Verfahren zur Herstellung von Dibenzo-cyclohepten-Derivaten
Die vorliegende Erfindung betrifft ein neues Verfahren zur Herstellung von Derivaten der Dibenzo-cycloheptene. Aus diesen Verbindungen können mit Aminopropyliden-substituierte Dibenzo-cycloheptene erhalten werden, welche letztere Verbindungen bei der Behandlung von Geisteskrankheiten nützlich sind, und als Antidepressants und zur Verbesserung der Gemütsverfassung oder zur psychischen Kräftigung verwendet werden. Diese Verbindungen werden vorteilhafterweise in Form ihrer sauren Salze verabreicht.
Das erfindungsgemässe Verfahren wird schematisch durch das folgende Fliessbild veranschaulicht, in welchem die gestrichelten Linien anzeigen, dass die Verbindung gesättigt (10,11-Dihydro) oder in den 10,11 Stellungen ungesättigt sein kann.

Diese Klasse von Verbindungen umfasst 5-(2'-Pro penyliden)-5H-dibenzo- [a, d]-cyclohepten, 5- (2'-Propen- yliden)-5H-dibenzo- [a, d]-l0, ll-dihydrocyclohepten und die 3-und 10-Chlor-Derivate der oben genannten Verbindungen.

Die Reaktion, wobei ein Phosphorpentahalogenid Additionskomplex, mit der dehydrogenierten Form des Ausgangsketons, gebildet wird, wird am zweckmässigsten so ausgeführt, dass man eine bestimmte Menge von Phos phorpentachlorid oder Pentabromid in trockenem Benzol zum Keton zusetzt und das Gemisch unter Rückfluss, während mehreren Stunden unter Abschluss gegen atmosphärische Feuchtigkeit, gerührt wird. Vorteilhafterweise wird eine kleine Menge an Phosphoroxychlorid in das Reaktionsgemisch einverleibt, um die Ausbeute an gewünschtem Produkt zu erhöhen. Nach Beendigung der Behandlung bei Rückfluss wird das Reaktionsgemisch auf 10 C abgeschreckt und der Additionskomplex durch Filtrieren isoliert.
Die an die obengenannte Reaktion anschliessende Hydrolyse wird vorteilhafterweise in einem wässrigen Medium durchgeführt, um das ungesättigte Keton in einer guten Ausbeute zu erhalten. In einem typischen Reaktionsgang wird der Additionskomplex durch tropfenweise Zugabe des Komplexes zu einer kräftig gerühr- ten Methanol-Wasserlösung zersetzt. Der Komplex wird vorteilhafterweise mit einer solchen Geschwindigkeit zugesetzt, welche es erlaubt, ein mildes Sieden der Lösung aufrechtzuerhalten. Schliesslich wird das kristalline Produkt bei etwa 10 C filtriert.
In der Erhitzungsstufe wird der Additionskomplex während einer bestimmten Zeitspanne, vorteilhafterweise während mindestens einer Stunde, bei erhöhter Temperatur, vorteilhafterweise bei etwa 100 C, in einem Vakuum erhitzt. Das gekühlte Reaktionsprodukt wird dann mit Essigsäure zerrieben, worauf sich das gewünschte 10-Chloro-5H-dibenzo- [a, d]-cyclohepten-5-on absetzt.
Das Produkt wird dann, zwecks Erhöhung seines Rein heitsgrades, umkristallisiert. Falls die X-und X'-Substi- tuenten andere sind als Wasserstoff, erhält man ein Gemisch der 10-und 11-Halogen-Derivate, welche dann, gewünschtenfalls mit Hilfe chromatographischer Techniken, separiert und isoliert werden können.
Das erfindungsgemässe Verfahren umfasst eine Konversion eines 5H-Dibenzo- [a, d]-cyclohepten-5-ons oder eines 5H-Dibenzo-[a, d]-10, 11-dihydrocyclohepten-5-ons in das entsprechende 5- (y-Oxypropyliden)-Derivat der Ausgangsverbindung. Das Keton wird in das entsprechende 5-Oxy-5-allyl-5H-dibenzo- [a, d]-cyclohepten oder das 10,11-Dihydro-Derivat desselben durch Behandlung mit Allylmagnesiumhalogenid überführt. Nach der Dehydration, wobei ein Molekül Wasser entfernt wird, entsteht das Dien, nämlich das 5- (2'-Propenyliden)-5H-di- benzo- [a, d]-cyclohepten oder das Dihydro-Derivat desselben.
