技术领域

背景技术
化学疗法仍然是许多癌症类型(包括黑色素瘤)的全身治疗的主要方法。大部分化疗剂对于肿瘤细胞仅具有轻微的选择性,并且对健康增殖细胞的毒性会很高(Allen TM.(2002)Cancer 2:750-763),常常需要减小剂量甚至中止治疗。理论上,克服化疗毒性问题以及提高药物疗效的一种途径是利用对于由癌细胞选择性表达(或过表达)的蛋白质特异性的抗体使化疗药物靶向肿瘤,以便将靶向药物吸引到肿瘤,由此改变化学治疗的生物分布并且使更多药物到达肿瘤从而减小对健康组织的影响。虽然有30年的研究,但在治疗中特异性靶向却很少成功。

发明内容

术语“冻干保护剂”包括在干燥或“脱水”过程(初次干燥循环和二次干燥循环)期间为蛋白质提供稳定性的试剂,推测这是通过提供无定型玻璃状基质并且通过经由氢键合与蛋白质结合从而代替在干燥过程期间被去除的水分子。这有助于保持蛋白质构象、使冷冻干燥循环期间的蛋白质降解最小化,并且改善长期产品。例子包括多元醇类或糖类(如庶糖和海藻糖)。
术语“药物制剂”是指这样的制剂,所述制剂处于允许活性成分是有效的形式中,并且不含有对于将接受制剂的受试对象有毒的另外组分。

“复原时间(reconstitution time)”是将冻干制剂再水合成为溶液所需的时间。

本文中所采用的术语“表位”是指被结合剂(例如抗体)所识别的抗原的部分。表位包括但不限于:使与蛋白质(例如,抗体)或配体的特异性相互作用成为可能的短氨基酸序列或肽(任选地被糖基化或者以另外方式被修饰)。例如,表位可以是结合剂的抗原结合部位所附着的分子的一部分。

如本文中所采用的术语“低聚物”或“低聚的”或“低聚化的”是指包含两种或更多种单体的低聚物。
融合蛋白是生物基因工程化的多肽,这些多肽将蛋白质或其片段(例如,抗体的可结晶片段(Fc)结构域)与另外的生物活性剂(例如,蛋白结构域、肽、或核酸或肽适体)连接而生成具有所需结构-功能特性和显著治疗潜力的分子。由于具有有利的特性(如效应子功能的征集和增加的血浆半衰期),γ免疫球蛋白(IgG)同种型常常被用作生成Fc融合蛋白的基础。考虑到可以被用作融合伴侣的一系列适体(肽和核酸两者),融合蛋白具有许多生物学和药学应用。
术语“无毒的纳米颗粒”(NTP)或“NTP复合物”是指包含载体蛋白(例如白蛋白)和毒性低于紫杉醇的紫杉醇衍生物的纳米颗粒。任选地,NTP或NTP复合物包含结合剂(例如抗体)和/或治疗剂(例如化疗剂)。
短语“毒性低于紫杉醇”是指与紫杉醇相比显示对细胞(包括癌细胞和正常细胞)降低的毒性(例如减少对所述细胞的杀伤)的紫杉醇衍生物。例如,紫杉醇的Meerwein产物(20-乙酰氧基-4-脱乙酰基-5-表-20,O-开环紫杉醇)具有显著降低的毒性,这有可能是由于紫杉醇的C-4,C-5氧杂环丁烷环的断裂所致。
术语“降低毒性的纳米颗粒”(RTP)或“RTP复合物”是指包含载体蛋白(例如白蛋白)和与相比减少量的紫杉醇(例如约9:1的白蛋白:紫杉醇比)的纳米颗粒。任选地,RTP或RTP复合物包含结合剂(例如抗体)和/或治疗剂(例如化疗剂)。

