in der RI eine Hydroxy, Acyloxy-, Benzyloxy-, Cyclo- penten-(1)-yloxy-, 1-Äthoxycyclopentyloxy- oder die Te- trahydropyranyloxygr-uppe, R2 Wasserstoff oder eine Al- kyl-,
Alkenyl- oder Alkinylgruppe oder R# und R,1 zu sammen ein Sauerstoffatom darstellen, bzw. von 6-De y- droderivaten davon.
Acyloxygruppen enthalten bevorzugt den Säurerest einer gesättigten oder ungesättigten aliphatischen oder cycloaliphatischen, einer draliphatischen oder aromati schen Carbonsäure mit<B>1</B> bis 20 C-Atomen. Beispiele solcher Säure sind:
Ameisensäure, Essigsäure, Propion- säure, Buttersäure, Capronsäure, önenthsäure, Palmitin- säure, Stearinsäure, Pivalinsäure, Bernsteinsäure, Malon- säure und Benzoesäure.
Die durch das Symbol R2 dargestellten Alkyl-, Alke- nyl- und Alkinylgruppen enthalten vorzugsweise<B><I>1-5</I></B> C- .Atome. Beispiele solcher Gruppen sind:
Methyl, Äthyl, Propyl, Isopropyl, Butyl, Isobutyl, Amyl, Vinyl, Allyl, <B>1-</B> und 2-Methallyi, Äthinyl und Propargyl.
Beispiele von erfindungsgemäss erhältlichen 9ss,10a- -Sterolden der allgemeinen Formel<B>1</B> sind: l#-Cyano-17a- -methyl-17#-acetoxy-9p,lOcc-androsta-4,6-dien-3-on und l#-Cyano-17,i-methyl-17#-acetoxy-9#,10,i-androst-4-en- -3-on. oder des sen 6-Dehydroderivat mit Cyanwasscrstoff oder einem Alkalicyanid umsetzt.
Die Anlagerung von Cyanwasserstoff an die -,\1-Dop- pelbindung des Ausgangssteroids kann mittels Alkali- cyaniden, wie Kaliumcyanid, in vorzugsweise polaren or- clanischen Lösungsmitteln, wie Alkoholen, Dioxan oder Dimethylformamid,
bei erhöhter Temperatur z.B. Rück- flusstemperatur (vgl. die amerikanischen Patentschriften Nr. <B>3 088 952</B> und<B>3</B> 054<B>809),
</B> ferner mit mit in situ ge bildeten Cyanwasserstoff z.B. durch Behandlung des Ausgangssteroids mit Acetoncyanhydrin in Geg" enwart einer Base z.B. eines Alkalihydroxids oder -carbonats, vorzugsweise unter Er-wärmen,
z.B. auf Rückflusstem- peratur erreicht werden [vgl. Bulletin Soc. des Chim. France<B>(1963)</B> 24711.
Die Verfahrensprodukte sind hormonal bzw. antihor monal wirksam. Sie können als Heilmittel in Form phar mazeutischer Präparate Verwendung finden, welche sie in Mischuna mit einem für die enterale oder parenterale Applikation geeigneten pharmazeutischen, organischen oder anoroanischen inerten Trägermaterial enthalten. Die pharmazeutischen Präparate können in fester Form oder in flüssiger Form vorliegen.
Gegebenenfalls enthalten sie Hilfsstoffe, wie Konservierungs-, Stabilisierungs-, Netz- oder Emulgiermittel, Salze zur Veränderung des osmoti- schen Druckes oder Puffer. Sie können auch noch ande re therapeutisch wertvolle Stoffe enthalten.
<I>Beispiel</I> Eine Mischung von<B>5,0 g</B> 17a-Methyl-17ss-acetoxy-9#,- lk-androsta-1,4,6-trien-3-on, 2 ml Acetoncyanhydrin, <B>10</B> ml Tetrahydrofuran, <B>100</B> ml Methanol und<B>5</B> ml ge sättigter, wässeriger Natriumcarbonatlösung wurde 2V2 Stunden unter Stickstoff zum Rückfluss erhitzt. Das Reaktionsgernisch wurde abgekühlt, auf Eiswasser<B>ge-</B> gossen und mit Äther extrahiert. Die Ätherextrakte wur den mit Wasser neutral gewaschen, mit Natriumsulfat getrocknet und eingedampft.
