Abstract
本发明涉及保健食品技术领域,公开了一种多糖组合物及其制备方法和应用。本发明所述多糖组合物由肉桂多糖、益智仁多糖、黑芝麻多糖、桑葚子多糖、枸杞多糖和黑木耳多糖组成。本发明选择食品资源中的六种多糖组成多糖组合物,其在减少剂量的前提下,可以显著改善阴虚模型大鼠的多动好斗、烦躁不安等症状、显著缓解下丘脑—垂体—肾上腺素轴(HPA)和下丘脑—垂体—性腺轴(HPG)的功能紊乱以及显著缓解cAMP和cGMP水平异常,与每种多糖单独使用比较,作用更全面,效果更显著。

技术领域
本发明涉及保健食品技术领域,特别涉及一种多糖组合物及其制备方法和应用。
背景技术
阴虚证是指体内阴液亏少而无以制阳,滋润、濡养等作用减退,临床常表现出形体消瘦,多动好斗、烦躁不安、口燥咽干,两颧潮红,无心烦热,潮热,盗汗,小便短黄,大便干结,舌红少津或少苔,脉细数等症状。
有文献报道指出亚健康以虚证为主,其中阴虚证最为常见。现代人工作压力大,经常熬夜,饮食起居没有规律,是导致阴虚症状多发的一个重要因素。
目前,改善阴虚症状的治疗方案多以中药为主,而且主要针对肾虚症状,这些药物组分数量普遍较多,中草药成分占多数,鲜有以普通食品为主的治疗方案。
中医历来强调药膳食疗,《黄帝内经素闻·五常政大论》:“谷肉果菜,食养尽之,无使过之,伤其正也”。《千金方·食治》:“夫为医者,当须先洞晓病源,知其所犯,以食治之;食疗不愈,然后命药”。由此可见对于具有阴虚症状的亚健康群体来说,药膳食疗能够在改善其阴虚症状、恢复机体健康状态的过程中起到至关重要的作用。
基于以上原因,发明一种普通食品来源的改善阴虚症状的产品显得尤为必要。
发明内容
有鉴于此,本发明的目的在于提供一种多糖组合物,使得所述多糖组合物各组分产生协同作用,在降低使用量的前提下提高对阴虚症状中多动好斗、烦躁不安、形体消瘦,口燥咽干以及大便干结的功效、提高缓解下丘脑—垂体—肾上腺素轴和下丘脑—垂体—性腺轴功能紊乱以及cAMP和cGMP水平异常的功效,同时提供所述多糖组合物的制备方法和在上述各方面的应用。
肉桂,为樟科常绿乔木植物肉桂的树皮,味辛、甘,性热,归脾、肾、心、肝经,有补火助阳、散寒止痛、温经通脉功效,主治肾阳衰弱的阳痿宫冷、虚喘心悸,心腹冷痈,寒疝作痛,寒痹腰痛,胸痹,阴疽,闭经,痛经等;
益智仁,为姜科植物益智的果实,味辛,性温,归脾、肾经,有温脾,暖肾,固气,涩精功效,主治冷气腹痛,中寒吐泻,多唾,遗精,小便余沥,夜多小便等;
黑芝麻,为胡麻科植物芝麻的黑色种子,味甘,性平,归肝、脾、肾经,有补益肝肾、养血益精、润肠通便功效,主治肾不足所致的头晕耳鸣、腰脚痿软、须发早发、肌肤干燥,以及肠燥便秘、妇人乳少、痈疮湿疹、风癞疬疡、小儿瘰疬、汤火伤、痔疮等;
桑葚子,为桑科植物桑的果穗,味甘、酸,性寒;归肝、肾、心经;有滋阴养血、生津、润肠功效,主治肾不足和血虚精亏的头晕目眩;腰酸耳鸣;须发早白;失眠多梦;津伤口渴;消渴;肠燥便秘等;
枸杞,茄科植物宁夏枸杞的果实,味甘性平,归肝、肾经;有滋补肝肾,益精明目功效;主治虚劳精亏、腰膝酸痛、眩晕耳鸣、内热消渴、血虚萎黄、目昏不明等;
黑木耳,为为木耳科植物木耳的子实体,味干性平,归肺、胃、肝、脾、肾、大肠经;有补气养血、润肺止咳、止血、降压、抗癌功效,主治气虚血亏、肺虚久咳、咳血、衄血、血痢、痔疮出血、妇女崩漏、高血压、眼底出血、子***、***癌、跌打伤痛等;
本发明精选上述六种中药原料,利用水提醇沉法提取多糖组分,组成多糖组合物。
为实现上述发明目的,本发明提供如下技术方案:
一种多糖组合物,由肉桂多糖、益智仁多糖、黑芝麻多糖、桑葚子多糖、枸杞多糖和黑木耳多糖组成。
作为优选,所述肉桂多糖、益智仁多糖、黑芝麻多糖、桑葚子多糖、枸杞多糖、黑木耳多糖的质量比为(1-2):(1-2):(2-3):(2-5):(3-5):(4-6);更优选地,所述肉桂多糖、益智仁多糖、黑芝麻多糖、桑葚子多糖、枸杞多糖、黑木耳多糖的质量比为1:1:2:5:3:6、2:2:3:2:5:6、1:1:2:3:5:6、1:1:3:5:4:6或1:1:2:4:3:6。
在本发明具体实施过程中,按照重量份数,所述多糖组合物可由肉桂多糖10~20份、益智仁多糖10~20份、黑芝麻多糖20~30份、桑葚子多糖20~50份、枸杞多糖30~50份、黑木耳多糖40~60份组成;所述重量份每一份可以表示任何重量单位形式下的任何数值,例如一重量份可表示为1g、0.5kg、10公斤、0.5斤等。
在本发明具体实施方式中,所述多糖组合物可具体如下:
(1)肉桂多糖1g、益智仁多糖1g、黑芝麻多糖2g、桑葚子多糖5g、枸杞多糖3g、黑木耳多糖6g;
(2)肉桂多糖2g、益智仁多糖2g、黑芝麻多糖3g、桑葚子多糖2g、枸杞多糖5g、黑木耳多糖6g;
(3)肉桂多糖1g、益智仁多糖1g、黑芝麻多糖2g、桑葚子多糖3g、枸杞多糖5g、黑木耳多糖6g;
(4)肉桂多糖1g、益智仁多糖1g、黑芝麻多糖3g、桑葚子多糖5g、枸杞多糖4g、黑木耳多糖6g;
(5)肉桂多糖1g、益智仁多糖1g、黑芝麻多糖2g、桑葚子多糖4g、枸杞多糖3g、黑木耳多糖6g。
本发明所述多糖组合物中各多糖组分可按照常规方法提取获得,但是在本发明具体实施过程中给出了如下获得方法:
将肉桂、益智仁、黑芝麻、桑葚子、枸杞或黑木耳加入其12倍量水煎煮1.0~2.0h过滤,得滤液和滤渣;滤渣加入其10倍量水煎煮0.5~1.0h,过滤,合并两次的滤液,在80℃条件下减压浓缩水提液,然后边搅拌边加入4倍体积60%乙醇,静置12h;分离沉淀,60%乙醇洗涤,真空冷冻干燥获得肉桂多糖、益智仁多糖、黑芝麻多糖、桑葚子多糖、枸杞多糖或黑木耳多糖。
本发明所述多糖组合物改善阴虚症状的作用主要体现在其可以改善多动好斗、烦躁不安、形体消瘦,口燥咽干(饮水量增多)以及大便干结;调节下丘脑—垂体—肾上腺素轴(HPA)和下丘脑—垂体—性腺轴(HPG)水平,改善功能紊乱;缓解cAMP和cGMP水平异常。由实验结果可以看出,多糖组合物与各多糖单独使用的剂量相同,但是在上述各方面均表现出更加优异的特性,这表明在大幅度减少多糖组合物中各多糖组分的使用量的前提下,对于上述各方面的功效反而得到提高,表现出明显的协同增效的作用。
基于上述优异的技术效果,本发明分别提出了所述多糖组合物在如下几个方面的应用:
(1)在制备改善阴虚症状药物中的应用;在该应用中,所述阴虚症状为多动好斗、烦躁不安、形体消瘦,口燥咽干以及大便干结;
(2)在制备改善下丘脑—垂体—肾上腺素轴和下丘脑—垂体—性腺轴功能紊乱的药物中的应用;
(3)在制备缓解cAMP和cGMP水平异常的药物中的应用。
同时,本发明还提供了所述多糖组合物的制备方法,包括:
将原料加入原料12倍量水煎煮1.0~2.0h过滤,得滤液和滤渣;滤渣加入其10倍量水煎煮0.5~1.0h,过滤,合并两次的滤液,在80℃条件下减压浓缩水提液,然后边搅拌边加入4倍体积60%乙醇,静置12h;分离沉淀,60%乙醇洗涤,真空冷冻干燥获得多糖提取物,所述原料为肉桂、益智仁、黑芝麻、桑葚子、枸杞或黑木耳;
