Abstract
本发明公开了一种黑木耳‑辣木复合压片糖果及其制备方法与应用,属于食品加工技术领域。本发明提供的糖果各组分及重量百分比为:黑木耳粉10%‑50%,辣木粉10%‑50%,甘露醇5%‑20%,低聚木糖5%‑20%,木糖醇5%‑30%,L‑***糖5%‑20%,以上述物料总重为基准,黑木耳提取液添加量为0.1mL/g‑0.5mL/g。本发明产品具有止咳祛痰的效果,用黑木耳提取液做粘合剂制备的糖果在营养成分和功效成分上具有比较突出的优势,产品中大大减少了食品添加剂用量、蛋白质等营养成分更足、黑木耳多糖等功效成分更高。

技术领域
本发明涉及一种黑木耳-辣木复合压片糖果及其制备方法与应用,属于食品加工技术领域。
背景技术
黑木耳富含多糖、黑色素、腺苷、蛋白质、粗纤维、胶质、铁、钙、磷和B族维生素等,其“性平、味甘”,归肺、胃、肝、脾、肾和大肠经,“益气不饥,轻身强志”(明代李时珍《本草纲目》),“清肺润肺”、“利五脏、排毒气”(《食疗本草》)。
辣木树原产印度北部喜马拉雅山南麓,其根、叶和嫩果均可食用,性味辛、微温、无毒,含有丰富而惊人的营养,号称为高钙、高蛋白质、高纤维、高矿物质、高维生素、低脂质的“奇迹之树”,于2012年被***批准为新资源食品。辣木含有氨基甲酸酯类、多酚类、绿原酸类、长链葡萄糖苷、磷酸酯类、甾醇、三萜以及木脂素等活性成分。
黑木耳-辣木复合片具有以下功能作用:
1、高效补充铁、钙元素和维生素
缺铁性贫血与缺钙是世界卫生组织确认的比较严重和广泛的营养缺乏症,二者往往相伴而生。研究证明,钙、铁等矿物质对预防高血压、高血脂、高血糖等慢性疾病有独特作用。黑木耳是含铁量最高的食物(185mg/100g),是猪肝7倍,是菠菜和芹菜的20多倍,而且黑木耳主要含二价铁离子,能直接被人体吸收,优于多数食物中的三价铁离子,非常适合缺铁性贫血人群。辣木叶其所含的钙质(2000.3mg/100g)是牛奶的4倍,铁是菠菜的3倍,VC是柑桔的7倍,VA是胡萝卜的4倍,是天然优质的钙、铁、钾和维生素补充剂。人体对矿物质的吸收率一般为20-30%,配方中的低聚木糖配合钙源及维生素,可以调节机体肠道健康,从调节吸收环境、增加吸收动力和开启吸收通道(钙结合蛋白表达量增加,激活了钙扩散通道)三个方面提高机体对钙、铁等的矿物质的吸收率。
2、改善微循环、清肺、清肠胃
传统医学认为黑木耳清肺润肺,补血活血,强志益气,镇静止痛。《食疗本草》记载黑木耳“利五脏、排毒气”,对异物具有消融分解及促进体内和肺部外来纤维和粉尘排出体外的作用,被称为“呼吸道和胃肠道的清洁工”(高艳荣,2011;张介驰,2011),是纺织、矿山、理发等空气污染严重工种工人的日常保健品。黑木耳黑色素与多糖共同作用,可以改善肺泡内血液循环,铁离子可提升血氧输送能力,帮助肺部排出异物;辣木具有抗菌、抗炎、抗病毒作用,二者复合可治疗呼吸道炎症,增强清肺润肺作用,减轻空气污染对人体伤害,是当之无愧的“人体清道夫”。
3、显著降血脂、显著降血糖、抗氧化、提升细胞修复能力、有力保障心脑血管健康。
发明内容
为解决现有技术的不足,本发明提供了一种黑木耳-辣木复合压片糖果及其制备方法与应用,采用的技术方案如下:
本发明的目的在于提供一种黑木耳-辣木复合压片糖果,该糖果各组分及重量百分比为:黑木耳粉10%-50%,辣木粉10%-50%,甘露醇5%-20%,低聚木糖5%-20%,木糖醇5%-30%,L-***糖5%-20%,以上述物料总重为基准,黑木耳提取液添加量为0.1mL/g-0.5mL/g。
优选地,所述黑木耳提取液,固形物含量为4.0%-5.0%。
优选地,所述黑木耳提取液,固形物含量为4.5%。
优选地,所述糖果,各组分及重量百分比为:黑木耳粉37.5%,辣木粉12.5%,甘露醇10%,低聚木糖10%,木糖醇20%,L-***糖10%,以上述物料总重为基准,黑木耳提取液添加量为0.3mL/g。
优选地,所述黑木耳提取液,制备方法为:将黑木耳加水泡发、清洗,加入40倍~60倍水,匀浆0.5min~3min,于60~90℃水浴提取两次,每次提取60min~120min,在8000rpm/min~10000rpm/min下离心,取上清液合并,向每升上清液中滴加50μL~150μL的含有维生素E的橄榄油进行旋转蒸发浓缩,浓缩至固形物含量为4.0%~5.0%,获得黑木耳提取液;所述维生素E,浓度为100mg/mL。
本发明还提供一种如权利要求1-5所述任一糖果的制备方法,其特征在于,步骤如下:
1)将黑木耳加水泡发、清洗,加入40倍~60倍水,匀浆0.5min~3min,于60~90℃水浴提取两次,每次提取60min~120min,在8000rpm/min~10000rpm/min下离心,取上清液合并,向每升上清液中滴加50μL~150μL的含有维生素E的橄榄油进行旋转蒸发浓缩,浓缩至固形物含量为4.0%~5.0%,获得黑木耳提取液;
2)以步骤1)获得的黑木耳提取液、木糖醇、低聚木糖和L-***糖混合,加热至沸,搅拌溶解,放冷,过滤,滤液做为粘合剂,将黑木耳粉,辣木粉和甘露醇过筛后混匀制粒,烘干、整粒,压片获得黑木耳-辣木复合压片糖果。
优选地,步骤1)所述黑木耳,品种为黑木耳2号,黑威10号,黑威15,黑威单片中的一种或几种。
更优选地,所述黑木耳粉的来源品种为黑木耳“黑29”。
本发明方法适用于所有黑木耳品种,优选为黑木耳2号,黑威10号,黑威15,黑威单片,更优选为黑木耳2号,黑木耳2号(黑龙江省科学院微生物研究所通过富集沙棘黄酮黑木耳栽培技术生产,黑木耳“黑29”菌株是通过国家食用菌品种认定(认定证书编号:国品认菌2007018),品种名称:黑木耳2号(黑29),该品种生产的黑木耳子实体色黑肉厚,营养足,品质佳)。
优选地,步骤1)所述维生素E,浓度为100mg/mL。
优选地,所述方法,具体步骤为:
1)将黑木耳加水泡发、清洗,加入50倍水,匀浆2min,于80℃水浴提取两次,每次提取90min,在8000rpm/min~10000rpm/min下离心,取上清液合并,向每升上清液中滴加100μL的含有维生素E的橄榄油进行旋转蒸发浓缩,浓缩至固形物含量为4.5%,获得黑木耳提取液;
2)将黑木耳粉,辣木粉和甘露醇过筛后混匀,然后以步骤1)获得的黑木耳提取液、木糖醇、低聚木糖和L-***糖混合做为粘合剂,混合溶解后加热至沸,过滤后制粒,烘干后整粒,压片获得黑木耳-辣木复合压片糖果。
以上所述任一糖果在制备糖果、保健品、药品中的应用。
