本发明公开了一种双靶向多肽及其在抗肿瘤以及抑制肿瘤血管生成中的应用,属于生物技术领域,本发明的一种双靶向多肽由具有聚集发光效应的荧光单元双芘(BP)、可自组装成β‑sheet纳米纤维的氨基酸序列和可靶向识别CD 105蛋白的氨基酸序列组合而成。本发明的双靶向多肽可靶向识别肿瘤干细胞膜上特异性表达的CD 105蛋白,在肿瘤干细胞表面自组装变构形成稳定水不溶的纳米纤维,粘附在肿瘤干细胞表面并破坏肿瘤干细胞,进而发挥抑制肿瘤的增殖与转移的作用。该双靶向多肽还可同时靶向于肿瘤新生血管,发生原位自主装聚集在细胞表面,产生J型聚集发射荧光信号并形成纳米纤维,封堵内皮细胞间的间隙,降低肿瘤血管通透性、抑制血管新生,最终发挥抑制肿瘤增殖和转移的作用。
Description
技术领域
本发明涉及一种双靶向多肽及其在抗肿瘤以及抑制肿瘤转移中的应用。本发 明属于生物技术领域。
背景技术
肾癌位居发达国家恶性肿瘤前十位,占所有新发癌症病例的3.7%。世界范围 内发病率持续呈上升趋势,且预后较差。肾透明细胞肾癌是最常见的肾癌类型, 约占全部病例的80%,具有高血管密度和高转移特性,在确诊时约有30%患者已 发生转移。肾癌已经成为危害人类健康的突出问题。
肿瘤干细胞是肿瘤内的一类恶性程度高的细胞亚群,具有极强的转移能力, 并且可以通过旁分泌的方式促进肿瘤细胞发生远处转移,目前肿瘤干细胞被视为 是肿瘤转移发生的始动因素。同时在肿瘤血管新生方面,由于内皮细胞的快速增 殖,导致肿瘤血管的发育并不成熟、内皮细胞间隙相对较大,因此肿瘤细胞较易 透过血管进入血液循环进而发挥肿瘤转移。肿瘤的新生血管的高通透性打开了肿 瘤转移的大门。因此,同时抑制肾癌干细胞和肿瘤血管对拮抗肾癌的增殖与转移 具有重大意义。
CD105在肾癌干细胞和新生血管内皮细胞膜上特异性高表达,是肿瘤新生血 管和肾癌干细胞共同的生物标记物。因此本发明构建了一种可靶向识别并结合 CD105的多肽,命名为TDS(Transformable dual-inhibited system),TDS对肾癌干 细胞和肿瘤血管内皮细胞具有双靶向作用,并且可以通过变构和活体自组装形成 水不溶性纳米纤维长期滞留在细胞膜外,具有抗肿瘤和抑制血管生成的双重作 用。
发明内容
本发明的目的在于提供一种双靶向多肽及其在抗肿瘤以及抑制肿瘤转移中 的应用。
为了达到上述目的,本发明采用了以下技术手段:
本发明的一种双靶向多肽(命名为TDS),可识别并结合CD105蛋白形成水 不溶性纳米纤维,所述的双靶向多肽由以下三部分组成:
1)可自组装成β-sheet纳米纤维的氨基酸序列,其氨基酸序列如SEQ ID NO.1 所示(FFVLK);源自β淀粉样蛋白中的FFVLK肽基序列,由于氢键的相互作用 可自组装成β-sheet二级结构的水不溶性纳米纤维;
2)靶向识别CD105蛋白的氨基酸序列,其氨基酸序列如SEQ ID NO.2所示(AHKHVHHVPVRL);可靶向识别CD105蛋白的序列,其在TDS中充当靶头、 与靶标CD105特异性结合;
3)荧光单元双芘(BP)。双芘是典型的AIE效应(聚集诱导发光)荧光分 子,可用于观察TDS聚集、自组装、变构的过程。
其中,优选的,所述的多肽具有如下式所示的结构:
