背景技术
随着老龄化社会的来临,在世界的健康生态***中,患病人数将逐渐增加,其中大多数疾病属于慢性病。因此,在世界范围内,健康领域正在发生着新的变化,即由疾病到来时采取治疗手段而逐渐转移至治未病。在营养食品领域也是一样,为患者提供经过科学论证的营养配方,与药品共同辅助疾病治疗,能加快人体技能的恢复,这一理念已经在医疗体系中扮演越来越重要的角色。根据病人病情选用饮食营养、肠内或肠外营养支持、或营养补充剂等方法,使病人获得适宜的营养,是减少并发症和患者死亡的有效办法。从市场情况来看,肠内营养品在欧美等发达国家都很受重视,并且有着非常可观的市场份额。我国的肠内营养品市场份额还处于较低水平,但随着对营养知识的不断认识以及政策性的引导,我国肠内营养食品的消费额也在逐年上升,但与发达国家比还有很大差距。从现有的碳水化合物类肠内营养食品的品种和数量来看,有全营养型的、组件型的等几种类型,品种和数量少,且多靠进口。因此,我国肠内营养食品有着很大的市场潜力。
通过营养支持改善临床患者的的营养状况已具有至关重要的作用,但目前市场上可供选择的肠内营养食品还比较缺乏,因此开发一种稳定的、便于携带和使用、制备方法相对简单的适合患者口服的全营养组合物,用来为对营养素没有特殊要求的临床患者提供全面而均衡的营养支持尤为重要。
发明内容
本发明主要涉及一种口服全营养组合物及其制备方法,具体如下:
一种口服全营养组合物,其特征在于所述组合物包含脂溶性成分、非脂溶性成分和纯化水;每1000mL所述组合物含有的非脂溶性成分为蛋白质40-60g、碳水化合物50-120g、钠0.5-2g、氯0.4-2.0g、镁0.01-0.6g、磷0.5-1.5g、钾1-4g、钙0.3-2g、铁0.005-0.05g、锌0.001-0.05g、铜0-0.01g、锰0-0.01g、硒0.01-0.5mg、碘0-1.5mg、水溶性维生素0.1-2g,含有的脂溶性成分为脂肪40-120g、脂溶性维生素0.02-1g。
进一步地,上述组合物还包含乳化剂、复配乳化增稠剂及甜味剂。
进一步地,所述蛋白质选自乳清蛋白、酪蛋白、水解乳清蛋白或其混合;碳水化合物选自麦芽糊精、木薯淀粉、葡萄糖或其混合;脂肪选自葵花籽油、大豆油、红花油或其混合,优选葵花籽油、大豆油或其混合;水溶性维生素选自泛酸、生物素、维生素K1、叶酸、维生素C、维生素B1、维生素B2、烟酸、维生素B6、维生素B12或其混合;脂溶性维生素选自维生素E、维生素A、维生素D或其混合;所述甜味剂选自三氯蔗糖、甜菊糖苷或其混合;所述乳化剂选自单双酸甘油酯、柠檬酸脂肪酸甘油酯或其混合,优选柠檬酸脂肪酸甘油酯;所述复配乳化增稠剂为食品用乳化剂和增稠剂的混合,优选微晶纤维素、单/双甘油脂肪酸酯、结冷胶、硬酯酰乳酸钠、双乙酰酒石酸单双甘油酯、羧甲基纤维素钠的混合,进一步优选丹尼斯克复配乳化增稠剂9566-C。
