Description

Callicarpa nudiflora extract, preparation method and anti-cervicitis application thereof
Technical Field
The invention relates to the field of traditional Chinese medicine extraction, in particular to a preparation method of a callicarpa nudiflora extract and application thereof in cervicitis resistance.
Background
The world health organization's survey of chinese women showed that: about 41 percent of women in childbearing age in China have gynecological inflammatory diseases with different degrees, and the incidence rate of married women is higher to 70 percent. According to statistics, the cervicitis accounts for about 40-50% of the total number of outpatients in obstetrics and gynecology department, and the chronic cervicitis accounts for 10-25% of outpatients, so the development of the anti-cervicitis preparation with definite curative effect is an urgent requirement of most patients.
Callicarpa nudiflora is dry leaf of Callicarpa of Verbenaceae, and is from Verbenaceae, and recorded in Chinese medicinal dictionary, Callicarpa davidii is Callicarpa pallidiflora and Callicarpa chinensis of Callicarpa, and has effects of removing toxic substance, treating sore, astringing, stopping bleeding, dispelling pathogenic wind, and eliminating dampness, and can be used for treating scald, burn, traumatic hemorrhage, etc.
The existing research shows that the callicarpa nudiflora extract has the pharmacological activities of promoting hemostasis, inhibiting thrombosis, inhibiting bacteria and cell toxicity, resisting inflammation, enhancing immunity and the like.
The nakedflower beautyberry bolt, the nakedflower beautyberry tablet and the nakedflower beautyberry capsule which take the nakedflower beautyberry as the main medicine are widely applied to the treatment of gynecological diseases and skin diseases and have obvious effect. Although the efficacy of the callicarpa nudiflora as a compound medicine is exact, the research on the aspect of treating cervicitis by using a single medicine of the callicarpa nudiflora does not exist, so that the invention takes the callicarpa nudiflora as a research object to research the anti-inflammation, hemostasis and the effect of the callicarpa nudiflora on a phenol cervicitis rat model, and discloses the action mechanism of the callicarpa nudiflora for resisting cervicitis.

TNF-alpha and PGE of three concentration groups of nakedflower beautyberry n-butanol extract2All had significant reduction (P) compared with the model group<0.05); IL-1 levels in the medium and high concentration groups (i.e., A2 and A3) of n-butanol extract of Callicarpa nudiflora were reduced compared to the model group, and P was found to be lower than that in the model group<0.05, has statistical difference, and shows that the n-butyl alcohol extract of the lithospermum nudiflorum has an anti-inflammatory effect. TNF-alpha, IL-1 and PGE compared with the model group in the Fuyanjie group2Slightly lower level, P<0.05, with statistical differences. TNF-alpha, IL-1, PGE, compared to n-butanol2Close to the horizontal, P>0.05, no statistical significance.

(1) Pathological histological examination: the pathological histology examination process of female SD rat is shown in figure 1
(2) The research results are as follows: FIGS. 2-8 are the pathological section views of the cervical tissue of rats in each group,
as can be seen from FIG. 2, the tissue structure of the cervical mucosa of the rats in the blank group is clear and complete, the structure of the squamous columnar epithelial transition part is normal, and lymphocytes and eosinophils are sporadically scattered in the cervical mucosa and submucosal tissues.
As can be seen from FIG. 3, the epithelial tissues of the cervical mucosa of the rats in the model group are obviously degenerated and necrosed, partially exfoliated, and the glands are destroyed, so that a large amount of lymphocytes and neutrophil infiltration can be seen in the cervical mucosa and the interstitium, and interstitial fibroplasia and a large amount of inflammatory exudates can be seen in the uterine cavity.
Fig. 4 shows that the pathological changes of rats in the low concentration drug group are reduced, but are not obvious, a large amount of inflammatory cells such as eosinophils and lymphocyte infiltration are still remained from the mucous membrane to the stroma, the inflammatory exudate and necrotic tissues in the uterine cavity are obviously reduced, and part of the cervical mucous membrane is close to normal.
Fig. 5 shows that the pathological changes of the cervical tissues of rats in the medium concentration group of the drug group are obviously reduced compared with the model group, the epithelium of the cervical mucosa is not recovered to be normal, but no obvious squamous epithelial hyperplasia and degeneration necrosis are generated, a little inflammatory cell infiltration is occasionally seen from the mucosa to the interstitium, and no obvious inflammatory exudation is seen in the uterine cavity.
As can be seen in fig. 6, the cervical mucosal epithelium of rats in the high concentration group of the drug group had been substantially restored, some of the cervical mucosa had been nearly normal, a small amount of inflammatory cell infiltration was occasionally seen from mucosa to interstitium, and no inflammatory exudate was seen in the cavity.
Fig. 7 shows that the pathological changes of the cervical tissues of the rats in the Fuyanjie group are reduced compared with the cervical tissue of the model group, the epithelium of the cervical mucosa is not completely repaired, the part is close to normal, a small amount of inflammatory cell infiltration is occasionally seen from the mucosa to the stroma, and no inflammatory exudate and necrotic tissues are seen in the uterine cavity.
As can be seen in FIG. 8, the lesion of the cervical tissue of the rat with n-butanol group was reduced as compared to the model group.
Therefore, the n-butyl alcohol extract of the callicarpa nudiflora has obvious positive effect on repairing cervical tissues.
Finally, it should be noted that: although the present invention has been described in detail with reference to the foregoing embodiments, it will be apparent to those skilled in the art that changes may be made in the embodiments and/or equivalents thereof without departing from the spirit and scope of the invention. Any modification, equivalent replacement, or improvement made within the spirit and principle of the present invention should be included in the protection scope of the present invention.
