本发明提供了一种植物油中维生素D3的鉴别前处理方法,其步骤如下:称取植物油‑维生素D3油状物适量,加乙醇适量,剧烈振摇,离心,吸出上层溶液,在冰盐浴条件下放置10min,取出离心,吸出上层溶液,于30~40℃水浴下用氮气流吹干,即得。为植物油中维生素D3的化学鉴别提供了一种简便快速、专属性好、灵敏度高、重现性好、显色明显的前处理方法。

一种植物油中维生素D3的鉴别前处理方法
技术领域
本发明属于药物制剂技术领域,涉及一种植物油中维生素D3的鉴别前处理方法。
背景技术
维生素D3是人体必需的重要维生素,具有非常重要的生理功能,如能提高肌体对钙、磷的吸收,使血浆钙和血浆磷的水平达到饱和程度;能促进生长和骨骼钙化,促进牙齿健全;能通过肠壁增加磷的吸收,并通过肾小管增加磷的再吸收;能维持血液中柠檬酸盐的正常水平;还能防止氨基酸通过肾脏损失等等。维生素D3缺乏不仅会导致佝偻病、骨质疏松等骨骼***疾病,大量临床试验还表明,肺部疾病、代谢性疾病、心血管疾病、免疫***疾病等均与维生素D3缺乏有关。因此,维生素D3对人类健康发展是非常重要的。
维生素D3是一种脂溶性维生素,化学性质不稳定,遇空气等容易发生降解,从而影响产品质量,甚至产生对人体有害的降解产物。因此,常将维生素D3溶入大量的植物油,充分避免与空气接触,从而形成稳定的植物油-维生素D3油状物。
目前,维生素D3的鉴别方法有显色法和高效液相色谱法,显色法相比高效液相色谱法更简便、快速,因此,显色法常作为维生素D3迅速诊断的方法。而对于鉴别溶于植物油中的维生素D3来讲,由于大量植物油的存在,干扰维生素D3的显色,使普通的显色方法显色效果极不明显,高效液相色谱法存在植物油干扰维生素D3色谱峰的分离、速度慢、成本高、极容易毁坏色谱柱等问题。因此,我们对植物油中维生素D3的鉴别前处理方法进行了大量试验摸索,成功发明了一种有效的鉴别前处理方法,弥补了该技术领域的空白,为植物油中维生素D3的鉴别提供了一种简便快速、专属性好、灵敏度高、重现性好、显色明显的前处理方法。
发明内容
本发明提供了一种植物油中维生素D3的鉴别前处理方法。所述的植物油中维生素D3的鉴别前处理方法,步骤如下:称取植物油-维生素D3油状物适量,加乙醇适量,剧烈振摇,离心,吸出上层溶液,在冰盐浴条件下放置10min,取出离心,吸出上层溶液,于30~40℃水浴下用氮气流吹干,即得。
本发明的植物油中维生素D3的鉴别前处理方法的创新点为采用一定比例的乙醇并结合冰盐浴条件将植物油中的维生素D3提取出来,其优势有:1、该方法能去除大量对维生素D3显色鉴别有干扰的植物油,使颜色反应明显,操作简便、重现性好,灵敏度极高;2、所用试剂方便易得、价格低廉、不会污染环境。
具体实施方式
实施例1 称取维生素D3油状物适量,按体积比1:2加乙醇适量,剧烈振摇2min,离心5min(4000r/min),吸出上层溶液,在冰盐浴(约-20℃)条件下放置10min,取出离心5min(4000r/min),吸出上层溶液,于30~40℃水浴下用氮气流吹干,即得。
实施例2 称取维生素D3油状物适量,按体积比1:3加乙醇适量,剧烈振摇2min,离心5min(4000r/min),吸出上层溶液,在冰盐浴(约-10℃)条件下放置10min,取出离心5min(4000r/min),吸出上层溶液,于30~40℃水浴下用氮气流吹干,即得。
实施例3 称取维生素D3油状物适量,按体积比1:3加乙醇适量,剧烈振摇1min,离心5min(4000r/min),吸出上层溶液,在冰盐浴(约-20℃)条件下放置10min,取出离心5min(4000r/min),吸出上层溶液,于30~40℃水浴下用氮气流吹干,即得。
实施例4 称取维生素D3油状物适量,按体积比1:4加乙醇适量,剧烈振摇1min,离心5min(4000r/min),吸出上层溶液,在冰盐浴(约-15℃)条件下放置10min,取出离心5min(4000r/min),吸出上层溶液,于30~40℃水浴下用氮气流吹干,即得。

1.一种植物油中维生素D3的鉴别前处理方法,其步骤如下:称取植物油-维生素D3油状物适量,加乙醇适量,剧烈振摇,离心,吸出上层溶液,在冰盐浴条件下放置10min,取出离心,吸出上层溶液,于30~40℃水浴下用氮气流吹干,即得。
2.如权利要求1所述的植物油中维生素D3的鉴别前处理方法,其特征在于,采用乙醇将植物油-维生素D3油状物中的维生素D3提取出来。
3.如权利要求1所述的植物油中维生素D3的鉴别前处理方法,其特征在于,植物油-维生素D3油状物和乙醇的体积比为1:1~1:4。
4.如权利要求1所述的植物油中维生素D3的鉴别前处理方法,其特征在于,冰盐浴条件为-10℃~-20℃。