概述
用于可注射悬浮液的(用于可注射悬浮液的紫杉醇蛋白质结合颗粒)是具有大约130纳米的平均粒径的紫杉醇的白蛋白结合型形式。紫杉醇可以以非晶体、无定形的状态存在于颗粒中。可以被提供为例如冻干粉,用于在静脉内输注前用2.0mL的0.9%氯化钠注射液(USP)进行复原。一次性使用小瓶含有100mg的紫杉醇和大约900mg的人白蛋白。每毫升(mL)的经复原的悬浮物含有5mg紫杉醇。
纳米颗粒是稳定化的白蛋白结合型紫杉醇。紫杉醇与白蛋白的结合可以利用例如疏水性相互作用而实现。在一些实施例中,在130nm的平均尺寸,纳米颗粒是稳定的。也已知的是,其他疏水性药物(例如多西他赛、雷帕霉素)可以类似地通过与白蛋白结合而形成稳定化的纳米颗粒。
为了使常规ADC有效,关键的是,连接体足够稳定而不在体循环中解离,但是允许在肿瘤部位处进行充分的药物释放。Alley,S.C.等(2008)Bioconjug Chem 19:759-765。这已经被证明是开发有效药物缀合物中的主要障碍(Julien,D.C.等(2011)MAbs 3:467-478;Alley,S.C.等(2008)Bioconjug Chem 19:759-765);因此,纳米免疫缀合物的吸引人的特征是不需要生化连接体。
当前ADC的另一个缺点是在人类肿瘤中尚未充分证明药物向肿瘤中的渗透更高。在小鼠模型中对ADC的早期测试表明,使用抗体靶向肿瘤会使得肿瘤中活性剂的浓度更高(Deguchi,T.等(1986)Cancer Res 46:3751-3755);然而,这与人类疾病的治疗无关,这可能是因为人类肿瘤比小鼠肿瘤在渗透性方面更加异质。Jain,R.K.等(2010)Nat Rev ClinOncol 7:653-664。此外,纳米颗粒的尺寸对于从血管***外渗到肿瘤中是关键的。在使用人类结肠腺癌异种移植模型的小鼠研究中,血管孔隙对于高达400nm的脂质体是可渗透的。Yuan,F.等(1995)Cancer Res 55:3752-3756。关于肿瘤孔径和渗透性的另一项研究证实,这两种特征都取决于肿瘤位置和生长状态,其中消退中的肿瘤和颅内肿瘤对于小于200nm的颗粒是可渗透的。Hobbs,S.K.等(1998)Proc Natl Acad Sci U S A 95:4607-4612。本文所述的纳米免疫缀合物通过以下事实克服了这个问题,即在完整时小于200nm的大复合物在体循环中部分解离成能够容易地渗透肿瘤组织的更小的功能单元。此外,一旦缀合物到达肿瘤部位,就可以释放更小的毒性有效负载并且只有毒性部分而不是整个缀合物需要被肿瘤细胞摄取。
