Description
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von 5-Fluor-2-methyl-1 -(4' -methylsulfinylbenzyli den)-3-indenyl-essigsiiure der Formel I EMI1.1 Diese Verbindung weist entzündungshemmende und fiebersenkende sowie analgetische Aktivitiit auf und diese Verbindung ist zur Behandlung derartiger Krankheiten geeignet, welche sich dadurch äussern, dass sie Symptome wie z. B. Schmerzen, Fieber oder Entziindungsprozesse zeigen. Bei der Behandlung derartiger Krankheiten kann 5-Fluor-2-methyl-1 (4'-methylsulfinylbenzyliden)-3-indenyl-essigsäure topisch, oral. rektal oder parenteral verabreicht werden, und zwar in Dosierungsbereichen, welche von etwa 0,1 mg bis 50 mg/kg Körpergewicht pro Tag erreichen und insbesondere von etwa 1 mg bis etwa 15 mg/kg Körpergewicht pro Tag. Es sei festgestellt, dass die 5-Fluor-2-methyl-1 -(4' -me- thylsulfinylbenzyliden)-3-indenyl-essigsäure nach an sich bekannten Verfahren in die cis- bzw. trans-isomere Form umgewandelt werden kann. Das cis-Isomere ist diejenige isomere Form, in welcher die Benzylidenfunktion sich unterhalb des Phenylringes des Indenkernes befindet. Es sei somit festgestellt, dass dieses cis-Isomere sowohl chemisch stabiler ist und auch biologisch wirksamer als das entsprechende trans Isomere. Des weiteren sei festgestellt, dass das in der oben angefiihrten 5 -Fluor-2-methyl-1 -(4' -methylsulfinylbenzyliden)- 3-indenylessigsäure enthaltene Schwefelatom ein asymmetrisches Zentrum darstellt und dass die Verbindung mittels Verfahren, die wohi bekannt sind, in Form einer (+) und in Form einer (-) Form erhalten werden kann. Es sei dariiber hinaus festgestellt, dass diese Verbindung polymorph ist und mehr als eine kristalline Struktur aufweist, womit es gegeben ist, dass diese Verbindung je nach ihrem Herstellungsverfahren in festem Zustand verschiedene Schmelzpunkte aufweisen kann. Bisher wurden andere 1 -Benzyliden-3 4ndenylessigsiuren hergestellt, und zwar indem man substituierte Benzaldehyde mit substituierten Essigsiiureestern in einer Claisen-Reaktion kondensierte; ein anderes Herstellungsverfahren fir derartige Verbindungen beruhte darauf, dass man einen a -halogenierten Propionsäureester mit einem substituierten Benzaldehyd in einer Reformatsky-Reaktion umsetzte. Der erhaltene unge slttigte Ester wurde reduziert und hydrolysiert, wobei sich einess-Arylpropionsäure ergab, welche einem Ringschluss unterworfen wurde, worauf man ein Indanon erhielt. Die aliphatische Säure-Seitenkette wurde mittels einer Reformatsky- oder einer Wittig-Reaktion eingefiihrt und sodann wurde der 1 -Substituent in die resultierende Indenylessigsiure oder deren Ester durch Umsetzung des jeweiligen Säure- derivates mit einem aromatischen Aldehyd oder Keton der erwiinschten Formel und anschliessendes Dehydratisieren eingefiigt, um die erwiinschte Indenylessigslure zu erhalten. Ziel der Erfindung ist also ein neues Verfahren zur Herstellung von 5-Fluor-2-methyl-l -(4' -methylsulfinylbenzyli- den)-3-indenyl-essigsäure, das dadurch gekennzeichnet ist, dass man eine Verbindung der Formeln EMI1.2 in uelchen n = Null oder 1 ist, mit einem Alkalimetallhypohalogenit behandelt. Nachfolgend werden bevorzugte Ausfiihrungsformen zur Herstellung von im erfindungsgemiissen