本发明有益效果:
1、本发明采用黑龙江大兴安岭生产的色黑、肉厚、营养足、品质佳的“黑29”品种(国家农作物委员会品种认定)黑木耳和云南楚雄辣木叶为原料,通过添加低聚木糖和L-***糖科学配方,摒弃了传统压片的粘合剂(如聚维酮等)和赋形剂(如硬脂酸镁、滑石粉等),而采用高效提取的黑木耳提取液为粘合剂制备而成,使产品中的活性物质含量更高、效果更好,真正赋予了产品“天然、营养、健康、绿色有机”的理念。
2、本发明产品具有显著的止咳化痰作用,本品在生物活性方面协同增效,在营养素补充方面达到互补,实现铁、钙及多种维生素的高效吸收、促进排毒、预防“三高”。具有清肺清肠胃、降血脂、降血糖、抗氧化、防衰老及提升免疫力的功效。
3、本发明提高了黑木耳原料在加工产品中的占比,同时添加黑木耳多糖等生物提取的活性成分,实现了以黑木耳为主的产品深加工的开发宗旨。添加的黑木耳活性成分浓缩液赋予了产品更好地营养和功效,本发明使人们真正方便快捷的享用到天然营养健康的黑木耳深加工健康食品。
4、按照传统片剂制作技术,若要达到片剂紧致感觉需要添加化学粘合剂如聚维酮等才能实现,而本发明采用纯天然的黑木耳提取液作为生物粘合剂,由于黑木耳提取液含有黑木耳粘多糖等粘稠成分,粘合效果非常好,大大改变了压片需要加如化学粘合剂的制备工艺,且片剂成型效果较好。
5、在黑木耳提取工艺浓缩上解决了黑木耳提取液粘稠,减压浓缩会导致起泡沫,使得浓缩操作过程比较困难的技术瓶颈,使黑木耳提取液产业化成为可能。经反复试验本发明在每升黑木耳提取液旋转蒸发浓缩过程中滴加80μL、100mg/mL的维生素E橄榄油溶液效果最好,此项技术大大提高了黑木耳提取液旋转蒸发浓缩的效率。

下面结合具体实施例对本发明做进一步说明,但本发明不受实施例的限制。
实施例1:
本实施例提供了一种黑木耳-辣木复合压片糖果,具体配方如下:
黑木耳粉37.5g,辣木粉12.5g,甘露醇10g,低聚木糖10g,木糖醇20g,L-***糖10g,黑木耳提取液30mL。
黑木耳提取液固形物含量为4.5%。
黑木耳为黑木耳2号(黑29)。
按照上述配方制备黑木耳-辣木复合压片糖果的方法,具体步骤如下:
1)将黑木耳加水泡发、清洗,加入50倍水,匀浆2min,于80℃水浴提取两次,每次提取90min,在8000rpm/min~10000rpm/min下离心,取上清液合并,向每升上清液中滴加100μL的含有维生素E的橄榄油(维生素E含量100mg/ml)进行旋转蒸发浓缩,浓缩至固形物含量为4.5%,获得黑木耳提取液;
2)以步骤1)获得的黑木耳提取液、木糖醇、低聚木糖和L-***糖混合,加热至沸,搅拌溶解,放冷,过滤,滤液做为粘合剂,将黑木耳粉,辣木粉和甘露醇过筛后混匀制粒,60℃烘干、整粒,压片获得黑木耳-辣木复合压片糖果。每片0.40g,单片进行单独铝塑包装,装瓶,封口为产品成品。
实施例2

实施例5
按照实施例1配方(最优配方)制备黑木耳-辣木复合压片糖果的方法,具体步骤如下:
1)将黑木耳加水泡发、清洗,加入40倍水,匀浆3min,于60℃水浴提取两次,每次提取120min,在8000rpm/min下离心20min,取上清液合并,向每升上清液中滴加50μL的含有维生素E的橄榄油(维生素E含量100mg/ml)进行旋转蒸发浓缩,浓缩至固形物含量为5.0%,获得黑木耳提取液;