本发明的双靶向多肽TDS可靶向识别肿瘤干细胞膜上CD105蛋白,在肿瘤 干细胞表面自组装变构形成稳定水不溶的纳米纤维,粘附在肿瘤干细胞表面并破 坏肿瘤干细胞,进而发挥抑制肿瘤的增殖与转移的作用。经实验证明肾癌干细胞 干性被双靶向多肽TDS抑制了68±9.3%。肾癌干细胞的转移和浸润能力分别抑 制了55.5±1.8%和62.7±7.2%;另一方面,TDS可靶向识别肿瘤新生血管内皮细 胞膜上CD105蛋白,同样在血管内皮细胞表面变构、活体自组装形成稳定水不 溶的纳米纤维,粘附包封在血管内皮细胞表面,使原本通透性强的肿瘤血管致密 度增强,阻止肿瘤细胞透过内皮细胞发生癌转移。同时,体内外实验也证明双靶 向多肽TDS具有较强的抗血管新生作用。实验数据显示双靶向多肽TDS使得血 管内皮细胞通透性降低了33.0±4.7%,抑制了38±4.0%的血管新生。因此,本 发明的双靶向多肽TDS可通过抗肿瘤和抗肿瘤血管生成两方面来发挥抗癌作用。
进一步的,本发明还提出了所述的双靶向多肽在制备抗肿瘤药物中的应用, 所述的肿瘤为有CD105膜表面标志物的肿瘤。
其中,优选的,所述的肿瘤包括肾癌、口腔癌、卵巢癌。更优选的,所述的 肿瘤为肾癌。
其中,优选的,所述的双靶向多肽具有抗肿瘤细胞增殖、转移的作用。
更进一步的,本发明还提出了所述的双靶向多肽在制备抑制肿瘤血管生成药 物中的应用,在用于抑制肿瘤血管生成时,所针对的肿瘤可以是有CD105膜表 面标志物的肿瘤,或是没有CD105膜表面标志物的肿瘤。
其中,优选的,所述的双靶向多肽具有降低肿瘤新生血管的通透性、抑制肿 瘤血管新生的作用。
其中,优选的,所述的肿瘤包括肾癌、口腔癌、卵巢癌。更优选的,所述的 肿瘤为肾癌。
相较于现有技术,本发明的有益效果是:
本发明的一种双靶向多肽(命名为TDS)由具有聚集发光效应的荧光单元双 芘(BP)、可自组装成β-sheet纳米纤维的氨基酸序列和可靶向识别CD105蛋白的 氨基酸序列组合而成。TDS可在肿瘤干细胞和肿瘤血管内皮细胞表面变构、自组 装,包裹细胞。该可变形的双靶向抑制***(TDS)可同时靶向结合肿瘤干细胞 和肿瘤新生血管,发生原位自主装聚集在细胞表面,产生J型聚集发射荧光信号 并形成纳米纤维杀伤肿瘤干细胞,封堵内皮细胞间的间隙,降低肿瘤血管通透性、 抑制血管新生,最终发挥抑制肿瘤增殖和转移的作用。
此外,本发明注重临床转化应用的实际问题,以人源性肽分子为材料设计并 构建,因此TDS具有较强的生物安全性,临床应用潜力大。

具体实施方式
下面将结合实施例对发明做进一步说明,应该理解的是,这些实施例仅用于 例证的目的,绝不限制本发明的保护范围。
实施例1、双靶向多肽的制备及分子结构
1、双靶向多肽TDS(BP-FFVLK-AHKHVHHVPVRL)的制备
可识别并结合CD105蛋白形成水不溶性纳米纤维,所述的双靶向多肽由以 下三部分组成:
1)可自组装成β-sheet纳米纤维的氨基酸序列,其氨基酸序列如SEQ ID NO.1 所示(FFVLK);源自β淀粉样蛋白中的FFVLK肽基序列,由于氢键的相互作用 可自组装成β-sheet二级结构的水不溶性纳米纤维;
2)靶向识别CD105蛋白的氨基酸序列,其氨基酸序列如SEQ ID NO.2所示(AHKHVHHVPVRL);可靶向识别CD105蛋白的序列,其在TDS中充当靶头、 与靶标CD105特异性结合;
3)荧光单元双芘(BP)。为观察TDS聚集、自组装、变构的的过程,我们 选择双芘分子作为荧光信号分子,双芘是典型的AIE效应(聚集诱导发光)荧光 分子。
2、不可变构自组装的对照组多肽TDS-C(BP-AHKHVHHVPVRL)
TDS和TDS-C均通过固相合成法制备。
多肽TDS及TDS-C的分子结构模式图如图1所示。
实施例2、双靶向多肽结合CD105后变构、自组装成水不溶性纳米纤维
