技术领域
本发明涉及MXene量子点,具体地,涉及一种具有过氧化物模拟酶性质的MXene量子点及其制备方法和检测谷胱甘肽的方法。
背景技术
现有的谷胱甘肽的检测方法存在的有荧光光谱法、吸光度法、高效液相色谱法、质谱法、比色法、吸光度法、毛细管法等,这些方法中吸光度法和比色法因其简单,成本低,方便等优点越来越受到人们的重视。
在上述诸多检测方法中,往往会使用到生物酶或者模拟酶。模拟酶是以纳米材料为基础的人工酶模拟物,由于与传统的生物酶相比具有显著的优势,模拟酶在过去十年一直备受关注。目前各种纳米材料如贵金属纳米粒子、金属氧化物和碳基纳米材料已经被开发为潜在的纳米酶。
但是现有的模拟酶的效果并不是十分理想,如PB(普鲁士蓝)材料,如文献“Colloids and Surfaces A Physicochemical and Engineering Aspects,2019:622-629”记载以PB作为原料合成的材料模拟酶应用到谷胱甘肽的检测中仍然存在缺陷;一是因为PB固有的蓝色会对目标物的比色检测产生强烈的背景干扰,二是由于PB的分散性差,在水溶液中聚集,进一步降低了催化活性。
发明内容
本发明的目的是提供一种具有过氧化物模拟酶性质的MXene量子点及其制备方法和检测谷胱甘肽的方法,该具有过氧化物模拟酶性质的MXene量子点能够作为模拟酶对谷胱甘肽进行定量检测,以提高谷胱甘肽的检测的准确性;同时该制备方法具有工序简单和条件温和的特性。
为了实现上述目的,本发明提供了一种具有过氧化物模拟酶性质的MXene量子点的制备方法,该制备方法包括:将Ti2C3于浓硝酸中进行回流处理,接着将体系的pH调节至中性或者碱性,然后后处理,以得到MXene量子点。
本发明还提供了一种具有过氧化物模拟酶性质的MXene量子点的制备方法,该MXene量子点通过上的制备方法制备而得。
本发明还提供了一种检测谷胱甘肽的方法,包括:
1)将不同量的谷胱甘肽分别加入相同体积的空白溶液中,得到多组待测溶液:其中空白溶液含有水、醋酸钠缓冲溶液、H2O2、TMB和Mxene量子点;
2)体系反应15-25min后,测定各组待测溶液的吸光度1;
3)以待测溶液中谷胱甘肽的最终浓度为横坐标(最终浓度指的是谷胱甘肽与空白溶液混合后的浓度),以测定的吸光度1为纵坐标,作线性拟合求出方程;
4)将含未知浓度的谷胱甘肽的待测样加入至空白溶液中,然后检测吸光度2,然后根据吸光度2计算出待测样中谷胱甘肽的浓度。
MXene是一种新型的二维过渡金属碳化物或氮化物纳米材料,具有良好的金属导电性、稳定性、表面积大、易功能化、亲水性和生物相容性等优点。并且它还具有类似石墨烯的层状结构,在能源、纳米医学、催化和传感等领域得到了广泛的探究。
在上述技术方案中,本发明通过将Ti2C3在浓硝酸中回流使得制得的Mxene量子点不仅具有尺寸优势、强的量子限域和边缘效应,而且还具有类似于普通碳量子点的优良发光性能;制得的Mxene量子点的表面具有丰富的含氧基团,如羟基、羧基基团,这种Mxene量子点作为过氧化物模拟酶在H2O2存在的条件下,可以催化氧化3,3′,5,5′-四甲基联苯胺(TMB),从而产生蓝色产物(OXTMB),使其能够作为过氧化物模拟酶使用。同时,该Mxene量子点是绿色无污染催化剂。此外,Mxene量子点还具有荧光量子产率高、分散性好、且可控制,生产成本低和重现性好的特点。
在本发明中,谷胱甘肽的检测原理如下:在辣根过氧化物酶或其他适当过氧化物酶的催化下,TMB会被过氧化氢氧化产生可溶性蓝色产物,而在本申请中,Mxene量子点便可胜任过氧化物模拟酶;一旦加入谷胱甘肽,由于谷胱甘肽的还原性,便可使得体系褪色,随着谷胱甘肽的浓度的不同,体系的褪色程度也不同,由此根据比色法便可对谷胱甘肽的浓度实施定量检测。

本发明提供了一种具有过氧化物模拟酶性质的MXene量子点的制备方法,该制备方法包括:将Ti2C3于浓硝酸中进行回流处理,接着将体系的pH调节至中性或者碱性,然后后处理,以得到MXene量子点。
