背景技术
蓝莓果实中含有丰富的营养成分,具有防止脑神经老化、保护视力、强心、抗癌、软化血管、增强人机体免疫等功能,营养成分高。蓝莓中含有的花青素,是其他水果如苹果、葡萄的几倍甚至几十倍,总氨基酸含量0.254%,比山楂还高。蓝莓花青素抗氧化性很强,对眼科和心脑血管疾病有较好的疗效,能起到美容保健等多种功效。
如专利申请号CN201310380698.X,一种缓解视力疲劳蓝莓含片及其制作方法,以榨汁后剩余的蓝莓果渣为主要原料,采用多种现代食品加工新技术,经真空冷冻干燥,超微粉碎后制成蓝莓果渣基料。将蓝莓果渣基料、枸杞粉、叶黄素、木糖醇、玉米淀粉和糊精等主要原料,经混合、造粒、干燥、压片等工序加工后,制成酸甜适口,具有抗氧化、缓解视力疲劳等保健功能的蓝莓含片。而且操作过程安全,工艺技术简单,成本低廉,无污染。专利申请号CN201410484723.3,蓝莓含片,公开了一种采用野生蓝莓果、野生五味子果、野生刺五加果为原料的含片,其特征是,计量按百分比计:野生蓝莓果40%、野生五味子果40%、野生刺五加果20%。具有很高的营养和药用价值,降酶保肝、保护中枢神经***、抗病毒、抗肿瘤、抗衰老、抑制胆固醇生物合成等作用。克服了现有含片,所含营养成份和功能单一的不足之处。

又如专利申请号CN201510888143.5,一种原花青素泡腾片及其制备方法,提供一种原花青素泡腾片及其制备方法,以花牛苹果原花青素提取物为主要原料,添加崩解剂、赋形剂、矫味剂和食用色素,所有添加剂均符合食品卫生和安全要求,采用粉末直接压片技术,形成具有一定形状的原花青素泡腾片。研究中优化苹果皮渣中原花青素提取工艺,并将其研制成原花青素泡腾片,该泡腾片克服了传统口服型保健品携带不便和食用口味不佳的缺点,实现资源的充分利用,而且可以增加一种新型保健食品,从而改善人们健康状况,提高生活质量。但其制片过程中容易发生粘冲,得到的含片稳定性不高。
发明内容
本发明的目的是提供一种方法简单、成本低、便于工业化生产的、所得产品含有更高的蓝莓花青素、具备更加明显的视力保护、提神醒目、补钙、增强机体免疫力等多重作用的复合含片。
本发明的另一个目的是提供一种上述蓝莓复合含片的制备方法。
本发明使用的米曲霉为A.orgzae SP-01。
具体的,本发明的技术方案如下:
一种蓝莓复合含片,由以下重量份数比的组分:蓝莓花青素提取粉40~60份、钙粉6~10份,海藻粉15~25份,薄荷油5~15份,胖大海粉1~5份,菊花粉1~5份,荔枝汁1~5份,大豆蛋白溶液50~70份,水适量。
优选的,该蓝莓复合含片包含下列重量分数比的组分:蓝莓花青素提取粉50份,钙粉8份,海藻粉20份,薄荷油10份,胖大海粉4份,菊花粉4份,荔枝汁4份,大豆蛋白溶液60。
由以下步骤制备而得:
(1)取优质大米60~80份,清水洗净,加水侵泡至大米达到无硬芯,将侵泡好的大米放入电饭锅中煮至大米熟而不粘,然后将煮熟的大米分装至容器,接种米曲霉并混匀,置于35℃、90%以上湿度条件下发酵4~6天;所述米曲霉接种量为2~4%;
(2)取发酵4~6天后的大米放入容器中,加入其质量10~20%的磷酸缓冲液,均质3min后,4200r/min下离心5min,取上清液备用;所述磷酸缓冲液pH为6.5~7.5;
优选的,取发酵4天后的大米进行提取。
(3)清洗蓝莓鲜果,用液氮对蓝莓鲜果进行速冻处理,快速将蓝莓捣碎,然后加入其质量5~10%的步骤(2)所得上清液,混合均匀,10~30min后用冷冻干燥机在-80℃冷冻干燥,制得蓝莓干粉;所述蓝莓干粉中的水分应低于5%;所述液氮温度为-30~-50℃,流量为4~5m/s;
