Verfahren zur Herstellung neuer 17-Cycloalkenyläther von 17ss-Hydrogysteroiden der Androstanreihe Es ist bekannt [Acta Chimica Academiae Scientia- rum Hungarice, Bd. 1 (1951), 281], dass man durch Behandlung eines 3-Hydroxysteroids mit einer funk tionellen Verbindung des Cyclohexanons,
wie Dialkyl- acetal oder Alkylenoläther, einen Steroid-3-(1'-cyclo- hexenyl)-äther erhält. Solche Verbindungen, die aus dem hormonalen Gesichtspunkt wenig interessant sind, wurden als Zwischenverbindungen zur Synthese von physiologisch wirksamen Substanzen, insbeson dere zur Herstellung des Testosterons, verwendet.
Es wurde nun gefunden, dass man durch Behand lung eines 17ss-Hydroxysteroids mit einer funktionel len Verbindung des Cyclopentanons oder des Cyclo- hexanons neue Steroidverbindungen erhält, die eine wertvolle, perorale, hormonale Wirkung besitzen, die die der entsprechenden 17ss-Hydroxysteroide oft über trifft.
Gegenstand vorliegender Erfindung ist ein Ver fahren zur Herstellung neuer 17-(1'-Cycloalkenyl)- äther von 17f-Hydroxysteroiden der Formel

Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet, dass man das entsprechende 17ss-Hydr- oxysteroid, worin die gegebenenfalls vorhandene 3-Hydroxygruppe in Ester- bzw.
Ätherform maskiert ist, mit einer funktionellen Verbindung des Cyclo- pentanons oder des Cyclohexanons bei einer Tempe ratur über 80 C umsetzt.
Als funktionelle Verbindung des Cyclopentanons bzw. Cyclohexanons kann man ein Dialkylacetal oder einen Alkylenoläther, vorzugsweise das Dünethyl- bzw. Diäthylacetal oder den Methyl- bzw. Äthylenol- äther oder auch Gemische derselben verwenden.
Sol che funktionelle Verbindungen können nach bekann ten Methoden hergestellt werden, indem man das Cyclopentanon oder Cyclohetanon mit den geeigneten Orthoameisensäurealkylestern umsetzt.
Die Verätherung in 17-Stellung kann sowohl in Gegenwart von geeigneten organischen Lösungs- mitteln, sowie auch in Abwesenheit von Lösungs mitteln durchgeführt werden. Besonders geeignete Lösungsmittel sind diejenigen mit Siedepunkt über 80 C, insbesondere Benzol, Toluol, Dioxan, Isooctan, Dimethylformamid.
Im allgemeinen ist die Reaktion verhältnismässig kurz und ist nach 15- bis 60-minütigem Erhitzen be endet. Die Umsetzung kann beschleunigt werden, indem man in Gegenwart eines sauren Katalysators, wie eine starke, organische Säure oder das Salz einer starken Säure mit einer organischen Basis, z. B. Ben zol- bzw. p-Toluolsulfonsäure oder Pyridinchlor- hydrat, arbeitet.
Der bei der Reaktion entstehende 17-Cyclo- alkenyläther kann durch vorzugsweise im Vakuum durchgeführtes Eindampfen des Lösungsmittels, ge gebenenfalls nach Abstumpfen des sauren Kataly- sators, isoliert werden; durch nachstehende Umkristal- lisierung kann man das Produkt in reinem Zustand erhalten.
Wenn das Endprodukt eine veresterte 3-Hydroxy- gruppe enthält, kann es gegebenenfalls einer Hydro- lyse im alkalischen Medium unterworfen werden;
unter solchen Bedingungen ist der 17-ständige ,Äther beständig, während der 3-ständige Ester hydrolysiert wird, wobei der 17ss-Cycloalkenyläther eines 3,17-Di- hydroxysteroids entsteht.
Die neuen, erfindungsgemäss herstellbaren, 17- Cycloalkenyläther besitzen wertvolle Hormonaleigen- schaften; solche Eigenschaften sind in den Cyclopen- tenyläthern besonders hervorragend.
Die 17-Cyclo- pentenyläther des Testosterons, Androstan-17ss-ol-3- ons, Androst-l-en-17ss-ol-3-ons, Androstan-3a,17ss- diols und Androst-4-en-3ss, 17ss-diols stellen beispiels weise stark androgene, per os wirksame Verbindungen dar; ihre Wirksamkeit ist gleich und oft auch über legen gegenüber der Wirkung des Methyltestosterons.
Die 17-Cyclopentenyläther des 19 Nortestosterons, 19-Norandrost 5(10)-en-17ss-ol-3-ons, Androsta-1,4- dien-17ss-ol-3-ons, 2-Hydroxymethylen-dihydrotesto- sterons und 17ss-Hydroxy-androstano[3,2c]isoxazols sind gegenüber den entsprechenden, freien 17ss-Hydr- oxysteroiden überlegen, weil sie, ausser einer erhöh ten peroralen Wirkung,
ein günstigeres Verhältnis zwischen der anabolischen und androgenen Wirkung besitzen; z. B. weisen die 17-Cyclopentenyläther des 19-Nortestosterons, 19 Norandrost-5(10)-en-17ss-ol- 3-ons und Androsta-1,4-dien-17ss-ol-3-ons ein etwa 4- bis 14-fach höheres Verhältnis Anabol/Androgen- Wirkung als Methyltestosteron nach;
ferner haben die neuenVerbindungen gegenüber dem Methyltestosteron den Vorteil einer reineren physiologischen Wirkung, die von der Abwesenheit der 17-ständigen Methyl- gruppe abhängt. <I>Beispiel 1</I> 150 cm' Cyclopentanon werden mit 285 cm' Orthoameisensäureäthylester, 306 cmg absolutem Äthanol und 1,425 g p-Toluolsulfonsäure eine Stunde unter Rückfluss gekocht, dann abgekühlt und mit 1,5g Kaliumcarbonat behandelt.
Durch Abdestillieren im Vakuum erhält man eine Mischung von Diäthyl- acetal und Äthylenoläther des Cyclopentanons mit Siedepunkt von<B>68-71'</B> C/20 Torr. 5 g Testosteron werden in 10 cm-3 der so erhaltenen Mischung suspen diert und bei 145 C 3 Stunden gekocht, dann wird die Temperatur auf 180-190 C gesteigert und das Kochen wird fortgesetzt, indem man gleichzeitig ab destilliert. Der Rückstand wird mit einigen Tropfen Pyridin enthaltendem Methanol aufgenommen.
Durch Eindampfen im Vakuum erhält man den 17-Cyclo- pentenyläther des Testosterons, der, nach Umkristalli- sierung aus Methanol bei 128-130 C schmilzt; [a]D = + 100 (Dioxan).
In der Tabelle I werden einige, wie oben herge stellte 17-Cyclopentenyläther von 17ss-Hydroxyste- roiden zusammengefasst.

