背景技术
铁是人体必需的微量元素中含量最多的一种,有半微量元素之称,总量约为4~5g。铁缺乏可使血红蛋白含量和生理活性降低,引起携带的氧明显减少,从而影响大脑中营养素和氧的供应。人体常期缺乏铁,会发生缺铁性贫血。表现症状为食欲减退、面色苍白、心悸头晕、免疫功能下降、容易疲乏、注意力不集中、记忆力减退等。在人的一生中,婴儿期和青春发育期是最需要铁的时期。若摄取的铁不足,就可使血红蛋白合成减少,以致通过血红蛋白输送到身体各组织的氧减少,使组织缺氧,从而引起多种器官和组织的生理功能异常。人体不能自主合成铁,只能通过食物来供给,但往往由于饮食搭配不合理,容易造成铁摄入量不足。
维生素B是B族维生素的总称,包括维生素B1、维生素B2、维生素B6、维生素B12、烟酸、泛酸等,是一类水溶性维生素,相互之间具有协同作用,对于调节人体新陈代谢,维持皮肤和肌肉的健康,增进免疫***和神经***的功能,促进细胞生长和***等具有重要的作用。维生素B是人体组织必不可少的营养素,但其在体内滞留的时间只有数小时,不会贮存于体内,会完全排出体外,因此必须每天通过食物来补充。但由于饮食搭配不合理,偏食等原因,常会导致儿童维生素B摄入量不足。维生素B缺乏将导致食欲减退、消化不良、碳水化合物、蛋白质和脂肪代谢障碍、细胞功能降低等,不利于儿童的健康成长。
儿童铁和维生素B摄入不足易导致食营养性贫血,食营养性贫血是指因缺乏生血所必须的营养物质,如铁、叶酸、维生素B等,使血红蛋白的形成或红细胞的生成不足,以致造血功能低下的一种疾病,多发于6个月至2岁的婴幼儿。目前除日常食品以外,儿童主要通过硫酸亚铁、富马亚铁、力蜚能、葡萄糖酸亚铁等铁剂复合维生素B等来补充所需的铁及维生素。但是这些复合补铁剂大多对胃肠刺激性较大,可能会产生恶心、呕吐、便秘、腹泻,牙齿、大便变黑等副作用,并且由于铁剂和B族维生素本身的特殊气味,这类复合补铁剂的口感较差,降低了儿童的服用意愿。
另外,补铁剂中的铁必须要保持二价铁离子的形式才容易被人体吸收,然而二价铁离子又极易被氧化,所以目前的补铁剂需遮光密封保存,特别是补铁口服液,其长期存放后二价铁含量下降明显,甚至会出现沉淀物。B族维生素也对光敏感,多种B族维生素容易见光分解。因此,目前铁剂复合维生素B的制剂需要遮光密封保存,口服液则需使用棕色瓶子封装,然而棕色瓶子的包装外观却难以吸引儿童的服用兴趣。
中国专利申请CN105596342A公开了一种儿童补铁与维生素的组合物,其通过玫瑰茄提取物和山楂提取物的加入提高了组合物中铁和维生素的生物利用度,降低副作用,还提高了组合物的稳定性,使得组合物长期存放二价铁含量稳定,延长保质期,同时玫瑰茄提取物和山楂提取物带有玫瑰茄和山楂的滋味,可改善产品口感。
L-蛋氨酰基甘氨酸(CAS号为14486-03-4)是由蛋氨酸和甘氨酸缩合得到的产物,其盐酸盐于2014年3月被批准作为食品添加剂的新品种,用作食品香料。中国专利申请105685968A公开了一种儿童补钙与维生素的组合物,其通过添加一定量的山楂提取物和L-蛋氨酰基甘氨酸盐酸盐,可以在丰富营养成分的同时,促进钙的吸收,改善产品口感,产品具有香味,且可显著提高产品的稳定性。
甜菜红(CAS号为7659-95-2)是以食用红甜菜(Betavulgarisvar.rubra)的根茎为原料,通过浸提、分离、浓缩、干燥而成的天然食用色素,用作食品着色剂,其染着性好,水溶液呈现红色至***,色泽鲜艳,但其耐热性较差,光和氧会促进其降解。
因此,针对目前补铁剂存在的口感和稳定性较差等问题,有必要研发一种组方合理、口感好、稳定性好、有效补充铁和维生素的儿童补铁与维生素的组合物。
发明内容
为了解决现有技术中存在的问题(口感较差,稳定性差等),本发明提供一种儿童补铁与维生素的组合物,针对儿童设计,通过添加六种维生素,起到了补铁的协同增效作用,具有补铁效果好、口感好、营养成分丰富、储存稳定性佳等优点。
