Abstract
本发明属于医用高分子材料技术领域,公开了一种含生长因子的分散体。该分散体包括固体分散相和分散介质;所述固体分散相包括生长因子和负载所述生长因子的透明质酸或透明质酸盐。采用透明质酸作为生长因子的载体材料,利用化学键对生长因子进行负载,在分散介质中制得含生长因子的分散体,不仅不影响生长因子的生物学活性,且能够延长其半衰期,实现对生长因子的保护。该分散体具有良好的稳定性,可以方便储存以及使用。该制备方法简单,有利于实现产业化。该含生长因子的分散体能够用于制备药物。
Description
技术领域
本发明属于医用高分子材料技术领域,具体涉及一种含生长因子的分散体及其制备方法与应用。
背景技术
生长因子是一类通过与特异的、高亲和的细胞膜受体结合,调节细胞生长与其他细胞功能等多效应的多肽类物质。因其具有促进皮肤细胞生长、增殖及分化的作用,被广泛应用于临床治疗创伤、烧伤、溃疡等皮肤损伤,以及作为化妆品用于除疤、去皱和其他皮肤老化问题。
现有技术中,生长因子的给药手段多为水剂以及喷雾剂,其中使用量较大的水剂多数是采用生长因子的冻干粉现用现配,这导致生长因子制剂的成本高,且冻干粉的现用现配给使用带来不便,也不易储存。更重要的是,水剂的给药方式存在着药物于创口处停留时间短、需多次给药,药物易受细菌和创口分泌物影响等诸多问题,给制剂的临床使用带来了限制。
因此,亟需提供一种含生长因子的制剂或分散体,具有良好稳定性,且不影响生长因子的生物学活性。
发明内容
本发明旨在至少解决上述现有技术中存在的技术问题之一。为此,本发明提出一种含生长因子的分散体,所述分散体具有良好的稳定性,且不影响生长因子的生物学活性。
本发明的发明构思:本发明采用透明质酸与生长因子反应,然后经疏水修饰得到的生长因子-透明质酸复合物,再将其分散于分散介质中,制得含生长因子的分散体。这种分散体是一种固油分散体,能够有效保持生长因子的生物活性,延长其半衰期,并可以方便地进行存储及使用。
本发明提供一种含生长因子的分散体,包括固体分散相和分散介质;所述固体分散相包括生长因子和负载所述生长因子的透明质酸或透明质酸盐。
优选的,在所述含生长因子的分散体中,所述固体分散相的含量为0.1-3mg/mL;所述固体分散相的粒径为0.1-20μm。
进一步优选的,在所述含生长因子的分散体中,所述固体分散相的含量为0.5-2mg/mL;所述固体分散相的粒径为0.2-10μm。
优选的,在所述固体分散相中,所述生长因子的负载率(质量分数)为0.2%-2%。
优选的,所述生长因子为表皮生长因子、成纤维细胞生长因子、角质细胞生长因子或***中的至少一种。
优选的,所述分散介质包含肉豆蔻酸异丙酯。肉豆蔻酸异丙酯不仅具有良好的分散效果,能够很好地分散生长因子-透明质酸复合物;而且肉豆蔻酸异丙酯是一种安全无害的渗透促进剂,能够帮助生长因子穿过磷脂双层膜,增加生长因子在皮肤组织的溶解度,从而达到促进生长因子等药物对皮肤的渗透的效果。
进一步优选的,所述分散介质还包括蔗糖酯。
本发明也提供一种含生长因子的分散体的制备方法。
所述含生长因子的分散体的制备方法,包括以下步骤:
(1)制备两亲性生长因子-透明质酸复合物;
(2)将步骤(1)制得的两亲性生长因子-透明质酸复合物加入到分散介质中,分散,得到所述含生长因子的分散体。
优选的,所述含生长因子的分散体的制备方法,包括以下步骤:
(1)向透明质酸盐或透明质酸中加入氧化剂,反应得到醛基透明质酸;将所述醛基透明质酸与有机碱反应,生成醛基透明质酸-有机盐,然后加入生长因子,反应,再加入还原剂还原,最后进行疏水处理,干燥,制得生长因子-透明质酸复合物。
(2)将步骤(1)所述的生长因子-透明质酸复合物与溶剂混合,得混合物,然后将所述混合物加入分散介质中,除去溶剂,分散,制得所述含生长因子的分散体。
进一步优选的,在步骤(1)中,所述氧化剂的摩尔量为所述透明质酸钠的重复单元的摩尔量的1-1.5倍。