抗体(即)包被的含有治疗剂(即)的白蛋白纳米颗粒的出现已经产生了将两种或更多种治疗剂定向递送到体内预定部位的新范例。参见PCT专利公开号WO 2012/154861和WO 2014/055415,它们中的每一件以引用的方式整体并入本文。
然而,考虑一些癌症不能通过抗体复合物中的紫杉醇进行治疗,和/或其他治疗剂(例如抗癌化疗剂)将更有效地治疗某些癌症。本文中公开了包含载体蛋白(例如白蛋白)和减少量的紫杉醇(例如与相比)的毒性降低的纳米颗粒(RTP);或者包含载体蛋白(例如白蛋白)和紫杉醇衍生物(比紫杉醇毒性小)的无毒纳米颗粒(NTP)。纳米颗粒还可包含治疗剂和/或结合剂(例如抗体)。
纳米颗粒和纳米颗粒组合物
在阅读本公开后,对于本领域技术人员显而易见的是,本公开涉及纳米颗粒和纳米颗粒的组合物,所述纳米颗粒含有载体蛋白(例如白蛋白)和紫杉醇衍生物(例如毒性低于紫杉醇的衍生物)或减少量的紫杉醇(例如与治疗量相比和/或与中的量相比)。在一些实施方案中,纳米颗粒也含有结合剂和/或治疗剂。在一些实施方案中,纳米颗粒或纳米颗粒组合物被冻干。
本发明还部分地基于纳米颗粒的形成,所述纳米颗粒包含载体蛋白及紫杉醇(与存在于中的量相比更低)或紫杉醇衍生物(其与紫杉醇相比(例如对细胞)毒性更低),任选地含有结合剂和/或治疗剂。不受理论所束缚,认为这类纳米颗粒提供对肿瘤的靶向治疗同时使对患者的毒性最小化,并且可以用于治疗通常对紫杉醇不敏感和/或更好地响应不同治疗剂的肿瘤。
在一些实施方案中,所述载体蛋白可以是白蛋白、明胶、弹性蛋白(包括弹性蛋白原)或弹性蛋白衍生多肽(例如α-弹性蛋白和弹性蛋白样多肽(ELP))、麦醇溶蛋白、豆球蛋白、玉米醇溶蛋白、大豆蛋白(例如大豆蛋白分离物(SPI))、乳蛋白(例如β-乳球蛋白(BLG)和酪蛋白)或乳清蛋白(例如乳清蛋白浓缩物(WPC)和乳清蛋白分离物(WPI))。在优选的实施方案中,所述载体蛋白是白蛋白。在优选的实施方案中,所述白蛋白是蛋清(卵清蛋白)、牛血清白蛋白(BSA)等。在甚至更优选的实施方案中,所述载体蛋白是人血清白蛋白(HSA)。在一些实施方案中,所述载体蛋白是重组蛋白(例如重组HSA)。在一些实施方案中,所述载体蛋白是由美国食品和药品管理局(United States Food and Drug Administration,FDA)批准的公认安全(GRAS)的赋形剂。
在一些实施方案中,所述结合剂是选自曲妥珠单抗-美坦新偶联物、阿仑单抗、阿特珠单抗、贝伐珠单抗、西妥昔单抗、地诺单抗、地努图希单抗、伊匹单抗、纳武单抗、奥滨尤妥珠单抗、奥法木单抗、帕尼单抗、派姆单抗、帕妥珠单抗、利妥昔单抗、阿维鲁单抗或德瓦鲁单抗、皮地利珠单抗、BMS 936559和曲妥珠单抗的抗体。在一些实施方案中,抗体选自表1中所列出的抗体。在一些实施方案中,所述抗体是在全部或部分纳米颗粒表面上基本上为单层的抗体。