本发明提供一种儿童补铁与维生素的组合物,包括以下含量的制备原料:葡萄糖酸亚铁0.9~1.8g/kg、维生素B115~40mg/kg、维生素B210~30mg/kg、维生素B618~40mg/kg、烟酸200~300mg/kg、泛酸40~100mg/kg、维生素E100~180mg/kg、L-赖氨酸40~160mg/kg、柠檬酸2~4g/kg、甜味剂200~600g/kg和辅料400~800g/kg。
优选地,所述儿童补铁与维生素的组合物包括以下含量的制备原料:葡萄糖酸亚铁1.5g/kg、维生素B130mg/kg、维生素B220mg/kg、维生素B630mg/kg、烟酸280mg/kg、泛酸80mg/kg、维生素E160mg/kg、L-赖氨酸120mg/kg、柠檬酸3g/kg、甜味剂300g/kg,余量为辅料。
优选地,所述甜味剂为葡萄糖、白砂糖和低聚异麦芽糖中的一种或多种。
优选地,所述辅料为淀粉、糊精、微晶纤维素、硬脂酸镁、滑石粉、羟丙基甲基纤维素、海藻酸钠和水中的一种或多种。
更优选地,所述儿童补铁与维生素的组合物还包括甜菜红200~280mg/kg和L-蛋氨酰基甘氨酸盐酸盐80~140mg/kg;所述辅料选自水。当辅料为水时,本发明提供的组合物为口服液制剂,通过添加甜菜红和L-蛋氨酰基甘氨酸盐酸盐,与本发明的其他成分协同,在本发明的配比下,可显著提高产品的稳定性,尤其是在自然光照下的稳定性,同时改善产品口感,使产品具有香味,且带有鲜艳的红紫色,提高儿童的服用意愿。
更优选地,所述儿童补铁与维生素的组合物由以下含量的原料制备而成:葡萄糖酸亚铁1.5g/kg、维生素B130mg/kg、维生素B220mg/kg、维生素B630mg/kg、烟酸280mg/kg、泛酸80mg/kg、维生素E160mg/kg、L-赖氨酸120mg/kg、柠檬酸3g/kg、甜味剂300g/kg、甜菜红240mg/kg和L-蛋氨酰基甘氨酸盐酸盐100mg/kg,余量为水。
相应的,本发明还提供了上述儿童补铁与维生素的组合物的制备方法,包括以下步骤:取葡萄糖酸亚铁、维生素B1、维生素B2、维生素B6、烟酸、泛酸、维生素E、L-赖氨酸、柠檬酸和甜味剂,分别粉碎后,混匀,过筛,加入辅料,混匀,得到儿童补铁与维生素的组合物。
优选的,本发明提供的儿童补铁与维生素的组合物的制备方法,包括以下步骤:取葡萄糖酸亚铁、维生素B1、维生素B2、维生素B6、烟酸、泛酸、维生素E、L-赖氨酸、柠檬酸、甜味剂、甜菜红和L-蛋氨酰基甘氨酸盐酸盐,分别粉碎后,混匀,过筛,加入水,混匀,得到儿童补铁与维生素的组合物。
此外,本发明还提供所述的儿童补铁与维生素的组合物在制备预防和改善营养性贫血保健品中的应用。本发明通过动物实验证明所述儿童补铁与维生素的组合物具有促进营养性贫血模型大鼠体重增长、提高血红蛋白含量以及降低红细胞内游离原卟啉(FEP)含量的作用,可使营养性贫血模型大鼠恢复健康状态,同时治疗期间大鼠均未出现呕吐、便秘和腹泻等情况,可作为预防和改善营养性贫血的保健品。
因此,与现有技术相比,本发明的优势在于:
(1)本发明补铁与维生素的组合物针对儿童设计,通过添加六种维生素,起到了补铁的协同增效作用,其口感好、营养成分丰富、储存稳定,为儿童有效地补充所需的微量元素铁和维生素B,可作为预防和改善营养性贫血的保健品;
(2)本发明补铁与维生素的组合物为口服液制剂时,口服适应性好,通过进一步添加一定量的甜菜红和L-蛋氨酰基甘氨酸盐酸盐,可以改善产品外观,使产品带有鲜艳的红紫色,同时使产品具有淡淡的香味,更容易吸引儿童的服用兴趣,提高了儿童的服用意愿,还可显著提高产品的稳定性,尤其是自然光照下的稳定性,可以无需使用棕色瓶封装。
