Traditional Chinese medicine composition for growing and nourishing hair and preparation thereof
Technical Field
The invention belongs to the field of beauty treatment and hairdressing, and particularly relates to a traditional Chinese medicine composition for growing and nourishing hair and a preparation thereof.
Background
Nowadays, the hair style and the hair length are vital to the life of people, but with the development of society and the increase of living pressure, the number of patients suffering from alopecia is increased, the patients tend to be younger, and the hair style and the hair length have great influence on the life quality of the patients. According to the reports of the literature, the incidence rate of alopecia reaches 30.2%, and the alopecia is in an ascending trend, has complex pathology and long course of disease, and is still a difficult problem in the medical field.
The hair grows from hair follicles, the growth of the hair follicles is periodic and can be divided into a growth period, a catagen period and a telogen period, the normal alopecia is mainly caused by the fact that the hair follicles are in the catagen period and the telogen period, but the hair follicles in all the periods are in dynamic balance, and for an alopecia patient, the hair follicles shrink and cannot enter the growth period, so that the hair is gradually sparse.
At present, a lot of traditional Chinese and western medicines for treating alopecia are available, western medicines are approved by FDA and comprise minoxidil and finasteride, but all the western medicines have defects, minoxidil is easy to relapse after being stopped, and finasteride can cause damage to the reproductive system after being used for a long time; the traditional Chinese medicine for treating alopecia has many medicines, but the effect is not ideal, the price is high, the treatment course is extremely long, and therefore, the traditional Chinese medicine is valuable for finding a hair restorer which has easily obtained materials, is convenient to use and has remarkable curative effect without rebound.
The traditional Chinese medicine has already been used for treating alopecia in traditional Chinese medicine, but a plurality of commonly used alopecia Chinese herbal medicines are available, and experimental researches show that some traditional Chinese medicines have good hair growth effects, and the introduction is as follows:
sanguisorba, originally recorded in Shen nong Ben Cao Jing (Shen nong's herbal), belongs to Rosaceae, is slightly cold in nature, bitter, astringent and sour, and has the effects of cooling blood and stopping bleeding. Modern medical research proves that the sanguisorba is rich in flavone and gallic acid, and has good antibacterial and anti-inflammatory effects.
The dried ginger belongs to the Zingiberaceae plant and has a long history of medication in China, and the traditional Chinese medicine theory considers that the dried ginger has pungent and hot taste, enters spleen, stomach, kidney, heart and lung channels and can warm the middle-jiao and dispel cold. Modern medical research proves that the dried ginger can play roles of increasing blood pressure, resisting blood coagulation and the like, and the gingerol in the dried ginger can activate transdermal capillaries, promote transdermal microcirculation and promote hair follicle nutrition.
The Chinese medicine theory believes that the cacumen biotae is bitter and astringent in taste and cold in nature and has the effects of cooling blood and stopping bleeding. Modern medical research proves that the flavone in the cacumen biotae has obvious antibacterial and anti-inflammatory effects;
folium Eriobotryae, the dried leaves of the plant of the genus lutea of the family Rosaceae of the genus Eriobotrya, is a commonly used Chinese medicine. The traditional Chinese medicine theory considers that the loquat leaves have the functions of clearing away the lung-heat, relieving cough and arresting vomiting, and have obvious effect on various cough with lung heat. Modern researches prove that the loquat leaf is rich in triterpenic acid, flavonoid, sesquiterpene and glycoside, and Japanese researchers also find that the triterpenic acid has anticancer activity.
Folium Ginkgo, which is the dry leaf of Ginkgo biloba of Ginkgoaceae, can be used for treating cough, various heart diseases, hypertension, hyperlipemia, etc. in the theory of traditional Chinese medicine. The ginkgo leaf extract contains flavonoids as main chemical components, and modern researches prove that the flavonoids and terpenoids in the ginkgo leaf extract can remove free radicals, resist cell aging and promote blood vessel microcirculation.
Dandelion, belonging to Compositae, is bitter, sweet and cold in nature. The dandelion tea has the effects of clearing away heat and toxic materials, removing swelling and removing blood stasis, and modern medical research proves that the dandelion is rich in various mineral substances such as iron, calcium and the like, and can better promote the nutrition of hair follicles;
the liquorice, also known as Chinese grass and sweet grass, belongs to leguminous perennial herbs, has strong tonifying property, can clear away heat and toxic material, eliminate phlegm and relieve cough and the like, and is a medicinal material widely used in the traditional Chinese medicine. The glycyrrhizic acid has antiinflammatory and antiallergic effects.