2)以步骤1)获得的黑木耳提取液、木糖醇、低聚木糖和L-***糖混合,加热至沸,搅拌溶解,放冷,过滤,滤液做为粘合剂,将黑木耳粉,辣木粉和甘露醇过筛后混匀制粒,60℃烘干、整粒,压片获得黑木耳-辣木复合压片糖果。每片0.40g,单片进行单独铝塑包装,装瓶,封口为产品成品。
实施例6

2)以步骤1)获得的黑木耳提取液、木糖醇、低聚木糖和L-***糖混合,加热至沸,搅拌溶解,放冷,过滤,滤液做为粘合剂,将黑木耳粉,辣木粉和甘露醇过筛后混匀制粒,60℃烘干、整粒,压片获得黑木耳-辣木复合压片糖果。每片0.40g,单片进行单独铝塑包装,装瓶,封口为产品成品。
实施例7
将实施例1-5所制备的糖果,对硬度、脆裂度、均匀度进行了比较,结果如表1所示。

实施例7
按照实施例1配方(最优配方),以不同品种的黑木耳为原料制备糖果,对硬度、崔烈度、均匀度进行了比较,结果如表2所示。
黑木耳品种:黑29,黑威单片,黑威10号,黑威15号,即就是“黑29”、黑威单片,黑威10号,黑威15号的黑木耳菌株通过栽培获得的黑木耳子实体干品制成粉作为本发明的原料。
配方:黑木耳粉37.5g,辣木粉12.5g,甘露醇10g,低聚木糖10g,木糖醇20g,L-***糖10g,黑木耳提取液30mL。黑木耳提取液固形物含量为4.5%。

由表2数据可知,与黑威单片,黑威10号,黑威15号这三个品种的黑木耳生产的产品比较,用“黑29“的黑木耳,利用“黑29”菌株通过栽培获得的黑木耳子实体制成粉作为原料制备的黑木耳片剂产品效果最佳,由于“黑29”黑木耳营养成分及功效成分如黑木耳多糖含量等较高,提取液粘稠,作为压片粘合剂,其粘合效果非常好,所以硬度评分为90分,脆裂度评分为92分,均匀度评分为90分,各个指标均比用黑威单片、黑威10号、黑威15号的黑木耳提取液压片效果好。实施例8
添加提取液的黑木耳-辣木压片糖果(实施例1)和用其他粘合剂的黑木耳-辣木经比较发现用黑木耳提取液做粘合剂的产品在产品天然健康、营养成分和功效成分上具有比较突出的优势。产品中食品添加剂用量大大减少、蛋白质等营养成分更足、黑木耳多糖等功效成分更高,而感官品质与传统压片方法获得的产品相当,结果见表3。

黑木耳提取液作压片粘合剂相比于聚维酮做压片粘合剂具有更好的营养成分和功效成分,蛋白质含量由9.8%提高到11.2%,提高了14%;黑木耳多糖含量由5.2%提高到6.8%,提高了30%;黑木耳腺苷含量由0.9%提高到1.2%,提高了33%;其他如黑色素、钙铁含量等均有提高。而且片剂产品经过质检表明复合片剂的质量标准。片剂质检包括检查硬度,脆碎度,崩裂时限,重量差异,均匀度,含量及微生物限度等。
硬度检查方法:在1米高处自然落下不裂即为合格。片剂脆碎度指在规定时间内不能掉渣,检查方法:片重为0.65g或以下者取若干片,使其总重约为6.5g;片重大于0.65g者取10片。用吹风机吹去脱落的粉末,精密称重,置圆筒中,转动100次。取出,同法除去粉末,精密称重,减失重量不得过1%,且不得检出断裂、龟裂及粉碎的片。用黑木耳提取液做压片粘合剂的产品片剂质检结果表明合格,结果见表4。

