches obtained in the step S4 into a culture bag of the hyphae of the cultivated species obtained in the step S3, extending the peeled pine branches to the outer side of the culture bag, fastening the opening of the culture bag, and culturing and propagating at the temperature of 15-30 ℃ for 10-15 days for later use;
s6, preparing the nutrition packet: mixing 20% of pine sawdust, 25% of pine chips, 15% of pine wool, 15% of corn meal, 20% of wheat bran, 1% of brown sugar, 1% of gypsum and 3% of soybean meal according to parts by weight, and then carrying out disinfection treatment to prepare the required nutrition bag;
s7, inoculating and culturing a nutrition package: inserting the strain bag prepared in the S5 step into the nutrition bag prepared in the S6 step under aseptic conditions, and culturing at 15-30 ℃ for 30-40 days;
s8, soil preparation and land selection: selecting a planting field with good drainage, excavating cultivation pits in the planting field in sandy soil with the soil mainly comprising yellow sandy soil and the pH value of 3-5, and spraying a sterilizing liquid to the cultivation pits to perform disinfection and insect killing treatment after excavation;
s9, grafting; when hypha begins to attach to the leading wood, 50 g of fresh poria cocos blocks are taken and inoculated on the cross section of the leading wood and wrapped by an agricultural film;
s10, planting: transplanting the nutrition bag and the strains cultured and propagated in the step S9 into sandy soil, and placing the nutrition bag below the soil;
s11, post management: 7-10 days after inoculation, the growth conditions of hyphae in each section are checked so as to supplement the seeds in time, if the poria cocos is exposed out of the ground, soil covering, weeding and insect killing are carried out in time, and drainage is carried out in rainy season.
Further optimizing the technical scheme, the nutrition bag prepared in the step S6 is prepared by adding water into pine wood chips, pine nuts, corn flour, wheat bran, brown sugar, gypsum and bean pulp, uniformly mixing, and controlling the water content to be 50-55%.
Further optimizing the technical scheme, the culture bag in the step S5 is cultured for 13-18 days at the constant temperature of 10-15 ℃, and then is kept at the constant temperature of 25-30 ℃ for 5-12 days until the culture bag is full of mycelia, so that the required cultivated species are obtained.
Further optimizing the technical scheme, in the step S7, in the process of inoculation treatment, an opening is cut in the strain bag, a new opening is sawed at one end of the nutrition bag close to the strain bag, and the opening of the strain bag is closely attached to the new opening of the nutrition bag.