一种口服全营养组合物,其特征在于所述组合物包含脂溶性成分、非脂溶性成分、甜味剂、乳化剂、复配乳化增稠剂和纯化水,每1000mL所述组合物含有的非脂溶性成分为乳清蛋白40-60g、麦芽糊精50-120g、磷酸氢二钾3-8g、柠檬酸钠2-7g、乳酸钙1.5-10g、氯化镁0.1-5g、焦磷酸铁15-160mg、硫酸锌4-222mg、***钠20-1000μg、碘化钾0-2mg、硫酸锰0-40mg、硫酸铜0-40mg、泛酸0-30mg、生物素0-20mg、叶酸0-20mg、维生素K1 0-2mg、维生素C100-300mg、维生素B11-5mg、维生素B21-5mg、烟酸10-20mg、维生素B60.4-10mg、维生素B120.0006-0.3mg,含有的脂溶性成分为葵花籽油20-60g、大豆油0-40g、维生素E30-80mg、维生素A 0.1-20mg、维生素D 20-60μg,甜味剂0-0.5g,乳化剂1-4g,复配乳化增稠剂1-4g。
进一步地,每1000mL所述组合物含有的非脂溶性成分为乳清蛋白55g、麦芽糊精87g、磷酸氢二钾4.5g、柠檬酸钠3.6g、乳酸钙2.2g、氯化镁2.3g、焦磷酸铁50mg、硫酸锌50mg、***钠400μg、碘化钾1.3mg、硫酸锰2.4mg、硫酸铜10mg、泛酸20mg、生物素10mg、叶酸10mg、维生素K1 0.5mg、维生素C 240mg、维生素B12mg、维生素B22mg、烟酸18mg、维生素B63mg、维生素B120.1mg,含有的脂溶性成分为葵花籽油25g、大豆油25g、维生素E40mg、维生素A2mg、维生素D 30μg,甜味剂0.2g,乳化剂3g,复配乳化增稠剂1g。
进一步地,每1000mL所述组合物含有的非脂溶性成分为乳清蛋白55g、麦芽糊精87g、磷酸氢二钾4.5g、柠檬酸钠3.6g、乳酸钙2.2g、氯化镁0.3g、焦磷酸铁50mg、硫酸锌60mg、***钠400μg、维生素C 240mg、维生素B12mg、维生素B22mg、烟酸18mg、维生素B61.6mg、维生素B120.00018mg,含有的脂溶性成分为葵花籽油49g、维生素E 40mg、维生素A1mg、维生素D 30μg,甜味剂0.2g,乳化剂3g,复配乳化增稠剂1g。
同时,本发明提供了上述组合物的制备方法,包含以下步骤:
步骤1:配料
a)按配方分别称量非脂溶性成分、复配乳化增稠剂及甜味剂,待用;
b)按配方分别称量脂溶性成分及乳化剂,待用;
步骤2:溶解
c)将称得的脂溶性成分及乳化剂加热搅拌至全溶,得到油相溶液;
d)将称得的非脂溶性成分蛋白质及碳水化合物干混,用配制总量60%的水50-80℃水温剪切搅拌至全溶;加入复配乳化增稠剂,剪切搅拌至全溶;
e)向步骤d)溶液中加入称得的除水溶性维生素外的剩余非脂溶性成分,搅匀;
步骤3:以高剪切转速向步骤e)溶液中缓慢加入步骤c)溶液,继续循环剪切30min,剪切结束前5min加入用水溶解的水溶性维生素,剪切均匀;
步骤4:磷酸氢二钾调节产品pH为6.0-7.0;
步骤5:定容至配制总体积,制得初乳;
步骤6:将步骤4制得的初乳自循环;定容至总体积,冷却到20℃以下;
步骤7:灌装,灭菌,温度≥100℃,灭菌时间≥8分钟,或UHT 130~135℃灭菌10~30秒,灌装。
另外,本发明还涉及所述组合物用于对营养素没有特殊要求的患者提供全面而均衡的营养支持的应用。
本发明在研发过程中发现,乳化剂的选择对产品稳定性影响很大,乳化剂选择不当则会产生大量沉淀和或浮油,出现分层现象,严重则导致产品变性凝固,因此对乳化剂的选择进行了实验研究:
将筛选出的乳化剂柠檬酸脂肪酸甘油酯和单双酸甘油酯,分别和复配的乳化增稠剂一起使用,结果显示柠檬酸脂肪酸甘油酯和复配乳化增稠剂一起使用时产品状态更稳定,加速(温度40℃,湿度:70%)放置6个月产品呈均一液体,底部有少量沉淀,但摇晃即消失。
另外,钙的来源选择对产品的稳定性影响也很大,对氯化钙、碳酸钙、磷酸氢钙、乳酸钙进行制样筛选:
将筛选出的乳酸钙和磷酸氢钙,制备样品,加速(温度40℃,湿度:70%)放置6个月,其中选择乳酸钙的产品状态均一,包装瓶底部仅有少量沉淀,稍一晃动即消失,比加磷酸氢钙的产品更稳定。
同时,配方中磷酸二氢钾的含量选择较为重要,要保证磷及钾的含量,同时要使产品pH在规定范围6.0-7.0以内。
本发明所涉及的口服全营养组合物,在对患者的治疗中提供营养支持的同时能更好地帮助病人康复,减少并发症的发生,是适用于大多数临床患者的全营养配方组合物,通过口服或鼻饲等方式为病人提供全面、充足机体所需的各种营养物质,以达到预防或纠正热量、蛋白质缺乏所致的营养不良的目的,同时起到增强病人对严重创伤的耐受力,促进病人康复的作用。
本发明组合物营养全面均衡,适合绝大多数对营养素没有特殊需求的病人,富含高单不饱和脂肪酸葵花籽油、大豆油、红花油或其混合,有效改善糖脂代谢;蛋白含量达到4%,能够促进细胞与组织的修复。本发明中组合物中各成分含量符合规定;由于选择了柠檬酸脂肪酸甘油酯作为乳化剂,乳酸钙作为钙的来源,对磷酸氢二钾的量进行控制,使产品稳定性高;同时本产品便于携带和使用,制备方法简单,成本低,便于工业化大生产。
具体实施方式
下面将通过实施例对本发明作进一步的描述,这些描述并不是对本发明内容作进一步的限定。本领域的技术人员对发明内容所作的等同替换或改进,仍属于本发明的保护范围之内。
实施例1:
制备工艺:
步骤1:配料
1)按配方分别称量好各非脂溶性物料乳清蛋白、麦芽糊精、柠檬酸钠、氯化镁、磷酸氢二钾、乳酸钙、焦磷酸铁、硫酸锌、硫酸铜、硫酸锰、***钠、碘化钾、泛酸、生物素、维生素K1、叶酸、维生素C、维生素B1、维生素B2、烟酸、维生素B6、维生素B12、维生素A、维生素D、维生素E、复配乳化增稠剂、三氯蔗糖,待用;
2)按配方分别称量好脂溶性物料葵花籽油、大豆油、乳化剂柠檬酸脂肪酸甘油酯、维生素A、维生素D、维生素E,待用;
3)于配料罐中加入配制总量60%水,待用;
步骤2:溶解
1)将称好的葵花籽油、大豆油、乳化剂柠檬酸脂肪酸甘油酯、维生素A、维生素D、维生素E加热搅拌至全溶,得到油相溶液;
2)将称好的蛋白、麦芽糊精干混好,投入配料罐,70℃左右水温剪切搅拌至全溶;
3)再将复配乳化增稠剂、三氯蔗糖投入配料罐,分别剪切搅拌至全溶;
4)将称好备用的氯化镁、柠檬酸钠、乳酸钙、硫酸锰、硫酸铜、碘化钾、硫酸锌、***钠、焦磷酸铁、磷酸氢二钾,再分别加入上述水相溶液中,搅匀;
步骤3:将水相置剪切机下,以高剪切转速缓慢加入油相,油相加入完毕后继续循环剪切约30min,在剪切结束前5min将所有的水溶性维生素用水溶好,投入配料罐,剪切均匀;
步骤4:定容至配制总体积,搅匀制得初乳;
步骤5:将制得的初乳先在配料罐中自循环,待均质参数调好后,再均质到另一个罐子,将罐中剩余未均质到的料液排净;之后将上述料液定容至总体积,冷却到20℃以下;