实施例9:止咳化痰动物实验
1、片剂产品对氨水引咳小鼠的止咳作用
80只小鼠随机分成8组:片剂产品1(实施例1,黑木耳提取物作压片粘合剂)和片剂产品2(聚维酮作压片粘合剂)低、中、高剂量组(剂量分别为50、100、200mg/kg),阳性对照组可待因组(30mg/kg)和空白对照组。以20mL/kg体积给小鼠分别进行连续10d的灌胃给药,1次/d,给予可待因组和空白对照组小鼠相同体积的溶媒(0.5%CMC-Na),在最后一次给药1h后进行实验。实验前0.5h给可待因组注射磷酸可待因(0.03g/kg)。
在棉球中注入0.2mL的浓氨水,放入1L倒扣的钟罩内,随后将小鼠快速置于钟罩内。观察小鼠的咳嗽动作(张大嘴,腹肌收缩,有时会有咳嗽声),记录小鼠的咳嗽潜伏期(小鼠放入至出现咳嗽的时间)及在3min内咳嗽的次数。咳嗽抑制率(%)=(空白对照组平均咳嗽数-给药组平均咳嗽数)/空白对照组平均咳嗽数×100%;止咳活性(%)=给药组咳嗽抑制率/可待因组咳嗽抑制率×100%。
止咳效果见表5。咳嗽相关指标包括咳嗽潜伏期、咳嗽次数、咳嗽抑制率及止咳活性。与空白对照组比较,片剂产品1中剂量组和高剂量组小鼠的咳嗽次数均减少(中剂量组P<0.05;高剂量组P<0.01),而咳嗽潜伏期延长(中剂量组P<0.05;高剂量组P<0.01);片剂产品1的中、高剂量组小鼠的潜伏期、咳嗽次数与对照组比较差异有统计学意义(中剂量组P<0.05;高剂量组P<0.01),低剂量组小鼠的潜伏期最短,咳嗽次数最多;而高剂量组小鼠的潜伏期最长,咳嗽次数最少。片剂产品1中剂量组能明显降低氨水导致的小鼠咳嗽次数,止咳活性为45.1%,咳嗽抑制率为25.3%。片剂产品1高剂量组能显著降低氨水导致的小鼠的咳嗽次数、延长引咳潜伏期,止咳活性为55.5%,咳嗽抑制率为31.2%。而片剂产品2的高剂量组止咳活性为43.8%,咳嗽抑制率为24.6%。因此片剂产品1在止咳试验效果上远远好于片剂产品2。

2、片剂产品对小鼠酚红排泌反应的影响
小鼠分组及灌胃方式同上。实验前12h小鼠禁食不禁水,最后一次给药0.5h后,按照10mL/kg体积注射5%的酚红溶液于各组小鼠的腹腔内。0.5h后处死,解剖,剥去气管周围的组织,将甲状软骨下至气管分支处一段气管剪下放入试管中,注入0.1mL的1mol/L NaOH和2mL生理盐水(注意使体积完全一致),盖盖,充分振荡,在546nm处测定上清液吸光度(A)值。
8组小鼠的酚红排泌量比较见表6。结果表明,片剂产品1高剂量组小鼠的酚红排泌量明显高于空白对照组(中剂量组P<0.05,高剂量组P<0.01);而片剂产品2的低、中剂量组与空白对照组比较差异无统计学意义,高剂量组小鼠的酚红排泌量高于空白对照组(P<0.05)。在促进酚红***量方面,片剂产品1的效果好于片剂产品2。

片剂产品1的高剂量组能显著抑制小鼠咳嗽次数(P<0.01),具有使气管段的酚红排泌量显著上升的作用(P<0.01),起重要的止咳和化痰作用。本发明优选了合适范围黑木耳提取液,黑木耳提取液中主要活性成分(黑木耳多糖)作为免疫活性多糖,调节机体免疫功能,本发明产品具有止咳祛痰效果。