(4)取蓝莓干粉,按照1g:10~30ml的料液比将蓝莓干粉和醋酸溶液混合均匀,用柠檬酸调节pH至2~4,在40~60℃下,420w功率微波处理20s,提取60~130min,得到花青素粗提液;所述醋酸浓度为3~4mol/L;
(5)取花青素粗提液,加入其体积5~8%的柠檬酸,加入其体积5~8%的步骤(2)所得上清液,用乙酸乙酯萃取3次,非酯层加入到D101大孔树脂柱中,先用超纯水洗净脱,再用乙醇-水体系洗脱,收集50%的乙醇洗脱液,减压浓缩,冷冻干燥,得到精制花青素粉。所述柠檬酸pH为3~5,浓度为0.3~0.5g/ml;
(6)取精制花青素粉40~60份、海藻粉15~25份,薄荷油5~15份,胖大海粉1~5份,枸杞粉1~5份,荔枝汁1~5份,钙粉5~10份,碳酸氢钾粉16~32份混合均匀,加入5%无水乙醇湿润制软料,然后于40~50℃鼓风干燥机中干燥2~3h,过100目筛整粒得到原料粒;所述钙粉与碳酸氢钾粉份数比为(20~25):80;
(7)将原料粒与大豆蛋白溶液100~200份混合,500rpm搅拌均匀,再加入转谷氨酰胺酶10~20份,并快速振荡使此混合液分散均匀,然后将此混合液体系在900rpm,40℃的磁力搅拌器作用下反应120~150min,然后在500rpm下离心1min,去除油相并用生理盐水洗涤2次,在500rpm下离心1min收集微胶囊颗粒;
(8)在70~75℃下将聚乙二醇溶解,加入无水柠檬酸粉末混合均匀,冷却后将混合物粉碎,过80目筛得到酸源;
(9)将所得微胶囊颗粒与酸源1:1混合均匀,按质量比加入2~3%L-亮氨酸和3~4%聚乙二醇6000粉末,混合均匀后送入旋转压片机制片,得到蓝莓复合含片。
作为本发明的进一步改进,所述蓝莓花青素的提取中,添加米曲霉发酵大米提取物,利用其活性物质抑制多酚氧化酶,从而提高花青素的提取率。
大米含有游离态的香草酸、紫丁香酸、阿魏酸、香豆酸以及苯乙烯酸,同时也含有结合态的紫丁香酸、阿魏酸、香豆酸和苯乙烯酸,其中游离态和结合态的阿魏酸为主要酚类物质,酚类物质具有很强的抗氧化作用,能中和人体内的自由基,对人体健康具有很好的保护作用。
利用米曲霉A.orgzae SP-01发酵大米,然后提取发酵大米中的酚类物质,其主要酚类物质为曲酸和阿魏酸,相比于同浓度的曲酸和阿魏酸,从曲霉发酵大米提取物中获得的曲酸的阿魏酸具有更强的抗氧化作用。利用该物质抑制多酚氧化酶,能提高蓝莓花青素的提取率。
泡腾片是以适宜的酸和碱为崩解剂制成的一种片剂,泡腾片入水后会产生大量二氧化碳气体从而迅速溶解,药物起效迅速,生物利用度高,携带方便且成本低,由于泡腾制剂中含有酸源和碱源,只要有微量水的存在,很容易引发酸碱反应,一旦发生反应就难以控制,严重影响制剂的稳定性。为了提高泡腾制剂的稳定性,用微胶囊将碱源与活性物质包裹,利用PEG对酸源进行包裹,再制粒,是酸碱分开。
作为本发明的进一步改进,所述蓝莓复合含片采用泡腾片制作技术原理,同时利用微胶囊技术包裹碱源,提高含片的稳定性。
微胶囊技术作为药物释放体系,在医学界研究已久,在本发明中,蓝莓花青素等药物经过微胶囊化后,不仅能提高其抗氧化性,同时不受体内环境对活性成分的影响,更好的发挥药用疗效。微胶囊包裹的碱源,在蓝莓复合含片中能避免与酸源接触,提高了蓝莓复合含片的稳定性。
海藻粉富含海藻多糖、甘露醇、氨基酸、蛋白质、维生素和钾、铁、钙、磷、碘、硒、钴等微量元素,维生素A与蛋白质是合成视紫质的主要物质。
薄荷油富含醇类物质和酮类物质,能提高人体免疫力。在本发明中,利用薄荷油作为矫味剂,提高蓝莓复合含片的风味,还是蓝莓复合含片的润滑剂,减少在制片过程中发生粘冲,提高生产效率。