进一步优选的,在步骤(1)中,所述还原剂的摩尔量为所述透明质酸钠的重复单元的摩尔量的1-2倍;更优选的,所述还原剂的摩尔量为所述透明质酸钠的重复单元的摩尔量的1-1.5倍。
进一步优选的,在步骤(1)中,采用有机胺化合物进行疏水处理;更优选的,所述有机胺化合物为直链脂肪胺;进一步优选的,在所述直链脂肪胺中碳原子的个数为8-14。
优选的,所述有机胺化合物的摩尔量为所述透明质酸钠的重复单元的摩尔量的1.5-3倍;进一步优选的,所述有机胺化合物的摩尔量为所述透明质酸钠的重复单元的摩尔量的1.8-2.5倍。
优选的,在步骤(1)中,所述氧化剂为高碘酸盐。
优选的,在步骤(1)中,所述有机碱为四丁基氢氧化铵。
优选的,在步骤(1)中,所述还原剂为氰基硼氢化钠。
优选的,在步骤(2)中,所述混合物与所述分散介质的体积比为(5-20):1;进一步优选的,所述混合物与所述分散介质的体积比为(8-15):1。
具体,所述含生长因子的分散体的制备方法,包括以下步骤:
(1)将透明质酸钠溶解于蒸馏水中,得到1-5wt%的透明质酸钠水溶液,然后加入高碘酸钠反应2-6小时,再向体系中加入过量的乙二醇反应1-4小时,除去未反应的高碘酸钠,将所得溶液在去离子水中透析得到醛基透明质酸。随后向醛基透明质酸中加入四丁基氢氧化铵,搅拌2小时后,冷冻干燥得到醛基透明质酸-四丁基铵盐。将醛基透明质酸-四丁基铵盐与生长因子溶解于N-甲基吡咯烷酮中,在室温下搅拌1-4个小时,使醛基透明质酸中的醛基与生长因子的氨基端基形成席夫碱。向体系中加入氰基硼氢化钠,反应2-6个小时对席夫碱进行还原。随后向体系中加入直链脂肪胺(n=8-14),与未反应的醛基进行结合实现疏水修饰。最后通过超滤离心除去未反应的生长因子以及其他杂质。将所得溶液进行冷冻干燥得到生长因子-透明质酸复合物。
(2)将蔗糖酯(HLB=2-4)加热溶解于肉豆蔻酸异丙酯中,在冷却、过滤后得到蔗糖酯饱和的肉豆蔻酸异丙酯溶液。将生长因子-透明质酸复合物加入到无水乙醇中,得到5~20mg/mL生长因子-透明质酸复合物的乙醇溶液(混合物)。在室温条件下,将蔗糖酯饱和的肉豆蔻酸异丙酯溶液与生长因子-透明质酸复合物的乙醇溶液按体积比(5-20):1的比例进行混合,再使用旋转蒸发仪除去其中的乙醇,对其进行超声分散,得到含生长因子的分散体。
本发明还提供所述含生长因子的分散体在制备药物中的应用。如在制备治疗创伤、烧伤油剂中的应用。
一种药物,包含所述含生长因子的分散体。所述药物包括多肽类药物或蛋白类药物。
透明质酸是一种天然的高分子量直链粘多糖,由重复的双糖单位组成,双糖单位由D-葡糖醛酸与D-N-乙酰氨基葡萄糖组成。由于透明质酸不具抗原性、无过敏性、不致炎、不发生免疫反应,并且能被生物体内的透明质酸酶降解。作为生长因子的载体,无副作用,负载性能良好。
蛋白/多肽固-油分散体系能够有效地隔绝其中蛋白/多肽组分与水的接触,能够提高水溶液中易降解蛋白/多肽的稳定性,并减少水相中其他物质对蛋白/多肽的影响。本发明提供的含生长因子的分散体即为一种透明质酸负载生长因子的固油分散体系,其采用透明质酸为生长因子的载体材料,利用化学键对生长因子进行负载,分散于分散介质中形成固油分散体系,能够提高生长因子的稳定性,有效延长生长因子的半衰期,且化学修饰不会影响生长因子的生物活性。
相对于现有技术,本发明的有益效果如下:
(1)本发明采用透明质酸为生长因子的载体材料,利用化学键对生长因子进行负载,在分散介质中制得含生长因子的分散体,不仅不影响生长因子的生物学活性,且能够延长其半衰期,实现对生长因子的保护。
(2)本发明所述含生长因子的分散体具有良好的稳定性,可以方便储存以及使用。
(3)本发明提供的含生长因子的分散体,其载体材料易得,成本较低,制备方法简单,有利于实现产业化。

具体实施方式
为了让本领域技术人员更加清楚明白本发明所述技术方案,现列举以下实施例进行说明。需要指出的是,以下实施例对本发明要求的保护范围不构成限制作用。
