目前***的治疗方式主要采取的是手术治疗、外照射放疗、腔内放疗及辅助化疗。早期***通常选择手术治疗,手术方式选择根治性子宫切除加盆腔***清扫术,治疗效果尚可;当不符合手术指征时,可以采用放射治疗,***通常对放疗比较敏感,但对于晚期及复发性的***,其预后相对较差。尽管手术治疗和辅助治疗取得了一定的进展,但疾病的发展和复发继续困扰着患有***的女性,因此寻找有效的治疗方法十分关键,特别是对中晚期及复发患者,有效的化疗尤为重要。抗血管生成(贝伐珠单抗)靶向治疗已改善了复发转移***患者的生存质量,而其他新的分子靶向药物及免疫检查点抑制剂的应用也在积极探索中。
发明内容
本发明所要解决的技术问题就是针对现有技术的不足,提供一种治疗***的药物组合物及其制药用途。本发明将甘草查尔酮和VEGFR抑制剂联合用于治疗***,大大提高了单药的使用效果。通过实验证明,甘草查尔酮和VEGFR抑制剂联合用药能够显著抑制***细胞的增殖。
为解决上述技术问题,本发明采用的技术方案是:
一种治疗***的药物组合物,包含有效量的甘草查尔酮和VEGFR 抑制剂。
优选地,所述甘草查尔酮和VEGFR抑制剂的摩尔比为(1-5):(5-10)。
优选地,所述甘草查尔酮和VEGFR抑制剂的摩尔比为1:3。
优选地,所述甘草查尔酮为甘草查尔酮B。
优选地,所述VEGFR抑制剂选自阿帕替尼、伊马替尼、拉帕替尼中的一种或多种。
优选地,所述VEGFR抑制剂选自阿帕替尼。
优选地,所述***为宫颈鳞状细胞癌。
进一步地,本发明还提供了一种具有治疗***作用的药物制剂,包括上述的药物组合物和药剂学常规辅料。
优选地,所述制剂选自片剂、胶囊剂、颗粒剂、注射剂中的一种或几种。
进一步地,本发明还提供了所述药物组合物在制备治疗***药物中的用途。
查尔酮类化合物分子能与不同的生物受体结合,对肿瘤细胞增殖具有显著的抑制作用,其作用机制包括:抑制微管蛋白聚合、抗血管生成、诱导凋亡、抗***以及逆转多药耐药性。研究表明,查尔酮是具有应用前景的新型抗肿瘤药物或抗肿瘤辅助药物的重要化合物类型。甘草查尔酮是新发现的一种***黄酮,又可分为多种不同类型。甘草查尔酮B(Lico B) 是从甘草属植物中得到的一种查尔酮类化合物,存在于豆科植物甘草的根及茎,为黄色针状结晶,难溶于水。研究报道,Lico B具有抗炎、抗菌(对革兰氏阳性菌具有较强的抑制活性)、抗氧化、抗阿尔茨海默病、保护肝脏等作用。最新的研究报道,Lico B也具有显著的抗肿瘤活性,它能抑制肿瘤细胞的增殖并诱导肿瘤细胞的细胞周期停滞和细胞凋亡。
血管生成异常是包括肿瘤在内的多种疾病发生的关键因素,该过程较为复杂,涉及多个信号分子的高度调节的相互作用。促血管生成信号分子如VEGF及VEGFR-2等,在多种人类肿瘤中高表达,且在肿瘤新血管生成中具有重要作用。由于血管***在肿瘤的发生中具有重要作用,因而其已成为肿瘤治疗的靶点之一。抗血管生成疗法可以通过阻止肿瘤脉管***的扩张,使肿瘤脉管***正常化,进而改善肿瘤周围血运状况,使细胞毒性药物能够均匀地递送至肿瘤组织核心,增强药物的抗肿瘤效果。另外,抗血管生成疗法还可以通过改善缺氧微环境和减少间质压力,降低肿瘤细胞的化疗耐药性。上述研究提示,采用血管生成抑制剂靶向血管生成级联中的多个分子,优化对血管生成的抑制,可能改善***患者的临床治疗效果。阿帕替尼是我国自主研发的新型小分子VEGFR-2酪氨酸激酶抑制剂,其主要作用机制是通过抑制VEGFR-2酪氨酸激酶活性,阻断VEGF 与其受体结合后的信号转导,抑制肿瘤血管生成。
