所选黄精为九华山多花黄精,属十大皖药之一,为安徽道地药材。《青阳县志》:“黄精虽处处有之,唯九华山者为上。”黄精(学名:Polygonatum sibiricum)是百合科黄精属的植物。《别录》记载:“主补中益气,除风湿,安五脏。”具有补气养阴,健脾,润肺,益肾。用于脾胃气虚,体倦乏力,胃阴不足,口干食少,肺虚燥咳,劳嗽咳血,精血不足,腰膝酸软,须发早白,内热消渴等症状。黄精的化学成分主要包括多糖、甾体皂苷、三萜、生物碱、木脂素、黄酮、植物甾醇及挥发油等,其中黄精多糖为其主要药效成分。现代药理研究发现,黄精具有降低血糖,增强免疫,抗病毒,抗炎,延缓衰老,改善学习记忆等功效。
枸杞是茄目茄科枸杞属(Lycium)的植物。《本草纲目》中记载枸杞“久服坚筋骨,轻身不老,耐寒暑。”中医常用它来治疗肝肾阴亏、腰膝酸软、头晕、健忘、目眩、目昏多泪、消渴等病症。枸杞的主要化学成分为枸杞多糖,现代药理学研究证实枸杞子可调节机体免疫功能、能有效抑制肿瘤生长和细胞突变、具有延缓衰老、抗脂肪肝、调节血脂和血糖、促进造血功能等方面的作用。
红枣是鼠李科枣属植物枣Ziziphus jujuba Mill.var.inermis(Bunge)Rehd.的干燥成熟果实。《本经》:“主心腹邪气,安中养脾,助十二经。平胃气,通九窍,补少气、少津液,身中不足,大惊,四肢重,和百药。”补中益气,养血安神。用于脾虚食少,乏力便溏,妇人脏躁。现代药理研究证实大枣具有增强免疫力,降血糖,抗肿瘤,延缓衰老,改善心肌功能,抗病毒等功效。
玉竹为百合科植物玉竹Polygonatum odoratum(MilL)Druce的干燥根茎。《本经》:“主中风暴热,不能动摇,跌筋结肉,诸不足。久服去面黑野,好颜色,润泽。”养阴润燥,生津止渴。用于肺胃阴伤,燥热咳嗽,咽干口渴,内热消渴。玉竹的主要化学成分为多糖类、甾体皂苷类、高异黄酮类、氨基酸及挥发油类等化合物。现代药理研究证实玉竹具有抗肿瘤作用,抗Ⅰ型***反应的作用,以及对中枢神经有抑制作用。
目前市场以中药材为原料发酵的保健酒就多为低度酒,口感和色泽较差,不适合大众消费需求;且加工工艺简单,中药活性成分含量低,保健效果不明显等。
发明内容
本发明的目的在于提供一种黄精酒及其制备方法,以获得一款富含降糖和免疫调节活性的植物多糖的健康白酒产品。本发明原材料为黄精、构杞、玉竹、红枣等药食同源中药,药食同源中药既具备食物的丰富营养及食用安全性,又兼顾有药物的治疗效果,具有药用保健价值。
本发明黄精酒是由包括如下质量份的原料加工获得:
原酒56-68份,黄精提取液20-30份,枸杞提取液10-15份,红枣提取液10-15份,玉竹提取液10-15份。
所述原酒为浓香型古井贡酒原酒。
所述黄精提取液是由包括如下步骤的方法制备获得:
将干燥的黄精生药材用中药粉碎机粉碎5-10min,随后过40-60目筛,按1g:10-15mL的比例加入蒸馏水,同时加入1-2%纤维素酶,在50℃下酶解5h,酶解液调节pH至5.0-5.5,然后进行超声提取,提取温度50-55℃,超声功率300W,超声时间30-40min,过滤,得黄精提取液。
所述枸杞提取液是由包括如下步骤的方法制备获得:
将干燥的枸杞药材用中药粉碎机粉碎5-10min,然后过40-60目筛;以蒸馏水为提取溶剂,于50-55℃下超声提取20-30min,料液比1g:15-20mL,超声功率300W;超声提取结束后将体系转移至回流提取装置中,沸腾回流提取30-45min,过滤,得枸杞提取液。
所述红枣提取液是由包括如下步骤的方法制备获得:
将去核的干红枣用中药粉碎机粉碎5-10min,然后过40-60目筛;以蒸馏水为提取溶剂,于50-55℃下超声提取20-30min,料液比1g:15-20mL,超声功率300W;超声提取结束后将体系转移至回流提取装置中,沸腾回流提取30-45min,过滤,得红枣提取液。
所述玉竹提取液是由包括如下步骤的方法制备获得:
将干燥的玉竹药材用中药粉碎机粉碎5-10min,然后过40-60目筛;以蒸馏水为提取溶剂,于50-55℃下超声提取20-30min,料液比1g:6-10mL,超声功率300W;超声提取结束后将体系转移至回流提取装置中,沸腾回流提取30-45min,过滤,得玉竹提取液。
本发明黄精酒的制备方法包括如下步骤:
步骤1:将黄精提取液、枸杞提取液、红枣提取液以及玉竹提取液按照配比量混合,向混合提取液中添加20%-30%的葡萄糖,采用巴氏杀菌法进行灭菌,灭菌时间20-25min,灭菌温度90-95℃;
步骤2:向步骤1获得的灭菌液中接种酿酒酵母B0213发酵,接种量为3-5%,发酵时间5-8天,发酵温度25-30℃;
步骤3:将步骤2获得的发酵液调pH至5.0-5.5,然后送入孔径30nm的无机陶瓷超滤膜进行过滤,过滤压力为0.2-0.3MPa,得到发酵澄清液;
步骤4:将步骤3获得的发酵澄清液和原酒按比例混合调配成酒精度36-38°的黄精白酒,随后置于阴凉避光处密封静置,20-25℃避光陈酿50-60天。
步骤2发酵后获得的发酵液中总多糖含量不低于0.7%。
与现有技术相比,本发明的有益效果体现在:
1、本发明采用超声辅助萃取中药提取液,提取液中总多糖含量达39.36%,有效提高活性物质的含量;
2、本发明提取液、发酵液采用巴氏杀菌法灭菌,避免活性成分在杀菌过程中被破坏;
3、本发明据经典中药配方,并进行了改进,在体外实验中,该黄精白酒对α-葡萄糖苷酶和α-淀粉酶活性的抑制率分别为(84.5±0.574)%和(85.6±2.34)%,与甘氨胆酸钠和牛磺胆酸钠的结合率分别为90.5%和91.7%,在胃环境和肠环境对胆固醇的吸附量分别为0.014mg/g和0.015mg/g。推测该酒具有一定调节血糖、调节血脂等功效。并且保持了白酒的特征和口感。

具体实施方式
本实施例在以本发明技术方案为前提下进行实施,给出了详细的实施方式和具体的操作过程,但本发明的保护范围不限于下述的实施例。
实施例1:
1、提取液的制备:
A.黄精提取液制备:将干燥黄精生药材用中药粉碎机打成粉末,粉碎时间为5min,粉碎后的黄精过40目筛。以蒸馏水为提取溶剂按1g:10mL的比例提取,同时加入1%纤维素酶进行酶解,调节至pH5.0进行超声提取,提取温度50℃,超声功率300W,超声时间30min。过滤,得黄精提取液。
B.红枣提取液制备:将去核的干红枣用中药粉碎机打成粉末,粉碎时间为5min,粉碎后的红枣过40目筛。以蒸馏水为提取溶剂,料液比1g:15mL,置于超声波设备中于50℃,超声20min,超声功率300W;然后放置到回流提取装置中,沸腾回流提取30min;提取完成后,过滤,得红枣提取液。
C.玉竹提取液的制备:干燥玉竹药材用中药粉碎机打成粉末,粉碎时间为5min,粉碎后的玉竹过40目筛。以蒸馏水为提取溶剂,料液比1g:6mL,置于超声波设备中于50℃,超声提取20min,超声功率300W;然后置于回流提取装置中,沸腾回流提取30min;提取完成后,过滤,得玉竹提取液。
D.枸杞提取液的制备:干燥枸杞药材用中药粉碎机打成粉末,粉碎时间为5min,粉碎后的枸杞过40目筛。以蒸馏水为提取溶剂,料液比1g:15mL,置于超声波设备中50℃,超声20min,超声功率300W;然后置于回流提取装置中,沸腾回流提取30min;提取完成后,过滤,得枸杞提取液。
2、将黄精提取液20份、枸杞提取液10份、红枣提取液10份、玉竹提取液10份混合均匀,在混合提取液中添加20%葡萄糖,采用巴氏杀菌法进行灭菌,灭菌时间20min,灭菌温度90℃,灭菌液接种酿酒酵母B0213发酵,酵母菌接种量3%、发酵时间5天,发酵温度25℃。
3、将发酵液调pH至5.0,然后将其送入无机陶瓷超滤膜进行过滤,过滤压力为0.2MPa,得到发酵澄清液。
4、将发酵澄清液和56份原酒调成酒精度约37°的黄精白酒,置于阴凉避光处密封静置进行陈酿。
实施例2:
1、提取液的制备:
A.黄精提取液制备:将干燥黄精生药材用中药粉碎机打成粉末,粉碎时间为10min,粉碎后的黄精过60目筛。以蒸馏水为提取溶剂按1g:15mL的比例提取,同时加入2%纤维素酶进行酶解,调节至pH5.5进行超声提取,提取温度55℃,超声功率300W,超声时间40min。过滤,得黄精提取液。
B.红枣提取液制备:将去核的干红枣用中药粉碎机打成粉末,粉碎时间为10min,粉碎后的红枣过60目筛。以蒸馏水为提取溶剂,料液比1g:20mL,置于超声波设备中于55℃,超声30min,超声功率300W;然后放置到回流提取装置中,沸腾回流提取45min;提取完成后,过滤,得红枣提取液。