在上述制备方法中,浓硝酸的浓度可以在宽的范围内选择,但是为了提高制得的Mxene量子点得催化性能,优选地,浓硝酸的浓度为60-75重量%。
在上述制备方法中,各物料的用量可以在宽的范围内选择,但是为了提高制得的Mxene量子点得催化性能,优选地,Ti2C3、浓硝酸的用量分别为0.1g:140-160mL。
在上述制备方法中,回流处理的条件可以在宽的范围内选择,但是为了提高制得的Mxene量子点得催化性能,优选地,回流处理至少满足以下条件:温度为110-150℃,时间为36-60h;更优选地,回流处理至少满足以下条件:温度为120-140℃,时间为45-50h。
在上述制备方法中,回流处理后体系的pH可以在宽的范围内选择,但是为了提高制得的Mxene量子点得催化性能,优选地,在回流处理后,体系的pH调节至7-8。
在上述实施方式中,体系的pH的调节方式可以在宽的范围内选择,但是为了进一步提高调节效果,优选地,体系的pH调节通过添加碱性化合物进行;更优选地,碱性化合物选自氢氧化钾、氢氧化钠和氨水中的至少一者。
在本发明中,Ti2C3可以是市售品,也可以是自行制备,为了提高Ti2C3的纯度,优选地,Ti2C3通过以下方法制备而得:将Ti2AlC3和HF进行接触反应,然后离心、取沉淀并用水清洗至pH为7,接着依次经过冰水浴超声、真空干燥。
在上述Ti2C3的制备方法中,为了进一步提高产率,优选地,Ti2AlC3和HF的摩尔比为1:200-250;更优选地,接触反应至少满足以下条件:温度为60-80℃,时间为15-30h。
本发明还提供了一种具有过氧化物模拟酶性质的MXene量子点的制备方法,该MXene量子点通过上的制备方法制备而得。
本发明还提供了一种检测谷胱甘肽的方法,包括:
1)将不同量的谷胱甘肽分别加入相同体积的空白溶液中,得到多组待测溶液:其中空白溶液含有水、醋酸钠缓冲溶液、H2O2、TMB和Mxene量子点;
2)体系反应15-25min后,测定各组待测溶液的吸光度1;
3)以待测溶液中谷胱甘肽的最终浓度为横坐标,以测定的吸光度1为纵坐标,作线性拟合求出方程;
4)将含未知浓度的谷胱甘肽的待测样加入至空白溶液中,然后检测吸光度2,然后根据吸光度2计算出待测样中谷胱甘肽的浓度。
在上述检测方法中,空白溶液中各组分的含量可以在宽的范围内选择,但是为了提高检测的准确性,优选地,在空白溶液中,醋酸钠缓冲溶液、H2O2、TMB、Mxene量子点和水的用量比为1mL:0.9μmol:0.3-0.5μmol:15-25μg:700-900μL;
在上述检测方法中,空白溶液中各组分可以通过纯物质的形式提供,也可以通过其他形式提供,如溶液的形式,为了便于混合,优选地,醋酸钠缓冲溶液的pH为3-4,TMB由浓度为8-10mmol/L的溶液提供,Mxene量子点由350-450μg/mL的溶液提供,H2O2由浓度为8-10mol/L的溶液提供。
在本发明中,为了进一步提高谷胱甘肽的检测的精准度,优选地,在步骤3)中,线性拟合求得的方程为:y=0.02755x-0.0284,x为谷胱甘肽的摩尔浓度,y为吸光度值。
以下将通过实例对本发明进行详细描述。以下实例中,Ti2C3采用文献“AdvanceMaterials,2017,29(15):1604847.1-1604847.6”记载的方法制备而得,具体为:取Ti2AlC3和HF(摩尔比为1:200)溶解在聚四氟乙烯反应釜中,60℃下搅拌24h后取出,去离子水洗涤离心直至pH值为7,然后冰水浴超声、真空干燥,得到Ti2C3固体粉末。
实施例1
称取0.1g的Ti2C3加入150mL浓硝酸(68重量%)中,25℃下搅拌20min,在130℃条件下回流48h,自然冷却后用NaOH溶液(8mol/L)调节pH至7.5,纯化用0.22μm微孔膜过滤之后透析12h得到溶液,旋蒸浓缩之后真空干燥得到固体颗粒,定量稀释备用。
实施例2
按照实施例1的方法进行,所不同的是,将回流温度设为110℃,浓硝酸的用量为160mL,其他条件不变。
实施例3
按照实施例1的方法进行,所不同的是,将回流温度设为120℃,浓硝酸的用量为155mL,其他条件不变。