以下实施例中所用的透明质酸钠(MW=100kDa),购买于天津希恩思生化科技有限公司;表面生长因子(EGF),由暨南大学生物医药基地提供;其他试剂购买自上海阿拉丁公司。磁力搅拌器,购买于巩义市予华仪器有限责任公司;冷冻干燥机,购买于宁波新芝生物科技股份有限公司;超声波细胞粉碎仪,购买于上海乔跃电子有限公司。其他原料、试剂或装置如无特殊说明,均可从常规商业途径得到,或者可以通过现有已知方法得到。
实施例1
向100mL含有1wt%的透明质酸水溶液中加入530mg的高碘酸钠,在室温下反应4小时,然后再向溶液中加入1mL乙二醇反应2个小时,所得溶液透析3天后得到醛基透明质酸水溶液。向得到的醛基透明质酸水溶液加入1.6mL 40%的四丁基氢氧化铵水溶液,在室温下搅拌2小时,随后冻干得到醛基透明质酸-四丁基铵盐。
将100mg醛基透明质酸-四丁基铵盐与1mg表皮生长因子加入到10mLN-甲基吡咯烷酮中,于室温条件下搅拌2个小时。向溶液中加入20mg氰基硼氢化钠,于室温条件下搅拌4个小时。随后向反应体系中加入50μL正辛胺,反应12小时。将所得的溶液滴入如无水***中进行沉淀,后将沉淀用蒸馏水复溶并使用截留分子量为10000Da的超滤离心管进行纯化,最后溶液经冷冻干燥后得到表皮生长因子-透明质酸复合物。
利用超声将10mg表皮生长因子-透明质酸复合物均匀分散于2mL无水乙醇当中,随后将其缓慢地滴入饱和蔗糖酯(HLB=3)的肉豆蔻酸异丙酯溶液中。在40℃的条件下,使用旋转蒸发仪除去体系中的乙醇。最后使用超声波细胞粉碎仪对此体系进行分散,即可得到含表皮生长的分散体,其中表皮生长因子的溶度为0.8μg/mL,负载量为0.8%。
实施例2
向100mL含有3wt%的透明质酸水溶液中加入530mg的高碘酸钠,在室温下反应4小时,然后再向溶液中加入1mL乙二醇反应2个小时,所得溶液透析3天后得到醛基透明质酸水溶液。向得到的醛基透明质酸水溶液加入2mL 40%的四丁基氢氧化铵水溶液,在室温下搅拌2小时,随后冻干得到醛基透明质酸-四丁基铵盐。
将100mg醛基透明质酸-四丁基铵盐与18mg表皮生长因子加入到15mLN-甲基吡咯烷酮中,于室温条件下搅拌2个小时。向溶液中加入20mg氰基硼氢化钠,于室温条件下搅拌4个小时。随后向反应体系中加入50μL正辛胺,反应12小时。将所得的溶液滴入如无水***中进行沉淀,后将沉淀用蒸馏水复溶并使用截留分子量为10000Da的超滤离心管进行纯化,最后溶液经冷冻干燥后得到表皮生长因子-透明质酸复合物。
利用超声将18mg表皮生长因子-透明质酸复合物均匀分散于2mL无水乙醇当中,随后将其缓慢地滴入饱和蔗糖酯(HLB=3)的肉豆蔻酸异丙酯溶液中。在40℃的条件下,使用旋转蒸发仪除去体系中的乙醇。最后使用超声波细胞粉碎仪对此体系进行分散,即可得到含表皮生长因子的分散体,其中表皮生长因子的浓度度为1.5μg/mL,负载量为1.5%。
产品效果测试
(1)测试实施例(1)制得的含表皮生长因子的分散体中表皮生长因子的浓度随时间变化情况
将实施例(1)制得的含表皮生长因子的分散体、与实施例(1)含相同表皮生长因子浓度的的生理盐水溶液置于相同的室温环境中,以考察不同体系中表皮生长因子的浓度变化。于预先设定好的时间分别取10μL液体,使用表皮生长因子ELISA试剂盒对表皮生长因子的浓度进行测算,绘制表皮生长因子浓度曲线。图1为表皮生长因子浓度随时间的变化图,其中I代表了表皮生长因子水溶液中表皮生长因子浓度百分比的变化,II代表了实施例(1)制得的含表皮生长因子的分散体中表皮生长因子浓度百分比的变化。由图1可知,在含表皮生长因子的分散体中,表皮生长因子浓度降低的速度明显小于表皮生长因子水溶液中表皮生长因子浓度降低的速度。在含表皮生长因子的分散体中的表皮生长因子具有更强的稳定性,该分散体可以有效地延长表皮生长因子的半衰期。
(2)将实施例(1)制得的含表皮生长因子的分散体进行促细胞增殖实验
