Abstract
本发明涉及一种具养颜美容的草本萃取发酵果蔬液的制备方法,包括选择发酵菌、将果蔬草本组合物提取发酵液中所用的蔬菜和水果原料依照不同比例进行清洗、杀菌、打浆;然后加入中药草本提取发酵液再次植菌进行二次发酵;将二次发酵得到的发酵液经过过滤及压榨除去固态杂质,即得具养颜美容的草本萃取发酵果蔬液原浆。本发明通过结合食品可用中药草本配方以不同的比例调整来改善其美容养颜效果,白芷及茯苓透过热水提取后再植菌发酵所得到的发酵液更能有效抑制黑色素的形成;此外将中药草本提取发酵液结合各种比例不同的蔬果及美容圣品胶原蛋白创造出的新组合配方更可以针对在养颜美容达到更有效的成果。

本发明涉及一种具养颜美容的草本萃取发酵果蔬液的制备方法,属于食品加工技术领域。
背景技术
酵素又称为“酶”,是一种具有功能性的微小蛋白质,也是一种生物催化剂。酵素承担调控人体内新陈代谢中各种化学反应,如增加组织各种生化反应(氧化、还原、分解、合成、转化)的速率的工作。人体内凡是细胞的代谢、新生、分解、消化、合成等的化学作用,都依靠酵素来完成。在自然界中,小生物体,如细菌和霉菌等微生物,因无法直接摄取大分子有机物作为营养来源,故必须先借由分泌可分解大分子有机物的消化酵素,将其成为小分子单位如单糖、甘油、短脂肪酸、氨基酸等,再进行吸收利用。此外,微生物也需依靠酵素来局部分解寄主细胞。对大型生物体,如人类与其他动物种类而言,也需要利用酵素帮助食物消化与产生能量的转化。但酵素不是药品,无任何副作用,是大自然恩赐的营养食品。
目前市场上果蔬发酵原液都标榜具有养颜美容的效果,但实际状况效果不是很明显。所以透过建立一套完整的中药提取发酵液与各种果蔬调整成适合在养颜美容的果蔬的配方,是可以有效改善未来保健食品产品开发上的主轴。果蔬发酵原液成分大多来自蔬菜(萝卜、甜菜根、西红柿、黄瓜、菠菜、苋菜…等)及水果(波罗、木瓜、柠檬、猕猴桃、苹果、诺丽果、西瓜…等)原料,其中波萝含丰富的维生素B群能有效地滋养肌肤,防止皮肤干裂,滋润头发的光亮,同时也可以消除身体的紧张感和增强免疫力;木瓜含有丰富木瓜酶,维生素C、B及钙、磷及矿物质,营养丰富,果实含大量丰富的胡萝卜素、蛋白质、钙盐、蛋白酶、柠檬酶等,对美容护肤养颜的功效极大帮助,至于其他蔬菜和水果也都是含有养颜美容成分的维生素、矿物质及胺基酸;以上是目前广泛用于发酵果蔬液产品常出现的水果大多是含有维生素C、B、E等营养元素及胺基酸可以具有皮肤修补其抗氧化能力的效果。白芷来源于伞形科多年生草本植物白芷或杭白芷的根。现代药理作用表明白芷中的香豆素类具有解热、镇痛抗炎作用,扩血管、抗血小板凝集、降血压以及光敏作用等;茯苓,俗称云苓、松苓、茯灵,为寄生在松树根上的菌类植物,形状像甘薯,外皮黑褐色,含多糖类成分,包括β-茯苓聚糖、茯苓次聚糖等;三萜类成分,包括松苓酸、茯苓酸、块苓酸、齿孔酸、去氢齿孔酸、7,9(11)-去氢茯苓酸、茯苓酸甲酯、β-香树脂醇乙酸酯、去氢乙酰茯苓酸、3β-羟基羊毛甾三烯酸、麦角甾二烯-3β-醇、3-氢化松苓酸、茯苓新酸A-D、G-H、AM、DM。人参所含人参皂苷按其苷元的化学结构可分为三类:人参二醇系皂苷,人参三醇系皂苷,齐墩果酸。