Schliesslich wird das Dien in die entsprechende 5-y-Oxypropyliden-Verbindung, durch Behandlung mit einem gegebenenfalls substituierten Borwasserstoff konvertiert, wobei ein Wassermolekül unter Sprengung der Enddoppelbindung addiert wird. Sobald die Oxyverbindungen erhalten werden, können sie in die nützlichen Aminopropyliden-Verbindungen durch Halogenierungsund Aminierungsstufen konvertiert werden.
Die Konversionsstufe umfasst eine neue Kondensation des 5-Ketons mit einem Grignard-Reagens, hergestellt von einem Allyl oder einem substituierten Allylhalogenid, wie z. B. Allylbromid, unter Bildung der entsprechenden 5-Oxy-5-allyl-Zwischenverbindung. In einem typischen Gang wird 5H-Dibenzo- [a, d]-10,11-dihydrocyclohepten mit dem Grignard-Reagens, dem Allyl magnesiumbromid, in trockenem Äthyläther bei Zimmertemperatur umgesetzt. Nach einer geeigneten Reaktionsdauer wird das Reaktionsgemisch hydrolysiert und die Oxyverbindung mittels einer Extraktionstechnik isoliert.
In der nächsten Stufe wird die 5-Oxy-5-allyl-Verbindung dehydratisiert, wobei sich die entsprechende 5- (2'- Propenyliden)-Zwischenverbindung, z. B. 5- (2'-Propen- yliden)-5H-dibenzo- [a, d]-10,11-dihydrocyclohepten, bildet. Am geeignetsten kann die Entfernung des Wassers so erfolgen, dass man einfach die Oxy-Zwischenverbindung im Vakuum destilliert, obgleich auch andere Techniken, wie eine sauer katalysierte Dehydratation mit Jod oder p-Toluolsulfonsäure in einem inerten Lösungsmittel, wie Benzol, gleichfalls verwendet werden kann.
Das Propenyliden-Zwischenprodukt wird dann in das entsprechende 5-(y-Oxypropyliden)-cyclohepten durch Behandlung mit Borwasserstoff des Propenyliden-Zwischenproduktes konvertiert. Zum Beispiel gelangt man nach Behandlung von 5- (2'-Propenyliden)-5H-dibenzo- [a, d]-10,11-dihydrocyclohepten in Tetrahydrofuran-Lö- sung mit einem Mol-Äquivalent eines Borans und nachfolgender oxydativer Hydrolyse, zum entsprechenden 5- (y-Oxypropyliden)-5H-dibenzo-[a,d]-10,11-dihydrocyclohepten. Geeignete Borane enthalten mindestens eine B-H-Bindung im Molekül, wie z. B. Diboran, Aminborane, Alkylborane, Arylborane, Alkyl-Arylborane und Boranaluminiumalkoxyde.
Beispiel 1 5- (y-Oxypropyliden)-5H-dibenzo- [a, d]-
10,11-dihydrocyclohepten
In einen 125 ml-Dreihalskolben, der ausgeflammt und unter trockenem Stickstoff gekühlt wurde, und mit einem Rührer, Zuflusstrichter und einem Kondensator vom Ather-Typus versehen war, wurden 4,8 g reiner Magnesiumspäne und 15 ml trockener Athyläther gegeben. Danach wurden 17 g Allylbromid in 10 ml trokkenem Ather unter Rühren mit genügender Geschwindigkeit zutropfen gelassen, um das Gemisch bei mildem Rückfluss aufrechtzuerhalten. Das Rühren und die Rück- flussbehandlung wurden bis zum vollständigen Auflösen des Metalls fortgesetzt.
Danach wurde das Reaktionsgemisch unterhalb dem Rückflusspunkt gekühlt, aber nicht so tief, um das Grignard-Reagens ausfallen zu lassen, wonach 10 g 5H-Dibenzo- [a, d]-10,11-dihydrocyclohepten-5-on in 20 ml trockenem Ather unter Rühren während 15 Minuten zugefügt werden. Das Reaktions genrsch wird dann gerührt, während einer halben Stunde bei Zimmertemperatur reagieren gelassen, dann in einem Eisbad abgeschreckt und mit 45 ml gesättigter Ammo niumchloridlösung behandelt. Die Schichten werden hernach separiert und es wird gerade so viel Wasser zugesetzt, um die festen Salze in der wässrigen Schicht zu lösen.