将肉桂多糖、益智仁多糖、黑芝麻多糖、桑葚子多糖、枸杞多糖和黑木耳多糖混合,获得所述多糖组合物。
由以上技术方案可知,本发明选择食品资源中的六种多糖组成多糖组合物,其在减少剂量的前提下,可以显著改善阴虚模型大鼠的多动好斗、烦躁不安等症状、显著缓解下丘脑—垂体—肾上腺素轴(HPA)和下丘脑—垂体—性腺轴(HPG)的功能紊乱以及显著缓解cAMP和cGMP水平异常,与每种多糖单独使用比较,作用更全面,效果更显著。
具体实施方式
本发明公开了一种多糖组合物及其制备方法和应用,本领域技术人员可以借鉴本文内容,适当改进工艺参数实现。特别需要指出的是,所有类似的替换和改动对本领域技术人员来说是显而易见的,它们都被视为包括在本发明。本发明所述组合物及应用已经通过实施例进行了描述,相关人员明显能在不脱离本发明内容、精神和范围内对本文所述的组合物和应用进行改动或适当变更与组合,来实现和应用本发明技术。
在具体实施方式中,本发明所述多糖组合物中各多糖组分均采用水提醇沉法提取获得,具体为将干燥药材加入原生药的12倍量水煎煮1.0~2.0h,过滤,得滤液;第二次加入原生药的10倍量水煎煮0.5~1.0h,过滤,合并两次的滤液。在80℃条件下减压浓缩水提液,然后边搅拌边加入4倍体积60%乙醇,静置12h;分离沉淀,60%乙醇洗涤沉淀3次,真空冷冻干燥获得多糖样品。
在具体实施方式中,各试验试剂、仪器、动物、原料等均可由市场购得。其中,实验动物为雄性Wistar大鼠,体重160~180g,由湖北省疾病预防与控制中心提供;甲状腺片,40mg/片,上海长城药业有限公司,批号:20150704,国药准字H31022151;电子分析天平,BS124S,德国Startorius公司;超低温冷冻冰箱,美国Thermo Fisher Scientific;超纯水机,Mill-QⅡ,Milipore,Bedford,MA,USA.。
如未特别说明,在试验中所有多糖组分均采用相同方法在相同环境下提取获得,即来源相同。
以下就本发明所提供的一种多糖组合物及其制备方法和应用做进一步说明。
实施例1:本发明所述多糖组合物
按照水提醇沉法制备各多糖组分,称取肉桂多糖1g、益智仁多糖1g、黑芝麻多糖2g、桑葚子多糖5g、枸杞多糖3g、黑木耳多糖6g,混合制得多糖组合物。
实施例2:本发明所述多糖组合物
按照水提醇沉法制备各多糖组分,称取肉桂多糖2g、益智仁多糖2g、黑芝麻多糖3g、桑葚子多糖2g、枸杞多糖5g、黑木耳多糖6g,混合制得多糖组合物。
实施例3:本发明所述多糖组合物
按照水提醇沉法制备各多糖组分,称取肉桂多糖1g、益智仁多糖1g、黑芝麻多糖2g、桑葚子多糖3g、枸杞多糖5g、黑木耳多糖6g,混合制得多糖组合物。
实施例4:本发明所述多糖组合物
按照水提醇沉法制备各多糖组分,称取肉桂多糖1g、益智仁多糖1g、黑芝麻多糖3g、桑葚子多糖5g、枸杞多糖4g、黑木耳多糖6g,混合制得多糖组合物。
实施例5:本发明所述多糖组合物
按照水提醇沉法制备各多糖组分,称取肉桂多糖1g、益智仁多糖1g、黑芝麻多糖2g、桑葚子多糖4g、枸杞多糖3g、黑木耳多糖6g,混合制得多糖组合物。
实施例6:本发明所述多糖组合物功效试验
1、动物分组及处理