截至目前为止,人参皂苷已鉴定者有三十余种,此外所富含的人参多糖也是常见用来做保健的功效,较常被研究其药理作用包含了心血管、糖代谢、中枢神经、性腺、抗衰老等与荷尔蒙相关之研究方针,其中人参因含有人参皂苷,能促进血液循环,防止肌肤老化与皱纹产生,增加肌肤的营养供应,发挥补气养颜的效果。金银花主要药效成分含绿原酸、异绿原酸和咖啡酸、环己方醇、黄酮类、肌醇、皂苷、鞣质等等有具有抗氧化、提高免疫力、抗发炎的作用。
发明内容
本发明针对现有技术存在的不足,提供了一种具养颜美容的草本萃取发酵果蔬液的制备方法,具体技术方案如下:
一种具养颜美容的草本萃取发酵果蔬液的制备方法,包括以下步骤:
步骤一、选择发酵菌
发酵所用菌包含植物乳杆菌、嗜酸乳杆菌、干酪乳杆菌、鼠李糖乳杆菌、长双岐杆菌、啤酒酵母菌、醋酸杆菌,以菌液的形式加入;
步骤二、备料
将果蔬草本组合物提取发酵液中所用的蔬菜和水果原料依照不同比例进行清洗、杀菌、打浆;然后加入中药草本提取发酵液再次植菌进行二次发酵;
步骤三、发酵
按配比将菌液、果蔬草本组合物提取发酵液混和均匀在25-30℃的温度下发酵45-60天得发酵液;
四、压榨和过滤
经过步骤三所得到的发酵液经过过滤及压榨除去固态杂质,即得具养颜美容的草本萃取发酵果蔬液原浆。
上述技术方案的进一步优化,步骤一中,发酵所用菌,以菌液的形式加入,菌液浓度为105-108cfu/mL。
上述技术方案的进一步优化,步骤二中,在二次发酵过程中,将发酵液的pH值调整为3.5-4.0。
上述技术方案的进一步优化,步骤三中,在发酵过程中同时还进行搅拌,搅拌的速率为30-80rpm/min。
上述技术方案的进一步优化,所述果蔬草本组合物提取发酵液的原料包括以下组分:菠萝30±3质量份、木瓜25±2质量份、中药草本提取发酵液10±1质量份、苹果5质量份、胶原蛋白5质量份、红枣5质量份、蓝莓2质量份、葡萄2质量份、猕猴桃2质量份、柠檬2质量份、番石榴2质量份、青梅2质量份、甜菜根2质量份、萝卜2质量份、西红柿2质量份、黄瓜1质量份、薏仁1质量份。
上述技术方案的进一步优化,所述中药草本提取发酵液以如下方法制备:将中药草本组合物进行热水提取30分钟,加入2-10质量份红糖、2-10质量份蜂蜜,放冷至摄氏25-30℃,然后再植入乳酸菌、醋酸菌、酵母菌发酵处理;配制浓度为105-108cfu/mL的菌液,在25-30℃的温度下发酵30天;最后再经过滤除去滤渣即得所述中药草本提取发酵液。
上述技术方案的进一步优化,所述中药草本组合物包括以下重量份数的中药草本:
白芷20±2质量份、人参10±1质量份、茯苓20±2质量份、金银花5质量份;所述中药草本组合物与40±4质量份的热水进行提取。
上述技术方案的进一步优化,所述中药草本组合物与热水在100℃的温度下提取30分钟。
本发明的有益效果:
本发明针对现有技术中纯蔬果发酵原液对养颜美容效果较不明显,尤其是在美白效果更没有明显的效益。本发明通过结合食品可用中药草本配方以不同的比例调整来改善其美容养颜效果;与一般热水提取方式相比较,白芷及茯苓通过热水提取后再植菌发酵后的白芷及茯苓提取发酵液,能有效抑制黑色素的形成。此外将中药草本提取发酵液结合各种比例不同的蔬果及美容圣品胶原蛋白创造出的新组合配方更可以针对在养颜美容达到更有效的成果。

