Abstract

一种消灭核酸污染的方法:利用盐酸的蒸气熏蒸实验仪器、器皿、耗材、实验场地,以彻底消灭污染的DNA和RNA。然后利用洁净空气将盐酸带走,酸吸收装置将空气净化后循环使用。本方法可保证PCR、RT-PCR检测技术不会因实验仪器、器皿、耗材、实验场地存在核酸污染而出现假阳性结果,亦可使生物芯片的重复使用成为可能。

Description

一种消灭核酸污染的方法
技术领域
本发明涉及分子生物学领域,具体是一种消灭核酸污染以确保PCR、PT-PCR检测技术准确性的措施。
背景技术
PCR(polymerase chain reaction,聚合酶链反应)及其衍生的RT-PCR(reverse transcription-PCR,逆转录聚合酶链反应)是极其灵敏的分子生物学检测技术,理论上可以检测到单个的核酸分子(DNA、RNA)。但与此同时,仪器、器皿、耗材、实验场地等存在的极微小的核酸污染都可能造成假阳性结果。而PCR、RT-PCR本身又依赖于对目的核酸片段进行数十亿倍扩增,这进一步增加了污染的可能性。
现有消除核酸污染的方法主要有紫外线照射法、酸浸泡法和大量清水无限稀释法。紫外线照射法的缺点是紫外线直线传播注定了会留下大量死角。酸浸泡法的缺点是对金属腐蚀严重;结束时必须辅以水洗,而水洗可能带来新的核酸污染;废水排放后会污染环境;整个过程难以实现自动化。大量清水无限稀释法的缺点是浪费水资源;且难以保证彻底清除核酸污染。目前,我国禁止将PCR方法用于临床诊断,污染问题难以解决是主要原因之一。
发明内容
本发明的目的在于有效消灭核酸污染,从而保证PCR、RT-PCR检测技术的准确性。主要方法采用盐酸蒸气熏蒸。
具体实施方式
首先,利用盐酸的蒸气在常压、室温或稍加热(≤100℃)条件下熏蒸实验仪器、器皿、耗材、实验场地等,彻底消灭污染的DNA(脱氧核糖核酸)和RNA(核糖核酸)。本人通过实验发现:常压、室温下37.5%盐酸的饱和蒸气作用15秒即可彻底消灭污染的DNA和RNA。
然后,吹入经紫外线照射并过滤获得的洁净空气或氮气彻底带走酸熏蒸后实验仪器、器皿、耗材、实验场地残留的盐酸及其蒸气。本步骤亦可在常压、室温或稍加热(≤100℃)条件下进行。
最后,利用水吸收法或碱吸收法(碱石灰等)处理带酸的空气,使其重复利用,或多次除酸,保证无酸后放空。
经以上处理后的实验仪器、器皿、耗材、实验场地等可直接投入使用,无须进一步水洗。如有必要水洗时,可先进行水洗然后利用本发明的步骤消灭核酸污染。
本发明的优点在于:
1.盐酸蒸气可以到达每一个角落,不留死角。
2.消灭核酸污染效率高。
3.适用范围广,对于被熏蒸的物品耐高温性、耐腐蚀性要求不高。
4.适用于大型仪器、器皿、耗材、房间等核酸污染的清除,也适用于微型的生物芯片。
5.步骤简单、避免二次核酸污染。
6.易于实现自动化。
7.成本低廉。

1. 一种消灭核酸污染的方法:其特征是利用盐酸的蒸气熏蒸实验仪器、器皿、耗材、实验场地,彻底消灭污染的DNA和RNA,以确保PCR、RT-PCR检测技术的准确性。