Die letztere wird dann mit 2 X 25 ml Äther extrahiert und die vereinigten organischen Schichten mit 25 ml gesättigter Salzlösung gewaschen und über Magnesiumsulfat getrocknet ; nach Entfernung des Lösungs- m tels im Vakuum erhält man das Oxy-Zwischenprodukt : A CH301I 240u, E /o 495. max
Dieses 5-Oxy-5-allyl-5H-dibenzo- [a, d]-10,11-dihydrocyclohepten wird im Hochvakuum bei 1 mm destil- liert und man erhält das 5- (2'-Propenyliden)-5H-dibenzo [a, d]-10,11-dihydrocyclohepten als viskose Flüssigkeit :

Eine Lösung von 0,5 g des oben erzeugten Diens in 15 ml Tetrahydrofuran wurde mit einem Moläquivalent von Bis-3-methyl-2-butylboran bei 0-5 C während 3 Stunden behandelt. Nach Abschluss dieser Zeitspanne fügte man 3 ml Wasser, hernach 8 ml 2,5-n Natriumhydroxyd zu und liess anschliessend 6-7 ml von 30"/o- igem Wasserstoffperoxyd tropfenweise zufliessen. Die wässrige Phase wird dann mit Kaliumcarbonat ausgesalzen, die Tetrahydrofuranschicht separiert, über Magnesiumsulfat getrocknet und das Lösungsmittel im Vakuum abgedampft. Der Rückstand wird aus Ather-Petrol äther kristallisiert und liefert die gewünschte y-Oxy-Verbindung mit F = 88-89 C.
Beispiel 2
Unter Befolgung des oben im einzelnen beschriebenen Verfahrens und unter Verwendung gleichwertiger Mengen an 5H-Dibenzo-[a, d]-cyclohepten-5-on gelangt man zum entsprechenden 5-Oxy-5-allyl-5H-dibenzo- [a, d]-cyclohepten.
Gemäss dem oben beschriebenen Verfahren und unter Benützung gleichwertiger Mengen an 5H-Dibenzo [a, d]-cyclohepten-5-onen und 5H-Dibenzo- [a, d]-10,11 dihydrocyclohepten-5-onen, die mit X, X'und Y-Substituenten besetzt sind, und deren Bedeutung oben angeführt wurde, werden auch andere entsprechende 5-Oxy 5-allyl-cycloheptene erzeugt.
Nach Destillieren im Hochvakuum bei 1 mm, z. B. von 5-Oxy-5-allyl-5H-dibenzo-[a, d]-cyclohepten, erhält man durch Behandlung mit Borwasserstoff gemäss Bei spiel 1, unter Verwendung äquivalenter Mengen obiger Verbindung, das entsprechende 5- (y-Oxypropyliden)-5H- dibenzo- [a, d]-cyclohepten.

The present invention relates to a new process for the preparation of derivatives of the dibenzo-cycloheptenes. The reaction, in which a phosphorus pentahalide addition complex is formed with the dehydrogenated form of the starting ketone, is most conveniently carried out by adding a certain amount of phosphorus pentachloride or pentabromide in dry benzene to the ketone and refluxing the mixture for several hours Completion against atmospheric moisture, is stirred. The product is then recrystallized to increase its degree of purity. If the X and X 'substituents are other than hydrogen, a mixture of the 10- and 11-halogen derivatives is obtained, which can then be separated and isolated, if desired with the aid of chromatographic techniques.
The inventive method comprises a conversion of a 5H-dibenzo- [a, d] -cyclohepten-5-one or of a 5H-dibenzo- [a, d] -10, 11-dihydrocyclohepten-5-one into the corresponding 5- (y -Oxypropylidene) derivative of the starting compound. The ketone is converted into the corresponding 5-oxy-5-allyl-5H-dibenzo- [a, d] -cycloheptene or the 10,11-dihydro derivative thereof by treatment with allyl magnesium halide. After dehydration, in which one molecule of water is removed, the diene is formed, namely 5- (2'-propenylidene) -5H-dibenzo [a, d] -cycloheptene or the dihydro derivative thereof.
Finally, the diene is converted into the corresponding 5-γ-oxypropylidene compound by treatment with an optionally substituted hydrogen boride, a water molecule being added with the end double bond being broken. Once the oxy compounds are obtained, they can be converted to the useful aminopropylidene compounds through halogenation and amination steps.
The conversion step involves a new condensation of the 5-ketone with a Grignard reagent made from an allyl or a substituted allyl halide such as e.g. B. allyl bromide, with formation of the corresponding 5-oxy-5-allyl intermediate. In a typical course, 5H-dibenzo- [a, d] -10,11-dihydrocycloheptene is reacted with the Grignard reagent, the allyl magnesium bromide, in dry ethyl ether at room temperature. After a suitable reaction time, the reaction mixture is hydrolyzed and the oxy compound is isolated using an extraction technique.