具体实施方式
为了使本发明的目的、技术方案及优点更加清楚明白,以下结合附图及实施例,对本发明进行进一步详细说明。应当理解,此处所描述的具体实施例仅仅用以解释本发明,并不用于限定本发明。
实施例1
具养颜美容的草本萃取发酵果蔬液的制备方法,如图1所示,包括以下步骤:
步骤一、选择发酵菌
发酵所用菌包含植物乳杆菌、嗜酸乳杆菌、干酪乳杆菌、鼠李糖乳杆菌、长双岐杆菌、啤酒酵母菌、醋酸杆菌,以菌液的形式加入,菌液浓度为105-108cfu/mL。
步骤二、备料
将果蔬草本组合物提取发酵液中所用的蔬菜和水果原料依照不同比例进行清洗、杀菌、打浆;然后加入中药草本提取发酵液再次植菌进行二次发酵;将发酵液的pH值调整为3.5-4.0。
其中,所述中药草本组合物的组分为:白芷20±2kg、人参10±1kg、茯苓20±2kg、金银花5kg;所述中药草本组合物与40±4kg的热水进行提取,在100℃的温度下提取30分钟;然后,加入2-10kg红糖、2-10kg蜂蜜,放冷至摄氏25-30℃,然后再植入乳酸菌、醋酸菌、酵母菌发酵处理;配制浓度为105-108cfu/mL的菌液,在25-30℃的温度下发酵30天;最后再经过滤除去滤渣即得中药草本提取发酵液。
所述果蔬草本组合物提取发酵液的原料包括以下组分:菠萝30±3kg、木瓜25±2kg、中药草本提取发酵液10±1kg、苹果5kg、胶原蛋白5kg、红枣5kg、蓝莓2kg、葡萄2kg、猕猴桃2kg、柠檬2kg、番石榴2kg、青梅2kg、甜菜根2kg、萝卜2kg、西红柿2kg、黄瓜1kg、薏仁1kg。
步骤三、发酵
按配比将菌液、果蔬草本组合物提取发酵液混和均匀在25-30℃的温度下发酵45-60天得发酵液;其中,在发酵过程中同时还进行搅拌,搅拌的速率为30-80rpm/min。
四、压榨和过滤
经过步骤三所得到的发酵液经过过滤及压榨除去固态杂质,即得具养颜美容的草本萃取发酵果蔬液原浆。
实施例2
抑制酪氨酸酶活性试验
酪氨酸酶抑制活性是以分光亮度方式,在96孔盘上分析其在492nm时的吸亮度,而熊果素与曲酸被用来作为正控制组。将预测物质溶解于DMSO水溶液中,然后与L-酪胺酸(2.5mg/mL)一起加于50mmol/L磷酸盐缓冲液中(pH=6.8)。接下来,再加入在同一种缓冲液中的mushroom tyrosinase,然后将混合物培养于37℃,三十分钟。酪氨酸酶抑制剂于492nm时的活性可由下式测得:
Inhibition=100{[(A-B)-(C-D)]/(A-B)};Inhibition的单位符号为%;
A:未加入预测物质时的光学浓度(OD492);
B:未加入预测物质时的光学浓度(OD492),但有加酪氨酸酶;
C:已加入预测物质的光学浓度(OD492);
D:已加入预测物质的光学浓度(OD492),但没有加酪氨酸酶。
具体分组如下:
空白组:水;
熊果素:positive control;
白芷萃取液:20%±2%、热水80%;
茯苓萃取液:20%±2%、热水80%;
人参萃取液:20%±2%、热水80%;