在一个方面,所述组合物内少于约0.01%的纳米颗粒具有大于200nm、大于300nm、大于400nm、大于500nm、大于600nm、大于700nm或大于800nm的粒度。在一个方面,所述组合物内少于约0.001%的纳米颗粒具有大于200nm、大于300nm、大于400nm、大于500nm、大于600nm、大于700nm或大于800nm的粒度。在一个优选的实施方案中,所述组合物内少于约0.01%的纳米颗粒具有大于800nm的粒度。在一个更优选的实施方案中,所述组合物内少于约0.001%的纳米颗粒具有大于800nm的粒度。
在一个优选的方面,本文所述的尺寸和尺寸范围与复原的冻干纳米颗粒组合物的粒度有关。也就是说,在将冻干纳米颗粒重悬在水溶液(例如水、其它药学上可接受的赋形剂、缓冲剂等)中之后,粒径或平均粒径在本文所述的范围内。
在一个方面,至少约50%、60%、70%、80%、90%、95%、96%、97%、98%、99%、99.5%或99.9%的纳米颗粒作为单个纳米颗粒存在于复原的组合物中。也就是说,少于约50%、40%、30%等的纳米颗粒二聚化或多聚化(低聚化)。
在一些实施方案中,所述组合物中的纳米颗粒具有以数量计少于20%的二聚化、以数量计少于10%的二聚化并且优选地少于5%的二聚化。
在一些实施方案中,通过调整载体蛋白与紫杉醇或紫杉醇衍生物的量(例如比率)、和/或载体蛋白-紫杉醇(或紫杉醇衍生物)纳米颗粒与结合剂的比率,可以控制纳米颗粒的尺寸。纳米颗粒的尺寸和尺寸分布也是重要的。本发明的纳米颗粒可以根据它们的尺寸而表现不同。在大尺寸下,团聚可能会阻塞血管。因此,纳米颗粒的团聚可以影响组合物的性能和安全性。另一方面,更大的颗粒在某些条件下(例如不静脉内施用时)可能更具治疗性。
由于所述纳米颗粒被用于治疗中,因此提出了进一步的挑战。
虽然纳米颗粒中组分的重排可以通过组分之间的共价键来减轻,但是这些共价键对纳米颗粒在癌症治疗中的治疗用途提出了挑战。结合剂、载体蛋白以及治疗剂通常在肿瘤中的不同位置处并且经由不同的机制起作用。非共价键容许纳米颗粒的组分在肿瘤处解离。因此,虽然共价键对于冻干可能是有利的,但是它对于治疗用途可能是不利的。
纳米颗粒的尺寸和尺寸分布也是重要的。纳米颗粒可以根据它们的尺寸而表现不同。在大尺寸下,纳米颗粒或颗粒的团聚可能会阻塞血管,其中任何一种都可能影响组合物的性能和安全性。
紫杉醇衍生物
在本发明中所使用的紫杉醇衍生物优选地比紫杉醇的毒性低。也就是说,与紫杉醇相比,紫杉醇衍生物对细胞的毒性较小(例如,也不杀伤癌症和/或正常细胞和/或也不抑制细胞增殖)。紫杉醇衍生物可包括紫杉醇的任何衍生物或前体。例子包括但不限于:20-乙酰氧基-4-脱乙酰基-5-表-20,O-开环紫杉醇(紫杉醇的Meerwein产物)、或巴卡亭III((2β,5α,7α,10α,13β)-4,10-二乙酰基-1,7,13-三羟基-9-氧代-5,20环氧紫杉-11-烯-2-基苯甲酸酯)。
例如,与紫杉醇相比,大部分的C-13简化紫杉醇衍生物和具有不同立体化学的衍生物显示降低的活性。2'-羟基和3'-苯甲酰氨基对于生物活性不是必不可少的,但对于牢固的微管结合和细胞毒性却是重要的。醚,如烷基醚(例如甲基醚)和烷基硅烷基醚(例如叔丁基二甲基硅烷基醚),在2'-羟基处的形成降低了细胞毒性。2'-羟基(例如2'-乙酰基紫杉醇)的酰化也导致活性的损失。还发现2'-羟基被卤素(例如氟)取代显著地降低细胞毒性。6α-羟基-7-表-紫杉醇的四醋酸铅氧化导致C-去甲紫杉醇(C-nor-paclitaxel)和C-开环紫杉醇衍生物(C-seco-paclitaxel)的形成。6α-羟基-7-表-紫杉醇衍生物的四丙基过钌酸铵(TPAP)氧化导致产生6-甲酰基-C-去甲紫杉醇衍生物。6α-O-三氟甲磺酰基-7-表-紫杉醇衍生物与DMAP的反应获得20-O-乙酰基-4-脱乙酰基-5,6-脱氢-6-甲酰基-C-去甲紫杉醇衍生物。C-去甲紫杉醇类似物的活性低于紫杉醇。因此,该紫杉醇衍生物可包括在这些部位中的任意一个或多个部位处的修饰,或者利用这些方法中的一种或多种方法而形成。参见例如Liang等人Tetrahedron,53(10):3441-3456(1997);US 5,756,301、US 5,981,777;以上各文献的全部内容以参考的方式并入本文中。
制备纳米颗粒和纳米颗粒复合物的方法
在一些方面,本发明涉及制备本文中所描述的纳米颗粒组合物的方法。在一些实施方案中,本公开涉及制备纳米颗粒组合物的方法,其中所述方法包括:在将允许所需纳米颗粒形成的条件和各组分的比率下,使载体蛋白与紫杉醇或紫杉醇衍生物接触。在一些实施方案中,所述方法还涉及在形成纳米颗粒复合物的条件下,使纳米颗粒与结合剂和/或治疗剂接触。
在一些实施方案中,通过将载体蛋白与紫杉醇或紫杉醇衍生物混合并且在高压下进行均质化以形成稳定的纳米颗粒而制备纳米颗粒。用于使白蛋白和紫杉烷(例如,紫杉醇)均质化的非限制性方法可以参见例如实施例1,以及美国专利号5,916,596、6,506,405和6,537,579,以上各专利的全部内容以参考的方式并入本文中。