So erhält man den Cyclo- pentenyläther des 9a-Fluor-11ss-hydroxy-testosterons, F.
200-204 C, [a]D = +93 (Dioxan). <I>Beispiel 3</I> Eine Mischung von 3 g 19-Nortestosteron und 10 cm3 der wie im Beispiel 1 erhaltenen Mischung von Diäthylacetal-Äthylenoläther des Cyclopentanons wird 30 Minuten bei 145 C erwärmt, dann steigert man die Temperatur auf 180-190 C und destilliert, bis man einen Rückstand erhält, der mit verdünntem Methanol aufgenommen wird.
Die so erhaltene Mi schung liefert den 17-Cyclopentenyläther des 19- Noriestosterons; F. 102-105 C, [a]D = +50 (Di- oxan).
Auf gleiche Weise erhält man den Cyclopentenyl- äther des 19-Norandrost-5(10)-en-17ss-ol-3-ons; F. 152-154 C, [a]D = +163 (Dioxan).
<I>Beispiel 4</I> 3 g 2-Hydroxymethylen-5a-androstan-17ss-ol-3- on werden mit 5 cm3 Cyclopentanondimethylacetal in 5 cm3 Dimethylformamid in Gegenwart von 30 mg p. Toluolsulfonsäure umgesetzt. Das Reaktions- gemisch wird eine Stunde bei 180 C gekocht, dann mit einigen Tropfen Pyridin neutralisiert und im Va kuum eingedampft.
Der aus Methanol umkristalli- sierte Rückstand liefert den 17-Cyclopentenyläther des 2-Hydroxymethylen-5a-androstan-17ss-ol-3-ons, F. 168-173 C, [ab = +58 (Dioxan). <I>Beispiel 5</I> 3 g 17ss-Hydroxy-androstano[3,2c]isoxazol wer den mit 5 cm3 Cyclopentanondiäthylacetal in 5 cm3 Dimethylformamid, in Gegenwart von 30 mg p-Tolu- olsulfonsäure behandelt.
Man kocht die Mischung 30 Minuten bei 180-181' C, dann neutralisiert mit wenigem Pyridin, dampft im Vakuum ein und nimmt den Rückstand mit einer Mischung Methylenchlorid- Methanol auf. So erhält man den 17-Cyclopentenyl- äther des 17-ss-Hydroxy-androstano[3,2c]isoxazols, F.
126-128 C, [a]D = +64 (Dioxan). <I>Beispiel 6</I> 3 g 3a-Acetoxy-5a-androstan-17ss-ol werden mit 5 cm3 Cyclopentanondiäthylacetal in 5 cm3 Dimethyl- formamid in Gegenwart von 40 mg Benzolsulfon- säure umgesetzt. Man kocht 30 Minuten bei 180 C, dann neutralisiert mit wenigem Pyridin, dampft im Vakuum ein und nimmt den Rückstand mit einer Mischung Methylenchlorid-Methanol auf.
So erhält man den 17-Cyclopentenyläther des 3a-Acetoxy- androstan-17ss-ols, F. 134-136 C, [a]D = +32 (Dioxan). Auf gleiche Weise erhält man den 17-Cy- clopentenyläther des 3a-Propionoxy-5a-androstan- 17ss-ols, F. 91-92 C, [a]D = +35 (Dioxan).
1,5 g des wie oben erhaltenen 17-Cyclopentenyl- äthers des 3a-Acetoxy-5a-androstan-17ss-ols werden in 50 cm3 Methanol gelöst und 2 Stunden in Gegen wart von 0,5 g Kaliumcarbonat in 5 cm3 Wasser ge kocht.
Die im Vakuum eingedampfte Lösung liefert den 17-Cyclopentenyläther des 5a-Androstan-3a,17ss- diols, F. 145-147 C, [a]D = +30 (Dioxan). Durch Arbeiten unten den oben beschriebenen Bedingungen wird der 17-Cyclopentenyläther des 3ss- Propionoxy-androst-5-en-17ss-ols, F.
77-79 C, [a.]D = -34,5 (Dioxan) hergestellt, der, durch Hydrolyse in alkalischem Medium, den 17-Cyclopentenyläther des Androst-5-en-3ss,17ss-diols, F. 115-117 C, [a]D _ -32 (Dioxan), liefert.
Durch Arbeiten unter den oben beschriebenen Bedingungen erhält man neue 17-Cyclopentenyläther, die in der Tabelle II zusammengefasst sind.