金银花萃取液:20%±2%、热水80%;
中药草本提取发酵液:白芷20±2%、茯苓20±2%、人参10±1%、金银花5%、水40±4%、红糖2-10%、蜂蜜2-10%,加热到100℃提取30分钟冷却。
一般果蔬配方发酵液:菠萝40±3%、木瓜25±2%、苹果5%、胶原蛋白5%、红枣5%、蓝莓2%、葡萄2%、猕猴桃2%、柠檬2%、番石榴2%、青梅2%、甜菜根2%、萝卜2%、西红柿2%、黄瓜1%、薏仁1%。
养颜美容的草本萃取发酵果蔬液:菠萝30±3%、木瓜25±2%、中药草本提取发酵液10±1%、苹果5%、胶原蛋白5%、红枣5%、蓝莓2%、葡萄2%、猕猴桃2%、柠檬2%、番石榴2%、青梅2%、甜菜根2%、萝卜2%、西红柿2%、黄瓜1%、薏仁1%。
以上组分中的百分比均为质量百分比;热水的温度均为100℃。
试验结果如图2所示,通过分析可知:一般果蔬配方发酵液对酪氨酸酶的活性抑制效果非常差。白芷萃取液、茯苓萃取液、人参萃取液、金银花萃取液等对酪氨酸酶的活性抑制效果虽然比一般果蔬配方发酵液有所提高,但是提高幅度有限。而将白芷、茯苓、人参、金银花等提取发酵制成的中药草本提取发酵液,中药草本提取发酵液比其中单一成分对酪氨酸酶的活性抑制效果要好的多。
本发明所述具养颜美容的草本萃取发酵果蔬液对酪氨酸酶的活性抑制效果最好,其抑制效果接近熊果素;由于能抑制酪氨酸酶的活性,进而阻碍黑色素的形成,因此对美白颇具功效。
现代多数在中药材提取方式多以传统方法(水萃取、有机萃取..等)或是现代方法(膜提取分离、超临界萃取…等)来萃取所需的有效成分。
在本发明中,先通过利用传统的热水提取破坏植物体组织架构后在利用植入优势益生菌群来进行发酵方式;由抑制酪氨酸酶活性试验可得知:其中药(白芷、人参、茯苓、金银花)草本提取发酵液的美白效果最为有效,最后再经有共发酵的方式将蔬菜、水果和草本提取发酵液进行一套标准生产流程可得到一种具养颜美容的草本萃取发酵果蔬液的制备方法。
实施例3
抑制酪氨酸酶活性的对照实验,其实验步骤与实施例2中的抑制酪氨酸酶活性试验相同,具体如下:
空白组:水;
熊果素:positive control;
中药提取混和液白芷20±2%、茯苓20±2%、人参10±1%、金银花5%、水40±4%、红糖2-10%、蜂蜜2-10%;
中药草本提取发酵液:白芷20±2%、茯苓20±2%、人参10±1%、金银花5%、水40±4%、红糖2-10%、蜂蜜2-10%,加热到100℃提取30分钟冷却。
以上组分中的百分比均为质量百分比;热水的温度均为100℃。
实验结果如图3所示。
通过分析实施例2和实施例3可知:将白芷萃取液、茯苓萃取液、人参萃取液、金银花萃取液只是通过简单的混合得到混合液,该混合液对酪氨酸酶活性的抑制效果明显比中药草本提取发酵液对酪氨酸酶活性的抑制效果差。这也就说明,虽然白芷萃取液、茯苓萃取液、人参萃取液、金银花萃取液等都具有一定的对酪氨酸酶活性抑制效果;但是如果只通过简单的混合技术手段,其对酪氨酸酶活性的抑制效果提升并不明显;而如果采用混合发酵的方法,则能显著提高其对酪氨酸酶活性的抑制效果。