荔枝汁含葡萄糖、蔗糖、蛋白质、脂肪以及维生素A、B、C等,并含叶酸、精氨酸、色氨酸等各种营养素,对人体健康十分有益。在本发明,利用荔枝汁作为矫味剂,提高蓝莓复合含片的甜味;另一方面,荔枝汁中维生素A是视紫质主要合成物质之一,添加荔枝汁能提高蓝莓复合含片的功效。
钙粉,主要物质为CaCO3,在本发明中,作为钙源补充人体的钙,另一方面,与碳酸氢钾作为碱源制备蓝莓复合含片,特定钙粉与碳酸氢钾的配比能提高含片的稳定性。
胖大海、菊花粉、枸杞粉能提高人体免疫力,对视力保护具有很好的功效。
本发明制备而得的蓝莓复合含片,既能直接含服,也可放于水中冲服。
本发明的有益效果是:
1、米曲霉发酵大米产物中主要含有曲酸和阿魏酸,能有效抑制蓝莓中多酚氧化酶活性,利用液氮处理将蓝莓果破碎,有效地降低了氧化多酚酶的活性,提高后续花青素的提取率;
2、添加海藻粉、钙粉、薄荷油、胖大海粉、枸杞粉、荔枝汁等原料,提高含片中的保健成分;海藻粉含有丰富的蛋白质、维生素以及微量元素,荔枝汁含有丰富的维生素A,与蛋白质共同作用,促进视紫质合成,胖大海粉、菊花粉对清热降火,平肝明目,促进皮肤新陈代谢,延缓衰老,改善皮肤生理状态和功能障碍具有很好的作用。进一步提高该含片视力保护、提神醒目的作用,薄荷油、钙粉、等均是具有药用功效的天然材料,提高含片的保健功效。
3、米曲霉固体发酵大米提取液中,含有丰富的多酚类物质,能够有效地与亚油酸自氧化产生的自由基反应保护β-胡萝卜素,有利于视紫质的合成,进而保护视力;同时,提取液中的活性成分曲酸和阿魏酸具有高抗氧化活性,与纯曲酸和阿魏酸相比,能更有效地清除DPPH自由基,保护人体健康;
4、利用大豆蛋白将含片保健活性成分微胶囊化,能有效控制活性成分在人体中的释放,提高活性成分的作用效率,同时微胶囊能保护蓝莓花青素不被氧化,提高其活性保持时间。将含片制备成泡腾片,不仅可以口服,还可以溶于水送服,方便食用。

下面结合具体实施例对本发明进一步说明,而不作为对本发明的限制。
实施例1
(1)取优质大米60份,清水洗净,加水侵泡至大米达到无硬芯,将侵泡好的大米放入电饭锅中煮至大米熟而不粘,然后将煮熟的大米分装至容器,接种米曲霉并混匀,置于35℃、90%以上湿度条件下发酵4天;所述米曲霉接种量为2%;
(2)取发酵4天后的大米放入容器中,加入其质量10%的磷酸缓冲液,均质3min后,4200r/min下离心5min,取上清液备用;所述磷酸缓冲液pH为6.5;
(3)清洗蓝莓鲜果,用液氮对蓝莓鲜果进行速冻处理,快速将蓝莓捣碎,然后加入其质量5%的步骤(2)所得上清液,混合均匀,10min后用冷冻干燥机在-80℃冷冻干燥,制得蓝莓干粉;所述蓝莓干粉中的水分应低于5%;所述液氮温度为-50℃,流量为4m/s;
(4)取蓝莓干粉,按照1g:10ml的料液比将蓝莓干粉和醋酸溶液混合均匀,用柠檬酸调节pH至4,在40℃下,420w功率微波处理20s,提取60min,得到花青素粗提液;所述醋酸浓度为3mol/L;
(5)取花青素粗提液,加入其体积5%的柠檬酸,加入其体积5%的步骤(2)所得上清液,用乙酸乙酯萃取3次,非酯层加入到D101大孔树脂柱中,先用超纯水洗净脱,再用乙醇-水体系洗脱,收集50%的乙醇洗脱液,减压浓缩,冷冻干燥,得到精制花青素粉。所述柠檬酸pH为3,浓度为0.3g/ml;
(6)取精制花青素粉40份、海藻粉15份,薄荷油5份,胖大海粉1份,枸杞粉1份,荔枝汁1份,钙粉5份,碳酸氢钾粉16份混合均匀,加入5%无水乙醇湿润制软料,然后于40℃鼓风干燥机中干燥2h,过100目筛整粒得到原料粒;所述钙粉与碳酸氢钾粉份数比为2:8;
(7)将原料粒与大豆蛋白溶液50份混合,500rpm搅拌均匀,再加入转谷氨酰胺酶10份,并快速振荡使此混合液分散均匀,然后将此混合液体系在900rpm,40℃的磁力搅拌器作用下反应120min,然后在500rpm下离心1min,去除油相并用生理盐水洗涤2次,在500rpm下离心1min收集微胶囊颗粒;