Experimental animals: 60C 57 mice, male, 18-20g in weight, SPF grade, provided by Shanghai Sphere-Bikeh animal experiments, Inc., animal production license number: SCXK (Shanghai) 2013-0016
The experimental method comprises the following steps:
modeling a mouse alopecia model:
1) depilatory mice were anesthetized by intraperitoneal injection of 4% chloral hydrate (0.1ml/10g), and 1.5 × 1.0cm was shaved off with a razor while the mice were under anesthesia2The top hair is left with only short skin. 0.6ml of 6 percent sodium sulfide is uniformly coated on the unhairing area of the neck of each mouse, and after the sodium sulfide stays on the neck of the mouse for 150 seconds, the hair on the neck is quickly washed off, so that the skin on the neck of the mouse is exposed.
2) The imiquimod induced alopecia model selects mice with pale red top skin, applies imiquimod ointment to unhairing areas of the mice, 0.1g of the imiquimod ointment is applied to the unhairing areas once a day every one week, three weeks, five days and four days, the top skin of the mice is cleaned by physiological saline before the imiquimod to remove the residual imiquimod at the previous time, after four weeks, the top skin of the mice has obvious horny and hard and hairless areas, the modeling is successful, and the application of the imiquimod is stopped.
Grouping and administration: dividing the mice successfully modeled into 8 groups, wherein each group comprises 6 mice, namely a blank control group, a positive group and an observation group (6 groups); wherein the blank control group: smearing blank gel, smearing 0.2 ml/one commercially available chemical hair growth promoting agent minoxidil tincture on positive group, and respectively smearing hydrogel prepared from Chinese medicinal composition 1-6 on the other 6 groups; the dosage is 0.2 g/time, 2 times daily, and is administered at 10 am and 4 pm (administration is timed to ensure stability of the experiment).
And (3) determining an experimental end point: the end point of the experiment was the complete restoration of hair to the normal length of the unhaired area in the positive group of mice (about 30 days).
Note that: a: after depilation, the skin color was observed and only red-skinned mice had their hair in the resting stage, so red-skinned mice were selected. B: the administration is gentle and lasting for 2-3min to allow the medicine to be fully absorbed, and the medicine is returned to the mouse cage about 15min after administration to prevent the pad from drying.
Detection indexes are as follows: A. mouse hair growth scoring: observing the skin and hair growth condition of each group of mice every 4 days, comparing the mice with the same initial top color of different groups, and scoring the skin and hair growth condition of the mice according to the following scoring standards:
Figure BDA0002256863230000051
irregular hair growth was calculated by area ratio, e.g. 50% red, 50% grey, then 0.5 points were recorded.
B. Record of new hair growth: drawing the longest tuft of hair in the hair removal treatment area at the end point of the experiment, selecting the longest tuft of hair of 3 tufts, measuring the length of the tuft of hair by using a vernier caliper, recording and calculating a mean value;
C. newly born hair weight recording: end of experiment the same area of new hair was cut with scissors along the hair roots in the epilation area, weighed on an analytical balance and recorded.
The experimental results are as follows:
comparison of mouse hair length and weight:
table 2 comparison of hair length and weight (n ═ 6, x ± SD)
Figure BDA0002256863230000052
Figure BDA0002256863230000061
Remarking: the value of the hair weight/hair length is positively correlated with the number of hair roots per unit area (density of hair) in the case of the same hair thickness;
because the hair loss patients need to increase the number of hairs in unit area rather than the length, the increase of the hair weight/hair length value is more realistic than the simple increase of the hair length.
The results show that the hair length and the hair weight/hair length of the mice in the blank control group are lower than those of the positive drug minoxidil group by comparing the hair weight/hair length values of the mice in each group, which indicates that the modeling is successful. The Chinese medicinal composition of the invention in 2-6 groups is higher than that in a blank control group in different degrees, and the hair growth promoting effect is proved. The density of the hair of the composition groups 2-6 is higher than that of the composition group 1 (namely patent CN201810728202.6 group), which shows that the composition of the invention achieves better hair growth effect compared with the prior traditional Chinese medicine composition. After the ginkgo leaves and the dandelion are added, the hair growth density of the mouse is further increased, and the mouse has more remarkable hair growth and hair growth effects.
