Abstract
本发明涉及一种具有抗疲劳功效的组合物及制备方法和用途。所述的组合物包括玫瑰花提取物、茯苓提取物以及大枣提取物。所述制备方法包括加入溶剂、浸泡、提取、浓缩以及干燥。本发明制备的组合物具有显著的抗疲劳作用,可用于制备治疗或预防疲劳症状的药品、食品或保健食品。
Description
技术领域
本发明属于健康产品领域,涉及一种以天然植物为原料加工成组合物的制备方法,特别是一种抗疲劳组合物。本发明的组合物可以作为食品、保健食品、或药品使用。
背景技术
疲劳是机体经过复杂的生理生化变化过程所导致的结果。从中枢神经***到骨骼肌细胞再到细胞内的物质代谢过程,中间任何一个环节或者过程的综合变化都可造成疲劳。疲劳不但降低了机体原有的工作能力,而且提示着机体发展到一定程度的伤病状态。人体产生疲劳状态后,假如得不到及时的治疗和消除,将会逐渐积累,最终导致过劳,出现慢性疲劳综合征。
随着社会的发展,人们工作、生活节奏的加快,疲劳已成为21世纪人类健康的大敌。调查显示,我国约80%的白领处于过劳状态,而且白领阶层“过劳死”现象已开始蔓延。因长期疲劳导致的慢性疲劳综合征正慢慢向忙碌的都市人群逼近。由于人们对该病的认识不足,常被忽视,时间久了可导致多种***功能下降,继而引发呼吸、消化、免疫等***性疾病,重者可导致猝死。
目前,针对疲劳的消除仍未发现理想的手段。在西药方面,多数通过补充糖类、蛋白质等能源物质、清除代谢产物、补充抗氧化剂、使用强壮剂等来抗疲劳,虽具起效快、作用明显,但往往含有违禁成分,被国际奥委会所禁用。而中草药在此方面具有的一定的优势,具有良好的调节身体生理机能,作用靶点多,并且不含违禁成分等优点,备受预防医学和运动医学领域的青睐,从中草药中寻求具有抗疲劳活性的有效成分已成为目前研究的热点。
本发明旨在寻找一种具有较好抗疲劳功效的中草药组合物,是以药膳同源的大枣、玫瑰花和茯苓为原料。大枣含有蛋白质、糖类、有机酸、脂肪、人体所需的18种氨基酸以及铁、锌、磷、钙、硒等,同时还含有丰富的维生素A,B2,C及多种生物活性成分。它可以提高机体血糖水平,增强运动耐力。玫瑰花中丰富的多酚、黄酮、原花青素等抗氧化因子可清除体内自由基可以延缓疲劳的出现。茯苓中的三萜成分具有改善大脑机能以及缓解大脑疲劳等功效。本发明将这三种植物经过特殊工艺制备得到具有显著抗疲劳功效的组合物,并经生物活力实验得以确证。
发明内容
本发明要解决的技术问题是:克服现有技术的不足,提供了一种高效抗疲劳的组合物,该组合物可以用作食品、保健食品以及药品,本发明另一个目的是提供了一种抗疲劳组合物的制备方法,该方法简单、绿色环保,适合大规模生产。
本发明是通过下述技术方案实现的:
一种具有抗疲劳功效的组合物,所述组合物是由玫瑰花提取物,大枣提取物以及茯苓提取物组成。
优选地,所述组合物中各提取物质量百分比是:玫瑰花提取物15~80wt%,茯苓提取物1~6wt%,大枣提取物15~80wt%。
优选地,所述组合物中各提取物质量百分比是:玫瑰花提取物20~75wt%,茯苓提取物1~6wt%,大枣提取物20~79wt%。。
优选地,所述组合物中各提取物质量百分比是:玫瑰花提取物43wt%,茯苓提取物4wt%,大枣提取物53wt%。
优选地,所述组合物中各提取物质量百分比是:玫瑰花提取物、茯苓提取与大枣提取物含量分别为42.8wt%、4.0wt%和53.2wt%。
优选地,所述组合物中各提取物质量百分比是:玫瑰花提取物、茯苓提取与大枣提取物含量分别为42.6wt%、4.1wt%和53.3wt%。
上述组合物是由下述方法制备得到的:
所述玫瑰花提取物是由下述方法制备得到的:将玫瑰花瓣或者花蕾,与溶剂按质量比为1∶15~1∶30混合,在50~90℃下搅拌提取1~2h/次,浓缩提取液至50~60℃至密度为1.01~1.04g/mL;干燥即可。
