技术领域
本发明属于微生物技术领域,特别涉及一种具有美容养颜和通便功能的益生菌酵素。
背景技术
随着人民生活水平的逐步提升,女性朋友对于美丽容颜有着美好的向往和追求。但是由于工作压力大,长期处于电子辐射及加班状态,人们往往会出现代谢循环缓慢、皮肤干燥等皮肤问题。为解决这些问题,市场上出现较多保健品以期解决问题,但是大多产品效果不明显,服用后存在副作用,给人们健康带来隐患。
便秘是较为常见的肠道疾病之一,主要表现为排便每周小于3次,并伴有排便不畅,排便量小,排便时***肿痛等症状。若人体长期便秘,体内毒素将无法及时排出,造成毒素被人体重新吸收,导致器官病变,严重影响人们的健康。
引起便秘的主要原因是肠道内菌群失调,补充益生菌酵素调节肠道微生物是改善便秘的有效途径之一。益生菌进入肠道内可促进胃肠蠕动,改善肠道内微生物环境,加速粪便及毒素的排出,有效缓解便秘,润肠通便,起到美容养颜的作用。本发明在原料选择及技术制备上针对性缓解便秘问题,且为美容养颜提供一款全新的酵素产品。

针对现有问题,本发明提供一种具有美容养颜和润肠通便功能的益生菌酵素的制备方法。
本发明采用的技术方案,一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素,包括如下原料:水85-95g/L,玫瑰茄1.0-2.0g/L,桑椹0.5-1.5g/L,白芷0.5-1.5g/L,葡萄籽0.5-1.5g/L,干姜0.2-0.4g/L,银耳0.05-0.3g/L,大枣0.05-0.3g/L,丁香0.05-0.3g/L,沙棘0.05-0.3g/L,茯苓0.05-0.3g/L,白果0.05-0.3g/L,低聚异麦芽糖1.0-3.0g/L,胶原蛋白肽0.001-
0.002g/L。
优选地,上述的一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素,包括如下原料:水92.599g/L、玫瑰茄1.5g/L,桑椹1.0g/L,白芷1.0g/L,葡萄籽1.0g/L,干姜0.3g/L,银耳0.1g/L,大枣0.1g/L,丁香0.1g/L,沙棘0.1g/L,茯苓0.1g/L,白果0.1g/L、低聚异麦芽糖2.0g/L、胶原蛋白肽0.001g/L。
上述的一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素的制备方法,包括如下步骤:
1)原料预处理:将葡萄籽破壳粉碎,茯苓粉碎至10-20目;白果去壳,白果仁粉碎至10-20目;
2)按比例称取原料,将玫瑰茄、葡萄籽、沙棘、茯苓、白果平均分装至蒸煮袋中投入提取罐,白芷、干姜、桑椹、银耳、大枣直接投入多功能提取罐中,罐内物料浸没水中浸泡4~6h,投入分装蒸煮袋的丁香;
3)蒸煮:在提取罐内蒸煮已投物料进行蒸煮,出现浓郁的白芷味,且伴有微酸、微辣、微苦,液体颜色为红棕色,此时停止蒸煮;
4)收集:过滤,收集滤液备用;
5)发酵液培养基制备:将步骤4)所得料液搅拌均匀打入发酵罐中,投入低聚异麦芽糖、胶原蛋白肽,搅拌均匀后进行灭菌及冷却处理;
6)菌种活化:培养基pH值调节为6.60,将培养基灭菌后冷却,接种植物乳杆菌HCS03-001、罗伊氏乳杆菌HCS02-001、鼠李糖乳杆菌HCS01-013,37℃条件下培养16±1h;
7)发酵:将活化的菌种接种到步骤5)所得的培养基上投入发酵罐中,接种量为5%进行发酵。
8)发酵液离心:将步骤7)中所得发酵液进行离心,离心后产品外观保持红棕色,溶液均一,澄清,无杂质。
9)超高温瞬时灭菌:将步骤8)中得到的发酵液灭菌,要求温度131℃,时间15s,料液出口温度<30℃;以未灭菌的样品作为参考,颜色、滋气味及状态无明显差别,检测无杂菌即为杀菌结束。
10)无菌灌装:将步骤9中得到的料液冷却至室温进行灌装;
11)巴氏杀菌:将步骤10)的罐装产品进行巴氏杀菌,得具有美容养颜和润肠通便功能葡萄籽白芷桑椹酵素。
优选地,上述的一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素的制备方法,步骤3)中,所述的蒸煮为温度达到100℃时开始计时40~50min。
优选地,上述的一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素的制备方法,步骤5)中,所述的灭菌是115℃灭菌30min。