C.玉竹提取液的制备:干燥玉竹药材用中药粉碎机打成粉末,粉碎时间为10min,粉碎后的玉竹过60目筛。以蒸馏水为提取溶剂,料液比1g:10mL,置于超声波设备中于55℃,超声提取30min,超声功率300W;然后置于回流提取装置中,沸腾回流提取45min;提取完成后,过滤,得玉竹提取液。
D.枸杞提取液的制备:干燥枸杞药材用中药粉碎机打成粉末,粉碎时间为10min,粉碎后的枸杞过60目筛。以蒸馏水为提取溶剂,料液比1g:20mL,置于超声波设备中55℃,超声30min,超声功率300W;然后置于回流提取装置中,沸腾回流提取45min;提取完成后,过滤,得枸杞提取液。
2、将以上所得四种提取液按权利要求1的比例混合,在混合提取液中添加25%葡萄糖,采用巴氏杀菌法进行灭菌,灭菌时间25min,灭菌温度95℃,灭菌液接种酿酒酵母B0213发酵,酵母菌接种量4%、发酵时间7天,发酵温度27℃。
3、将发酵液调pH至5.5,然后将其送入无机陶瓷超滤膜进行过滤,过滤压力为0.3MPa,得到发酵澄清液。
4、将发酵澄清液和原酒按一定的比例调成酒精度约37°的黄精白酒,置于阴凉避光处密封静置进行陈酿。
实施例3:
1、提取液的制备:
A.黄精提取液制备:将干燥黄精生药材用中药粉碎机打成粉末,粉碎时间为7min,粉碎后的黄精过50目筛。以蒸馏水为提取溶剂按1g:12mL的比例提取,同时加入1.5%纤维素酶进行酶解,调节至pH5.2进行超声提取,提取温度52℃,超声功率300W,超声时间35min。过滤,得黄精提取液。
B.红枣提取液制备:将去核的干红枣用中药粉碎机打成粉末,粉碎时间为7min,粉碎后的红枣过50目筛。以蒸馏水为提取溶剂,料液比1g:17mL,置于超声波设备中于52℃,超声25min,超声功率300W;然后放置到回流提取装置中,沸腾回流提取42min;提取完成后,过滤,得红枣提取液。
C.玉竹提取液的制备:干燥玉竹药材用中药粉碎机打成粉末,粉碎时间为7min,粉碎后的玉竹过50目筛。以蒸馏水为提取溶剂,料液比1g:8mL,置于超声波设备中于52℃,超声提取25min,超声功率300W;然后置于回流提取装置中,沸腾回流提取38min;提取完成后,过滤,得玉竹提取液。
D.枸杞提取液的制备:干燥枸杞药材用中药粉碎机打成粉末,粉碎时间为7min,粉碎后的枸杞过50目筛。以蒸馏水为提取溶剂,料液比1g:12mL,置于超声波设备中52℃,超声25min,超声功率300W;然后置于回流提取装置中,沸腾回流提取38min;提取完成后,过滤,得枸杞提取液。
2、将以上所得四种提取液按权利要求1的比例混合,在混合提取液中添加30%葡萄糖,采用巴氏杀菌法进行灭菌,灭菌时间22min,灭菌温度92℃,灭菌液接种酿酒酵母B0213发酵,酵母菌接种量5%、发酵时间8天,发酵温度30℃。
3、将发酵液调pH至5.2,然后将其送入无机陶瓷超滤膜进行过滤,过滤压力为0.25MPa,得到发酵澄清液。
4、将发酵澄清液和原酒按一定的比例调成酒精度约37°的黄精白酒,置于阴凉避光处密封静置进行陈酿。

1.一种黄精酒,其特征在于由包括如下质量份的原料加工获得:
原酒56-68份,黄精提取液20-30份,枸杞提取液10-15份,红枣提取液10-15份,玉竹提取液10-15份。

6.权利要求1-5中所述的任一种黄精酒的制备方法,其特征在于包括如下步骤:
步骤1:将黄精提取液、枸杞提取液、红枣提取液以及玉竹提取液按照配比量混合,向混合提取液中添加20%-30%的葡萄糖,采用巴氏杀菌法进行灭菌,灭菌时间20-25min,灭菌温度90-95℃;
步骤2:向步骤1获得的灭菌液中接种酿酒酵母B0213发酵;
步骤3:将步骤2获得的发酵液调pH至5.0-5.5,然后送入孔径30nm的无机陶瓷超滤膜进行过滤,得到发酵澄清液;
步骤4:将步骤3获得的发酵澄清液和原酒按比例混合调配成酒精度36-38°的黄精白酒,随后置于阴凉避光处密封静置,20-25℃避光陈酿50-60天。
7.根据权利要求6所述的制备方法,其特征在于:
步骤2中,酿酒酵母的接种量为3-5%,发酵时间5-8天,发酵温度25-30℃。
8.根据权利要求6或7所述的制备方法,其特征在于:
步骤2发酵后获得的发酵液中总多糖含量不低于0.7%。
9.根据权利要求6所述的制备方法,其特征在于:
步骤3中,过滤压力为0.2-0.3MPa。