将3T3细胞分别于与表皮生长因子水溶液(表皮生长因子浓度为5ng/mL、10ng/mL、50ng/mL)、实施例(1)制得的含表皮生长因子的分散体(表皮生长因子浓度为5ng/mL、10ng/mL、50ng/mL)共培养24小时后,利用Cell Counting Kit-8(CCK-8)测定各组3T3细胞的细胞数。最后通过与空白对照组(表皮生长因子浓度为5ng/mL、10ng/mL、50ng/mL的生理盐水)对比,计算得到各组3T3细胞的存活率百分比。
细胞存活率%=(给药组细胞数/空白对照组细胞数)*100%
图2为不同体系表皮生长因子促细胞增殖情况图,I为与表皮生长因子水溶液共培养的3T3细胞的存活率,II为与含表皮生长因子的分散体共培养的3T3细胞的存活率。由图2可以看出,在相同的表皮生长因子浓度下,表皮生长因子水溶液与含表皮生长因子的分散体促进3T3细胞增殖的能力相当,可见化学修饰并没有对表皮生长因子的生物活性产生影响。
(3)采用实施例2制得的含表皮生长因子的分散体进行创面愈合试验
建立背部创伤大鼠模型,对经麻醉的大鼠的背部进行脱毛处理并进行消毒,制造4个大小相近的创面,清洗并吸干创面上的残留液体。对背部创伤大鼠模型的4个创面分别施以生理盐水(空白对照组)、透明质酸固油分散体、表皮生长因子水溶液(表皮生长因子含量为1.5μg)以及含表皮生长因子的分散体(表皮生长因子含量为1.5μg)。待创面基本干燥后,进行包扎。实验大鼠在相同的环境中进行饲养,让其自由进水、进食,观察大鼠背部创面愈合情况,分别在3、7、10、14天进行观察,并对创面面积进行统计。
其中,相对创面面积%=(t时间创面面积/初始创面面积)*100%。
图3为小鼠的相对创面面积图,其中I为空白对照组的相对创面面积变化,II为透明质酸固油分散体组的相对创面面积变化,III为表皮生长因子水溶液组的相对创面面积变化,IV为含表皮生长因子的分散体组的相对创面面积变化。由图3可以看出,透明质酸固油分散体组和空白对照组的相对创面面积变化基本一致,说明透明质酸固油分散体没有产生毒性且不具有促进创面愈合效果,而含表皮生长因子的分散体组与表皮生长因子水溶液组对促进创面愈合有着相似的效果,且效果明显。
Claims (10)
1.一种含生长因子的分散体,其特征在于,包括固体分散相和分散介质;所述固体分散相包括生长因子和负载所述生长因子的透明质酸或透明质酸盐。
2.根据权利要求1所述的含生长因子的分散体,其特征在于,在所述含生长因子的分散体中,所述固体分散相的含量为0.1-3mg/mL;所述固体分散相的粒径为0.1-20μm。
3.根据权利要求1所述的含生长因子的分散体,其特征在于,在所述固体分散相中,所述生长因子的负载率为0.2%-2%。
4.根据权利要求1-3中任一项所述的含生长因子的分散体,其特征在于,所述生长因子为表皮生长因子、成纤维细胞生长因子、角质细胞生长因子或***中的至少一种。
5.根据权利要求1所述的含生长因子的分散体,其特征在于,所述分散介质包含肉豆蔻酸异丙酯。
6.权利要求1-5中任一项所述的含生长因子的分散体的制备方法,其特征在于,包括以下步骤:
(1)向透明质酸盐或透明质酸中加入氧化剂,反应得到醛基透明质酸;将所述醛基透明质酸与有机碱反应,生成醛基透明质酸-有机盐,然后加入生长因子,反应,再加入还原剂还原,最后进行疏水处理,干燥,制得生长因子-透明质酸复合物;
(2)将步骤(1)所述的生长因子-透明质酸复合物与溶剂混合,得混合物,然后将混合物加入到分散介质中,除去溶剂,分散,制得所述含生长因子的分散体。
7.根据权利要求6所述的制备方法,其特征在于,在步骤(1)中,所述氧化剂为高碘酸盐;所述有机碱为四丁基氢氧化铵;所述还原剂为氰基硼氢化钠。
8.根据权利要求6所述的制备方法,其特征在于,在步骤(2)中,所述混合物与所述分散介质的体积比为(5-20):1。
9.权利要求1-5中任一项所述的含生长因子的分散体在制备药物中的应用。
10.一种药物,其特征在于,包含权利要求1-5中任一项所述的含生长因子的分散体。