本发明与现有技术相比,具有以下有益效果:
1、本发明酵素原料应用药食同源原料,选用的乳酸菌均分离自人源粪便,应用安全性高。
2、本发明酵素应用水提技术,设置特定的温度、时间等参数,提高了提取效率,并将得到的滤液利用益生菌进行发酵,有效的保留活性成分,丰富产品风味。
3、本发明酵素原料采用葡萄籽,其中含有葡萄多酚类物质,主要有儿茶素类和原花青素类,葡萄籽中还含有粗蛋白、氨基酸、和维生素A、D、E、K及多种微量元素。除此之外,葡萄籽还含有丰富的亚油酸、维生素A和维生素E等营养物质,具有抗氧化、抗衰老、降血脂等作用。
4、本发明酵素原料采用白芷,其性味辛、苦、温,有解表散寒、祛风止痛、宜通鼻窍、燥湿止带、消肿排脓的功效,同时有消淤、止血、通便的作用。
5、本发明酵素原料采用桑椹,其含有多种微量元素,促进红细胞再生,具有补血的功效,富含桑椹多酚,是一种天然的抗氧化剂,具有超强的清除自由基和抵抗自由基的能力,可加速细胞新陈代谢的速度,使面部的皮肤可以变得更加有弹性、嫩滑。除此之外,桑椹可以减少脸部皱纹以及色斑的形成,具有十分明显的美白抗氧化的效果。

实施例1一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素
1、原料预处理:将葡萄籽破壳粉碎,茯苓粉碎至10目左右;白果去壳,白果仁粉碎至10目左右;
2、投料:按比例称取原料,将玫瑰茄、葡萄籽、沙棘、茯苓、白果平均分装至蒸煮袋中(物料占蒸煮袋体积的四分之一以下)投入提取罐,白芷、干姜、桑椹、银耳、大枣直接投入多功能提取罐中,罐内物料浸没水中浸泡4~6h,投入分装蒸煮袋的丁香;
3、蒸煮:在提取罐内蒸煮已投物料,温度达到100℃时开始计时40~50min,并由经过筛选和培训的评价员对蒸煮液进行感官评定,出现浓郁的白芷味,且伴有微酸、微辣、微苦,液体颜色为红棕色,此时停止蒸煮;
优选的,所述蒸煮时间为每次蒸煮重量≤0.5t,40~45min;每次蒸煮重量>0.5t,45~50min;
4、收集:使用过滤器过滤,收集滤液备用;
5、发酵液培养基制备:将步骤4所得料液搅拌均匀打入发酵罐中,投入低聚异麦芽糖、胶原蛋白肽,搅拌均匀后进行灭菌及冷却处理;
优选的,培养基制备过程中不调节pH值,115℃灭菌30min,物料通过夹套升温加热,严禁向物料内通蒸汽;
6、菌种活化:培养基pH值调节为6.60,将培养基灭菌后冷却,接种植物乳杆菌HCS03-001、罗伊氏乳杆菌HCS02-001、鼠李糖乳杆菌HCS01-013(质量比为18:1:1),接种量为0.06%,37℃培养16±1h;
优选的,培养基配方为水93.743g/L、无水葡萄糖3.0g/L、酵母浸出物1.5g/L、酵母蛋白胨1.0g/L、L-苹果酸0.3g/L、一水柠檬酸0.2g/L、磷酸二氢钾0.2g/L、氯化钙0.05g/L、硫酸镁0.006g/L、硫酸锰0.001g/L;
7、发酵:将活化的菌种接种到步骤5所得的培养基上投入发酵罐中,接种量为5%,37℃下发酵72±2h;
优选的,发酵罐参数:装液量为50~75%;搅动培养,搅拌速度为80-100r/min;通气量为0,初始气压0.02-0.03MPa;
8、发酵液离心:将步骤7中所得发酵液进行离心,离心机转速为13000r/min(频率43.95Hz),上料速度1.5~2.0L/min;离心后产品外观保持红棕色,溶液均一,澄清,无杂质。
9、超高温瞬时灭菌:将步骤8中得到的发酵液灭菌,要求温度131℃,时间15s,料液出口温度<30℃;以未灭菌的样品作为参考,颜色、滋气味及状态无明显差别即为杀菌结束。
10、无菌灌装:将步骤9中得到的料液冷却至室温(≤25℃)进行灌装;
11、巴氏杀菌:将步骤10的罐装产品进行巴氏杀菌,得具有美容养颜和润肠通便功能葡萄籽白芷桑椹酵素。