步骤6:灌装,灭菌,温度≥100℃,灭菌时间≥8分钟,或UHT 130~135℃灭菌10~30秒,灌装。

实施例2
制备工艺:
步骤1:配料
1)按配方分别称量好各非脂溶性物料乳清蛋白、麦芽糊精、柠檬酸钠、氯化镁、磷酸氢二钾、乳酸钙、焦磷酸铁、硫酸锌、***钠、维生素C、维生素B1、维生素B2、烟酸、维生素B6、维生素B12、维生素A、维生素D、维生素E、复配乳化增稠剂、三氯蔗糖,待用;
2)按配方分别称量好脂溶性物料葵花籽油、乳化剂柠檬酸脂肪酸甘油酯、维生素A、维生素D、维生素E,待用;
3)于配料罐中加入配制总量60%水,待用;
步骤2:溶解
1)将称好的葵花籽油、乳化剂柠檬酸脂肪酸甘油酯、维生素A、维生素D、维生素E加热搅拌至全溶,得到油相溶液;
2)将称好的蛋白、麦芽糊精干混好,投入配料罐,70℃左右水温剪切搅拌至全溶;
3)再将复配乳化增稠剂、三氯蔗糖投入配料罐,分别剪切搅拌至全溶;
4)将称好备用的氯化镁、柠檬酸钠、乳酸钙、硫酸铜、硫酸锌、***钠、焦磷酸铁、磷酸氢二钾,再分别加入上述水相溶液中,搅匀;
步骤3:将水相置剪切机下,以高剪切转速缓慢加入油相,油相加入完毕后继续循环剪切约30min,在剪切结束前5min将所有的水溶性维生素用水溶好,投入配料罐,剪切均匀;
步骤4:定容至配制总体积,搅匀制得初乳;

Claims (10)
1.一种口服全营养组合物,其特征在于所述组合物包含脂溶性成分、非脂溶性成分和纯化水;每1000mL所述组合物含有的非脂溶性成分为蛋白质40-60g、碳水化合物50-120g、钠0.5-2g、氯0.4-2.0g、镁0.01-0.6g、磷0.5-1.5g、钾1-4g、钙0.3-2g、铁0.005-0.05g、锌0.001-0.05g、铜0-0.01g、锰0-0.01g、硒0.01-0.5mg、碘0-1.5mg、水溶性维生素0.1-2g,含有的脂溶性成分为脂肪40-120g、脂溶性维生素0.02-1g。
2.如权利要求1所述的组合物,其特征在于还包含乳化剂、复配乳化增稠剂及甜味剂。
3.如权利要求1或2所述的组合物,其特征在于所述蛋白质选自乳清蛋白、酪蛋白、水解乳清蛋白或其混合;碳水化合物选自麦芽糊精、木薯淀粉、葡萄糖或其混合;脂肪选自葵花籽油、大豆油、红花油或其混合;水溶性维生素选自泛酸、生物素、维生素K1、叶酸、维生素C、维生素B1、维生素B2、烟酸、维生素B6、维生素B12或其混合;脂溶性维生素选自维生素E、维生素A、维生素D或其混合;所述甜味剂选自三氯蔗糖、甜菊糖苷或其混合;所述乳化剂选自单双酸甘油酯、柠檬酸脂肪酸甘油酯或其混合;所述复配乳化增稠剂为食品用乳化剂和增稠剂的混合。
4.如权利要求3所述的组合物,其特征在于所述蛋白质为乳清蛋白;所述脂肪为葵花籽油、大豆油或其混合;所述乳化剂为柠檬酸脂肪酸甘油酯。