分组:雄性Wistar大鼠(体重160-180g)随机分为正常对照组(简称正常组)、阴虚模型组(简称模型组)、肉桂多糖组、益智仁多糖组、黑芝麻多糖组、桑葚子多糖组、枸杞多糖组、黑木耳多糖组、多糖组合I组、多糖组合II组、多糖组合III组、多糖组合IV组、多糖组合V组。其中,多糖组合I组(即实施例1,按照肉桂多糖、益智仁多糖、黑芝麻多糖、桑葚子多糖、枸杞多糖、黑木耳多糖比例为1:1:2:5:3:6的配比获得)、多糖组合II组(即实施例2,按照肉桂多糖、益智仁多糖、黑芝麻多糖、桑葚子多糖、枸杞多糖、黑木耳多糖比例为2:2:3:2:5:6的配比获得)、多糖组合III组(即实施例3,按照肉桂多糖、益智仁多糖、黑芝麻多糖、桑葚子多糖、枸杞多糖、黑木耳多糖比例为1:1:2:3:5:6的配比获得)、多糖组合IV组(即实施例4,按照肉桂多糖、益智仁多糖、黑芝麻多糖、桑葚子多糖、枸杞多糖、黑木耳多糖比例为1:1:3:5:4:6的配比获得)、多糖组合V组(即实施例5,按照肉桂多糖、益智仁多糖、黑芝麻多糖、桑葚子多糖、枸杞多糖、黑木耳多糖比例为1:1:2:4:3:6的配比获得)、多糖组合VI组(即对比例1,按照肉桂多糖、益智仁多糖、熟地多糖、天冬多糖、知母多糖、菟丝子多糖比例为1:1:3:2:1:1的配比获得)。每组动物10只。
处理:1)造模:除正常组灌胃等体积生理盐水,其他组别灌胃甲状腺片混悬液(45mg/100g),每日1次,自由饮食,连续15天。2)干预:正常组每日灌胃等体积生理盐水;模型组在造模同时每天灌胃等体积生理盐水;药物组造模的同时每天灌胃相应的多糖溶液,给药剂量为65mg/kg。共干预4周。
整个实验过程中控制好给药时间,注意保持实验环境的安静和实验装置的清洁,从而减少外界环境对实验结果的影响。
2、数据分析
实验结果皆以均数±标准差(±s)表示,均采用SPSS 20.0统计软件进行统计学处理。先进行正态性检验,正态分布数据两组以上的比较,采用单因素方差分析,组间两两比较,方差齐性的采用其项下的LSD检验;方差不齐的则采用其项下的Dunnett’s T3(3)检验。非正态分布数据采用非参数检验,两组以上的比较,采用Kruskal-Wallis 1-way ANOVA(ksamples)进行检验,组间两两比较,采用其项下的Stepwise step-down检验。以上均采用Levene法进行方差齐性检验。
假设检验采用双侧检验,检验水准为0.05。显著意义标识为P≤0.05。
3、结果
(1)对阴虚模型大鼠一般情况的影响
与正常对照组比较,各造模组大鼠从造模第6天开始出现活动频繁、多动好斗、烦躁不安等症状,伴体重增长缓慢,饮水量增多,大便干燥等表现。至实验结束时,模型组大鼠大多明显消瘦,毛无光泽,大便干燥,而其他多糖组情绪稳定,饮食、饮水、活动情况均较好,与正常组差异不明显。
(2)对阴虚模型大鼠体重的影响
实验结束后称取动物体重,结果见表1。与正常组比较,模型组体重显著降低(P<0.01);与模型组比较,除肉桂多糖组、益智仁多糖组、黑芝麻多糖组、多糖组合VI组外,其他多糖组体重显著增加(P<0.05),5个多糖组合(多糖组合I组、多糖组合II组、多糖组合III组、多糖组合IV组、多糖组合V组)对大鼠体重增长的恢复作用明显强于多糖单独使用及多糖组合VI组。

(3)对阴虚大鼠下丘脑—垂体—肾上腺素轴(HPA)和下丘脑—垂体—性腺轴(HPG)的影响
实验结束后称取动物体重,水合氯醛麻醉后心尖取血至抗凝管,静置30分钟,3000r/min离心10分钟,取上层血浆,置于-80℃冰箱中保存备用。测定血浆中环腺苷酸(cAMP)、环磷鸟嘌呤核苷(cGMP)、促肾上腺皮质激素释放激素(CRH)、促肾上腺皮质激素(ACTH)含量、血清皮质醇(CORT)、睾酮(TT)、***(LH)、促卵泡激素(FSH)含量。