所述茯苓提取物是由下述方法制备得到的:将茯苓与溶剂按质量比为1∶6~1∶12混合并浸泡3~5h后,回流提取3~4h/次,浓缩提取液至50~60℃至密度为1.03~1.08g/mL;干燥即可。
将大枣与纯化水或者自来水按质量比为1∶15~1∶30混合,在80~95℃下搅拌提取60~120min/次,浓缩提取液至50~60℃至固含为0.15~0.20g/mL。
本发明还提供了一种具有抗疲劳功效组合物的制备方法,其包括下述步骤:首先制备玫瑰花提取物,其次为茯苓提取物,最后为大枣提取物,按其优选百分配比,在大枣提取物中加入玫瑰花提取物和茯苓提取物,搅拌干燥,
优选地,玫瑰花提取物、茯苓提取物以及大枣提取物的制备方法如下:
(1)将玫瑰花瓣或者花蕾洗净,与溶剂按质量比为1∶15~1∶30混合,在50~90℃下搅拌提取1~2h/次,优选提取三次,合并三次提取液,浓缩至50~60℃至密度为1.01~1.04g/mL,干燥即可,收率为15~25%。所述溶剂可为水、有机溶剂中的一种或者它们的混合物,所述有机溶剂可以为甲醇、乙醇、异丙醇、正丁醇中的一种或者它们的混合物,优选地使用食品级乙醇,其体积浓度为70~75%。
(2)将茯苓洗净、粉碎,与溶剂按质量比为1∶6~1∶12混合并浸泡3~5h后,回流提取3~4h/次,优选提取两次,合并两次提取液,浓缩至50~60℃至密度为1.03~1.08g/mL,干燥即可,收率为0.3~0.6%。所述有机溶剂可以为甲醇、乙醇、异丙醇、正丁醇中的一种或者它们的混合物,优选地使用食品级乙醇,其体积浓度为70~75%。
(3)将大枣洗净、切片,与纯化水或者自来水按质量比为1∶15~1∶30混合,在80~95℃下搅拌提取60~120min/次,优选提取两次,合并两次提取液,减压浓缩至50~60℃至至固含为0.15~0.20g/mL,并准确测定,根据固含和浓缩液的体积计算浓缩液中固体的含量,收率为30~35%。最后按所需产品配比,加入玫瑰花提取物、茯苓提取物到大枣浓缩液中搅拌混匀,干燥即可。
优选地,所述的干燥方法可以为常压干燥、减压干燥、冷冻干燥以及喷雾干燥。
本发明还提供了一种药物组合物,其含有上述组合物和,可药用或食用辅料。
优选地,所述药物组合物的剂型可以为片剂、分散片、胶囊剂、软胶囊剂、微囊剂、颗粒剂、注射剂、粉针剂、冻干粉针剂、微丸剂、滴丸剂、糖浆剂、散剂、浸膏剂、煎膏剂、口服液体制剂及其他药用或食用上可以接受的剂型。
上述抗疲劳功效组合物和药物组合物可用于制备治疗或预防疲劳症状的药品、食品或保健食品,并在斑马鱼抗疲劳药效实验中得以确证。

实施例
下面通过实施例对本发明作进一步说明。应该理解的是,本发明实施例所述方法仅仅是用于说明本发明,而不是对本发明的限制,在本发明的构思前提下对本发明制备方法的简单改进都属于本发明要求保护的范围。实施例中用到的所有原料和溶剂均购自Sigma Biochemical and Organic Compounds for Research and Diagnostic ClinicalReagents公司。
实施例1:
将25kg干燥的玫瑰花蕾洗净,加入其质量为30倍体积浓度为70%乙醇溶液,在60℃下搅拌提取2h,提取三次,合并三次提取液,浓缩至50℃至密度为1.04g/mL,干燥即可,产率为18.1%。
将100kg干燥的茯苓洗净,粉碎过24目筛,加入其质量为10倍体积浓度为75%乙醇溶液浸泡5h后,回流提取4h,提取两次,合并两次提取液,浓缩至50℃至密度为1.05g/mL,减压干燥,产率为0.42%。