5.一种口服全营养组合物,其特征在于所述组合物包含脂溶性成分、非脂溶性成分、甜味剂、乳化剂、复配乳化增稠剂和纯化水,每1000mL所述组合物含有的非脂溶性成分为乳清蛋白40-60g、麦芽糊精50-120g、磷酸氢二钾3-8g、柠檬酸钠2-7g、乳酸钙1.5-10g、氯化镁0.1-5g、焦磷酸铁15-160mg、硫酸锌4-222mg、***钠20-1000μg、碘化钾0-2mg、硫酸锰0-40mg、硫酸铜0-40mg、泛酸0-30mg、生物素0-20mg、叶酸0-20mg、维生素K1 0-2mg、维生素C100-300mg、维生素B11-5mg、维生素B21-5mg、烟酸10-20mg、维生素B60.4-10mg、维生素B120.0006-0.3mg,含有的脂溶性成分为葵花籽油20-60g、大豆油0-40g、维生素E 30-80mg、维生素A 0.1-20mg、维生素D 20-60μg,甜味剂0-0.5g,乳化剂1-4g,复配乳化增稠剂1-4g。
6.如权利要求5所述的组合物,其特征在于每1000mL所述组合物含有的非脂溶性成分为乳清蛋白55g、麦芽糊精87g、磷酸氢二钾4.5g、柠檬酸钠3.6g、乳酸钙2.2g、氯化镁2.3g、焦磷酸铁50mg、硫酸锌50mg、***钠400μg、碘化钾1.3mg、硫酸锰2.4mg、硫酸铜10mg、泛酸20mg、生物素10mg、叶酸10mg、维生素K1 0.5mg、维生素C 240mg、维生素B12mg、维生素B22mg、烟酸18mg、维生素B6 3mg、维生素B120.1mg,含有的脂溶性成分为葵花籽油25g、大豆油25g、维生素E 40mg、维生素A 2mg、维生素D 30μg,甜味剂0.2g,乳化剂3g,复配乳化增稠剂1g。
7.如权利要求5所述的组合物,其特征在于每1000mL所述组合物含有的非脂溶性成分为乳清蛋白55g、麦芽糊精87g、磷酸氢二钾4.5g、柠檬酸钠3.6g、乳酸钙2.2g、氯化镁0.3g、焦磷酸铁50mg、硫酸锌60mg、***钠400μg、维生素C 240mg、维生素B12mg、维生素B22mg、烟酸18mg、维生素B6 1.6mg、维生素B120.00018mg,含有的脂溶性成分为葵花籽油49g、维生素E40mg、维生素A 1mg、维生素D 30μg,甜味剂0.2g,乳化剂3g,复配乳化增稠剂1g。
8.权利要求1-7任一项组合物的制备方法,其特征在于包含以下步骤:
步骤1:配料
a)按配方分别称量非脂溶性成分、复配乳化增稠剂及甜味剂,待用;
b)按配方分别称量脂溶性成分及乳化剂,待用;
步骤2:溶解
c)将称得的脂溶性成分及乳化剂加热搅拌至全溶,得到油相溶液;
d)将称得的非脂溶性成分蛋白质及碳水化合物干混,用配制总量60%的水50-80℃水温剪切搅拌至全溶;加入复配乳化增稠剂,剪切搅拌至全溶;
e)向步骤d)溶液中加入称得的除水溶性维生素外的剩余非脂溶性成分,搅匀;
步骤3:以高剪切转速向步骤e)溶液中缓慢加入步骤c)溶液,继续循环剪切30min,剪切结束前5min加入用水溶解的水溶性维生素,剪切均匀;
步骤4:用碱调节产品pH为6.0-7.0;
步骤5:定容至配制总体积,制得初乳;
步骤6:将步骤4制得的初乳自循环;定容至总体积,冷却到20℃以下;
步骤7:灌装,灭菌,温度≥100℃,灭菌时间≥8分钟,或UHT 130~135℃灭菌10~30秒,灌装。
9.根据权利要求8所述的制备方法,其特征在于步骤4所述碱为磷酸氢二钾。
10.权利要求1-8中任一项所述的组合物在对营养素没有特殊要求的患者提供全面而均衡的营养支持的应用。