将CD105试剂加入到TDS和TDS-C多肽溶液中,分别于0小时和12小时 使用透射电镜观察TDS和TDS-C多肽溶液样品。结果如图2所示。从该结果可 以看出多肽TDS在CD105蛋白水溶液中可变构、自组装形成疏水性纳米纤维, 而TDS-C在CD105蛋白水溶液中不可变构、不能自组装形成疏水性纳米纤维。
实施例3、细胞实验
1、给药方法
将TDS和TDS-C多肽溶于DMSO溶剂中,配置溶液浓度为4mM的多肽纳 米材料溶液。采用状态良好呈对数生长的实验细胞,随机分为TDS、TDS-C和 PBS(磷酸盐缓冲液)组,将TDS、TDS-C及PBS溶液以100uM的浓度缓缓滴 入到培养基中,分别验证TDS、TDS-C及PBS溶液对细胞生存状态的影响。
2、多肽在血管内皮细胞和肾癌干细胞表面发生纤维转变
HEK293(人胚胎肾细胞)、人源肿瘤的肾癌干细胞、HUVEC(人脐静脉血 管内皮细胞)以105个/mL在培养皿中培养12小时。TDS、TDS-C和PBS溶液 与细胞在37℃培养基中孵育12小时和24小时,然后用PBS洗涤3次进行激光 扫描共聚焦显微镜测量。样品在405nm激光下用40×浸泡物镜进行检测。同时, 104个细胞在96孔板中培养12小时,加入TDS、TDS-C或PBS溶液,培养24 小时。使用PBS洗涤三次。然后加入硫代黄素T(ThT)共培养30分钟,使用PBS洗涤三次。在荧光酶标仪下测量荧光强度。
结果如图3所示。从该结果可以看出多肽TDS可识别聚集在肾癌干细胞 (CSCs)和血管内皮细胞膜表面。
实施例4双靶向多肽对血管内皮细胞和肾癌干细胞的杀伤作用及生物安全 性
1、细胞给药方法
同实施例3。
2、人源肿瘤的肾癌干细胞获取
人透明细胞癌组织标本被切成1mm3的立方体块,酶解成单细胞。然后,用 抗CD105抗体偶联磁珠分离CD105+细胞。在CCRCC干细胞培养基中培养 CD105+细胞。最后将细胞置于37℃、5%CO2、饱和湿度的细胞培养箱中。
3、多肽对血管内皮细胞和肾癌干细胞的杀伤作用及生物安全性
采用状态良好呈对数生长的HEK293(人胚胎肾细胞)、人源肿瘤来源的肾癌 干细胞、HUVEC(人脐静脉血管内皮细胞)以每孔8×103个细胞、总体积100 μl加入到96孔板中,置于37°孵箱中,在24小时后,将细胞随机分为TDS、 TDS-C和PBS组,并以10μM、20μM、50μM、100μM、200μM的浓度分别 加入TDS、TDS-C和PBS溶液,置于37°孵箱中,在24小时后弃去培养基并加 入配好的CCK-8溶液,置于37°孵箱中1-4小时,测量吸光度。三组小鼠分别 给予TDS、TDS-C、PBS,每2天一次,静脉给药7次,取出,器官标本后进行 H&E染色检查。
结果如图4所示。从该结果可以看出,TDS对于人源肿瘤来源的肾癌干细胞 以及HUVEC均有杀伤作用,器官标本的H&E染色检查结果表明,三组小鼠不 存在显著差异,说明TDS以及TDS-C具有生物安全性。
实施例5、双靶向多肽对肾癌干细胞的成球、浸润、迁移的抑制作用

同实施例3。
2、人源肿瘤的肾癌干细胞获取
同实施例4。
3、双靶向多肽对肾癌干细胞的成球、浸润、迁移的抑制作用
