本发明公开了一株贝莱斯芽胞杆菌(Bacillus siamensis)菌株HYNM‑1,己于2017年10月12日保藏于中国普通微生物中心,保藏编号为CGMCC No.14804。该菌株具有抑制马铃薯疮痂病能力。

一株贝莱斯芽胞杆菌及其应用
技术领域
本发明属于生物工程技术领域,尤其涉及贝莱斯芽胞杆菌(Bacillusvelezensis)及其用于制备防治马铃薯疮痂病菌剂中的应用。
背景技术
马铃薯疮痂病,是一种危害马铃署块茎植物病害。表现为块茎表面出现近圆形至不定形木栓化疮痂状淡褐色病斑或斑块,手摸质感粗糙。一般分为两种发病症状,分别是网纹状病斑和裂口状病斑。通常病斑虽然仅限于皮层,但被害薯块质量和产量仍可降低,不耐贮藏,且病薯外观不雅,商品品级大为下降,招致一定的经济损失。以前在我国境内由于发病率不高,为害面积不大,所以没有引起人们的重视。但近些年来,在内蒙、河北、山东等地区的发病面积越来越大,引起的经济损失也非常可观。因此,迫切需要对这种病害进行科学防治。
根据人们长期对病原微生物的分离鉴定研究,已经确定引起马铃薯疮痂病的病原菌主要为疮痂链霉菌等。并且这些致病菌属于条件致病菌,即在土壤环境偏碱性条件下容易生长和引起马铃薯病害,而在偏酸性土壤环境中则很少出现。
防治方法主要有以下几种:(1)、选用无病种薯;(2)、多施有机肥或绿肥,可部分抑制发病;(3)、与葫芦科、豆科、百合科蔬菜进行5年以上轮作;(4)选择保水好的菜地种植,结薯期遇干旱应及时浇水;(5)、化学预防:一些化学农药可以用来防治疮痂病;(6)生物防治法:筛选对疮痂病病原菌有良好抑制效果且定植能力强的微生物,在生产种薯过程中及农田种植过程中施用,达到抑制病原菌生长的效果。
发明内容
本发明提供了能有效防治马铃薯疮痂病的贝莱斯芽胞杆菌(Bacillusvelezensis)菌株HYNM-1。
本发明所提供的贝莱斯芽胞杆菌(Bacillus velezensis)菌株HYNM-1,已于2017年10月12日保藏在中国微生物菌种保藏管理委员会普通微生物中心(简称为:CGMCC),保藏号为:CGMCC No.14804,保藏单位地址为:北京市朝阳区北辰西路1号院3号。

具体实施方式:
菌种从内蒙马铃薯疮痂病土采集土样,分离鉴定而得。
菌落形态:马铃薯培养基上培养2天菌落中间有凸起,白色,表面褶皱不规则无光泽,边缘不整齐;菌株的形状为:菌株呈杆状,具有鞭毛,宽度为0.76-0.94μm,长度为1.76-2.94μm,好氧,产芽孢,革兰氏染色阳性。
细菌菌株鉴定方法:16srRNA基因序列分析,经鉴定为贝莱斯芽胞杆菌(Bacillusvelezensis)。
细菌培养方法:
贝莱斯芽胞杆菌在马铃薯固体培养基上活化3次。然后取1环于50ml马铃薯液体培养基,28℃,摇床培养2天。发酵液经12,000r/min离心1min钟后,单独收集上清,沉淀悬浮于同体积的无菌水中,制备细胞悬液。同时菌液与上清液也一同用于试验。
菌种用途和效果:
疮痂病拮抗菌筛选材料与方法:试验病原为疮痂病链霉菌(Streptomycesscabiei)4.1765.土豆汁培养基(g/L):马铃薯浸取液1L,葡萄糖20g,KH2PO4 3g,MgSO4.7H2O1.5g,维生素B1微量。pH6.固体培养基加入15g琼脂。
病原菌培养方法:病原菌在马铃薯培养基上培养4-5天,28℃;用0.2ml无菌水刮取孢子和菌丝体,制成一定浓度的病原体悬液;要求100ul体积涂布平板时菌丝生长后可以铺满平板表面。
抑菌试验方法:100ul病原菌涂布于平板上(PDA),在平板局部点接10ul细菌菌液、上清、和细胞悬浮液。28℃培养。3天以后观察。通过对疮痂病链霉菌(Streptomycesscabiei)4.1765试验,发现细菌菌液和细胞悬浮液都能有效抑制这些病原微生物生长,上清没有抑制效果。
结果:细菌对病原菌有效果(图1,图2)。
