与传统的抗生素不同,两栖类皮肤抗菌肽是通过干扰原核细胞膜结构导致质膜通透性增大,从而引起抑菌或杀菌作用,病原体对其不易产生抗药性。两栖类抗菌肽除了具有高效、广谱且不易产生耐药性的抗菌活性外,还具有抗肿瘤,病毒,原虫等活性,且对人体正常细胞几乎无作用。因此,在耐抗生素病原菌株不断出现,病毒和肿瘤等疾病对类健康造成极具威胁的今天,抗菌肽将有望成为新一代的抗病原微生物及肿瘤的药物。据国内外文献报道,已从各种生物来源分离得到不同的活性多肽,而且有些已进入临床治疗。如从爪蟾(Xenopus laevis)皮肤分泌液获得的活性多肽Magainin具广谱抗微生物作用,同时具有抗肿瘤活性,在美国已获准作为广谱抗菌药物,其基因工程产品已进入三期临床;Intrabiotics公司生产的活性多肽IB-367已获准用于治疗癌症病人因多种细菌感染而引起的口腔溃疡一期临床试验;Applied Microbiology公司与Astra公司合作用活性多肽nisin治疗胃溃疡的临床试验也取得良好的疗效。
我国拥有丰富的两栖类动物资源,它是中国生物资源的重要组成部分,其中很多物种具有食用或药用等价值,具有很大的开发利用潜力。两栖动物由于长期生活在潮湿、阴暗、微生物大量滋生的环境下,在长期自然进化过程中形成了三套防御机制:(1)物理屏障,粘液腺分泌大量糖蛋白和蛋白聚糖,包裹在皮肤外表面,阻碍病原菌的侵入;(2)获得性免疫***,两栖动物皮肤中含有抗体IgG;(3)先天免疫***,主要由大量的抗菌肽构成。目前,对两栖类中蛙类抗菌肽尤其是皮肤和肠道抗菌肽研究的最清楚。自Zasloff从水蛙皮肤中分离到Magainins后,相继从不同的蛙类中分离到许多具有广谱的抑菌活性的抗菌肽家族(如 Dermaseptins、Temporins、Ranatuerins、Brevinins、Tigerinins),一些还具有独特的抗真菌活性。我国对两栖类药物的应用有悠久的历史,但对其活性成分和药理性质的研究主要集中于生物碱等有机小分子,对其皮肤活性肽类物质的研究还鲜有报道。海南湍蛙(Amolops hainanensis)为蛙科湍蛙属的两栖动物,是中国的特有物种,分布于海南等地。
本发明的海南湍蛙(Amolops hainanensis)环状抗菌肽Hainanenin-5分子量小,结构简单,仅含一个二硫键,方便化学合成及基因工程制备。相比较两栖动物来源的已报道的其他家族抗菌肽,Hainanenin-5抗微生物活性更强,且具有广谱的抗微生物活性,对***、革兰氏阴性细菌和真菌均有活性,其中包括大量临床分离耐药菌株,此外还具有溶血性低有益特点,均使Hainanenin-5成为外用抗菌药物开发的优良模板。
发明内容
本发明提供一种具有很强的抗微生物活性的来源于海南湍蛙(Amolops hainanensis)的一种抗微生物肽Hainanenin-5及其基因、分离纯化、化学合成分析和应用。
为了实现本发明的目的,本发明提供了如下技术方案:
海南湍蛙抗微生物肽Hainanenin-5是从两栖类海南湍蛙皮肤分泌液中先经Sephadex G-50凝胶过滤,然后反相高压液相层析(RP-HPLC)分离得到的一种C端含有七元Rana Box的环状多肽,分子量为2337.91道尔顿,等电点为10.11。多肽全序列一级结构为:苯丙氨酸 丙氨酸 亮氨酸 甘氨酸 丙氨酸 缬氨酸 苏氨酸 赖氨酸 精氨酸 亮氨酸 脯氨酸 丝氨酸 亮氨酸 苯丙氨酸 半胱氨酸 亮氨酸 异亮氨酸 苏氨酸 精氨酸 赖氨酸 半胱氨酸。含有4个碱性氨基酸残基(2个精氨酸和2个赖氨酸),无酸性氨基酸残基,静电荷为+4,说明它是一种碱性多肽。第15位和第21位的的半胱氨酸形成分子内二硫键。
构建了海南湍蛙(Amolops hainanensis)皮肤cDNA文库,从中筛选得到Hainanenin-5的前体序列,基因测序结果表明编码海南湍蛙(Amolops hainanensis)Hainanenin-5前体的基因由321个核苷酸组成,自5’端至3’端序列为:

Hainanenin-5的化学合成方法:根据编码海南湍蛙(Amolops hainanensis)抗菌肽hainanenin-5成熟肽的氨基酸序列,用自动多肽合成仪合成其全序列;通过HPLC反相柱层析脱盐纯化,并确定其纯度大于95%;用基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)测定其分子量。
化学合成得到的高纯度Hainanenin-5抗微生物肽可以溶于灭菌超纯水,用于药理活性检测。