制备毒性降低的纳米颗粒

应当指出的是,当使用例如具有约1升溶液的典型的静脉注射袋时,与在1mL中使用的量相比,将需要使用1000×量的载体蛋白/载体蛋白-治疗剂和抗体。因此,不能在标准静脉注射袋中形成本发明的纳米颗粒。此外,当将组分以本发明的治疗量添加到标准静脉注射袋中时,所述组分不会自组装形成纳米颗粒。

在一个实施方案中,本文中所描述的纳米颗粒组合物不包含结合剂。冻干
虽然包含单一来源蛋白质的蛋白质组合物通常以冻干形式(其中它们表现出相当长的保存期的)储存,但是这样的冻干组合物不含通过疏水性-疏水性相互作用整合在一起的两种不同蛋白质的自组装纳米颗粒。此外,纳米颗粒构型(其中结合剂的大部分结合部分暴露在纳米颗粒的表面上)使其自身易于因原本将被认为是温和的条件而发生移位或重构。举例来说,在冻干期间,蛋白质上的离子电荷被脱水,从而暴露下面的电荷。暴露的电荷允许两种蛋白质之间的电荷-电荷相互作用,这可以改变每一种蛋白质对另一种的结合亲和力。此外,在溶剂(例如水)被去除时,纳米颗粒的浓度显著增加。这样的增加的纳米颗粒浓度可能引起不可逆的低聚化。低聚化是蛋白质的已知特性,与单体形式相比,其降低了低聚物的生物学特性并且增加颗粒的尺寸,有时会增加到超过1微米。
另一方面,对于其中需要至少3个月的保存期并且大于6个月或9个月的保存期是优选的临床用途和/或商业用途,需要稳定形式的纳米颗粒组合物。这样的稳定组合物必须易于静脉内注射,必须在静脉内注射后保持它的自组装形式以将所述纳米颗粒引导到体内预定部位,必须具有小于1微米的最大尺寸以在递送到血流中时避免任何缺血事件,并且最终必须与用于注射的含水组合物相容。
冻干或冷冻干燥从组合物中去除水。在所述过程中,首先将待干燥的材料冷冻,然后通过在真空环境中升华来去除冰或冷冻溶剂。在冻干前制剂中可以包括赋形剂以增强在冷冻干燥过程期间的稳定性和/或提高冻干产品在储存时的稳定性。Pikal,M.Biopharm.3(9)26-30(1990)以及Arakawa等,Pharm.Res.8(3):285-291(1991)。
虽然可将蛋白质冻干,但是冻干和复原的过程可能影响蛋白质的特性。由于蛋白质比传统的有机药物和无机药物更大并且更复杂(即除了复杂的三维结构之外,还具有多个官能团),因此这样的蛋白质的制剂带来了特殊的问题。为了使蛋白质保持生物活性,制剂必须保持蛋白质氨基酸的至少核心序列的构象完整性是完整无缺的,同时保护蛋白质的多个官能团免于降解。蛋白质的降解途径可以涉及化学不稳定性(即涉及通过键形成或裂解修饰蛋白质,从而产生新的化学实体的任何过程)或物理不稳定性(即蛋白质的更高阶结构的变化)。化学不稳定性可以由脱酰胺、外消旋化、水解、氧化、β消除或二硫键交换引起。物理不稳定性可以由例如变性、聚集、沉淀或吸附引起。三种最常见的蛋白质降解途径是蛋白质聚集、脱酰胺以及氧化。Cleland等,Critical Reviews in Therapeutic DrugCarrier Systems 10(4):307-377(1993)。
最后,在冻干过程起辅助作用的冷冻保护剂和试剂对于治疗用途而言必须是安全、可耐受的。
本发明的冻干组合物是通过标准冻干技术在存在或不存在稳定剂、缓冲剂等的情况下制备的。令人惊讶的是,这些条件不会改变纳米颗粒的相对脆弱的结构。此外,最好,这些纳米颗粒在冻干后保持它们的尺寸分布,并且更重要的是,可以以如同新鲜制备的基本上相同的形式和比率复原用于体内施用(例如静脉内递送)。

