Test example 1 evaluation of efficacy in inhibiting Propionibacterium acnes
1. Preparation of Chinese medicine extract sample
The traditional Chinese medicine compound extract with the acne removing and red blood streak removing effects prepared in the examples 1-3 and the extracts prepared in the comparative examples 1-6 are prepared into sample aqueous solutions with the mass fractions of 0.5%, 1%, 2%, 3%, 4% and 5% for later use.
2. Method for testing inhibition capacity of propionibacterium acnes
1mL of each of the 9 sample aqueous solutions was added to a sterile glass tube, and 8mL of a previously prepared modified GAM broth (Nissui, Japan, No. ZZM-05433) was poured. Taking out the purified CO and keeping the temperature of the CO constant at 35 DEG C2Culturing clinical Propionibacterium acnes in an incubator for 48h, respectively injecting 1mL of bacterial suspension into the modified GAM broth test tube containing the sample aqueous solution, and using a glass test tube containing no bacterial suspension as a negative control. Inoculating Propionibacterium acnes in each sample culture medium from low concentration, and placing at constant temperature of 35 deg.C CO as soon as possible after inoculation2The incubator is used for 48 hours. After the culture, the liquid in the improved GAM broth culture medium was first extracted for preliminary verification experiments, and then the kill of Propionibacterium acnes was determined by a VITEK full-automatic microbiological analysis system and an ANI anaerobe identification card, with the results as shown in Table 1 below.

The comparison results of the examples 1-3 and the comparative examples 1-5 show that the traditional Chinese medicine compound extract prepared by the invention has an obvious inhibition effect on propionibacterium acnes, and has a much better effect than the single traditional Chinese medicine extract prepared by the comparative examples 1-4 and the composite extract of sophora flower bud and xanthium sibiricum prepared by the comparative example 5, which shows that the traditional Chinese medicine components have a remarkable synergistic effect after the compound.
The result of comparison between the example 1 and the comparative example 6 shows that the inhibition effect of the traditional Chinese medicine compound extract prepared by the extraction solvent adopted by the invention on propionibacterium acnes is better than that of the traditional Chinese medicine compound extract prepared by the solvent adopted by the comparative example 6, which indicates that the effect of the extraction solvent adopted by the invention is better.
Test example 2 radical scavenging test
1. Preparation of the test specimens
The traditional Chinese medicine compound extract with the acne removing and red blood streak removing effects prepared in the examples 1-3 and the extracts prepared in the comparative examples 1-6 are prepared into sample aqueous solutions with the mass fraction of 0.5%, and the following experiments are carried out.
2. Evaluation of hydroxyl radical scavenging ability
Sequentially adding 2mmol/L FeSO into a 25mL colorimetric tube4 3mL,1mmol/L H2O23mL, shaking up, then adding 3mL of salicylic acid with the concentration of 6mmol/L, shaking up, heating in a water bath at 37 ℃ for 15min, taking out, and measuring the absorbance; respectively adding 0.5mL of the solution to be tested, shaking, heating in water bath for 15min, and taking out to test its absorbance. The clearance of hydroxyl radical is calculated according to formula (1):
hydroxyl radical clearance (%) ([ a 1-a 2- (a 1-A3) ]/a1 × 100% (1)
Wherein A1 is the absorbance value of the reaction system before adding the sample; a2 is the absorbance value of the system after removing OH from the sample; a3 is the absorbance value of the system after blank control elimination of OH.
3. Results of the experiment
The results of the free radical scavenging experiments are shown in table 2.