Verfahren einsetzbaren Ausgangsverbindungen sowie des erfindungsgemässen Verfahrens beschrieben: Die Kondensation einer Verbindung der Formel II EMI2.1 in welcher X Wasserstoff bedeutet und n = 1 ist, mit Chloraceton unter üblichen Bedingungen ergibt das Indenyl(2propanon). Dieses Propanon geht eine tibliche Haloformreaktion unter Behandlung mit Natriumhypochlorit, Hypobromit oder Hypojodit ein, und zwar in einem Lösungsmittel wie z. B. Dioxan, Tetrahydrofuran oder Dimethylsulfoxyd, wobei die erwünschte Indenylessigsäure gemäss des folgenden Formelschemas gebildet wird: EMI2.2 Die folgenden Beispiele sollen Ausfuhrungsformen des vorliegenden erfindungsgemässen Verfahrens niter erläutern. Beispiel 1 Schritt A Herstellung von a -Methyl -3 -nitrozimtsiure Eine Mischung von 150 g (1 Mol) 3-Nitrobenzaldehyd, 200 g Propionsiureanhydrid und 96 g (1 Mol) Natriumpropionat werden bei 1700 C während 5 Stunden mit gelegentlichem Rfihren belassen. Das Produkt wird abgekiihlt und in 1 Liter Wasser eingegossen und durch portionsweise Zugabe von Natriumcarbonat unter Rühren stark alkalisch gemacht. Die dunkle Flussigkeit wird mit Aktivkohle gekocht bis eine abzentrifugierte Probe eine farblose aberstehende Lösung ergibt, sodann wird filtriert und mit Chlorwasserstoff siure angesiuert. Der erhaltene Niederschlag an a-Methyl-3 nitrozimtslure wird gesammelt, getrocknet und aus Ethanol umkristallisiert. Schritt B Herstellung von a -Methyl-3 -aminohydrozimtsHure Das Produkt aus Schritt A wird in 2500 ml absolutem Äthanol gelöst und bei Raumtemperatur unter einem Druck von 2,8 Atmosphären Wasserstoff reduziert, wobei ein Katalysator von Palladium auf Aktivkohle angewandt wird, und die Reduktion wird so lange fortgefiihrt, bis ein Aquivalent von 4 Molen Wasserstoff absorbiert ist. Nach Entfernung des Katalysators wird das Athanol unter vermindertem Druck abgedampft, wonach die Aminosiure kristallisiert. Schritt C Herstellung von α-Methyl-3-fluor-hydrozimtsäure 179 g (1 Mol) von a-Methyl-3-aminohydrozimtsiure werden in 2 1 4normaler Chlorwasserstoffsiure suspendiert und sodann wird auf 50 C gekiihlt. Es schliesst sich nun die Diazotierung unter Riihren an und sie wird mit einer L5- sung von 88 g Natriumnitrit in 250 ml Wasser ausgefiihrt, wobei das Reagens in einem derartigen Mass zugesetzt wird, dass jede Temperaturerhöhung vermieden wird. Zu der kla- ren Lösung von -5 C Temperatur werden 350 ml Hexafluor phosphorsaure zugegeben, und zwar in einem Zuge, und gleichzeitig etwas Trockeneis, um die Temperatur auf dem gewünschten Niveau zu halten. Der Niederschlag an Diazoniumhexafluorphosphat wird in einem Filter gesammelt und gut mit Wasser gewaschen und im Vakuum bei Raumtemperatur getrocknet. Das Salz wird in Portionen von 20 bis 30 g in 200 ml Xylol bei 125 C pyrolysiert, bis die Gasentwicklung aufhört. Die gekühlte Mischung wird sodann mit Natriumbicarbonatlösung extrahiert, um die a-Methyl-3-fluor hydrozimtsäure von neutralen Nebenprodukten abzutren nen, und der Extrakt wird mit Aktivkohle entfirbt. Aus dem farblosen Filtrat wird die niedrigschmelzende angestrebte Sauce durch Ansäuern bei 0 C und Kratzen der Gefiisswande, bis der anfänglich align Niederschlag kristallisiert, gewonnen. Schritt D Herstellung