(3)本发明补铁与维生素的组合物的制备方法操作简单,工艺稳定,可工业化生产,有利于推广应用。

以下通过具体实施方式进一步描述本发明,但本发明不仅仅限于以下实施例。在本发明的范围内或者在不脱离本发明的内容、精神和范围内,对本发明进行适当改进、替换功效相同的组分,对于本领域技术人员来说是显而易见的,它们都被视为包括在本发明的范围之内。
实施例1、本发明儿童补铁与维生素的组合物
配方:葡萄糖酸亚铁0.9g/kg、维生素B115mg/kg、维生素B210mg/kg、维生素B618mg/kg、烟酸200mg/kg、泛酸40mg/kg、维生素E100mg/kg、L-赖氨酸40mg/kg、柠檬酸2g/kg、白砂糖200g/kg,辅料补足1kg。所述辅料由淀粉、糊精和硬脂酸镁按照重量比50:40:1组成。
制备方法:取葡萄糖酸亚铁、维生素B1、维生素B2、维生素B6、烟酸、泛酸、维生素E、L-赖氨酸、柠檬酸和白砂糖,分别粉碎后,混匀,过筛,加入辅料,混匀,干燥,得到儿童补铁与维生素的组合物,灌装胶囊,得到胶囊剂。
实施例2、本发明儿童补铁与维生素的组合物
配方:葡萄糖酸亚铁1.8g/kg、维生素B140mg/kg、维生素B230mg/kg、维生素B640mg/kg、烟酸300mg/kg、泛酸100mg/kg、维生素E180mg/kg、L-赖氨酸160mg/kg、柠檬酸4g/kg、葡萄糖600g/kg,辅料补足1kg。所述辅料由淀粉和羟丙基甲基纤维素按照重量比80:20组成。
制备方法:取葡萄糖酸亚铁、维生素B1、维生素B2、维生素B6、烟酸、泛酸、维生素E、L-赖氨酸、柠檬酸和葡萄糖,分别粉碎后,混匀,过筛,加入辅料,混匀,干燥,得到儿童补铁与维生素的组合物,干法制粒,整粒,得到颗粒剂。
实施例3、本发明儿童补铁与维生素的组合物
配方:葡萄糖酸亚铁1.5g/kg、维生素B130mg/kg、维生素B220mg/kg、维生素B630mg/kg、烟酸280mg/kg、泛酸80mg/kg、维生素E160mg/kg、L-赖氨酸120mg/kg、柠檬酸3g/kg、葡萄糖300g/kg,纯水补足1kg。
制备方法:取葡萄糖酸亚铁、维生素B1、维生素B2、维生素B6、烟酸、泛酸、维生素E、L-赖氨酸、柠檬酸和葡萄糖,分别粉碎后,混匀,过筛,加入纯水,混匀,得到儿童补铁与维生素的组合物,灭菌和灌装后,得到口服液。

制备方法:取葡萄糖酸亚铁、维生素B1、维生素B2、维生素B6、烟酸、泛酸、维生素E、L-赖氨酸、柠檬酸、葡萄糖、甜菜红和L-蛋氨酰基甘氨酸盐酸盐,分别粉碎后,混匀,过筛,加入纯水,混匀,得到儿童补铁与维生素的组合物,灭菌和灌装后,得到口服液。。

制备方法:取葡萄糖酸亚铁、维生素B1、维生素B2、维生素B6、烟酸、泛酸、维生素E、L-赖氨酸、柠檬酸、葡萄糖、甜菜红和L-蛋氨酰基甘氨酸盐酸盐,分别粉碎后,混匀,过筛,加入纯水,混匀,得到儿童补铁与维生素的组合物,灭菌和灌装后,得到口服液。

配方:葡萄糖酸亚铁1.5g/kg、维生素B130mg/kg、维生素B220mg/kg、维生素B630mg/kg、烟酸280mg/kg、泛酸80mg/kg、维生素E160mg/kg、L-赖氨酸120mg/kg、葡萄糖300g/kg、甜菜红240mg/kg和L-蛋氨酰基甘氨酸盐酸盐100mg/kg,纯水补足1kg。
制备方法同实施例6。
试验例一、本发明儿童补铁与维生素的组合物的安全性试验
将实施例1~5制得的儿童补铁与维生素的组合物分别进行急性毒性试验、30天喂养试验和90天喂养试验,结果显示:本发明提供的儿童补铁与维生素的组合物属实际无毒类,喂养动物的生长情况良好,血液学和理化检查正常,未发现便秘的现象。因此,本发明提供的儿童补铁与维生素的组合物适宜长期服用。