(8)在70℃下将聚乙二醇溶解,加入无水柠檬酸粉末混合均匀,冷却后将混合物粉碎,过80目筛得到酸源;
(9)将所得微胶囊颗粒与酸源1:1混合均匀,按质量比加入2%L-亮氨酸和3%聚乙二醇6000粉末,混合均匀后送入旋转压片机制片,得到蓝莓复合含片。
本实施例中,蓝莓花青素提取率为340mg/100g蓝莓果;蓝莓复合含片在水中完全溶解时间为72s。
实施例2
(1)取优质大米80份,清水洗净,加水侵泡至大米达到无硬芯,将侵泡好的大米放入电饭锅中煮至大米熟而不粘,然后将煮熟的大米分装至容器,接种米曲霉并混匀,置于35℃、90%以上湿度条件下发酵6天;所述米曲霉接种量为4%;
(2)取发酵6天后的大米放入容器中,加入其质量10~20%的磷酸缓冲液,均质3min后,4200r/min下离心5min,取上清液备用;所述磷酸缓冲液pH为7.5;
(3)清洗蓝莓鲜果,用液氮对蓝莓鲜果进行速冻处理,快速将蓝莓捣碎,然后加入其质量5~10%的步骤(2)所得上清液,混合均匀,30min后用冷冻干燥机在-80℃冷冻干燥,制得蓝莓干粉;所述蓝莓干粉中的水分应低于5%;所述液氮温度为-50℃,流量为5m/s;
(4)取蓝莓干粉,按照1g:10~30ml的料液比将蓝莓干粉和醋酸溶液混合均匀,用柠檬酸调节pH至4,在60℃下,420w功率微波处理20s,提取60~130min,得到花青素粗提液;所述醋酸浓度为4mol/L。
(5)取花青素粗提液,加入其体积8%的柠檬酸,加入其体积8%的步骤(2)所得上清液,用乙酸乙酯萃取3次,非酯层加入到D101大孔树脂柱中,先用超纯水洗净脱,再用乙醇-水体系洗脱,收集50%的乙醇洗脱液,减压浓缩,冷冻干燥,得到精制花青素粉。所述柠檬酸pH为5,浓度为0.5g/ml。
(6)取精制花青素粉60份、海藻粉25份,薄荷油15份,胖大海粉5份,枸杞粉5份,荔枝汁5份,钙粉10份,碳酸氢钾粉32份混合均匀,加入5%无水乙醇湿润制软料,然后于50℃鼓风干燥机中干燥3h,过100目筛整粒得到原料粒;所述钙粉与碳酸氢钾粉份数比为3:8;
(7)将原料粒与大豆蛋白溶液70份混合,500rpm搅拌均匀,再加入转谷氨酰胺酶20份,并快速振荡使此混合液分散均匀,然后将此混合液体系在900rpm,40℃的磁力搅拌器作用下反应120~150min,然后在500rpm下离心1min,去除油相并用生理盐水洗涤2次,在500rpm下离心1min收集微胶囊颗粒;
(8)在70~75℃下将聚乙二醇溶解,加入无水柠檬酸粉末混合均匀,冷却后将混合物粉碎,过80目筛得到酸源;
(9)将所得微胶囊颗粒与酸源1:1混合均匀,按质量比加入2~3%L-亮氨酸和3~4%聚乙二醇6000粉末,混合均匀后送入旋转压片机制片,得到蓝莓复合含片。
本实施例中,蓝莓花青素提取率为327mg/100g蓝莓果;蓝莓复合含片在水中完全溶解时间为76s。
实施例3
(1)取优质大米70份,清水洗净,加水侵泡至大米达到无硬芯,将侵泡好的大米放入电饭锅中煮至大米熟而不粘,然后将煮熟的大米分装至容器,接种米曲霉并混匀,置于35℃、90%以上湿度条件下发酵4天;所述米曲霉接种量为3%;
(2)取发酵4天后的大米放入容器中,加入其质量10~20%的磷酸缓冲液,均质3min后,4200r/min下离心5min,取上清液备用;所述磷酸缓冲液pH为7;
(3)清洗蓝莓鲜果,用液氮对蓝莓鲜果进行速冻处理,快速将蓝莓捣碎,然后加入其质量8%的步骤(2)所得上清液,混合均匀,20min后用冷冻干燥机在-80℃冷冻干燥,制得蓝莓干粉;所述蓝莓干粉中的水分应低于5%;所述液氮温度为-40℃,流量为4m/s;