将状态良好呈对数生长的人源肿瘤肾癌干细胞置于含有20ng/ml EGF、 20ng/mlbFGF和B27的DMEM/F12培养基中培养7-10天获得肿瘤干细胞球,重 力法收集细胞球,酶解10-15min以获得单个肿瘤干细胞。然后以每孔5000细胞 的密度铺于低粘度六孔板中进行干细胞培养,并每两天加入TDS、TDS-C和PBS 溶液,7天后统计大于50nm的细胞球数目。在迁移和侵袭试验中,在Transwell (8μm孔隙大小、聚碳酸酯过滤器,6.5毫米直径;康宁)的上室涂上或不涂一层基质 胶(BD生物科学,富兰克林湖,新泽西,美国)来分别验证细胞的侵袭和迁移。将细胞 分别置于含有TDS、TDS-C或PBS溶液的培养基中。将含有50ng/ml表皮生长 因子(EGF)的1ml DMEM/F12培养基置于下室中。在37℃恒温培养48h后,对下 层被侵细胞进行染色计数。
结果如图5所示,从该结果可以看出多肽TDS具有显著抑制肾癌干细胞的 增殖、迁移及浸润能力的作用。
实施例6、双靶向多肽对血管内皮通透性及血管拟态生成的抑制作用。
细胞给药方法如实施例3。采用状态良好呈对数生长的人脐静脉内皮细胞 HUVEC,以每孔104的细胞浓度加入预先铺好50ul基质胶的Transwell上室中, 置于37℃孵箱中孵育,待细胞融合为单层后,PBS清洗Transwell上下室,吸出 PBS后分别在上室中加入含有TDS-C、TDS和PBS溶液的培养液,孵箱孵育24h 后,吸出上室液体,以PBS清洗,在上室加入200ul FITC-Dextran(异硫氰酸荧 光素-右旋糖酐),下室加入200ul PBS溶液,孵箱孵育3h后取上下室各100ul置 于96孔板中,利用荧光酶标仪进行内皮细胞通透性检测。同时,采用状态良好 呈对数生长的人脐静脉内皮细胞HUVEC,以每孔104的细胞浓度加入预先铺好50ul基质胶的96孔板中,将细胞随机分为TDS、TDS-C和PBS组,并均以100uM 的浓度分别加入TDS、TDS-C和PBS溶液,置于37°孵箱中,在24小时后拍照 观察并利用image J进行统计分析。
结果如图6所示。从该结果可以看出多肽TDS能够降低血管内皮细胞通透 性,并可抑制其增殖。
实施例7、双靶向多肽在小鼠体内分布、代谢情况。
本实验采用Balb/c裸小鼠,转移性肾癌患者的肿瘤组织细胞被接种于小鼠的 右侧肩部。当肿瘤体积达50mm3,小鼠分别通过静脉输液注射100μl TDS(400μM /鼠),TDS-C(400μM/鼠),PBS(400μM/鼠)。在注射后1,12,24,48小时使用多光 谱荧光小动物体内成像***监测小鼠的荧光成像。
结果如图7所示。从该结果可以看出,TDS多肽主要分布于肿瘤组织周围, 说明TDS多肽具有良好的靶向性,并且可被机体代谢出去。
实施例8、双靶向多肽与小鼠肿瘤血管共定位情况及对肿瘤血管生成的抑制 作用。
本实验采用Balb/c裸小鼠,转移性肾癌患者的肿瘤组织细胞被接种于小鼠的 右侧肩部。当肿瘤体积达50mm3,小鼠分别通过静脉输液注射100μl TDS(400μM /鼠),TDS-C(400μM/鼠),PBS(400μM/鼠)。并于注射后12小时,取瘤进行免疫荧 光和免疫组会检测,验证双靶向多肽与小鼠肿瘤血管共定位和对肿瘤血管的抑制 作用。
结果如图8所示。从该结果可以看出,多肽TDS可与肿瘤血管共定位并抑 制肿瘤血管生成。
实施例9、双靶向多肽在小鼠体内抑制肿瘤生长和肺部转移情况
本实验采用Balb/c裸小鼠,转移性肾癌患者的肿瘤组织细胞被接种于小鼠的 右侧肩部。当肿瘤体积达50mm3,小鼠分别通过静脉输液注射100μl TDS(400μM/ 鼠),TDS-C(400μM/鼠),PBS(400μM/鼠),每2天一次,静脉给药7次。在实验 过程中,每2天测量一次肿瘤体积和体重。取出标本后进行免疫组化和肺转移灶 测定。
结果如图9所示。从该结果可以看出,多肽TDS在体内能够抑制肿瘤增殖 以及血管新生和转移。