Auf gleiche Weise erhält man den 17-Cyclo- hexenyläther des 3ss-Acetoxy-androst-5(6)-en-17ss- ols, F. 142-144 C, [a]D = -26,4 (Dioxan), der, durch Hydrolyse mit wässrigem Kaliumcarbonat, wie im Beispiel 6 beschrieben, den 17-Cyclohexenyläther des Androst-5(6)-en-3ss,17ss-diols, F. 152-155 C, [a]D = -19 (Dioxan), liefert.
<I>Beispiel 8</I> 5 g Androst-2-en-17ss-ol werden mit 10 cm3 Cy- clopentanondiäthylacetal in 10 cm3 Dimethylform- amid in Gegenwart von 50 mg Pyridinchlorhydrat umgesetzt. Die Mischung wird eine Stunde bei 180 C erhitzt,
dann mit wenigem Pyridin neutralisiert und im Vakuum eingedampft. Der aus verdünntem Me- thanol aufgenommene Rückstand liefert den 17-Cy- clopentenyläther des Androst-2-en-17ss-ols.
<I>Beispiel 9</I> Eine Mischung von 2 g 19-Norandrost-4-en-17ss- ol, 5 cm3 Cyclopentanondiäthylacetal, 25 mg p-Tolu- olsulfonsäure und 15 cm3 Dimethylformamid wird 30 Minuten bei 190 C erhitzt, dann mit wenigem Py- ridin neutralisiert und im Vakuum eingedampft.
Der aus verdünntem Methanol aufgenommene Rückstand liefert den 17-Cyclopentenyläther des 19-Norandrost- 4-en-17ss-ols.

