Description
Method of making new . 17- (1'-Cycloalkenyl) -ether of 17fl-hydroxysteroids of the androstane series It is known (Acta Chimica Academiae Scientiarum Hungaricae, Vol. 1, 195 1, p. 28 1) that by treating a 3-hydroxysteroids with a functional Compound of cyclohexanone, such as a dialkyl acetal or alkylenol ether, a steroid 3- (1'-cyclohexenyl) ether is obtained. Such compounds, which are of little interest with regard to their hormonal action, were used as intermediate compounds for the synthesis of physiologically active substances, in particular for the production of tegtosterone.
It has now been found that by treating a 17β-hydroxysteroids with a functional compound of cyclopentanone or cyclohexanone new Receives steroid compounds that have a valuable, peroral, hormonal effect, which often exceeds that of the corresponding 17β-hydroxysteroids.
The present invention relates to a process for the preparation of the new 17- (1'-cycloalkenyl) ethers of 17β-hydroxysteroids of the general formula in which X is a methylene or ethylene group and S is the radical denotes where Y is hydrogen or methyl, R hydrogen, an alkyl or acyl group and the dashed lines denote the possible presence of one or at most two double bonds. The process according to the invention consists in reacting the corresponding 17fl-hydroxysteroid, in which the optionally present 3-hydroxy group is expediently masked, with a functional compound of cyclopentanone or cyclohexanone at a temperature above 80 ° C , optionally a 3- Acyloxy compound hydrolyzed in an alkaline medium or, if appropriate, a 3-ketone compound obtained is reduced with a metal hydride.
As a functional compound of cyclopentanone or cyclohexanone using a dialkyl acetal or an alkylenol ether, preferably the dimethyl or diethylacetal or the methyl or. Ethylenol ether or mixtures thereof. Such functional compounds can be produced by known methods, by mixing the cyclopentanone or cyclohexanone with the appropriate alkyl orthoformate implements.
The etherification in the 17-position, which proceeds according to known methods, can be carried out either in the presence of suitable organic solvents or in the absence of solvents. Particularly suitable solvents are those with a boiling point above 80 ° C., in particular benzene, toluene, dioxane, isooctane, dimethylformamide.
In general, the reaction is relatively short and is complete after 15 to 60 minutes of heating. The reaction can be accelerated by reacting in the presence of an acidic catalyst such as a strong organic acid or a salt of a strong acid with an organic base, e.g. B. benzene or p-toluenesulfonic acid or pyridine chlorohydrate works.
The 17-cycloalkenyl ether formed at the end of the reaction is through Evaporation of the solvent, preferably in vacuo, if necessary after blunting of the acidic catalyst is isolated, by subsequent recrystallization the Product received in the pure state.
The new 17-cycloalkenyl ethers of the present invention have valuable hormonal properties, in particular they have a superior androgenic and, i or anabolic effect compared to the corresponding 17fl-hydroxysteroids or . a more favorable ratio between the anabolic and androgenic effects when they are also administered orally. Such properties are particularly excellent in the cyclopentenyl ethers.
der Methode von H e r s b e r g e r (Proceedings of the Society for
Experimental Biology and Medicine, Bd. 83, 1953, S. 175) berechnet wurden;
Anabol-Androgen-Wirkung in der Kolonne B/A gegeben.
B. British Medical Journal, Bd. 1, 1959, S. 259, Gastroenterology, Bd. 31, 1956, S. 384; Journal of the American Medical Association, Bd. 169, 1959, S. 1453). Another advantage of the new compounds obtainable according to the invention over methyltestosterone and other orally active compounds is the lack of the 17a methyl group. It is known that methyltestosterone can cause functional disorders, particularly of the liver, disorders that are consistently attributed to the 17α-methyl group (see, e.g., British Medical Journal, Vol. 1, 1959, p. 259, Gastroenterology, Vol . 31, 1956, p. 384; Journal of the American Medical Association, Vol. 169, 1959, p. 1453). The following examples explain the process according to the invention. Protection is not sought in the context of the present invention for the preparation of the derivatives of cyclopentanone or cyclohexanone used as starting compounds.
Durch Eindampfen im Vakuum erhält man den 17-Cyclopentenyläther des Testosterons, der, nach Umkristallisieren aus Methanol, bei 128 bis 130' C schmilzt, [al # = + 100' (Dioxan).Example 1 150 cm3 of cyclopentanone are refluxed for 1 hour with 285 cm3 of ethyl orthoformate, 306 cm3 of absolute ethanol and 1.425 g of p-toluenesulfonic acid, then cooled and treated with 1.5 g of potassium carbonate. Distilling off in vacuo gives a mixture of diethyl acetal and ethylenol ether of cyclopentanone with a boiling point of 68 to 71 ° C./20 Torr. 5 g of testosterone are suspended in 10 cm3 of the mixture obtained in this way and boiled at 145 ° C. for 3 hours, then the temperature is increased to 180 to 190 ° C. and the boiling is continued by distilling off at the same time.