The comparison results of examples 1-3 and comparative examples 1-5 show that the traditional Chinese medicine compound extract prepared by the invention has a much better effect on removing hydroxyl radicals than the single traditional Chinese medicine extract prepared by the comparative examples 1-4 and the sophora flower bud and xanthium sibiricum compound extract prepared by the comparative example 5, and the synergistic effect among the traditional Chinese medicine components after the compound is carried out is obvious.
The result of comparison between the example 1 and the comparative example 6 shows that the hydroxyl radical scavenging effect of the traditional Chinese medicine compound extract prepared by the extraction solvent adopted by the invention is better than that of the traditional Chinese medicine compound extract prepared by the solvent adopted by the comparative example 6, which indicates that the effect of the extraction solvent adopted by the invention is better.
Test example 3 irritation evaluation test
The irritation of the Chinese medicinal extract is evaluated by a rabbit erythrocyte hemolysis experiment. The specific operation is as follows: respectively taking 500 mu L of the traditional Chinese medicine extracts prepared in the examples 1-3 and the comparative examples 1-5, placing the extracts in a 2mL centrifuge tube, respectively adding 500 mu L of 2% rabbit red blood cell (commercially available) suspension, shaking gently and mixing uniformly, placing the mixture in a constant temperature incubator at 37 ℃ for incubation for 3h, centrifuging the mixture at 2000r/min for 3min, absorbing supernatant, taking physiological saline as negative control and distilled water as positive control, measuring absorbance at the wavelength of 414nm, and calculating the hemolysis according to a formula. The results are shown in Table 3.

The comparison of the experimental results of the examples 1-3 and the comparative examples 1-5 shows that the hemolysis ratio of the traditional Chinese medicine compound extract is much lower than that of the extract prepared by the comparative examples 1-5, and the irritation of the traditional Chinese medicine extract after the compound is obviously reduced.
Application example 1 essence for removing acne and red blood streak
The acne-removing and redness-removing essence containing a traditional Chinese medicine compound extract with the effects of removing acne and redness-removing is shown in a formula shown in table 4.

The preparation method comprises the following steps:
(1) dispersing xanthan gum and sodium hyaluronate in glycerol in advance, then adding the dispersed xanthan gum and sodium hyaluronate, dipotassium glycyrrhizinate and hydroxyethyl cellulose into the traditional Chinese medicine compound extract (I) together, and stirring and dissolving until the dispersion is uniform and transparent;
(2) heating and dissolving p-hydroxyacetophenone in 1, 3-propylene glycol and 1, 2-hexanediol in advance, putting the mixture into the solution in the step (1), and stirring and dissolving the mixture uniformly;
(3) and (5) inspecting the physicochemical indexes such as color, pH, fragrance and appearance, filtering with 200 meshes after passing inspection, and discharging.
Application example 2 emulsion for removing acne and red blood streak
The acne-removing and redness-removing lotion contains a traditional Chinese medicine compound extract with the effects of removing acne and redness-removing, and the formula is shown in table 5.

The preparation method comprises the following steps:
(1) heating the raw material of the component A to 80 ℃, and stirring and dispersing uniformly for later use;
(2) heating the raw material of the component B to 80 ℃, and stirring and dissolving uniformly;
(3) pumping the raw materials of the component B into the phase A for homogenizing and emulsifying, and cooling after the emulsification is finished;
(4) cooling to 45 ℃, adding the raw material of the component C, and stirring and dispersing uniformly. Vacuumizing and defoaming are finished.
Application example 3 acne-removing and redness-removing mask
The acne-removing and redness-removing facial mask containing a traditional Chinese medicine compound extract with the effects of removing acne and removing redness has a formula shown in table 6.