The 17-cyclopentenyl ethers of 19 nortestosterone, 19-norandrost 5 (10) -en-17ss-ol-3-one, androsta-1,4-dien-17ss-ol-3-one, 2-hydroxymethylene dihydrotestosterone and 17ss-Hydroxy-androstano [3,2c] isoxazoles are superior to the corresponding, free 17ss-hydroxy steroids because, in addition to an increased oral effect, they

<I> Example 4 </I> 3 g of 2-hydroxymethylene-5a-androstan-17ss-ol-3-one are mixed with 5 cm3 of cyclopentanone dimethylacetal in 5 cm3 of dimethylformamide in the presence of 30 mg of p. Toluenesulfonic acid implemented. The reaction mixture is boiled for one hour at 180 C, then neutralized with a few drops of pyridine and evaporated in vacuo.

126-128 C, [a] D = +64 (dioxane). <I> Example 6 </I> 3 g of 3a-acetoxy-5a-androstan-17ss-ol are reacted with 5 cm3 of cyclopentanone diethyl acetal in 5 cm3 of dimethylformamide in the presence of 40 mg of benzenesulfonic acid. The mixture is boiled for 30 minutes at 180 ° C., then neutralized with a little pyridine, evaporated in vacuo and the residue is taken up in a methylene chloride-methanol mixture.
This gives the 17-cyclopentenyl ether of 3a-acetoxy-androstan-17ss-ol, mp 134-136 C, [a] D = +32 (dioxane). The 17-cyclopentyl ether of 3a-propionoxy-5a-androstan-17ss-ol, mp 91-92 C, [a] D = +35 (dioxane) is obtained in the same way.
1.5 g of the 17-cyclopentenyl ether of 3a-acetoxy-5a-androstan-17ss-ol obtained as above are dissolved in 50 cm3 of methanol and boiled in 5 cm3 of water for 2 hours in the presence of 0.5 g of potassium carbonate.
The solution evaporated in vacuo gives the 17-cyclopentenyl ether of 5a-androstane-3a, 17ss-diol, mp 145-147 C, [a] D = +30 (dioxane). By working under the conditions described above, the 17-cyclopentenyl ether of 3ss-propionoxy-androst-5-en-17ss-ol, F.
77-79 C, [a.] D = -34.5 (dioxane) produced, which, by hydrolysis in an alkaline medium, the 17-cyclopentenyl ether of androst-5-en-3ss, 17ss-diol, F. 115-117 C, [a] D_ -32 (dioxane) provides.
By working under the conditions described above, new 17-cyclopentenyl ethers are obtained, which are summarized in Table II.

The 17-cyclohexenyl ether of 3ss-acetoxy-androst-5 (6) -en-17ss-ol, m.p. 142-144 C, [a] D = -26.4 (dioxane), is obtained in the same way , by hydrolysis with aqueous potassium carbonate, as described in Example 6, the 17-cyclohexenyl ether of androst-5 (6) -en-3ss, 17ss-diol, F. 152-155 C, [a] D = -19 (dioxane) , delivers.
<I> Example 8 </I> 5 g of androst-2-en-17ss-ol are reacted with 10 cm 3 of cyclopentanone diethyl acetal in 10 cm 3 of dimethylformamide in the presence of 50 mg of pyridine chlorohydrate. The mixture is heated for one hour at 180 C,
then neutralized with a little pyridine and evaporated in vacuo. The residue taken up from dilute methanol yields the 17-cyclopentyl ether of androst-2-en-17ss-ol.
<I> Example 9 </I> A mixture of 2 g of 19-norandrost-4-en-17ss-ol, 5 cm3 of cyclopentanone diethyl acetal, 25 mg of p-toluenesulfonic acid and 15 cm3 of dimethylformamide is heated at 190 ° C. for 30 minutes, then neutralized with a little pyridine and evaporated in vacuo.
The residue taken up from dilute methanol gives the 17-cyclopentenyl ether of 19-norandrost-4-en-17ss-ol.