与现有技术相比,本发明的有益效果是:发明人意外地发现,甘草查尔酮和VEGFR抑制剂同时给药对HeLa S3细胞株具有协同的抑制作用,与单独使用其中一种活性成分成分相比,将二者联合使用时的抗癌效果更为显著。特别地,当甘草查尔酮B与阿帕替尼以摩尔比1:3混合时,抑制效果最佳,能够显著提高药物的治疗效果。

下面对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。所使用的材料、试剂等,如无特殊说明,为可从商业途径得到的试剂和材料。
实施例1
一种治疗***的药物组合物,包含有效量的甘草查尔酮B与阿帕替尼,二者之间的摩尔比为1:3。
实施例2
一种治疗***的药物组合物,包含有效量的甘草查尔酮B与伊马替尼,二者之间的摩尔比为1:3。
实施例3
一种治疗***的药物组合物,包含有效量的甘草查尔酮B与拉帕替尼,二者之间的摩尔比为1:3。
实施例4
一种治疗***的药物组合物,包含有效量的甘草查尔酮B与阿帕替尼,二者之间的摩尔比为1:10。
实施例5
一种治疗***的药物组合物,包含有效量的甘草查尔酮B与阿帕替尼,二者之间的摩尔比为1:1。
对比例1
一种治疗***的药物组合物,包含有效量的甘草查尔酮B。
对比例2
一种治疗***的药物组合物,包含有效量的阿帕替尼。
对比例3
一种治疗***的药物组合物,包含有效量的甘草查尔酮B与阿帕替尼,二者之间的摩尔比为2:1。
体外细胞实验
1、实验材料:
HeLa S3(人***细胞株)
2、实验方法:
将处于对数期生长的细胞株以定量(每孔约103个细胞)加入96孔培养板中,待24小时细胞完全贴壁后,加入不同浓度的实施例1-5、对比例1-3的药物组合物(0.125μg/ml,0.25μg/ml,0.50μg/ml,1.00μg/ml, 2.00μg/ml,4.00μg/ml,8.00μg/ml,16.00μg/ml,32.00μg/ml,64.00μg/ml),每个浓度设置4个复孔。阴性对照组加入完全培养基。在37℃培养箱中培养48h,每孔加入浓度为5mg/ml的MTT溶液10μl,继续培养4h后弃去上清液,每孔再加入150μl DMSO,充分震荡溶解后迅速用酶标仪 570nm下测定吸光度OD值。
3、计算方法:
生长抑制率(GI)=1-(药物组OD值/阴性对照组OD值)×100%。根据各实验组、阴性对照组抑制率用SPSS(logit法)计算得到药物作用 48小时的IC50。实验结果如表1所示。

由表1的实验结果可知,本发明的药物组合物对人***细胞株HeLa S3有较强的抑制作用。与单独使用甘草查尔酮(对比例1)或VEGFR 抑制剂(对比例2)相比,将二者联合使用时(实施例1-5)的肿瘤细胞生长抑制效果更为显著,对HeLa S3细胞株具有协同的抑制作用。具体地,当药物组合物中的VEGFR抑制剂为阿帕替尼时(实施例1),其抑制效果优于伊马替尼(实施例2)和拉帕替尼(实施例3),IC50值可低至 6.32μg/ml。此外,对甘草查尔酮与VEGFR抑制剂之间的适宜摩尔比例也做了研究(实施例1,4,5和对比例3),发现当甘草查尔酮B与阿帕替尼以摩尔比1:3混合时,肿瘤细胞生长抑制效果最佳,能够显著提高药物的治疗效果。
