Description

Traditional Chinese medicine composition fermentation liquor with anti-pollution effect for cosmetics and preparation and application thereof
Technical Field
The invention relates to a traditional Chinese medicine composition fermentation liquor, in particular to a traditional Chinese medicine composition fermentation liquor with an anti-pollution effect for cosmetics, and preparation and application thereof, and belongs to the field of cosmetics.
Background
In recent years, the number of skin patients has increased dramatically, and the reasons for this are many, among which air pollution is an important part, and the following skin problems are easily caused by air pollution: one is causing skin allergy: a large amount of dispersed floating matters in the air are allergic to cause blockage of skin pores and allergic symptoms, so that people feel pruritus, develop erythema and whelk, and even trigger urticaria and eczema. Secondly, the skin is aged: pollutants in the air, such as haze and the like, can cause the skin to generate an excessive amount of extremely unstable free radicals, and the free radicals can damage cell membranes and DNA (deoxyribonucleic acid) of human body self health, so that the aging of the body is accelerated, and the skin is aged, such as dull skin, pigmentation, easy relaxation, fine lines or wrinkles, dry skin and even skin cancerization. Whether it causes skin infection easily: haze causes excessive particles in the air, and the degree of light transmission is reduced, so that ultraviolet rays reaching the vicinity of the ground are weakened. Thus various viruses, bacteria and fungi floating in the air near the ground become less susceptible to being killed by ultraviolet rays. For people with poor physique, skin allergy and damaged skin, the microorganisms reach the skin along with dust particles in the air and are adhered to the surface of the skin, so that skin infection is easily caused, such as folliculitis, sweat stain, herpes simplex and the like, and people with skin rash are aggravated.
Therefore, to resist the damage of external pollution to skin, the treatment should be done with the aspects of expelling toxin, resisting allergy, scavenging free radicals, isolating ultraviolet rays, inhibiting bacteria, cleaning, etc.

1mL of each of the 7 kinds of aqueous solutions was added to a sterile glass tube, and 8mL of a previously prepared modified GAM broth (Nissui, Japan, No. ZZM-05433) was poured. Taking out the purified CO and keeping the temperature of the CO constant at 35 DEG C2Culturing clinical Propionibacterium acnes in an incubator for 48h, respectively injecting 1mL of bacterial suspension into the modified GAM broth test tube containing the sample aqueous solution, and using a glass test tube containing no bacterial suspension as a negative control. Inoculating Propionibacterium acnes in each sample culture medium from low concentration, and placing at constant temperature of 35 deg.C CO as soon as possible after inoculation2The incubator is used for 48 hours. After the culture, the liquid in the improved GAM broth culture medium was first extracted for preliminary verification experiments, and then the kill of Propionibacterium acnes was determined by a VITEK full-automatic microbiological analysis system and an ANI anaerobe identification card, with the results as shown in Table 1 below.

The comparison of the experimental results of the examples 1 to 3 and the comparative examples 1 to 5 shows that the fermentation liquor of the traditional Chinese medicine composition can effectively remove free radicals and has a good anti-aging function; compared with the single traditional Chinese medicine fermentation liquor prepared in the proportion of 1-5, the free radical removing effect is good, and the effect of synergy is achieved after optimized combination. Comparison of experimental results of examples 1-3 and comparative example 6 shows that the effect of scavenging free radicals of the traditional Chinese medicine composition is remarkably enhanced after fermentation.
Test example 3 evaluation of anti-allergic Effect
The anti-allergic effect was evaluated by an inhibition test on hyaluronidase. Hyaluronidase (commercially available) is a hydrolase of hyaluronic acid and is closely linked to the development of inflammation and allergy. The inhibition effect of the plant anti-allergy moisturizing composition on skin allergy can be indirectly determined by measuring the inhibition rate of the plant anti-allergy moisturizing composition on hyaluronidase.
Test protocol: the inhibition of hyaluronidase by the extract was determined by the Elson-Morgan modification assay. 0.1mL of 0.25mmol/L CaCl is taken2The solution and 0.5mL of hyaluronidase solution are cultured for 20min at 37 ℃ in an insulation way; respectively adding 0, 0.5, 1.0, 1.5 and 2.0mL of the fermentation liquid or the extracting solution prepared in the example 1 and the comparative examples 1-6, and continuing to culture at 37 ℃ for 20 min; adding 0.5mL sodium hyaluronate solution, keeping the temperature at 37 ℃ for 30min, and standing at normal temperature for 5 min; adding 0.1mL of 0.4mol/L NaOH solution and 0.5mL of acetylacetone solution, heating in a boiling water bath for 15min, and immediately cooling for 5min with ice water; adding 1mL of Ellisib reagent, diluting to 5mL with anhydrous ethanol, standing for 20min for color development, and measuring the absorbance value with a spectrophotometer.
The hyaluronidase inhibition of the extract was calculated as follows:
the inhibition ratio (%) - (A-B) - (C-D) ]/(A-B) × 100%
Wherein, A is the absorbance of a control solution (sodium hyaluronate + hyaluronidase + deionized water); b is the absorbance of a control blank solution (acetic acid buffer solution + deionized water); c is the absorbance of the sample solution (potassium hyaluronate + hyaluronidase + sample solution); d is the absorbance of the sample blank solution (acetic acid buffer solution + sample solution).
The test results are shown in Table 3.
TABLE 3 results of hyaluronidase inhibition
Figure BDA0001751821440000111
The comparison of the experimental results of the examples 1-3 and the comparative examples 1-5 shows that the traditional Chinese medicine composition fermentation liquor can strongly inhibit hyaluronidase and has a good anti-allergy function; compared with the single traditional Chinese medicine fermentation liquor prepared in the proportion of 1-5, the hyaluronidase inhibition effect is good, and the optimized combination plays a role in synergy. The comparison of the experimental results of the examples 1-3 and the comparative example 6 shows that the hyaluronidase inhibition effect of the traditional Chinese medicine composition is remarkably enhanced after fermentation.
Test example 4 irritation evaluation test

The results are shown in Table 4.
TABLE 4 results of hemolysis
Figure BDA0001751821440000121
The comparison of the experimental results of the examples 1 to 3 and the comparative examples 1 to 5 shows that the hemolysis ratio of the fermentation liquor of the traditional Chinese medicine composition is much lower than that of the single traditional Chinese medicine prepared by the comparative examples 1 to 5, and the irritation is much lower. The comparison of the experimental results of the examples 1-3 and the comparative example 6 shows that the irritation of the fermentation liquor of the traditional Chinese medicine composition is also obviously reduced after fermentation.