(4)取蓝莓干粉,按照1g:20ml的料液比将蓝莓干粉和醋酸溶液混合均匀,用柠檬酸调节pH至3,在50℃下,420w功率微波处理20s,提取70min,得到花青素粗提液;所述醋酸浓度为3mol/L;
(5)取花青素粗提液,加入其体积6%的柠檬酸,加入其体积6%的步骤(2)所得上清液,用乙酸乙酯萃取3次,非酯层加入到D101大孔树脂柱中,先用超纯水洗净脱,再用乙醇-水体系洗脱,收集50%的乙醇洗脱液,减压浓缩,冷冻干燥,得到精制花青素粉。所述柠檬酸pH为4,浓度为0.4g/ml;
(6)取精制花青素粉50份、海藻粉20份,薄荷油10份,胖大海粉4份,枸杞粉4份,荔枝汁4份,钙粉8份,碳酸氢钾粉32份混合均匀,加入5%无水乙醇湿润制软料,然后于40~50℃鼓风干燥机中干燥2~3h,过100目筛整粒得到原料粒;
(7)将原料粒与大豆蛋白溶液60份混合,500rpm搅拌均匀,再加入转谷氨酰胺酶12份,并快速振荡使此混合液分散均匀,然后将此混合液体系在900rpm,40℃的磁力搅拌器作用下反应130min,然后在500rpm下离心1min,去除油相并用生理盐水洗涤2次,在500rpm下离心1min收集微胶囊颗粒;
(8)在70~75℃下将聚乙二醇溶解,加入无水柠檬酸粉末混合均匀,冷却后将混合物粉碎,过80目筛得到酸源;
(9)将所得微胶囊颗粒与酸源1:1混合均匀,按质量比加入2%L-亮氨酸和3%聚乙二醇6000粉末,混合均匀后送入旋转压片机制片,得到蓝莓复合含片。
本实施例中,蓝莓花青素提取率为343mg/100g蓝莓果;蓝莓复合含片在水中完全溶解时间为71s。
实施例4
本实施例为对比例。
本实施例的配方为:蓝莓多糖10千克,维生素C5千克,淀粉7千克,柠檬酸15千克,碳酸氢钠20千克,聚乙二醇60002千克,羟丙基纤维素4千克。
制备方法为 :
(1)蓝莓多糖的制备:
将蓝鸟蓝莓果实真空低温干燥,超微粉碎机粉碎 ( 粉碎后粒度为200目),粉碎后的果粉加无水乙醇10倍体积,80℃水浴回流脱脂4h,挥干溶剂,重复提取2次,减压抽滤,滤渣挥干溶剂;加20倍体积蒸馏水,80℃水浴回流提取2h,滤纸过滤,再加同体积蒸馏水重复提取一次,两次滤液合并,真空浓缩(60℃,真空度0.095MPa)至原体积1/5,Sevage法除蛋白,反复进行至无蛋白层,加入无水乙醇使乙醇浓度达到80%,于4℃冰箱中过夜(12h),离心(4000r/min,10min),多糖沉淀依次用无水乙醇、丙酮、***各洗两次,60℃干燥至恒重,即得蓝莓多糖,备用。
(2)制片:取蓝莓多糖、维生素C和碳酸氢钠,进行干混;加入1千克聚乙二醇6000湿混,并制粒;加入柠檬酸和填充剂,进行干混;加入1千克聚乙二醇6000湿混,并制粒;冷风挥干;加入羟丙基纤维素,混匀,压片,即得。
本实施例中,蓝莓花青素提取率为205mg/100g蓝莓果;蓝莓复合含片在水中完全溶解时间为79s。

1.一种蓝莓复合含片,其特征在于,由以下重量份数比的组份:蓝莓花青素提取粉40~60份、钙粉6~10份,海藻粉15~25份,薄荷油5~15份,胖大海粉1~5份,菊花粉1~5份,荔枝汁1~5份,大豆蛋白溶液50~70份,水适量。
2.一种如权利要求1所述的蓝莓复合含片的制备方法,其特征在于,包括以下步骤:
(1)取优质大米60~80份,清水洗净,加水侵泡至大米达到无硬芯,将侵泡好的大米放入电饭锅中煮至大米熟而不粘,然后将煮熟的大米分装至容器,接种米曲霉并混匀,置于35℃、90%以上湿度条件下发酵4~6天;
(2)取发酵4~6天后的大米放入容器中,加入其质量10~20%的磷酸缓冲液,均质3min后,4200r/min下离心5min,取上清液备用;