药物组合物对HeLa S3(人***细胞)大鼠移植瘤的抑制作用
1、动物和细胞株的选取:
选取90只5周龄SPF级雌性SD大鼠,体质量(30±3)g。分9个笼子饲养,每个笼子10只。饲养环境温度为16-18℃,相对湿度为35-40%。 HeLa S3(人***细胞)购于中国科学院细胞库。
2、模型建立:
取对数生长期的HeLa S3细胞无血清培养基悬浮沉淀,显微镜下计数,调整细胞悬液浓度为(1.5-2)×108个/ml,染色观察活细胞数目大于 90%,在无菌环境下取0.2mlHeLa S3细胞悬浊液注入大鼠大腿根部皮下,建立人***HeLa S3细胞大鼠模型。
3、分组及药物干预:
第15天将建模成功的大鼠随机分为9组:模型组+实施例1-5+对比例 1-3,每组10只。各给药组腹腔注射给药,模型组腹腔注射等量生理盐水, 1次/天,连续给药14天。其中,各给药组每日给药剂量均为50mg/kg, VEGFR抑制剂的每日给药剂量可以通过二者间的摩尔比换算得到。
4、肿瘤抑制效果的检测:
第14天后,停药24h,脱颈处死大鼠,完整剥出瘤组织,观察各组瘤块情况及有无周围组织浸润,观察有无腹水及其他脏器转移,测量肿瘤质量并计算抑瘤率。实验结果如表2所示。

由表2的实验结果可知,与单独使用甘草查尔酮(对比例1)或VEGFR 抑制剂(对比例2)相比,将二者联合使用时(实施例1-5)的抗***效果更为显著,对HeLa S3细胞株具有协同的抑制作用。具体地,当药物组合物中的VEGFR抑制剂为阿帕替尼时(实施例1),其抗癌效果优于伊马替尼(实施例2)和拉帕替尼(实施例3)。此外,对甘草查尔酮与 VEGFR抑制剂之间的适宜摩尔比例也做了研究(实施例1,4,5和对比例 3),发现当甘草查尔酮B与阿帕替尼以摩尔比1:3混合时,抑制效果最佳,能够显著提高药物的治疗效果。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以对技术方案做出若干修改或等同替换,这些修改或等同替换也应视为本发明的保护范围。
Claims (10)
1.一种治疗***的药物组合物,其特征在于,包含有效量的甘草查尔酮和VEGFR抑制剂。
2.根据权利要求1所述的药物组合物,其特征在于,所述甘草查尔酮和VEGFR抑制剂的摩尔比为(1-5):(5-10)。
3.根据权利要求2所述的药物组合物,其特征在于,所述甘草查尔酮和VEGFR抑制剂的摩尔比为1:3。
4.根据权利要求1-3任一项所述的药物组合物,其特征在于,所述甘草查尔酮为甘草查尔酮B。
5.根据权利要求1-3任一项所述的药物组合物,其特征在于,所述VEGFR抑制剂选自阿帕替尼、伊马替尼、拉帕替尼中的一种或多种。
6.根据权利要求5所述的药物组合物,其特征在于,所述VEGFR抑制剂选自阿帕替尼。
7.根据权利要求1-3任一项所述的药物组合物,其特征在于,所述***为宫颈鳞状细胞癌。
8.一种具有治疗***作用的药物制剂,其特征在于,包括权利要求1-7任一项的药物组合物和药剂学常规辅料。
9.根据权利要求8所述的药物制剂,其特征在于,所述制剂选自片剂、胶囊剂、颗粒剂、注射剂中的一种或几种。
10.权利要求1-7中任一项所述的药物组合物在制备治疗***药物中的用途。