The preparation method comprises the following steps:
(1) dispersing xanthan gum and sodium hyaluronate in glycerol in advance, adding the dispersed xanthan gum and sodium hyaluronate, allantoin, panthenol, hydroxyethyl cellulose and dipotassium glycyrrhizinate into deionized water, heating to 80 ℃, stirring and dissolving until the mixture is uniform and transparent, and cooling;
(2) heating and dissolving p-hydroxyacetophenone in 1, 3-propylene glycol and 1, 2-hexanediol in advance until the p-hydroxyacetophenone is transparent, putting the p-hydroxyacetophenone into the solution obtained in the step (1) when the temperature of the system is reduced to 55 ℃, and stirring and dissolving the p-hydroxyacetophenone until the p-hydroxyacetophenone is transparent;
(3) when the temperature is reduced to 40 ℃, the traditional Chinese medicine compound extract (I) is put into the mixture and stirred uniformly until the mixture is completely dissolved;
(4) and (5) inspecting the physicochemical indexes such as color, pH, fragrance and appearance, filtering with 200 meshes after passing inspection, and discharging.
Effect example 1 evaluation of effects and evaluation of skin irritation
Evaluation target: 40 young people with whelk on the face (accompanied by a large amount of tiny acne) and the age of 15-30 years are selected, wherein 20 male people and 20 female people are selected.
The evaluation method comprises the following steps: the acne-removing and red blood streak-removing mask prepared in application example 3 is applied to the face in the morning and evening by a double-blind method, and the face is massaged after the mask is taken down until all mask liquid is absorbed by the skin.
Evaluation course of treatment: the acne removing effect evaluation takes 20 days as a treatment course, the micro powder and red blood streak removing effect evaluation takes 90 days as a treatment course, and the irritation evaluation takes 20 days as a treatment course.
Evaluation standard of acne removing effect: the method has the following advantages: acne eliminating rate over 70%; secondly, the method is effective: acne is eliminated by more than 30%; ③ invalid: acne is eliminated by less than 30%.
Evaluation standard of the effect of removing the micro-powder thorn: the method has the following advantages: micro acne is reduced by more than 70%; secondly, the method is effective: micro acne is reduced by more than 30%; ③ invalid: the micro acne is reduced by less than 30%.
Evaluation standard of effect of removing red blood streak: the red pigment values of the front and rear cheeks before and after use were tested with Mexamter MX18(CK Electronic GmH, Germany). The red blood streak removing effect is judged by testing the inhibition conditions (inhibition rate) of the red pigment before and after use, and the formula of the inhibition rate is as follows: inhibition (pre-use red pigment value-post-use red pigment value) × 100%/pre-use red pigment value; the method has the following advantages: the inhibition rate is more than or equal to 20 percent; the method has the following advantages: ② the inhibition rate is 5-20 percent; and (4) invalidation: the inhibition rate is less than 5%.
Irritation evaluation criteria: the examination was performed according to the criteria of Table 7.

Evaluation results of acne removal effects: 18 cases of obvious effect account for 45 percent; 14 cases are effective, accounting for 35 percent; and 8 ineffective cases account for 20 percent. The total effective rate reaches 80 percent, which shows that the traditional Chinese medicine compound extract has good acne removing effect.
Evaluation results of the effect of removing the micro-powder thorn: 12 cases of obvious effect account for 30 percent; 23 cases of effectiveness account for 57.5 percent; and 5 invalid cases account for 12.5 percent. The total effective rate reaches 87.5 percent, which shows that the traditional Chinese medicine compound extract has good micro-powder thorn removing effect.
Evaluation results of the effect of removing red blood streak: 15 cases of obvious effect account for 37.5 percent; the effective 18 cases account for 45%; and 7 ineffective cases account for 17.5 percent. The total effective rate reaches 82.5 percent, which shows that the traditional Chinese medicine compound extract has good effect of removing red blood streak.
Evaluation results of irritation: no allergic phenomenon was found in all volunteers.
The above embodiments are preferred embodiments of the present invention, but the present invention is not limited to the above embodiments, and any other changes, modifications, substitutions, combinations, and simplifications which do not depart from the spirit and principle of the present invention should be construed as equivalents thereof, and all such changes, modifications, substitutions, combinations, and simplifications are intended to be included in the scope of the present invention.

Claims (11)