试验例二、本发明儿童补铁与维生素的组合物对大鼠营养性贫血的影响
一、受试样品
本发明实施例3~6的口服液,葡萄糖酸亚铁糖浆(广西梧州制药(集团)股份有限公司,国药准字H10880040)
二、实验动物
54只SPF级雄性SD大鼠,体重70~90g,由湖南中医药大学提供。
三、主要仪器和试剂
1.仪器:RF-520荧光分光光度计、72-1型分光光度计、离心机、漩涡混匀器等。
2.试剂:碳酸氢钠、铁***、***、肝素抗凝剂、原卟啉、乙酸乙酯等。
四、实验方法
1.建立营养性贫血大鼠模型:将54只大鼠在SPF级动物实验室实验环境下适应5d后,饲予去离子水和低铁饲料[配方(g%):葡萄糖49.38,EDTA处理酪蛋白脱脐黄玉米粉15.00,明胶5.00,玉米油5.00,磷酸二氢钠2.00,碳酸钙2.00,氯化钾0.50,混合微量元素0.27,氯化胆碱0.15,混合纤维0.10,DL-蛋氨酸0.10],采用不锈钢笼和食罐,实验过程避免铁的污染,自第3周开始,每周选取部分大鼠采尾血测血蛋白(Hb)含量,直至大多数大鼠的血红蛋白含量低于100g/L时,测定全部大鼠的体重及血红蛋白的含量。
2.恢复实验:第6周采血测定血红蛋白含量后,从血红蛋白含量<100g/L的大鼠中选取含量最低的48只作为实验动物,根据贫血大鼠血红蛋白水平和体重,将其随机分为低铁对照组、4个实验组和阳性对照组,每组8只动物,各组均继续饲以低铁饲料以及去离子水,低铁对照组以纯水灌胃,4个实验组则分别以5.0ml/kg(相当于人体推荐用量的10倍)本发明实施例3~6的口服液灌胃,阳性对照组则以5.0ml/kg的葡萄糖酸亚铁糖浆灌胃,连续灌胃30d,试验期间观察各组大鼠的外观体征和行为活动。30d末测定大鼠体重、血红蛋白和红细胞内游离原卟啉含量。血红蛋白含量采用氰化高铁法测定,红细胞内游离原卟啉含量采用荧光分光光度计测定。
3.实验结果
(1)大鼠营养性贫血造模情况:试验大鼠实验室适应5d后饲予低铁饲料及去离子水,3周后发现部分大鼠开始脱毛、皮毛粗糙、稀疏,皮肤苍白,生长开始减慢;随着试验时间的延长,上述症状更加明显,食欲下降,身体消瘦,毛色失去光泽,出现板块脱落。耳、脚都出现明显苍白,表现倦态,活动减少,反应迟钝,喜欢缩成团,粪便较稀且黄。第6周采血测定血红蛋白含量,82%大鼠血红蛋白含量<100g/L。
(2)受试样品对大鼠外观体征和行为活动影响:
经观察,给予实施例3~6口服液的大鼠第6天开始摄食量与活动量明显增加,脱毛现象减少,随着治疗时间延长,脱毛处皮肤逐渐长出新毛并皮毛光滑,皮肤红润、神态自然,粪便呈灰褐色至黑色,同时治疗期间均未出现呕吐、便秘和腹泻等情况。
给予市售葡萄糖酸亚铁糖浆的大鼠第13天开始摄食量与活动量明显增加,随着治疗时间延长,逐渐恢复健康状态,治疗期间有3只大鼠出现呕吐,2只大鼠出现便秘,2只大鼠出现腹泻,大鼠粪便均呈黑色。
(3)受试样品对大鼠体重的影响:试验前各组大鼠体重基本一致,各组间无显著性差异。试验结束时,各受试样品组的大鼠体重和增重量均大于低铁对照组,其中实施例3~6组大鼠的增重量与低铁对照组比较具有显著性差异,表明实施例3~6口服液具有促进营养性贫血模型大鼠体重增长的作用,且其作用优于市售葡萄糖酸亚铁糖浆。
(4)受试样品对大鼠血红蛋白含量的影响:试验开始时各组大鼠血红蛋白含量基本一致,各组间无显著性差异。试验结束时,低铁对照组的血红蛋白含量较试验初降低,各受试样品组的血红蛋白含量均有升高,且与低铁对照组比较具有显著性差异,表明本发明实施例3~6口服液具有提高营养性贫血模型大鼠血红蛋白含量的作用,且其作用优于市售葡萄糖酸亚铁糖浆。