von 5-Fluor-2-methylindanon Das Produkt aus Schritt C wird mit einem lOfachen seines Gewichtes an Polyphosphorsäure bei 80 bis 90" C während 2 Stunden erhitzt. Die Fliissigkeit wird sodann in 1 Liter Eiswasser eingegossen und während 1/2 Stunde geruhrt und sodann mit drei Portionen von je 100 ml anther extrahiert. Die vereinigten Extrakte werden mit Wasser und Natrium bicarbonatlösung gewaschen bis die Atherphase neutral ist und sodann wird der Extrakt iiber Natriumsulfat getrocknet und konzentriert, wobei 5-Fluor-2-methylindanon als gelbliches ijl zurtickbleibt. Schritt E Herstellung von 5-Fluor-2-methyl -1 -(4'-methylthio- benzyl)-1 -indanol Eine Lösung von 4-Methylthiobenzyl-magnesiumbromid wird aus 45,8 g 4-Methylthiobenzylbromid (0,2 Mol) in 500 ml anther hergestellt. Diese Lösung wird tropfenweise zu einer intensiv geruhrten Lösung von 32,8 g (0,2 Mol) von 5-Fluor-2-methylindanon in 1 Liter wasserfreiem ither bei 0 C zugesetzt, wobei die Zusatzgeschwindigkeit eine derartige ist, dass der Gilman Test auf freies Grignardreagens niemals positiv ist. Schliesslich wird die kalte Mischung in 500 ml einer gesättigten wässrigen Ammoniumchloridlösung abgelöscht, die Atherphase wird abgetrennt und so lang mit Wasser gewaschen, bis sie frei von Ammoniumchlorid ist; sodann wird diese Phase tiber Magnesiumsulfat getrocknet und beim Konzentrieren unter vermindertem Druck erhält man das Indanol als Ruckstand. Schritt F Herstellung von 5-Fluor-2-methyl-1 -(4' -methyl- thiobenzyl) -inden Das Produkt aus Schritt E wird in einer Mischung von 600 ml wasserfreiem anther und 60 ml wasserfreiem Pyridin aufgenommen. Unter Rühren und Feuchtigkeitsausschluss wird die Lösung auf -10" C gekühlt und tropfenweise mit 31 g Thionylchlorid behandelt. Sobald die Reaktion voll shindig ist, lisst man die Mischung unter Rtihren sofort auf Raumtemperatur erwirmen und giesst sie sodann in 1 Liter Wasser ein. Nach innigem Vermischen wird die Atherphase abgetrennt. mit Wasser gewaschen, tiber Magnesiumsulfat getrocknet und konzentriert. Der Riickstand wird aus Hexan umkristallisiert. Schritt G Herstellung von 5-Fluor-2-methyl-1 -(4' -methyl- sulfinylbenzyliden) -3 -indenylessigsiure Eine Lösung von 7,61 g (0,1 Mol) kristalliner Glycolsiure in 120 ml Pyridin wird durch Zugabe einer 40%igen Lösung von Benzyltrimethylammoniumhydroxyd (Triton B) in Pyridin (ca. 42 ml) neutralisiert. Sodann wird 1 ml Triton B im Überschuss zugegeben und schliesslich werden 50 ml Benzol hinzugefügt. Die Lösung befindet sich in einem Reaktions gefiss, das mit einer Fraktionierkolonne und einem Stickstoffeinlass ausgestattet ist, und die erhaltene Lösung wird mit reinem Stickstoff sorgflätig von Luft befreit und schliesslich werden 33,04 g (0,1 Mol) an 5-Fluor-2-methyl-1-(4'-me- thylthiobenzyl)-inden zugegeben. Während man das Spülen mit Stickstoff aufrechterhilt, wird die Mischung so weit er wirmt, dass eine sehr langsame Destillation auftritt. Das Heteroazeotrop, das Benzol, Wasser und Pyridin enthält, kondensiert und scheidet sich in Phasen; das Erhitzen wird so lang aufrecht erhalten, bis kein Wasser mehr erscheint. Sodann wird das Benzol abgetrennt. etrennt. Die obige Lösung wird sodann auf Raumtemperatur abkiihlen gelassen, der