In the next step, the 5-oxy-5-allyl compound is dehydrated, whereby the corresponding 5- (2'-propenylidene) intermediate compound, e.g. B. 5- (2'-propenylidene) -5H-dibenzo- [a, d] -10,11-dihydrocycloheptene. Most conveniently, water removal can be accomplished by simply distilling the intermediate oxy compound in vacuo, although other techniques such as acid catalyzed dehydration with iodine or p-toluenesulfonic acid in an inert solvent such as benzene can also be used.
The propenylidene intermediate is then converted to the corresponding 5- (γ-oxypropylidene) cycloheptene by treatment of the propenylidene intermediate with hydrogen boride. For example, after treatment of 5- (2'-propenylidene) -5H-dibenzo- [a, d] -10,11-dihydrocycloheptene in tetrahydrofuran solution with one mole equivalent of a borane and subsequent oxidative hydrolysis, the corresponding 5- (γ-oxypropylidene) -5H-dibenzo- [a, d] -10,11-dihydrocycloheptene. Suitable boranes contain at least one B-H bond in the molecule, such as. B. diborane, amine boranes, alkylboranes, arylboranes, alkyl-arylboranes and borane aluminum alkoxides.
Example 1 5- (y-oxypropylidene) -5H-dibenzo- [a, d] -
10,11-dihydrocycloheptene
To a 125 ml three-necked flask which had been flame-out and cooled under dry nitrogen and fitted with a stirrer, feed funnel and an ether-type condenser was placed 4.8 g of pure magnesium turnings and 15 ml of dry ethyl ether. Thereafter, 17 g of allyl bromide in 10 ml of dry ether was added dropwise with stirring at a rate sufficient to maintain the mixture under mild reflux. Stirring and reflux treatment were continued until the metal was completely dissolved.
The reaction mixture was then cooled to below reflux, but not so low as to precipitate the Grignard reagent, after which 10 g of 5H-dibenzo- [a, d] -10,11-dihydrocyclohepten-5-one in 20 ml of dry ether be added with stirring over 15 minutes. The reaction is then stirred, allowed to react for half an hour at room temperature, then quenched in an ice bath and treated with 45 ml of saturated ammonium chloride solution. The layers are then separated and just enough water is added to dissolve the solid salts in the aqueous layer.
The latter is then extracted with 2 X 25 ml of ether and the combined organic layers washed with 25 ml of saturated brine and dried over magnesium sulfate; after removal of the solvent in vacuo, the intermediate oxy product is obtained: A CH301I 240u, E / o 495. max
This 5-oxy-5-allyl-5H-dibenzo- [a, d] -10,11-dihydrocycloheptene is distilled in a high vacuum at 1 mm and the 5- (2'-propenylidene) -5H-dibenzo [ a, d] -10,11-dihydrocyclohepten as a viscous liquid:

A solution of 0.5 g of the diene produced above in 15 ml of tetrahydrofuran was treated with one molar equivalent of bis-3-methyl-2-butylborane at 0-5 ° C. for 3 hours. At the end of this period of time, 3 ml of water were added, then 8 ml of 2.5N sodium hydroxide and then 6-7 ml of 30% hydrogen peroxide were added dropwise. The aqueous phase is then salted out with potassium carbonate and the tetrahydrofuran layer is separated , dried over magnesium sulfate and the solvent evaporated in vacuo. The residue is crystallized from ether-petroleum ether and gives the desired γ-oxy compound with F = 88-89 C.

By following the procedure detailed above and using equivalent amounts of 5H-dibenzo- [a, d] -cyclohepten-5-one, the corresponding 5-oxy-5-allyl-5H-dibenzo- [a, d] -cyclohepten.
According to the method described above and using equivalent amounts of 5H-dibenzo [a, d] -cyclohepten-5-ones and 5H-dibenzo- [a, d] -10,11 dihydrocyclohepten-5-ones, which are denoted with X, X 'and Y substituents are occupied, and the meaning of which has been given above, other corresponding 5-oxy 5-allyl-cycloheptenes are also produced.
After distilling in a high vacuum at 1 mm, e.g. B. of 5-oxy-5-allyl-5H-dibenzo- [a, d] -cyclohepten, is obtained by treatment with hydrogen boride according to Example 1, using equivalent amounts of the above compound, the corresponding 5- (y-oxypropylidene) -5H- dibenzo [a, d] cycloheptene.