将16kg干燥的大枣洗净,切片(厚度为5~7mm),与纯化水按质量比为1∶30混合,在80℃下搅拌提取100min,提取两次,合并两次提取液,减压浓缩至55℃至固含为0.18g/mL,浓缩液体积为28.3L,计算所得浓缩液中的固体含量为5.1kg,加入玫瑰花和茯苓提取物的质量分别为4.1kg和0.38kg,搅拌均匀喷雾干燥后得到7.2kg药品组合物(批号为Z-1),其中玫瑰花提取物、茯苓提取与大枣提取物质量百分比分别为42.8wt%、4.0wt%和53.2wt%。
实施例2:
将25kg干燥的玫瑰花蕾洗净,加入其质量为30倍体积浓度为70%乙醇溶液,在80℃下搅拌提取1h,提取三次,合并三次提取液,浓缩至60℃至密度为1.02g/mL,干燥即可,产率为19.6%。
将100kg干燥的茯苓洗净,粉碎过24目筛,加入其质量为8倍体积浓度为70%乙醇溶液浸泡5h后,回流提取3h,提取两次,合并两次提取液,浓缩至55℃至密度为1.07g/mL,减压干燥,产率为0.40%。
将16kg干燥的大枣洗净,切片(厚度为5~7mm),与纯化水按质量比为1∶30混合,在90℃下搅拌提取100min,提取两次,合并两次提取液,减压浓缩至55℃至固含为0.16g/mL,浓缩液体积为31.2L,计算所得浓缩液中的固体含量为5.0kg,加入玫瑰花和茯苓提取物的质量分别为4.0kg和0.38kg,搅拌均匀冷冻干燥后得到8.9kg药品组合物(批号为Z-2)其中玫瑰花提取物、茯苓提取与大枣提取物质量百分比分别为42.6wt%、4.1wt%和53.3wt%。
实施例3:组合物对斑马鱼运动的影响评价
斑马鱼抗疲劳药效评价模型出自文献:范能全,彭兰,杨武,张璞,江静子.红景天对斑马鱼游泳和耐低氧能力的影响的初步研究,《药物分析杂志》,2015,8:1342-1345。其全部内容在此引入并作参考。
实验动物为野生型AB品系,每个实验组为均30尾斑马鱼,在28℃培养箱中孵育。正常对照组为养鱼水处理正常斑马鱼;用亚硫酸钠诱导斑马鱼,以此作为斑马鱼缺氧疲劳模型;取浓度为5、17和50μg/mL组合物(批号为Z-1)进行评价,是以水溶给样方式处理模型组斑马鱼,并以中华跌打丸为阳性对照组。处理结束后,每个实验组随机选择的10尾斑马鱼,测定斑马鱼的运动总距离(S)(实验结果如表1),对运动能力影响评价的计算公式如下:

注:与模型对照组比较:p<0.001;*:显著性差异.
模型对照组斑马鱼总运动距离(4261mm)与正常对照组(10599mm)比较P<0.001,提示模型构建成功;阳性药中华跌打丸组(1000μg/mL),斑马鱼总运动距离为9262mm,与模型对照组比较P<0.001,运动改善作用为79.9%,提示中华跌打丸对亚硫酸钠诱发的斑马鱼缺氧疲劳具有极显著的抗疲劳作用。供试品5、17和50μg/mL浓度组斑马鱼总运动距离分别为:7172、8454和9939,其运动改善作用分别为48.4%、67.7%和90.0%,各组与模型对照组比较P<0.001,提示组合物各浓度对亚硫酸钠诱发的斑马鱼缺氧疲劳具有显著的抗疲劳作用,且呈现浓度相关性增加。
实施例4:组合物对斑马鱼乳酸代谢的影响评价
实验动物为野生型AB品系,每个实验组为均80尾斑马鱼,在28℃培养箱中孵育。正常对照组为养鱼水处理正常斑马鱼;用亚硫酸钠诱导斑马鱼,以此作为斑马鱼缺氧疲劳模型;取浓度为5、17和50μg/mL组合物(Z-1)进行评价,是以水溶给样方式处理模型组斑马鱼,并以中华跌打丸为阳性对照组。处理结束后,测定每个实验组斑马鱼体内的乳酸含量(C)(实验结果如表2),对乳酸代谢改善作用计算公式如下:

模型对照组斑马鱼体内乳酸含量(0.917mmol/gprot)与正常对照组(0.338mmol/gprot)比较P<0.001,提示模型构建成功;阳性药中华跌打丸组(1000μg/mL),斑马鱼体内乳酸含量为0.531mmol/gprot,与模型对照组比较p<0.001,其乳酸代谢促进作用为66.7%,提示中华跌打丸显著促进亚硫酸钠诱发的斑马鱼缺氧疲劳斑马鱼体内的乳酸代谢。组合物5、17和50μg/mL浓度组斑马鱼体内乳酸含量分别为0.814、0.655和0.355mmol/gprot,乳酸代谢促进作用分别为17.8%、45.2%和97.0%,各组与模型对照组比较p<0.001,提示组合物各浓度均可显著促进亚硫酸钠诱发的斑马鱼缺氧疲劳斑马鱼体内的乳酸代谢,且呈现浓度相关性增加。
Claims (11)
1.一种具有抗疲劳功效的组合物,其特征在于:是由玫瑰花提取物、茯苓提取物以及大枣提取物组成。
2.根据权利要求1所述的组合物,其特征在于:玫瑰花提取物15~80wt%,茯苓提取物1~6wt%,大枣提取物15~80wt%。
3.根据权利要求1所述的组合物,其特征在于:玫瑰花提取物20~75wt%,茯苓提取物1~6wt%,大枣提取物20~79wt%。
4.根据权利要求1所述的组合物,其特征在于:玫瑰花提取物43wt%,茯苓提取物4wt%,大枣提取物53wt%。
5.根据权利要求1所述的组合物,其特征在于:
所述玫瑰花提取物是由下述方法制备得到的:将玫瑰花瓣或者花蕾,与溶剂按质量比为1∶15~1∶30混合,在50~90℃下搅拌提取1~2h/次,浓缩提取液至50~60℃至密度为1.01~1.04g/mL;
所述茯苓提取物是由下述方法制备得到的:将茯苓与溶剂按质量比为1∶6~1∶12混合并浸泡3~5h后,回流提取3~4h/次,浓缩提取液至50~60℃至密度为1.03~1.08g/mL;
将大枣与纯化水或者自来水按质量比为1∶15~1∶30混合,在80~95℃下搅拌提取60~120min/次,浓缩提取液至50~60℃至固含为0.15~0.20g/mL。
6.权利要求1-5任一所述组合物的制备方法,其特征在于:依次制备玫瑰花提取物、茯苓提取物和大枣提取物,按百分配比,在大枣提取物中加入玫瑰花提取物和茯苓提取物,搅拌干燥即可。
7.根据权利要求6所述的制备方法,其特征在于:
制备玫瑰花提取物的步骤为:将玫瑰花瓣或者花蕾与溶剂按质量比为1∶15~1∶30混合,在50~90℃下搅拌提取1~2h/次,浓缩提取液至50~60℃至密度为1.01~1.04g/mL;所述溶剂选自水、有机溶剂中的一 种或者它们的混合物,所述有机溶剂选自甲醇、乙醇、异丙醇、正丁醇中的一种或者它们的混合物,优选地使用食品级乙醇,其体积浓度为70~75%;
制备茯苓提取物的步骤为:将茯苓洗与溶剂按质量比为1∶6~1∶12混合并浸泡3~5h后,回流提取3~4h/次,浓缩提取液至50~60℃至密度为1.03~1.08g/mL;所述有机溶剂选自甲醇、乙醇、异丙醇、正丁醇中的一种或者它们的混合物,优选地使用食品级乙醇,其体积浓度为70~75%;
制备大枣提取物的步骤为:将大枣与纯化水或者自来水按质量比为1∶15~1∶30混合,在80~95℃下搅拌提取60~120min/次,浓缩提取液至50~60℃至固含为0.15~0.20g/mL。
8.根据权利要求6所述的制备方法,其特征在于:所述的干燥方法可选自常压干燥、减压干燥、冷冻干燥和喷雾干燥。
9.一种药物组合物,其含有权利要求1-5任一的组合物,和可药用或食用辅料。
10.根据权利要求9的药物组合物,其是片剂、分散片、胶囊剂、软胶囊剂、微囊剂、颗粒剂、注射剂、粉针剂、冻干粉针剂、微丸剂、滴丸剂、糖浆剂、散剂、浸膏剂、煎膏剂、口服液体制剂及其他药用或食用上可以接受的剂型。
11.权利要求1或9的组合物在制备治疗或预防疲劳症状的药品、食品或保健食品方面的用途。