实施例4
按照实施例1的方法进行,所不同的是,将回流温度设为140℃,浓硝酸的用量为145mL,其他条件不变。
实施例5
按照实施例1的方法进行,所不同的是,将回流温度设为150℃,浓硝酸的用量为140mL,其他条件不变。
检测例1
1)通过日立HitachiHT 7700透射电子显微镜对合成的样检测。对检测实施例1中Mxene量子点进行透射电子显微镜检测,结果见图1-1和图1-2。
由图可知:MXene量子点尺寸分散均匀,接近球形的颗粒物,平均尺寸为3.5nm左右,和普通碳纳米材料即碳量子点(1-10nm)尺寸分布特点相一致。
2)通过美国爱丁堡有限公司FS5荧光光谱仪对检测实施例1中Mxene量子点进行荧光激发检测,结果见图2,在图2中,由上到下代表的曲线分别为330nm、320nm、310nm、340nm、350nm、360nm、370nm、380nm。
由图2可知:Mxene量子点的最佳激发波长在330nm处。
3)通过日本日立公司Hitachi U-3010紫外-可见分光光度计和美国爱丁堡有限公司FS5荧光光谱仪对检测实施例1中Mxene量子点进行紫外吸收、荧光激发和发射检测,结果见图3,在图3中,由左到右代表的曲线分别为Mxene量子点的紫外吸收光谱,最佳激发波长和最佳发射波长。
由图3可知:Mxene量子点在紫外区域有很宽的光吸收,由荧光光谱可以看出此量子点最佳激发波长和最佳发射波长分别在330nm和430nm处。
4)通过日本岛津傅里叶红外光谱仪IR-21对检测实施例1中Mxene量子点进行红外光谱表征,结果见图4。
由图4可知:Mxene量子点的红外光谱图中的3445cm-1归属于O-H的伸缩振动,1638cm-1归属于C=O的振动,1385cm-1归属于C-O的伸缩振动。
5)通过德国布鲁克公司的X-射线粉末衍射仪D8 Adance对检测实施例1中Mxene量子点和Ti2C3固体粉末进行X射线衍射表征,结果见图5。
由图5可知:Mxene量子点含有不饱和碳峰,而且前驱体经过HF酸处理特征峰明显变弱并且39°峰基本消失,可以判断层间Al消失表明刻蚀成功,随着碳化程度升高,到Mxene量子点时基本上只能看到无定性碳的峰。
6)通过美国热电有限公司X射线光电子能谱仪ESCALAB250对检测实施例1中Mxene量子点和Ti2C3固体粉末进行X射线光电子能谱表征,结果见图6。
由图6可知Mxene量子点主要含有C、Ti、O三种元素,其含量分别为57.88%、0.15%、41.97%。
按照上述相同的方法对实施例2-5的产物进行表征,表征结果与实施例1的产物的表征结果基本一致。
应用例1
取800μLH2O、1mL pH=3.5的HAc/NaAc缓冲溶液、50μL的TMB(9mmol/L),100μLH2O2(9mmol/L),50μL(400μg/mL)MXene量子点(实施例1制得的),反应20min,改变不同的环境条件中,测得TMB在652nm处的吸光光谱,如图7-8所示,其中图7中的三条曲线分别代表在TMB+H2O2,TMB+Mxene量子点,TMB+H2O2+Mxene量子点这三种环境条件下时TMB吸光度变化情况,由图7-8可知:只有量子点和过氧化氢同时存在才能使TMB明显变色。
应用例2
先选取三个含谷胱甘肽的待测样制得空白液,测定空白溶液中谷胱甘肽的浓度(通过计算得到待测样中谷胱甘肽的浓度,记为第一浓度),然后测定空白液+谷胱甘肽标准溶液组成的混合液中的浓度(去除标准溶液中谷胱甘肽,计算得到待测样中的谷胱甘肽的浓度,记为第二浓度),从而计算回收率,每组试验进行三次平行实验,结果取平均值。
谷胱甘肽标准溶液中谷胱甘肽的浓度为为1μmol/L。
空白液由如下组分组成:1000μL HAc/NaAc(0.2mol/L)、100μL TMB(9mmol/L)、200μL H2O2(9mmol/L)、50μL MXene量子点(400μg/mL)、100μL待测样和550μLH2O。