Der Rückstand wurde auf Silicagel mit Methylenchlorid-Aceton <B>(98:2)</B> chromato- graphiert, wobei reines<B>1</B> #-Cyano- 17oc-methyl- 17p-acetoxy- -9ss,10z#-androsta-4,6-dien-3-on erhalten wurde. Schmelz punkt: 245-2470 (aus Aceton-Isopropyläther); [a]25D <B>-5180</B> (Dioxan). UV: <B>286</B> m#t; F<B>=</B> 24350.

Acyloxy groups preferably contain the acid radical of a saturated or unsaturated aliphatic or cycloaliphatic, a draliphatic or aromatic carboxylic acid with 1 to 20 carbon atoms. Examples of such acid are:
Formic acid, acetic acid, propionic acid, butyric acid, caproic acid, oenenthic acid, palmitic acid, stearic acid, pivalic acid, succinic acid, malonic acid and benzoic acid.
The alkyl, alkenyl and alkynyl groups represented by the symbol R2 preferably contain <B> <I> 1-5 </I> </B> C atoms. Examples of such groups are:
Methyl, ethyl, propyl, isopropyl, butyl, isobutyl, amyl, vinyl, allyl, 1- and 2-methallyi, ethynyl and propargyl.
Examples of 9ss, 10a- sterols of the general formula <B> 1 </B> obtainable according to the invention are: l # -cyano-17a- -methyl-17 # -acetoxy-9p, lOcc-androsta-4,6-diene- 3-one and l # -cyano-17, i-methyl-17 # -acetoxy-9 #, 10, i-androst-4-en- -3-one. The addition of hydrogen cyanide to the -, \ 1-double bond of the starting steroid can be carried out by means of alkali cyanides, such as potassium cyanide, in preferably polar solvents such as alcohols, dioxane or dimethylformamide,
at elevated temperature e.g. Reflux temperature (cf. American patent specifications <B> 3 088 952 </B> and <B> 3 </B> 054 <B> 809),
</B> also with hydrogen cyanide formed in situ, e.g. by treating the starting steroid with acetone cyanohydrin in the presence of a base, e.g. an alkali hydroxide or carbonate, preferably with heating,
e.g. can be reached at reflux temperature [cf. Bulletin Soc. of the Chim. France <B> (1963) </B> 24711.
The products of the process have a hormonal or anti-hormonal effect. They can be used as remedies in the form of pharmaceutical preparations containing them mixed with a pharmaceutical, organic or anoroanic inert carrier material suitable for enteral or parenteral administration. The pharmaceutical preparations can be in solid form or in liquid form.
They may contain auxiliaries, such as preservatives, stabilizers, wetting agents or emulsifiers, salts for changing the osmotic pressure or buffers. They can also contain other therapeutically valuable substances.
<I> Example </I> A mixture of <B> 5.0 g </B> 17a-methyl-17ss-acetoxy-9 #, -lk-androsta-1,4,6-trien-3-one, 2 ml of acetone cyanohydrin, 10 ml of tetrahydrofuran, 100 ml of methanol and 5 ml of saturated, aqueous sodium carbonate solution were refluxed under nitrogen for 2/2 hours. The reaction mixture was cooled, poured into ice water and extracted with ether. The ether extracts were washed neutral with water, dried with sodium sulfate and evaporated.
The residue was chromatographed on silica gel with methylene chloride-acetone <B> (98: 2) </B>, whereby pure <B> 1 </B> # -Cyano- 17oc-methyl- 17p-acetoxy- 9ss, 10z # -androsta-4,6-dien-3-one was obtained. Melting point: 245-2470 (from acetone isopropyl ether); [a] 25D -5180 (dioxane). UV: <B> 286 </B> m # t; F <B> = </B> 24350.
