Abstract
以灵芝、罗汉果为培养基制备虫草菌丝体,是用灵芝、罗汉果为培养基,将培养基灭菌后接入虫草菌种,接着培育至菌丝长满培养基,然后将长满菌丝的培养基烘干并粉碎得菌丝体。通过实施本发明,用生物技术把灵芝、罗汉果、北虫草组成另一个特色的结合体——灵芝罗汉果虫草菌丝体,产品的形成不但具有特色、新颖,而且使其营养更富集,而其功能作用及保健功效更加突出。是一种新型特色的保健品。同时,灵芝罗汉果虫草菌丝体的生产制备工艺具有培育周期短、生产制备工艺简单、产品的生产效率高及稳定等特点。此外,形成的产品用途广泛,可以作为制备成药品、保健品、特色食品添加剂的原料。不但有非常好的市场前景,而且产生的经济效益亦会非常显著。

技术领域
本发明属于生物技术领域,具体涉及到一种以灵芝、罗汉果为培养基制备虫草菌丝体。
背景技术
虫草,也叫北虫草,是一种富含虫草素、高蛋白、氨基酸的食用菌类,是上等的滋补佳品。目前人工栽培的北虫草主要是有两类:一类是人工饲养用虫蛹生菌而成,第二类是市场上最常见的,人工饲养用谷类、麦子发酵生菌而成。
北冬虫夏草草长5~8公分,金黄、桔黄色,与青海冬虫夏草同属于真菌门虫草属,有相似的药用和滋补功效。国内外对已经成为诱发虫草子实体的种类研究认为,北虫草食用和药用价值可与传统的冬虫夏草媲美,人工培养的北虫草虫草素含量高于传统冬虫夏草几十倍。
现代医药学研究证明,北冬虫夏草含有丰富的蛋白质、18种氨基酸、17种微量元素、12种维生素: A、B1、B2、B6、B12、C、D、E等含量高于菇类,具有滋肺补肾、止血化痰、扩张气管、镇静、抗各类细菌、降血压等功效。经香港、韩国、日本、加拿大、美国以及国内国十多个单位等世界各国的研究和检测证明,北虫草是传统冬虫夏草的最理想代用品。
将北虫草制备成茶饮料,具抗衰老、抗疲劳、抗癌、抗炎、抗菌、保心、保肝、润肺、补肾壮阳、美颜乌发等多种医疗保健作用。亦不同于一般营养保健食品只对某一方面营养素的不足进行补充和强化,而是在整体上双向调节人体机能平衡,调动机体内部活力,调节人体新陈代谢机能,提高自身免疫能力,促使全部的内脏或器官机能正常化。
随着技术研究不断的深入,通过生物深层发酵技术获得了全新的北虫草产品——北虫草菌丝体。经过相关部门检测:北虫草菌丝体具有北虫草相同的有效成分,其中粗多糖及北虫草多糖和蛋白质的含量远远高于天然北虫草。用生物深层发酵技术来生产北虫草菌丝体,其生产周期短、工艺简单、生产效率高、稳定等特点,是人工培养北虫草产品的主要发展方向。目前生产北虫草菌丝体所用的原料主要粮食的副产物或植物淀粉、大米粉、玉米粉、黄豆粉等,并在上述原料基础上添加些矿物元素为碳、氮源,菌丝体培育出来后再通过烘干并粉碎成粉状物。菌丝体通过烘干后其营养成分会有所流失。另外用这样的工艺生产出来的菌丝体产品除了产品本身具有的功能、功效外,其产品形成的形态差、口感不佳,甚至无任何味道,不能引起人们的注意,所以产品推广效果不佳。
发明内容
本发明的目的就是提供全新的一种以灵芝、罗汉果为培养基制备虫草菌丝体。具体包括:该菌丝体是用灵芝、罗汉果为培养基,将培养基灭菌后接入虫草菌种,接着培育至菌丝长满培养基,然后将长满菌丝的培养基烘干并粉碎得菌丝体。
所述的灵芝,是祖国中医药宝库中的珍品,素有"仙草"之美誉。古今药理与临床研究均证明,灵芝确有防病治病、延年益寿之功效。东汉时期的《神农本草经》、明代著名医药学家李时珍的《本草纲目》,都对灵芝的功效有详细并极为肯定的记载。现代药理学与临床实践进一步证实了灵芝的药理作用,并证实灵芝多糖是灵芝扶正固本、滋补强壮、延年益寿的主要成分。现在,灵芝作为药物已正式被国家药典收载,同时它又是国家批准的新资源食品,无毒副作用,可以药食两用。