本发明通过结合食品可用中药草本配方以不同的比例调整来改善其对养颜美容效果,其中也发现在白芷及茯苓透过热水提取后再植菌发酵与一般热水提取方式比较发酵过后的白芷及茯苓提取发酵液能有效抑制黑色素的形成。此外将中药草本提取发酵液结合各种比例不同的蔬果及美容圣品胶原蛋白创造出的新组合配方更可以针对在养颜美容达到更有效的成果。
以上所述仅为本发明的较佳实施例而已,并不用以限制本发明,凡在本发明的精神和原则之内所作的任何修改、等同替换和改进等,均应包含在本发明的保护范围之内。
Claims (8)
1.一种具养颜美容的草本萃取发酵果蔬液的制备方法,其特征在于包括以下步骤:
步骤一、选择发酵菌
发酵所用菌包含植物乳杆菌、嗜酸乳杆菌、干酪乳杆菌、鼠李糖乳杆菌、长双岐杆菌、啤酒酵母菌、醋酸杆菌,以菌液的形式加入;
步骤二、备料
将果蔬草本组合物提取发酵液中所用的蔬菜和水果原料依照不同比例进行清洗、杀菌、打浆;然后加入中药草本提取发酵液再次植菌进行二次发酵;
步骤三、发酵
按配比将菌液、果蔬草本组合物提取发酵液混和均匀在25-30℃的温度下发酵45-60天得发酵液;
四、压榨和过滤
经过步骤三所得到的发酵液经过过滤及压榨除去固态杂质,即得具养颜美容的草本萃取发酵果蔬液原浆。
2.根据权利要求1所述的一种具养颜美容的草本萃取发酵果蔬液的制备方法,其特征在于:步骤一中,发酵所用菌,以菌液的形式加入,菌液浓度为105-108cfu/mL。
3.根据权利要求1所述的一种具养颜美容的草本萃取发酵果蔬液的制备方法,其特征在于:步骤二中,在二次发酵过程中,将发酵液的pH值调整为3.5-4.0。
4.根据权利要求1所述的一种具养颜美容的草本萃取发酵果蔬液的制备方法,其特征在于:步骤三中,在发酵过程中同时还进行搅拌,搅拌的速率为30-80rpm/min。
5.根据权利要求1所述的一种具养颜美容的草本萃取发酵果蔬液的制备方法,其特征在于:
所述果蔬草本组合物提取发酵液的原料包括以下组分:菠萝30±3质量份、木瓜25±2质量份、中药草本提取发酵液10±1质量份、苹果5质量份、胶原蛋白5质量份、红枣5质量份、蓝莓2质量份、葡萄2质量份、猕猴桃2质量份、柠檬2质量份、番石榴2质量份、青梅2质量份、甜菜根2质量份、萝卜2质量份、西红柿2质量份、黄瓜1质量份、薏仁1质量份。
6.根据权利要求5所述的一种具养颜美容的草本萃取发酵果蔬液的制备方法,其特征在于:
所述中药草本提取发酵液以如下方法制备:将中药草本组合物进行热水提取30分钟,加入2-10质量份红糖、2-10质量份蜂蜜,放冷至摄氏25-30℃,然后再植入乳酸菌、醋酸菌、酵母菌发酵处理;配制浓度为105-108cfu/mL的菌液,在25-30℃的温度下发酵30天;最后再经过滤除去滤渣即得所述中药草本提取发酵液。
7.根据权利要求6所述的一种具养颜美容的草本萃取发酵果蔬液的制备方法,其特征在于:
所述中药草本组合物包括以下重量份数的中药草本:
白芷20±2质量份、人参10±1质量份、茯苓20±2质量份、金银花5质量份;所述中药草本组合物与40±4质量份的热水进行提取。
8.根据权利要求7所述的一种具养颜美容的草本萃取发酵果蔬液的制备方法,其特征在于:所述中药草本组合物与热水在100℃的温度下提取30分钟。