本发明的有益效果在于从海南湍蛙皮肤分泌物冻干粉中分离纯化得到抗微生物肽Hainanenin-5,从已构建海南湍蛙(Amolops hainanensis)cDNA文库中克隆得到编码抗微生物肽Hainanenin-5前体的基因,通过化学合成方法得到成熟肽Hainanenin-5。该环状抗菌肽hainanenin-5分子量小,结构简单,仅含一个二硫键,方便化学合成及基因工程制备。相比较两栖动物来源的已报道的其他家族抗菌肽,Hainanenin-5抗微生物活性更强,且具有广谱的抗微生物活性,对***、革兰氏阴性细菌和真菌均有活性,其中包括大量临床分离耐药菌株,此外还具有溶血性低有益特点,均使Hainanenin-5成为外用抗菌药物开发的优良模板。

具体实施方式
下面结合技术方案详细叙述本发明的具体实施例,但本发明的内容并不局限于此。
实施例1
Hainanenin-5的分离纯化:选取健康状况良好的成年海南湍蛙(A.hainanensis)(n=5),用超纯水淋洗背部。采用电刺激法收集皮肤分泌物,用电击器对蛙背部皮肤进行电刺激,电压I0V,电击时间3ms,经刺激后,将皮肤分泌物用0.9%的生理盐水冲洗至干净的容器内,12000rpm离心20min,弃上清、冷冻干燥后保存。-20℃保存备用。
第一步Sephadex G-50凝胶过滤层析:0.9g A.hainanensis背部皮肤分泌物冻干粉用10ml0.1M磷酸盐(Na2HPO4-NaH2PO4,pH 6.0)缓冲液溶解,12000rpm离心10min,取上清液上样于已平衡好的Sephadex G-50凝胶排阻色谱柱(1.6cm x 90cm,Amersham Bioscience),用同样缓冲液洗脱,流速3mL/10min,用自动部分收集器收集3mL/管,220nm检测并收集各峰 检测抗菌活性,冻干备用。
第二步反相高压液相层析(RP-HPLC)为:Sephadex G-50凝胶排阻色谱分离得到的活性成分的峰重新溶解于纯水中,4℃,12000rpm离心15min,取上清液,用0.45μm滤膜过滤,收集滤液上样于经含1‰三氟乙酸的超纯水充分平衡的C18反相柱(Hypersil BDS C18,30cm x 0.46cm)用乙腈(含1‰三氟乙酸)构成的洗脱***进行梯度洗脱,215nm检测多肽浓度。收集得到的各峰,冻干浓缩,用灭菌的去离子水重新溶解并进行抗菌活性检测。
第三步一级结构分析纯化的Hainanenin-5分子量测定采用电喷雾四级杆飞行时间串联质谱法(ESI-Q-TOF-MS,Biosystems/MDS Sciex多伦多,加拿大)。等电聚焦电泳测定等电点,用自动氨基酸测序仪测定氨基酸序列结构。
实施例2
抗微生物肽Hainanenin-5前体基因的克隆及基因测序,包括:
采用AxyPrepTM Multisource Total RNA Miniprep Kit提取海南湍蛙(Amolops hainanensis)皮肤总RNA,利用CreatorTM SMARTTM cDNA library建库试剂盒构建海南湍蛙(Amolops hainanensis)皮肤cDNA文库。 Reverse Transcriptase反转录合成cDNA第一链,引物为:
正向SMARTer V Oligonucleotide:5′-AAGCAGTGGTATCAACGCAGAGTACXXXXX-3′(X=undisclosed base in the proprietary SMARTer oligo sequence)
反向3’In-Fusion SMARTer CDS Primer:
5’-CGGGGTACGATGAGACACCATTTTTTTTTTTTTTTTTTTTVN-3’(N=A,C,G,or T;V=A,G,or C)。
利用Advantage DNA Polymerase合成第二链,引物为:正向5’-primer:5′-AAGCAGTGGTATCAACGCAGAGT-3′。反向引物同为3’In-Fusion SMARTer CDS Primer。
设计一条特异性正向引物(P1)和一条反向非特异性通用引物(3’-primer),以海南湍蛙(Amolops hainanensis)皮肤cDNA文库为模板,PCR扩增Hainanenin-5前体的cDNA。
正向P1:5’-CCAAAGATGTTSMCCWYGAAG-3’(M=A or C;W=A or T;Y=C or T)
反向非特异性通用引物为3’-primer,其序列为:5′-CGGGGTACGATGAGACACCAT-3′所获阳性单克隆进行基因核苷酸序列测定,pMDTM19-T vector测序通用引物:
正向M13F:5′-CGCCAGGGTTTTCCCAGTCACGAC-3′;
反向M13R:5′-GAGCGGATAACAATTTCACACAGG-3′;