1. A traditional Chinese medicine compound extract with the effects of removing acne and red blood streak is characterized in that: the traditional Chinese medicine compound extract is prepared from the following traditional Chinese medicine components in parts by weight: 30-35 parts of ramie, 20-25 parts of root of canna edulis, 20-25 parts of sophora flower bud and 20 parts of xanthium sibiricum;
the preparation method of the traditional Chinese medicine compound extract with the effects of removing acne and red blood streak specifically comprises the following steps:
crushing the traditional Chinese medicine raw materials, respectively sieving the crushed raw materials by using a sieve with 20-120 meshes, and mixing the crushed raw materials to obtain a traditional Chinese medicine compound mixture;
secondly, adding an extraction solvent according to the liquid-material mass ratio of 2-20: 1, soaking for 0.5-6 h, and then extracting at normal temperature by using a high-pressure homogenizing extractor under the extraction pressure of 5-50 MPa to obtain an extracting solution; the extraction solvent is a mixed solution of PEG-40 hydrogenated castor oil, 1, 3-butanediol and water, and the mass fractions of the mixed solution are 0.5 percent, 35 percent and 64.5 percent respectively;
and thirdly, after centrifugal separation of the extracting solution, carrying out ultrafiltration treatment on the centrifugate, and carrying out nanofiltration treatment on the ultrafiltration permeating liquid to obtain nanofiltration trapped liquid, namely the traditional Chinese medicine compound extract.
2. The traditional Chinese medicine compound extract with the effects of removing acne and red blood streak according to claim 1, which is characterized in that:
screening by using a 60-100-mesh sieve;
the mass ratio of the liquid to the material in the second step is 5-15: 1;
the soaking time in the step II is 2 hours;
the extraction pressure in the step two is 9.5 MPa;
the radius of the centrifugation in the third step is 8-12 cm;
centrifuging at the normal temperature for 15-60 min at 500-4000 rpm;
the molecular interception of the ultrafiltration membrane used in the step (III) is 1000-2000 Da;
the molecular interception amount of the nanofiltration membrane used in the step (III) is 80-120 Da;
the normal temperature is 15-40 ℃.
3. The application of the traditional Chinese medicine compound extract with the effects of removing acne and red blood streak according to any one of claims 1-2 in preparing skin external products.

the addition amount of the traditional Chinese medicine compound extract with the acne removing and red blood streak removing effects in the skin external product is 0.1-50% by mass.

the skin external product comprises: emulsions, creams, gels, powders, masks, sprays, and essences.

the skin external product contains 0.1-50% of a traditional Chinese medicine compound extract with the effects of removing acne and red blood streak by mass fraction;
the skin external product comprises: aqueous solution and powder molding.
9. The skin external product with the effects of removing acne and red blood streak according to claim 6, wherein:
the skin external product is an essence for removing acnes and red blood streaks, and comprises the following components in parts by mass: 5.00 percent of glycerin, 4.00 percent of 1, 3-propylene glycol, 0.10 percent of sodium hyaluronate, 0.07 percent of xanthan gum, 0.05 percent of hydroxyethyl cellulose, 0.50 percent of dipotassium glycyrrhizinate, 12.00 percent of Chinese herbal compound extract, 0.80 percent of 1, 2-hexanediol, 0.50 percent of p-hydroxyacetophenone and 100 percent of deionized water.
10. The skin external product with the effects of removing acne and red blood streak according to claim 6, wherein:
the skin external product is an emulsion for removing acnes and red blood streaks, and comprises the following components in parts by mass: 0.02% of EDTA-2Na, 3.00% of dipotassium glycyrrhizinate, 0.50% of 1, 3-butanediol, 2.00% of 1, 2-propanediol, 0.80% of 1, 2-hexanediol, 0.50% of p-hydroxyacetophenone, 0.20% of carbomer, 0.05% of sodium hyaluronate, 2.50% of polyglycerol-3 methyl glucose distearate, 0.50% of sorbitan sesquioleate, 2.50% of isopropyl palmitate, 2.50% of meadowfoam seed oil, 1.50% of shea butter, 0.50% of tocopherol acetate, 3.50% of cyclopenta dimethyl siloxane, 15.00% of a traditional Chinese medicine compound extract, 0.10% of aminomethyl propanol, 0.10% of essence and 100% of deionized water.
11. The skin external product with the effects of removing acne and red blood streak according to claim 6, wherein:
the skin external product is a mask for removing acnes and red blood streaks, and comprises the following components in parts by mass: 7.00 percent of glycerin, 7.00 percent of 1, 3-propylene glycol, 0.05 percent of sodium hyaluronate, 0.10 percent of xanthan gum, 0.10 percent of hydroxyethyl cellulose, 0.25 percent of allantoin, 0.50 percent of panthenol, 0.50 percent of dipotassium glycyrrhizinate, 0.80 percent of 1, 2-hexanediol, 0.50 percent of p-hydroxyacetophenone, 25.00 percent of compound traditional Chinese medicine extract and 100 percent of deionized water.
