背景

方法
细胞培养和药物
使用下述人胶质瘤细胞系测试了TTFields和AZD1152的组合处理的效应:U87-MG(ATCC)、U-251 (ECACC)和U87-MGshp53(由Dr. Achim Temme提供)。所有细胞都在提供有5%CO2的加湿培养箱中生长。将细胞维持在EMEM中,所述EMEM补充有10% FBS、2 mmol/L谷氨酰胺、Pen-Strep溶液(100单位/ml青霉素和0.1 mg/ml链霉素)、1 mmol/L丙酮酸钠和1%NEAA。将U87-MGshp53细胞维持在用400 mg/mL遗传霉素的选择下。AZD1152得自以色列的Sigma。
细胞毒性测定和总体效应
使用Inovitro***,将TTFields (1.6 V/cm RMS,200 kHz)施加72小时。在处理结束时,基于细胞计数定量分析了肿瘤细胞生长的抑制。U87-MG、U87-MGshp53和U-251的克隆形成存活率通过在6孔皿中一式三份铺板300个细胞/皿进行测试。在2-3周后,将细胞用结晶紫染色,并且定量克隆数目。通过将处理结束时的存活细胞百分比乘以相对于对照形成的集落百分比来计算总体效应。
流式细胞术
对于细胞周期分析,细胞在72小时处理后立即用胰蛋白酶移出,用具有1% FBS的冰冷的PBS洗涤两次,并且用70%冰冷的乙醇固定30分钟。在固定后,将细胞用具有1% FBS的冰冷的PBS洗涤两次,形成团块,并且在含有10 µg/ml RNA酶和7.5 µg/ml 7-AAD (Sigma-Aldrich)的PBS中在37℃下温育30分钟。然后使用EC800流式细胞仪(Sony Biotechnology,日本)来定量细胞周期分布。
显微镜检查
在处理结束时,将细胞用100%甲醇固定,用0.5%结晶紫(Sigma)染色,并且在倒置显微镜(Nikon eclipse TS100)下成像。
统计分析
数据表示为平均值 ± SE,并且使用GraphPad Prism 6软件(GraphPadSoftware,La Jolla,CA)评价差异的统计显著性。组间的差异使用2因素ANOVA进行比较,并且在0.05 > *p > 0.01、**p < 0.01和***p < 0.001的值下视为显著的。
结果

值得注意的是,当AZD1152与TTFields组合时,相对于在不存在交变电场的情况下提供相同给定水平的总体效应的AZD1152剂量,获得给定水平的总体效应所需的AZD1152剂量经常可以减少至少50%。例如,图1D中的水平虚线显示了在不存在TTFields的情况下,需要70 nM的AZD1152剂量以达到38%的总体效应,而当AZD1152与TTFields组合时,只需要25nM的AZD1152剂量以达到38%的相同总体效应。
在用AZD1152处理后,U-251细胞的体积急剧增加,这使其无法使用流式细胞术进行计数。基于克隆形成测定确定对于U-251细胞的总体效应。对单独的TTFields施加的应答导致U-251细胞的克隆形成潜力的约50%降低(图1E)。TTFields和AZD1152的组合处理导致U-251集落数目的显著减少(2因素ANOVA,p<0.001) (图1E)。

另外的结果

这些结果确定了可以通过将Aurora B激酶抑制剂施用于癌细胞,并且将频率为100至300 kHz的交变电场施加于癌细胞,来降低癌细胞的生存力。在本文描述的方法的一些情况下,施加步骤的至少一部分与施用步骤的至少一部分同时执行。
在如本文所述测试的细胞中,与单独的每种处理相比,TTFields和AZD1125或MLN8237的组合导致细胞数目的显著减少。在所有测试的细胞系中,组合处理的总体效应明显高于单独的每种处理的效应。TTFields和MLN8337的组合导致多核和多倍体细胞数目的增加。
这些结果确定了可以通过将Aurora激酶抑制剂(例如,Aurora A激酶抑制剂、Aurora B激酶抑制剂)施用于癌细胞,并且将频率为100至300 kHz的交变电场施加于癌细胞,来降低癌细胞的生存力。在本文描述的方法的一些情况下,施加步骤的至少一部分与施用步骤的至少一部分同时执行。