具体步骤如下:
第一步、AxyPrepTM Multisource Total RNA Miniprep Kit提取海南湍蛙(Amolops hainanensis)皮肤总RNA(以下实验所用器具和试剂均经过处理,无RNase):
A.从新鲜宰杀的海南湍蛙(Amolops hainanensis)背部剪下一小块皮肤组织,放入液氮预冷的细胞冻存管中,然后迅速放入液氮中保存;
B.将保存在液氮中的组织材料取出,放入预冷的研钵内,迅速充分研磨,期间不断向研钵内加入少许液氮,研磨成粉末,加入400μl Buffer R-I,用注射器反复抽吸8-10次,转入1.5ml离心管(试剂盒提供)中。加入150μl Buffer R-II,漩涡振荡15-30s,12,000×g离心5min。
C.将离心后的上清转移至新的1.5ml离心管中,加入250μl异丙醇,混合均匀。将制备管置于2ml离心管(试剂盒提供)中,转移步骤4中的混合液到制备管中,6,000×g离心1min。弃滤液,将制备管置回到2ml离心管中,制备管中加入500μl Buffer W1A,12,000×g离心1min。弃滤液,将制备管置回到2ml离心管中,制备管中加入700μl Buffer W2,12,000×g离心1min;以同样的方法再用700μl Buffer W2洗涤一次。弃滤液,将制备管置回到2ml离心管中,12,000×g离心1min。将制备管放入一干净的1.5ml离心管(试剂盒提供)中,在制备管膜中央加70-100μl Buffer TE。室温静置1min,12,000×g离心1min,洗脱得RNA。
第二步、CreatorTM SMARTTM cDNA library建库试剂盒构建海南湍蛙(Amolops hainanensis)皮肤cDNA文库:
第一链的合成(mRNA反转录):
A.在新的0.2ml PCR管(无DNase和RNase)中配制下列混合液:
模板RNA 1μg
Oligo dT Primer(50μM) 1μl
dNTP Mixture(10mM each) 1μl
RNase free ddH2O补充至10μl,混匀后,短暂离心;PCR仪中65℃保温5min后,迅速在冰上急冷2min;短暂离心后,在上述管中配制如下反转录反应液:
RNase free dH2O补充至20μl体系,混匀后,短暂离心;在PCR仪中完成以下程序:42℃,60min;70℃,15min;4℃,保存。cDNA保存于-80℃。
第二链的合成:
第三步、海南湍蛙(Amolos hainanensis)抗菌肽hainanenin-5的基因克隆筛选
1、引物设计
根据已报道的蛙类抗菌肽前体的信号肽保守序列设计正向简并引物5’-CCAAAGATG TTSMCCWYGAAG-3’(M=A or C;W=A or T;Y=C or T)。引物使用前先12000rpm离心5min,然后根据标明的摩尔数加入相应体积的ddH2O溶解至20μM的浓度。
2、PCR扩增
以合成的皮肤cDNA为模板,用上述简并引物结合建库试剂盒提供的接头引物:3’-primer:5’-ATTCTAGAGGCCGAGGCGGCCGAC-3’,进行PCR扩增。在0.2ml PCR管中加入下列试剂(总体积20μl):
混匀后,短暂离心。PCR条件为:94℃变性5min;30个循环:94℃变性30s,56℃退火30s,72℃延伸40s;72℃延伸10min;4℃保存。反应结束后,取5μl产物于1%琼脂糖凝胶电泳检测目的条带。
3、目的片段回收
扩增完成后用胶回收试剂盒(天根生物)进行目的片段回收。将回收的目的DNA片段与测序载体pMDTM19-T vector连接,转化进CaCl2-MgCl2法制备好的DH5α感受态细胞。取100μl转化菌液均匀涂布在含有100μg/ml氨苄青霉素(Amp)的LB琼脂培养基平板上;表面晾干后,放在37℃恒温培养箱倒置培养12-16h,长出的菌落进行下一步菌落PCR测定。
4、目的片段序列测定
挑取单菌落做模板,以M13-F和M13-R为引物,菌落PCR检测单菌落中***片段大小。引物序列如下:
M13-F:5’-GTAAAACGACGGCCAGTG-3’
M13-R:5’-CAGGAAACAGCTATGACC-3’
挑选含有目的片段的阳性克隆送到DNA测序公司测序。测序结果与GeneBank数据库中序列进行比对。
第四步、海南湍蛙(Amolops hainanensis)抗菌肽Hainanenin-5前体的基因序列测定和结果:编码海南湍蛙(Amolops hainanensis)抗微生物肽Hainanenin-5前体的基因由321个核苷酸组成,自5’端至3’端序列为:

实施例3
抗微生物肽Hainanenin-5的化学合成方法:
1、根据推断的成熟肽Hainanenin-5氨基酸序列,用自动多肽合成仪(Applied Biosystems)合成其全序列,通过HPLC反相柱层析脱盐纯化。
2、分子量测定采用基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)。
实施例4
抗微生物肽Hainanenin-5的药理实验:
1.Hainanenin-5抗微生物活性检测:
1.1抑菌圈分析法:将化学合成的Hainanenin-5以2mg/ml的浓度溶解在无菌超纯水中。细菌接种于Luria-Bertani(LB)固体培养基上,37℃培养箱中倒置培养。待菌落长出后,用接种环挑取单菌落均匀涂布于LB固体培养基表面,将经过灭菌的直径为0.5cm的小滤纸片粘贴于培养基表面,取10μl待测样品滴加于滤纸片上。37℃培养箱中倒置培养18-20h,观察 抑菌圈形成与否,有抑菌圈形成则表示样品有抑菌活性。抑菌圈的大小可以衡量抗菌活性的强弱。将对hainanenin-5敏感的菌株记录下来。
1.2Hainanenin-5对敏感菌株最小抑菌浓度(Minimum Inhibitory Concentration,MIC)测定。
该实验以传统抗生素氨苄青霉素作阳性对照,无菌液体MH作阴性对照;最小抑菌浓度的定义为肉眼可以观察到的完全抑制微生物生长的最低多肽浓度,或者是光吸收值不高于阴性对照5%的最低浓度。
挑取新活化的微生物,接种至无菌液体MH培养基,37℃恒温振荡器中200rpm培养10-16h至对数生长期;用紫外分光光度计测菌液600nm光波处的吸光值,吸光值为1时,浓度大约为109CFU/ml,将菌液用无菌液体MH培养基稀释至106CFU/ml,冰上待用;在96微孔板上配制浓度梯度为200μg/ml、100μg/ml、50μg/ml、25μg/ml、12.5μg/ml、625μg/ml、3.13μg/ml、1.56μg/ml、0.78μg/ml、0.39μg/ml、0.20μg/ml的经0.22μm孔径膜过滤的hainanenin-5样品溶液,每孔50μl;每孔加入50μl上述稀释菌液;在恒温振荡器中37℃,100rpm振荡培养18h;使用酶标仪测600nm吸光值或肉眼观察;上述实验重复3次,取平均值。
由表1可见,Hainanenin-5对绝大多数测试菌株表现出非常强的抗菌活性,特别是对临床分离的耐药性菌株。在17种实验检测的菌株中,Hainanenin-5对绝大多数革兰氏阴性细菌(包括标准菌株和临床分离的耐药菌株)表现出极强的抗菌活性。在全部测试的5株革兰氏阳性菌(G+)菌株中,Enterococcus faecium 1299(IS),对Hainanenin-5极其敏感,最小抑菌浓度为4.69μg/ml,而阳性对照氨苄青霉素对该菌的MIC却大于100μg/ml。此外,Hainanenin-5还具有较强的抗真菌活性。对Candida albicans ATCC 2002和Slime mould 090413(IS)的最小抑菌浓度分别为2.34μg/ml和9.38μg/ml。

*MIC:最小抑菌浓度,浓度值为3次重复实验的平均值;>100μg/ml:2mg/ml剂量纸片抑菌试验有抑菌圈,100μg/ml剂量无抑菌活性;IS:临床分离菌株。以上结果为三次独立重复实验平均值。
2、Hainanenin-5的溶血活性分析
该实验以1%Triton X-100作阳性对照,以生理盐水作阴性对照。制备红细胞:取1ml人血,置于15ml离心管中,加入10ml生理盐水,混匀后2000rpm离心5min,用生理盐水重悬,重复离心至上清液不呈红色为止;用1ml生理盐水重悬红细胞沉淀,取少量重悬液加入小烧杯中,用生理盐水稀释至呈微红色;每种测试样品制备1000μl体系:
10μl多肽样品+990μl红细胞稀释液 Hainanenin-5(终浓度为20μg/ml)
10μl 1‰Triton X-100+990μl红细胞稀释液 阳性对照
10μl生理盐水+990μl红细胞稀释液 阴性对照
Hainanenin-5和阴、阳性对照各做3个重复体系。37℃温育30min,3000rpm离心5min;收集上清200μl,分别测540nm处光吸收值,计算3个重复的平均值;溶血率=(样品540nm光吸收值-阴性对照)×100%/(阳性对照-阴性对照)。
实验结果表明,在Hainanenin-5浓度为25μg/ml和50μg/ml(为最小抑菌浓度的5~10倍)时,其对人血红细胞的溶血率分别仅为13.31%和19.14%,显示其对微生物细胞的强选择性。