表2可以看出,与正常组比较,甲状腺混悬液可致模型组大鼠HPA轴功能亢进,即CRH、ACTH、CORT均显著升高(P<0.01),而使HP G轴功能下降,即FSH、LH、TT均显著下降(P<0.01)。各多糖组对HPA及HPG轴均有一定的改善作用,与模型组比较,各多糖组CRH、ACTH、CORT水平均明显下调,而使FSH、LH、TT水平显著上升,表明各多糖组可拮抗阴虚大鼠HPA轴亢进功能,而增强HPG功能。但是综合HPA轴及HPG轴数据,5个多糖组合(多糖组合I组、多糖组合II组、多糖组合III组、多糖组合IV组、多糖组合V组)对大鼠HPA轴及HPG轴功能的调节作用明显强于多糖单独使用及多糖组合VI组。

(4)对阴虚模型大鼠血管氧化相关因子cAMP、cGMP的影响
实验结束后称取动物体重,水合氯醛麻醉后心尖取血至抗凝管,静置30分钟,3000r/min离心10分钟,取上层血浆,置于-80℃冰箱中保存备用。测定血浆中环腺苷酸(cAMP)、环磷鸟嘌呤核苷(cGMP)含量。
由表3可知,与正常组比较,模型组大鼠血浆cAMP、cGMP、c AMP/cGMP均显著升高,各多糖组均能降低阴虚大鼠cAMP、cGMP,说明各多糖组均能缓解cAMP、cGMP紊乱引起的阴虚症状,但是综合cAMP、cGMP、c AMP/cGMP数据,5个多糖组合(多糖组合I组、多糖组合II组、多糖组合III组、多糖组合IV组、多糖组合V组)对大鼠cAMP、cGMP、c AMP/cGMP的调节作用明显强于多糖单独使用及多糖组合VI组。

以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。

1.一种多糖组合物,其特征在于,由肉桂多糖、益智仁多糖、黑芝麻多糖、桑葚子多糖、枸杞多糖和黑木耳多糖组成,所述肉桂多糖、益智仁多糖、黑芝麻多糖、桑葚子多糖、枸杞多糖、黑木耳多糖的质量比为1:1:2:5:3:6、2:2:3:2:5:6、1:1:2:3:5:6、1:1:3:5:4:6或1:1:2:4:3:6;所述肉桂多糖、益智仁多糖、黑芝麻多糖、桑葚子多糖、枸杞多糖或黑木耳多糖由以下制备方法获得:
将肉桂、益智仁、黑芝麻、桑葚子、枸杞或黑木耳加入其12倍量水煎煮1.0~2.0h过滤,得滤液和滤渣;滤渣加入其10倍量水煎煮0.5~1.0h,过滤,合并两次的滤液,在80℃条件下减压浓缩水提液,然后边搅拌边加入4倍体积60%乙醇,静置12h;分离沉淀,60%乙醇洗涤,真空冷冻干燥获得肉桂多糖、益智仁多糖、黑芝麻多糖、桑葚子多糖、枸杞多糖或黑木耳多糖。
2.权利要求1所述多糖组合物在制备改善阴虚症状药物中的应用。
3.根据权利要求2所述应用,其特征在于,所述阴虚症状为多动好斗、烦躁不安、形体消瘦,口燥咽干以及大便干结。
4.权利要求1所述多糖组合物在制备缓解下丘脑—垂体—肾上腺素轴和下丘脑—垂体—性腺轴功能紊乱的药物中的应用。
5.权利要求1所述多糖组合物在制备缓解cAMP和cGMP水平异常的药物中的应用。
6.权利要求1所述多糖组合物的制备方法,其特征在于,包括:
将原料加入原料12倍量水煎煮1.0~2.0h过滤,得滤液和滤渣;滤渣加入其10倍量水煎煮0.5~1.0h,过滤,合并两次的滤液,在80℃条件下减压浓缩水提液,然后边搅拌边加入4倍体积60%乙醇,静置12h;分离沉淀,60%乙醇洗涤,真空冷冻干燥获得多糖提取物,所述原料为肉桂、益智仁、黑芝麻、桑葚子、枸杞或黑木耳;
将肉桂多糖、益智仁多糖、黑芝麻多糖、桑葚子多糖、枸杞多糖和黑木耳多糖混合,获得所述多糖组合物。