空白液加谷胱甘肽标准溶液由如下组分组成:1000μL HAc/NaAc(0.2mol/L),100μL TMB(9mmol/L),200μLH2O2(9mmol/L),50μL MXene量子点(400μg/mL),100μL待测样,200μLGSH谷胱甘肽(1μmol/L)和350μLH2O。
两种溶液都是反应20min后测定其吸光度,再利用加标回收的公式计算其加标回收率。选取三个标样的浓度具体检测结果见表1。

由上表可知:采用标准加入法对标准样品中GSH含量进行了检测和验证,GSH测定的回收率在95-105%之间,相对标准偏差(RSD)小于2.5%。表明本方法可用于实验室待测样品中GSH的测定。
以上详细描述了本发明的优选实施方式,但是,本发明并不限于上述实施方式中的具体细节,在本发明的技术构思范围内,可以对本发明的技术方案进行多种简单变型,这些简单变型均属于本发明的保护范围。
另外需要说明的是,在上述具体实施方式中所描述的各个具体技术特征,在不矛盾的情况下,可以通过任何合适的方式进行组合,为了避免不必要的重复,本发明对各种可能的组合方式不再另行说明。
此外,本发明的各种不同的实施方式之间也可以进行任意组合,只要其不违背本发明的思想,其同样应当视为本发明所公开的内容。
Claims (11)
1.一种具有过氧化物模拟酶性质的MXene量子点的制备方法,其特征在于,所述制备方法包括:将Ti2C3于浓硝酸中进行回流处理,接着将体系的pH调节至中性或者碱性,然后后处理,以得到所述MXene量子点;
其中,所述Ti2C3、浓硝酸的用量比为0.1g:140-160mL;所述回流处理至少满足以下条件:温度为110-150℃,时间为36-60h;在所述回流处理后,所述体系的pH调节至7-8;所述体系的pH调节通过添加碱性化合物进行;所述碱性化合物选自氢氧化钾、氢氧化钠和氨水中的至少一者。
2.根据权利要求1所述的制备方法,其中,所述浓硝酸的浓度为60-75重量%。
3.根据权利要求1或2所述的制备方法,其中,所述回流处理至少满足以下条件:温度为120-140℃,时间为45-50h。
4.根据权利要求1或2所述的制备方法,其中,所述Ti2C3通过以下方法制备而得:将Ti2AlC3和HF进行接触反应,然后离心、取沉淀并用水清洗至pH为7,接着依次经过冰水浴超声、真空干燥。
5.根据权利要求4所述的制备方法,其中,所述Ti2AlC3和HF的摩尔比为1:200-250。
6.根据权利要求4所述的制备方法,其中,所述接触反应至少满足以下条件:温度为60-80℃,时间为15-30h。
7.一种具有过氧化物模拟酶性质的MXene量子点,其特征在于,所述MXene量子点通过权利要求1-6中任意一项所述的制备方法制备而得。
8.一种检测谷胱甘肽的方法,其特征在于,包括:
1)将不同量的谷胱甘肽分别加入相同体积的空白溶液中,得到多组待测溶液:其中空白溶液含有水、醋酸钠缓冲溶液、H2O2、TMB和如权利要求7所述的MXene量子点;
2)体系反应15-25 min后,测定各组待测溶液的吸光度;
3)以待测溶液中谷胱甘肽的最终浓度为横坐标,以测定的吸光度值为纵坐标,作线性拟合求出方程;
4)将含未知浓度的谷胱甘肽的待测样加入至空白溶液中,然后检测吸光度2,然后根据吸光度2计算出待测样中谷胱甘肽的浓度。
9.根据权利要求8所述的检测谷胱甘肽的方法,其中,在所述空白溶液中,所述醋酸钠缓冲溶液、H2O2、TMB、MXene量子点和水的用量比为1mL:0.9 μmol:0.3-0.5 μmol:15-25 μg:700-900 μL。
10.根据权利要求8所述的检测谷胱甘肽的方法,其中,所述醋酸钠缓冲溶液的pH为3-4,所述TMB由浓度为8-10 mmol/L的溶液提供,所述MXene量子点由350-450 μg/mL的溶液提供,所述H2O2由浓度为8-10 mol/L的溶液提供。
11.根据权利要求8所述的检测谷胱甘肽的方法,其中,在步骤3)中,线性拟合求得的方程为:y=0.02755x-0.0284,x为谷胱甘肽的浓度,y为吸光度值。