实施例2一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素有益成分分析
一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素的有益成分如表1所示。酵素中的有机酸具有具有抑菌、利胆、消炎、降血糖、抗氧化以及调节机体免疫等作用,能够软化血管、促进钙、铁元素的吸收,同时可刺激消化腺的分泌活动,帮助胃液消化脂肪,有增进食欲、帮助消化吸收及止渴解暑的功能,具有预防疾病和促进新陈代谢作用。游离氨基酸具有消除疲劳、保持精力旺盛、改善亚健康状态等功效,改善睡眠质量。多酚类化合物具有很强的抗氧化性能,可有效清除体内自由基,且含有抗凝因子,可以改善血液循环,对免疫***有一定的保护作用。SOD酶在生物体内的水平高低意味着衰老的直观指标,具有特殊的生理活性,是生物体内清除自由基的首要物质,具有抗衰老、免疫调节、调节血脂、抗辐射、美容等功能。

实施例3一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素功能小鼠试验
试验经口灌胃给予造模药物盐酸洛哌丁胺,建立小鼠便秘模型。给便秘小鼠饲喂不同剂量的葡萄籽白芷桑椹酵素,考察试验动物6h内排黑便粒数与首次排黑便时间。试验结果见表2,与空白对照组相比,模型对照组的6h内排黑便粒数显著降低(P<0.05),首次排黑便时间显著增加(P<0.05),说明小鼠便秘模型构建成功。经过服用受试物后,1.5和3.0mL/kg剂量组的6h排黑便粒数显著高于模型对照组(P<0.05),首次排黑便时间显著短于模型对照组(P<0.05),说明葡萄籽白芷桑椹酵素对盐酸洛哌丁胺的便秘模型有较好的排便效果。

试验筛选53人符合试验条件皮肤存在问题的成年人,作为试验评估样本,其中4人因中途退出,剩余可评估样本共计49人,具有皮肤暗淡、色斑、面部细纹等患者作为试验对象,详见表3。

一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素产品进行了4周人体功效试验,服用本发明酵素产品每日一次,每次20mL,记录试验对象皮肤暗淡、色斑、面部细纹改善情况,具体结果见表4。