Stickstoffzustrom wird unterbrochen und die Lösung wird unter Luftzutritt gerührt. Die Bildung von 3-Indenyl-peroxyd ftihrt sowohl zur Dehydrierung zu Fulven als auch zur Oxydation des Thioithers zu Sulfoxyd. Die Exponierung wird so lange fortschreiten gelassen, bis eine Testprobe unter Stickstoffatmosphire kein restliches Peroxyd mehr zeigt, wenn man den Jodstirketest anwendet. Das Pyridin wird sodann unter Vakuum entfernt und der Rtickstand wird mit 200 ml/einnormaler Essigsäure behandelt, wodurch rohe 5-Fluor-2-methyl-1 -(4'-methylsulfinylbenzyli- den) -3 -indenylessigsiure als Niederschlag gebildet wird, welche sodann durch Umkristallisierung gereinigt wird. Das gereinigte Produkt weist einen Schmelzpunkt von 184 bis 186"C auf. Beispiel 2 Schritt A Herstellung von 5-Fluor-2-methyl-1 -(4' -methyl- sulfinylbenzyl)-inden Zu einer Lösung von 0,1 Mol von 1-(4'-Methylthiobenzyl)-2-methyl-5-fluorinden in 100 ml Benzol werden tropfenweise unter Rühren 1086 g (0,1 Mol) von tert.-Butylhypochlorit zugesetzt. Wenn die anfinglich gelbe Färbung verschwunden ist, werden die Ldsungsmittel unter vermindertem Druck entfernt und der Sulfoxydriickstand wird direkt im nächsten Reaktionsschritt weiterverwendet. Schritt B Herstellung von 5-Fluor-2-methyl-1 -(4' -methyl- sulfinylbenzyliden) -3 -indenylessigsäure Es wird das in Beispiel 1 Schritt G angegebene Verfahren angewandt, jedoch ersetzt man die dortige Thiokomponente durch das Sulfoxyd, welches aus Beispiel 2 Schritt A erhalten worden ist, und behält im iibrigen die Arbeitsweise aus Beispiel 1 Schritt G bei. Auf diese Weise wird 5-Fluor 2 -methyl -1 -(4' -methylsulfinylbenzyliden) -3 -indenylessig siure hergestellt, und das Produkt weist einen Schmelzpunkt von 184 bis 186"Cauf. Beispiel 3 Schritt A Herstellung von 5-Fluor-2-methyl-1-(4'-methyl sulfinylbenzyl) -3 -(2' -propanon) -inden Die Kondensation von Chloraceton mit dem Produkt aus Beispiel 2 Schritt A in Gegenwart einer Base führt zum er wünschten Idenylpropanon. Schritt B Herstellung von 5-Fluor-2-methyl-1 -(4' -methyl- sulfinylbenzyliden) -3 -indenylessigsiure. 11,9 g (0,033 Mol) des Methylketons aus Schritt A werden in 100 ml Dioxah aufgenommen und mit einem 10 %igen Überschuss einer Hypochloritlösung, die 2,0 g Natriumhydroxyd enthalt, während 1 Stunde bei 35 C behandelt. Das ilberschtlssige Oxydationsmittel wird mit 10 %iger Na triumbisulfitlösung zerstört und die freie 3-Indenylessig sgure wird bei pH 2 mit Salzäure ausgefiillt. Umkristallisieren aus einem Lösungsmittelsystem, welches aus Essigsäureäthylester und Petrolliffier besteht, ergibt die Sure, welche einen Schmelzpunkt von 184 bis 186 C aufweist.
Claims (1)
- PATENTANSPRUCHVerfahren zur Herstellung von 5-Fluor-2-methyl-1-(pmethylsulfinylbenzyliden)-3-indenylessigsäure der Formel EMI4.1 dadurch gekennzeichnet, dass man eine Verbindung der Formeln EMI4.2 in welchen n = Null oder 1 ist, mit einem Alkalimetallhypohalogenit behandelt.Anmerkung des Eidg. Amtes fir geistiges Eigentum: Sollten Teile der Beschreibung mit der im Patentanspruch gegebenen Definition der Erfindung nicht in Einklang stehen, so sei daran erinnert, dass gemäss Art. 51 des Patentgesetzes der Patentanspruch für den sachlichen Geltungsbereich des Patentes massgebend ist.