虽然本发明已以较佳的实施例公开如上,但其并非用以限定本发明,任何熟悉此技术的人,在不脱离本发明的精神和范围内,都可以做各种改动和修饰,因此本发明的保护范围应该以权利要求书所界定的为准。
Claims (10)
1.一种黑木耳-辣木复合压片糖果,其特征在于,各组分及重量百分比为:黑木耳粉10%-50%,辣木粉10%-50%,甘露醇5%-20%,低聚木糖5%-20%,木糖醇5%-30%,L-***糖5%-20%,以上述物料总重为基准,黑木耳提取液添加量为0.1mL/g-0.5mL/g。
2.根据权利要求1所述的糖果,其特征在于,所述黑木耳提取液,固形物含量为4.0%-5.0%。
3.根据权利要求2所述的糖果,其特征在于,所述黑木耳提取液,固形物含量为4.5%。
4.根据权利要求1所述的糖果,其特征在于,各组分及重量百分比为:黑木耳粉37.5%,辣木粉12.5%,甘露醇10%,低聚木糖10%,木糖醇20%,L-***糖10%,以上述物料总重为基准,黑木耳提取液添加量为0.3mL/g。
5.根据权利要求1-4所述任一的糖果,其特征在于,所述黑木耳提取液,制备方法为:将黑木耳加水泡发、清洗,加入40倍~60倍水,匀浆0.5min~3min,于60~90℃水浴提取两次,每次提取60min~120min,在8000rpm/min~10000rpm/min下离心,取上清液合并,向每升上清液中滴加50μL~150μL的含有维生素E的橄榄油进行旋转蒸发浓缩,浓缩至固形物含量为4.0%~5.0%,获得黑木耳提取液;所述维生素E,浓度为100mg/mL。
6.一种如权利要求1-5所述任一糖果的制备方法,其特征在于,步骤如下:
1)将黑木耳加水泡发、清洗,加入40倍~60倍水,匀浆0.5min~3min,于60~90℃水浴提取两次,每次提取60min~120min,在8000rpm/min~10000rpm/min下离心,取上清液合并,向每升上清液中滴加50μL~150μL的含有维生素E的橄榄油进行旋转蒸发浓缩,浓缩至固形物含量为4.0%~5.0%,获得黑木耳提取液;
2)以步骤1)获得的黑木耳提取液、木糖醇、低聚木糖和L-***糖混合,加热至沸,搅拌溶解,放冷,过滤,滤液做为粘合剂,将黑木耳粉,辣木粉和甘露醇过筛后混匀制粒,烘干、整粒,压片获得黑木耳-辣木复合压片糖果。
7.根据权利要求6所述的方法,其特征在于,步骤1)所述黑木耳,品种为黑木耳2号,黑威10号,黑威15,黑威单片中的一种或几种。
8.根据权利要求6所述的方法,其特征在于,步骤1)所述维生素E,浓度为100mg/mL。
9.根据权利要求6所述方法,其特征在于,具体步骤为:
1)将黑木耳加水泡发、清洗,加入50倍水,匀浆2min,于80℃水浴提取两次,每次提取90min,在8000rpm/min~10000rpm/min下离心,取上清液合并,向每升上清液中滴加100μL的含有维生素E的橄榄油进行旋转蒸发浓缩,浓缩至固形物含量为4.5%,获得黑木耳提取液;
2)以步骤1)获得的黑木耳提取液、木糖醇、低聚木糖和L-***糖混合,加热至沸,搅拌溶解,放冷,过滤,滤液做为粘合剂,将黑木耳粉,辣木粉和甘露醇过筛后混匀制粒,烘干、整粒,压片获得黑木耳-辣木复合压片糖果。
10.权利要求1-5所述任一糖果在制备糖果、保健品、药品中的应用。