制剂
在一个方面,纳米颗粒组合物被配制用于全身递送,例如静脉内施用。
在一个方面,所述纳米颗粒组合物被配制用于直接注射到肿瘤中。直接注射包括注射到肿瘤部位中或肿瘤部位附近、灌注到肿瘤中等。由于纳米颗粒组合物不被全身施用,因此被配制用于直接注射到肿瘤中的纳米颗粒组合物可以包括任何平均粒径。不受理论所束缚,认为更大的颗粒(例如大于500nm、大于1μm等)更可能被固定在肿瘤内,从而提供被认为是更好的有益作用。
在另一个方面,本文提供了一种组合物,所述组合物包含本文提供的化合物和至少一种药学上可接受的赋形剂。

必要时,本发明的组合物可以存在于包装或分配器装置中,其容纳含有活性成分的一个或多个单位剂型。这样的包装或装置可以例如包括金属箔或塑料薄膜,例如泡罩包装(blister pack),或玻璃和橡胶塞,例如在小瓶中。所述包装或分配器装置可以附有用于施用的说明书。还可以制备包含配制于相容的药物载体中的本发明的化合物的组合物,将其放置在适当的容器中,并且贴标签用于所示病症的治疗。
治疗方法
如本文所述的纳米颗粒组合物可用于在哺乳动物中治疗癌细胞和/或肿瘤。在一个优选的实施方案中,所述哺乳动物是人类(即人类患者)。优选的是,在施用之前将冻干的纳米颗粒组合物复原(悬浮在水性赋形剂中)。
在一个方面,提供了一种用于治疗癌细胞的方法,所述方法包括使所述细胞与有效量的如本文所述的纳米颗粒组合物接触以治疗所述癌细胞。治疗癌细胞包括但不限于减少增殖、杀伤细胞、防止细胞转移等。
在一个方面,提供一种用于对需进行治疗的患者中的肿瘤进行治疗的方法,该方法包括向患者施用治疗有效量的本文中所描述的纳米颗粒组合物以***。在其中纳米颗粒包含包括抗原结合结合域的结合剂的一个实施方案中,该方法包括对患有表达抗原的癌症的患者筛选选择。在一个实施方案中,肿瘤的尺寸减小。在一个实施方案中,肿瘤尺寸在治疗期间和/或之后至少一段时间内不增加(即进展)。