试验结果如表4所示,食用酵素样品后面部问题得到改善,包括肤质改善、气色提升、皮肤光滑度改善、皮肤弹性改善均有所缓解,且效果明显。皮肤光滑度改善有效率最高,达到95.92%,说明试验酵素能够明显改善皮肤色泽,有效提升气色。
Claims (9)
1.一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素,其特征在于,包括如下原料:水85-95g/L,玫瑰茄1.0-2.0g/L,桑椹0.5-1.5g/L,白芷0.5-1.5g/L,葡萄籽0.5-1.5g/L,干姜0.2-0.4g/L,银耳0.05-0.3g/L,大枣0.05-0.3g/L,丁香0.05-0.3g/L,沙棘0.05-0.3g/L,茯苓0.05-0.3g/L,白果0.05-0.3g/L,低聚异麦芽糖1.0-3.0g/L,胶原蛋白肽0.001-0.002g/L。
2.根据权利要求1所述的一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素,其特征在于,包括如下原料:水92.599g/L、玫瑰茄1.5g/L,桑椹1.0g/L,白芷1.0g/L,葡萄籽1.0g/L,干姜0.3g/L,银耳0.1g/L,大枣0.1g/L,丁香0.1g/L,沙棘0.1g/L,茯苓0.1g/L,白果0.1g/L、低聚异麦芽糖2.0g/L、胶原蛋白肽0.001g/L。
3.权利要求1所述的一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素的制备方法,其特征在于,包括如下步骤:
1)原料预处理:将葡萄籽破壳粉碎,茯苓粉碎至10-20目;白果去壳,白果仁粉碎至10-20目;
2)按比例称取原料,将玫瑰茄、葡萄籽、沙棘、茯苓、白果平均分装至蒸煮袋中投入提取罐,白芷、干姜、桑椹、银耳、大枣直接投入多功能提取罐中,罐内物料浸没水中浸泡4~6h,投入分装蒸煮袋的丁香;
3)蒸煮:在提取罐内蒸煮已投物料进行蒸煮,出现浓郁的白芷味,且伴有微酸、微辣、微苦,液体颜色为红棕色,此时停止蒸煮;
4)收集:过滤,收集滤液备用;
5)发酵液培养基制备:将步骤4)所得料液搅拌均匀打入发酵罐中,投入低聚异麦芽糖、胶原蛋白肽,搅拌均匀后进行灭菌及冷却处理;
6)菌种活化:培养基pH值调节为6.60,将培养基灭菌后冷却,接种植物乳杆菌HCS03-001、罗伊氏乳杆菌HCS02-001、鼠李糖乳杆菌HCS01-013,37℃条件下培养16±1h;
7)发酵:将活化的菌种接种到步骤5)所得的培养基上投入发酵罐中,接种量为5%进行发酵。
8)发酵液离心:将步骤7)中所得发酵液进行离心,离心后产品外观保持红棕色,溶液均一,澄清,无杂质。
9)超高温瞬时灭菌:将步骤8)中得到的发酵液灭菌,要求温度131℃,时间15s,料液出口温度<30℃;以未灭菌的样品作为参考,颜色、滋气味及状态无明显差别,检测无杂菌即为杀菌结束。
10)无菌灌装:将步骤9中得到的料液冷却至室温进行灌装;
11)巴氏杀菌:将步骤10)的罐装产品进行巴氏杀菌,得具有美容养颜和润肠通便功能葡萄籽白芷桑椹酵素。
4.根据权利要求3所述的一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素的制备方法,其特征在于,步骤3)中,所述的蒸煮为温度达到100℃时开始计时40~50min。
5.根据权利要求3所述的一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素的制备方法,其特征在于,步骤5)中,所述的灭菌是115℃灭菌30min。
6.根据权利要求3所述的一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素的制备方法,其特征在于,步骤6)中,所述的培养基为水90~95g/L、无水葡萄糖2.0~4.0g/L、酵母浸出物1.0~2.0g/L、酵母蛋白胨0.5~1.5g/L、L-苹果酸0.2~0.4g/L、一水柠檬酸0.1~0.3g/L、磷酸二氢钾0.1~0.3g/L、氯化钙0.04~0.06g/L、硫酸镁0.005~0.007g/L、硫酸锰0.001~0.002g/L。
7.根据权利要求3所述的一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素的制备方法,其特征在于,步骤7)中,发酵时,发酵罐参数:装液量为50~75%;搅动培养,搅拌速度为80~100r/min;通气量为0,初始气压0.02-0.03MPa。
8.根据权利要求3所述的一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素的制备方法,其特征在于,步骤7)中,所述的发酵为37℃下培养72±2h。
9.根据权利要求3所述的一种具有美容养颜和润肠通便功能的葡萄籽白芷桑椹酵素的制备方法,其特征在于,步骤8)中,离心时,离心机转速为13000r/min,频率为43.95Hz,上料速度1.5~2.0L/min。