科学研究表明,灵芝的药理成分非常丰富,其中有效成份可分为十大类,包括灵芝多糖、灵芝多肽、三萜类、16种氨基酸(其中含有七种人体必需氨基酸)、蛋白质、甾类、甘露醇、香豆精苷、生物碱、有机酸(主含延胡索酸),以及微量元素Ge、P、Fe、Ca、Mn、Zn、等。灵芝对人体具有双向调节作用,所治病种,涉及心脑血管、消化、神经、内分泌、呼吸、运动等各个***,尤其对肿瘤、肝脏病变、失眠以及衰老的防治作用十分显著。
随着灵芝的药用价值被人们广泛认识,今深山老林的野生灵芝资源也在不断的减少。目前,市场上的灵芝产品大多采用人工栽培,通过人工培养的灵芝子实体其品形态跟天然的有变异,但其疗效是相同的。但人工栽培的缺点就是在栽培过程中不可避免的要用到农药与化学方法来培育菌种,栽培管理过程中要用到农药来控制病菌虫害的控制等等,给灵芝子实体带来实际性的化学污染。然而,化学污染往往是人类各种疾病的源泉。另外,灵芝的生物转化率一直都很低,但人工栽培灵芝的周期长,加上生产工艺及管理过程复杂且繁琐使生产成本过高低导致效益低下。
随着技术研究不断的深入,通过生物深层发酵技术获得了全新的灵芝产品——灵芝菌丝体。灵芝菌丝体是由灵芝孢子萌发形成菌丝,而灵芝子实体是通过菌丝不断地分解,吸收养份,在合适的光照,空气,温度等其它条件配合时形成的。子实体成熟后弹射孢子粉,也就形成了灵芝的生活史,灵芝子实体所需要的构成物质,有一半以上的营养物质来源于菌丝体中的贮存养份。经过相关部门检测:灵芝菌粉具有灵芝相同的有效成分,其中粗多糖及灵芝多糖和蛋白质的含量远远高于天然灵芝。所开发出来的菌丝体产品用于失眠健忘、身体虚弱、神经衰弱等作用效果和破壁灵芝孢子粉差不多;对于治疗高血压、冠心病、糖尿病、癌症、有显著效果,所以灵芝菌丝体被冠名为“健康长寿食品”。

所述的罗汉果,是葫芦科多年生藤本植物。被人们誉为“神仙果”。是桂林名贵的土特产。 其叶心形,雌雄异株,夏季开花,秋天结果。中医以其果实入药,含有罗汉果甜苷、多种氨基酸和维生素等药用成分,主治肺热痰火咳嗽、咽喉炎、扁桃体炎、急性胃炎、便秘等。罗汉果因其甜度高、热量低而作为甜味剂广泛使用,常作为肥胖者和糖尿病患者的代用糖。罗汉果是我国特有的珍贵葫芦科植物,素有良药佳果之称。果实中含有丰富的葡萄糖、果糖及多种维生素等,用途广泛,畅销国内外市场,在国际市场上享有很高的声誉。
功能主治:
清热润肺,止咳,利咽,滑肠通便。用于肺火燥咳,咽痛失音,肠燥便秘。
药理作用:
(1)止咳作用:D-甘露醇有止咳作用。又可用于脑水肿,能提高血液渗透压,降低颅内压,脱水作用强于尿素,且持续时间长。还可用于大面积烧伤和烫伤的水肿,防治急性肾功能衰竭病和降低眼球内压,治疗急性青光眼以及代替糖作糖尿病患者的甜味食品或调味剂。
(2)对肠管作用:罗汉果健身茶(含罗汉果77.5%,含茶叶15%,含7.5%罗汉果制剂)对小白鼠离体小肠自发活动无明显影响,但可加强家兔和狗离体小肠自发活动。对乙酰胆碱或氯化钡引起的肠管强直性收缩均有拮抗作用,使肠管松弛而解痉;对肾上腺素引起的肠管松弛也有拮抗作用,使肠管恢复自发性活动。表明罗汉果健身茶对肠管运动机能有双向调节作用。
(3)其它作用:罗汉果健身茶对正常家兔胃生物电无影响,即不影响正常的消化道运动机能。罗汉果健身茶给***灌胃,血压和心电图均无明显影响。
功效作用:
(1)罗汉果味甘性凉,归肺、大肠经,有润肺止咳,生津止渴的功效,适用于肺热或肺燥咳嗽,百日咳及暑热伤津口渴等,此外还有润肠通便的功效。