(5)受试样品对大鼠红细胞内游离原卟啉(FEP)含量的影响:试验结束时,各受试样品组大鼠红细胞内游离原卟啉(FEP)含量均显著低于低铁对照组,表明本发明实施例3~6口服液具有降低营养性贫血模型大鼠红细胞内游离原卟啉(FEP)含量的作用,且其作用优于市售葡萄糖酸亚铁糖浆。
试验例三、本发明儿童补铁与维生素的组合物的稳定性试验
将实施例3~6、对比例一~三制得的口服液封存于透明玻璃瓶中,在25±1℃下,于室内进行自然光照的稳定性考察,分别于0天、5天、10天和30天取样检测,结果如下表1所示。

从表1可知,本发明实施例4~6制得的组合物口服液的自然光照稳定性好,体系在自然光照下能保持鲜艳的红紫色且澄清、无沉淀,口服液中葡萄糖酸亚铁的含量未出现明显的变化;而实施例3、对比例一~三制得的组合物口服液在考察期内都出现了褐变,并伴有沉淀产生,口服液中葡萄糖酸亚铁的含量也显著降低。可见,同时添加一定量的甜菜红和L-蛋氨酰基甘氨酸盐酸盐,对本发明组合物的稳定性起到重要作用。甜菜红和L-蛋氨酰基甘氨酸盐酸盐的加入不仅使得产品带有鲜艳的红紫色和淡淡的香味,还起到了保护色的作用,使得本发明组合物中的葡萄糖酸亚铁、B族维生素等组分在没有遮光保存的情况下也能长期稳定、不易分解,可以无需使用棕色瓶封装,更容易吸引儿童的服用兴趣,提高了儿童的服用意愿。另外,试验还发现,柠檬酸作为酸性矫味剂,其不仅可以改善本发明组合物的口感,其还可使甜菜红和L-蛋氨酰基甘氨酸盐酸盐的组合稳定,共同提高组合物的稳定性。
此外,本发明还对组合物口服液(封存于透明玻璃瓶)的长期稳定性进行了考察,本发明实施例4~6制得的组合物保存12个月后,葡萄糖酸亚铁含量的降低值在20%以内,未出现沉淀现象,稳定性好,符合相关要求。而实施例3、对比例一至三制得的组合物保存12个月后几乎检测不到葡萄糖酸亚铁,且出现严重的沉淀现象。
上述实施例仅例示性说明本发明的原理及其功效,而非用于限制本发明。任何熟悉此技术的人士皆可在不违背本发明的精神及范畴下,对上述实施例进行修饰或改变。因此,举凡所属技术领域中具有通常知识者在未脱离本发明所揭示的精神与技术思想下所完成的一切等效修饰或改变,仍应由本发明的权利要求所涵盖。

1.一种儿童补铁与维生素的组合物,其特征在于,包括以下含量的制备原料:葡萄糖酸亚铁0.9~1.8g/kg、维生素B1 15~40mg/kg、维生素B2 10~30mg/kg、维生素B6 18~40mg/kg、烟酸200~300mg/kg、泛酸40~100mg/kg、维生素E 100~180mg/kg、L-赖氨酸40~160mg/kg、柠檬酸2~4g/kg、甜味剂200~600g/kg、辅料400~800g/kg、甜菜红200~280mg/kg和L-蛋氨酰基甘氨酸盐酸盐80~140mg/kg;所述辅料选自水。
2.根据权利要求1所述的儿童补铁与维生素的组合物,其特征在于,所述甜味剂为葡萄糖、白砂糖和低聚异麦芽糖中的一种或多种。
3.根据权利要求1所述的儿童补铁与维生素的组合物,其特征在于,由以下含量的原料制备而成:葡萄糖酸亚铁1.5g/kg、维生素B1 30mg/kg、维生素B2 20mg/kg、维生素B6 30mg/kg、烟酸280mg/kg、泛酸80mg/kg、维生素E 160mg/kg、L-赖氨酸120mg/kg、柠檬酸3g/kg、甜味剂300g/kg、甜菜红240mg/kg和L-蛋氨酰基甘氨酸盐酸盐100mg/kg,余量为水。
4.根据权利要求1所述的儿童补铁与维生素的组合物的制备方法,其特征在于,包括以下步骤:取葡萄糖酸亚铁、维生素B1、维生素B2、维生素B6、烟酸、泛酸、维生素E、L-赖氨酸、柠檬酸、甜味剂、甜菜红和L-蛋氨酰基甘氨酸盐酸盐,分别粉碎后,混匀,过筛,加入水,混匀,得到儿童补铁与维生素的组合物。
5.如权利要求1~4任一所述的儿童补铁与维生素的组合物在制备预防和改善营养性贫血保健品中的应用。