药剂(例如本发明的化合物)的有效量或治疗有效量或剂量指的是引起受试对象的症状改善或存活期延长的药剂或化合物的量。这样的分子的毒性和治疗功效可以通过标准药物程序在细胞培养物或实验动物中确定,例如通过确定LD50(对50%的群体致死的剂量)和ED50(在50%的群体中具有治疗有效的剂量)。毒性作用与治疗作用的剂量比是治疗指数,其可以被表示为比率LD50/ED50。表现出高治疗指数的药剂是优选的。

剂量和时间间隔可以单独调整以提供足以实现所期望的作用的活性部分的血浆水平;即最低有效浓度(MEC)。每一种化合物的MEC是不同的,但是可以根据例如体外数据和动物实验来估计。实现MEC所需的剂量将取决于个体特征和施用途径。在局部施用或选择性摄取的情况下,药物的有效局部浓度可能与血浆浓度无关。
在相关的实施方案中,治疗包括在施用纳米颗粒之前施用靶向结合剂。在一个实施方案中,在纳米颗粒施用前的约6和48小时之间、或者12和48小时之间,施用靶向结合剂。在另一个实施方案中,在纳米颗粒施用前的6和12小时之间施用靶向结合剂。在再一个实施方案中,在纳米颗粒施用前的2和8小时之间施用靶向结合剂。在其他的实施方案中,在纳米颗粒施用前的一周施用靶向结合剂。例如,在包含贝伐珠单抗或其他VEGF-结合抗体的纳米颗粒复合物施用之前的24小时,施用一定剂量的BEV。在另一个实施例中,在施用利妥昔单抗前,在含有利妥昔单抗的纳米颗粒复合物施用前。在纳米颗粒之前施用的结合剂可以以亚治疗的剂量(例如通常被认为具有治疗效果的量的1/2、1/10或1/20)被施用。因此,在人类中,用BEV预治疗可包括施用1mg/kg BEV,该剂量是常用剂量的1/10,接着施用纳米颗粒或纳米颗粒复合物。
实施例
利用包含作为核心的白蛋白结合型紫杉醇或紫杉醇衍生物、和作为抗体的贝伐珠单抗(即)或利妥昔单抗(即)的纳米颗粒来阐释本发明。
本领域技术人员将了解的是,制备和使用实施例的纳米颗粒仅仅是为了说明的目的,并且本公开不受该说明的限制。

实施例1:纳米颗粒制备
使紫杉醇与Meerwein试剂反应,根据由Samaranayake等人在“ModifiedTaxols.Reaction of Taxol with Electrophilic Reagents and Preperation of aRearranged Taxol Derivative with Tubulin Assembly Activity”(J.Org.Chem.1991,56,5114-5119)中所设计的反应方案而发生断裂,该文献的全部内容以参考的方式并入本文中。此反应使C-4,C-5氧杂环丁烷环发生断裂,从而严重地抑制毒性。反应和纯化由Organix Inc.(Woburn,MA)完成。将该反应示于图1中。
然后在高压下使紫杉醇的Meerwein产物(20-乙酰氧基-4-脱乙酰基-5-表-20,O-开环紫杉醇)与白蛋白一起均质化,以形成稳定的无毒纳米颗粒(NTP)。用于使白蛋白和紫衫烷(例如,紫杉醇)均质化的方法可以参见例如美国专利号5,916,596、6,506,405和6,537,579,以上各专利的全部内容以参考的方式并入本文中。