(2)罗汉果确实有清热润肺的功效,但中医文献中未记载罗汉果有养阴之功效。现代医学研究证实,罗汉果含一种比蔗糖甜300倍的甜味素,但它不产生热量,所以是糖尿病、肥胖等不宜吃糖者的理想替代饮料。
(3)罗汉果的一种名贵药材,性凉味甘,功能清肺润肠。主治百日咳,痰火咳嗽,血燥便秘等症;对于治疗急性气管炎,急性扁桃体炎,咽喉火,急性胃炎都有很好的疗效,用它的根捣碎,敷于患处,可以治顽癣,痈肿,疮疖等;果毛可作刀伤药;用罗汉果少许,冲入开水浸泡,是一种极好的清凉饮料,既可提神生津,又可预防呼吸道感染,常年服用,能延年益寿。罗汉果汁还可用于烹调,清香可口,罗汉果被人们誉为“神仙果”。对于治疗急性气管炎,急性扁桃体炎,咽喉火,急性胃炎都有很好的疗效。
罗汉果为***首批公布的药食两用名贵中药材,其所含罗汉果甜甙比蔗糖甜300倍,不产生热量,是饮料、糖果行业的名贵原料,是蔗糖的最佳替代品。常饮罗汉果茶,可防多种疾病,现代医学证明,罗汉果对支气管炎、高血压等疾病有显著疗效,还是起到防治冠心病、血管硬化、肥胖症的作用。

罗汉果是桂林的特产经济植物,为传统中药和出口商品之一,营养价值高,有清热解暑、化痰止咳、凉血舒骨、清肺润肠和生津止渴等功效;可治急慢性气管炎、咽喉炎、支气管哮喘、百日咳、胃热、便秘、急性扁桃体炎等症,糖尿病患者亦宜服用。用其果泡制的凉茶,久负盛名。还可作为调味品用于炖品、清汤及制糕点、糖果、饼干。除干果出口外,制品尚有冲剂、糖浆、果精、止咳露和浓缩果露等。
本发明是通过以下技术方案来实现的:
以灵芝、罗汉果为培养基制备虫草菌丝体,技术方案中在于:该菌丝体是用灵芝、罗汉果为培养基,将培养基灭菌后接入虫草菌种,接着培育至菌丝长满培养基,然后将长满菌丝的培养基烘干并粉碎得菌丝体。
具体包括如下步骤:
1、灵芝采集及处理:采集干透、无变质、色泽正常的灵芝为原料,接着将灵芝破碎成为玉米粒大小备用。
2、罗汉果采集及处理:采集熟透的罗汉果为原料,将罗汉果晒干至果实含水量低于8%,然后粉碎至能过10-30目备用。
3、制备培养基:培养基是由32%的灵芝、62%的罗汉果粉碎物、6%的玉米粉组成,先把上述各种原料混合起来并搅拌均匀得混合料,接着用3%的红糖水拌入混合料中,并把混合料的湿度含量调至55-60%,然后把混合物料分装于培养容器内,装入量为混合物料在培养容器内的厚度是2-3cm,最后按照常规方法灭菌后得生产用培养基;
4、接种:在无菌环境下把虫草菌种接入生产用培养基,接入固体虫草菌种时,要接入7-8个点的固体菌种,接入虫草液体菌种时,要接入培养基重量的10-12%液体菌种。
5、培菌:将接好种的培养容器移到培养室进行避光培养,在温度是20~23℃、相对湿度为60~65%的环境下培养至虫草菌丝长满培养基。
6、晒干:在温度低于50度的环境下将长满虫草菌丝的培养基晒至物料含水量低于8%。
7、成品:将晒干的培养基粉碎至能过120-200目得成品灵芝罗汉果虫草菌丝体。

Claims (1)
1.以灵芝、罗汉果为培养基制备虫草菌丝体,其特征在于:该菌丝体是用灵芝、罗汉果为培养基,将培养基灭菌后接入虫草菌种,接着培育至菌丝长满培养基,然后将长满菌丝的培养基烘干并粉碎得菌丝体;
具体包括如下步骤:
(1)灵芝采集及处理:采集干透、无变质、色泽正常的灵芝为原料,接着将灵芝破碎成为玉米粒大小备用;
(2)罗汉果采集及处理:采集熟透的罗汉果为原料,将罗汉果晒干至果实含水量低于8%,然后粉碎至能过10-30目备用;