就冻干的纳米颗粒而言,以适当的体积将溶液分入冷冻干燥瓶中从而允许产品充分冷冻干燥。将含有溶液的烧瓶在-80℃放置2-4小时。利用带-85℃冷凝器的台式冷冻干燥机,在<20毫托(mtorr),对烧瓶中的冻结溶液进行冻干。
实施例2:NTP与贝伐珠单抗在体外的结合
为了确定NTP与贝伐珠单抗(BEV)是否相互作用,利用Malvern NanoSight技术,单独地及在与4、6、8和10毫克每毫升(mg/mL)的贝伐珠单抗一起温育30分钟后,对在实施例1中所形成的NTP的尺寸进行了分析。
纳米颗粒直径随着BEV浓度的增加而增大(图2A-图2E),这表明含有BEV的NTP的非共价涂层的增加。利用Malvern NanoSight技术以1:300稀释度测量直径。
实施例3:贝伐珠单抗和利妥昔单抗与无毒纳米颗粒(NTP)结合的亲和力
利用生物膜层干涉技术(Bio Layer Interferometry Technology)测量了贝伐珠单抗(图3A)和利妥昔单抗(图3B)两者与NTP的结合亲和力(Kd)。利用胺偶联使这两种抗体生物素化,然后使其以100μg/mL单独地与BLITZ链霉亲和素探针结合。测定各种浓度的NTP针对探针的结合,其中结合时间和解离时间为120秒。利用BLITZ评估软件执行动力学分析。纳米-皮摩尔亲和力表明利妥昔单抗与NTP之间以及在贝伐珠单抗与NTP之间的强结合。
实施例4:与(ABX)相比在PBS和血清中无毒纳米颗粒(NTP)的稳定性
在PBS中将无抗体NTP和(ABX)溶液分别制备成30μg/mL的紫杉醇的Meerwein产物和紫杉醇。利用Malvern NanoSight技术获得每mL纳米颗粒的基线计数。然后制备两种溶液的一系列稀释溶液,并测量颗粒浓度。
图4A示出了ABX和NTP在盐水中的稳定性。在初始溶液的1:1.33稀释度附近Abraxane纳米颗粒开始分散开,直到它在大约1:2稀释度达到基线PBS水平。NTP随着稀释因子而线性地减小,这表明没有颗粒分散开。
图4B示出了ABX和NTP在血清中的稳定性。在血清中将NTP和ABX溶液分别制备成250μg/mL的紫杉醇的Meerwein产物和紫杉醇。利用Malvern NanoSight技术测量纳米颗粒的初始浓度,随后的测量结果被表示为初始浓度的百分比。在15、30、45、和60分钟后,在血清中测量了各溶液的浓度。
实施例5:利用NTP-利妥昔单抗复合物对Daudi细胞CD20的阻断
如上面或前面所描述地(参见例如PCT公开号WO 2014/055415,该专利公开的全部内容以参考的方式并入本文中),制备了ABX、AR160、无毒纳米颗粒(NTP)和结合利妥昔单抗的NTP。在室温下将纳米颗粒和未标记的利妥昔单抗与CD20+Daudi淋巴瘤细胞共温育达30分钟。将细胞清洗,接着用PE抗人CD20进行染色。将同种型(图5A)和抗CD20(图5B)染色的细胞分别用作阴性对照和阳性对照。
结果表明,AR160(图5D)、加载利妥昔单抗的NT颗粒(NTRit;图5F)和利妥昔单抗(图5G)阻断随后与PE抗人CD20的结合,而单独的ABX(图5C)和NT纳米颗粒(图5E)则不这样做。这表明,在AR160和加载利妥昔单抗的NT颗粒的环境中,利妥昔单抗保留其配体结合特性。
实施例6:含有减少的紫杉醇的白蛋白-紫杉醇纳米颗粒
形成与相比具有减少量的紫杉醇的白蛋白-紫杉醇纳米颗粒。
例如,将小于30mg(例如,1mg至25mg)的紫杉醇溶解于溶剂(例如,二氯甲烷、或者氯仿与乙醇)中。将该溶液添加到27.0mL的人血清白蛋白溶液(1%w/v)中。以低RPM将混合物均质化达5分钟,从而形成粗乳液,然后将其转移到高压均质机中。在9000-40,000psi下执行乳化,同时将乳液循环使用至少5个循环。将所形成的体系转移到旋转蒸发器中,在40℃下在减压(30mm Hg)下快速地去除二氯甲烷,持续20~30分钟。任选地,使分散体经过0.22微米滤膜而进行过滤。
