Process for the production of 17-propadiene steroids
This invention relates to the conversion of 17a-dihalocyclopropyl steroids into new steroids with useful hormonal effects, substituted in the 17sc-position by two-fold ethylenically unsaturated radicals.
Swiss Patent No. 492689 describes the production of new 17; / L-dihalocyclopropyl steroids by reacting a 17α-alkenyl steroid with a dihalocarbene made from sodium trichloroacetate, sodium chlorodifluoroacetate or sodium dichlorofluoroacetate.
The present invention relates to a method for
Production of steroids of the androstane, 19-nor-androstane or estran series substituted in the 17cc position by a 170c-propadienyl radical, characterized in that a corresponding 17-dihalocyclopropyl steroid is in an organic solvent which is inert with respect to the reaction with magnesium, potassium tert-butoxide, butyllithium or sodium methylsulfinylcarbanion.
The implementation can be represented by the following scheme:
EMI1.1
wherein each of the substituents X and Y is chlorine or fluorine.
In particular, according to the method, steroids of the formula: (11)
EMI1.2
produced, in which R2 is hydrogen, methyl, ethyl or n-propyl. Rs hydrogen, tetrahydropyran-2-yl, tetrahydrofuran-2-yl or an acyl group derived from a hydrocarbon carboxylic acid, R4 hydrogen, methyl or chlorine and A a group of the formula:
EMI2.1
where R is hydrogen or methyl, Rl is hydrogen, lower alkyl, tetrahydropyran-2-yl, tetrahydrofuran-2-yl, cyclopentyl, cyclohexyl or an acyl group derived from a hydrocarbon carboxylic acid and R5 is an oxygen atom or a group of the formula:
EMI2.2
wherein R6 is hydrogen, tetrahydropyran-2-yl, tetrahydrofuran-2-yl or an acyl group derived from a hydrocarbyl carboxylic acid.
The acyl group derived from a hydrocarbyl carboxylic acid and the acyloxy group in the preferred compounds contain fewer than 12 carbon atoms and can be straight, branched, cyclic or cycloaliphatic in structure. This structure can be saturated, unsaturated or aromatic and optionally substituted by functional groups such as hydroxyl, alkoxy containing up to 5 carbon atoms, acyloxy containing up to 12 carbon atoms, nitro, amino, halogen and the like. Typical ester groups are therefore acetate, propionate, enanthate, benzoate, trimethyl acetate, tert-butyl acetate, phenoxy acetate, cyclopentyl propionate, amino acetate, 13-chloropropionate. Adamantoate and the like.
The lower alkyl groups represented by R1 in the above formula may be straight or branched. Typical alkyl groups are methyl, ethyl, isopropyl, butyl and the like, which can contain up to about 7 carbon atoms.
In practicing the above method, a steroid having the dihalocyclopro acyl group at the 17sG position can be treated with magnesium in an inert reaction medium such as diethyl ether, tetrahydrofuran, dioxane, or the like to produce the 17oc-diene steroid. The reaction can be carried out at room temperature or higher for a period on the order of 2 to 24 hours, preferably at reflux temperature for about 2 hours.
The compounds can also be prepared by adding a 17ss-dihalocyclopropyl steroid at room temperature or higher for about 2 to 24 hours or more in an organic solvent which is inert with respect to the reaction, such as, for example, dimethyl sulfoxide, diethyl ether, tetrahydrofuran, dioxane or the like , treated with a molar excess, preferably with about 3 to 10 molar equivalents, of potassium tert-butoxide or sodium methylsulfinylcarbanion.
Another procedure for the preparation of the compounds consists in the 17, K-Dihalogencyclopro- py starting material at a temperature of about -400C to 0C for a period of the order of 1 to 24 hours or more, preferably at about -350C for about 1 hour, in an organic solvent which is inert with respect to the reaction, such as, for example, dioxane, tetrahydrofuran, hexane or the like, with a molar excess, preferably with about 2 to 10 molar equivalents, of butyllithium.
In practicing the above methods, the substituents on the steroid nucleus in positions 3 and 17 are preferably a hydroxyl group, an ether group, or an ester group. In the case of the preparation of the new A4 compounds of the formula II, the 3-keto-h4 starting material can be converted into the ketal or the enol ether before the conversion of the 17x-dihalocyclopropyl group into the 17a-propadienyl group and the ketal or the enol ether after Reaction can be converted into the 3-I (eto -A4 derivative by treatment with acid.
The substituent at the 3-position of the A4 and h5 "10" steroid nucleus can also be an ester group, e.g. an acetate group, a benzoate group, etc., or an ether group, e.g. a tetrahydropyran-2-yl-ether group or a tetrahydrofuran-2-yl-ether group. which are preferably formed before the conversion of the 17-dihalocyclopropyl group into the 17z-propadienyl group. The substituent at the 17 position of the steroid starting material may, prior to the preparation of the compounds of Formula II, be a hydroxyl group, an esterified hydroxyl group e.g. Acetoxy, benzoyloxy, etc., or an etherified hydroxyl group, e.g. Tetrahydropyran-2-yloxy or tetrahydrofuran-2-yloxy, preferably an esterified or etherified hydroxyl group.
The compounds of the formula II in which R is hydrogen, or in other words the 19-nor-A4- -17ez-dienes, can also be prepared by reacting the A5'1) -17¯-dienes of the formula II with a weak Treated acid or base to move the double bond from the 5 (10) position to the 4 position.
The new steroids of the formula II in which A is a group of the formula:
EMI2.3
represents, exhibit estrogenic activity and are useful for treating various conditions for which such agents are indicated, such as estrogen deficiency, climacteric, and the like. These compounds can also be used in veterinary medicine in the same way as known estromens and for contraception and fertility control. In addition, these agents show an anti-androgen effect. They can be administered in the usual pharmaceutical forms at dosages appropriate for the condition being treated.
The new steroids of the formula II in which A is a group of the formula:
EMI3.1
or the formula:
EMI3.2
represents, show corpus luteum hormone action and are useful for the treatment of various menstrual disorders and for contraception and fertility control. They can be administered in the usual pharmaceutical preparations at dosages that are suitable for the condition being treated.
The following examples illustrate the invention.
Example I.
To a suspension of 1217c - (2 ', 2'-dichlorocyclopropyl) -17) - (tetrahydropyran-2'-yloxy) oestr-4-en-3-one in 7.5 cm3 of anhydrous, peroxide-free dioxane 1.2 cm3 of freshly distilled ethyl orthoformate and 0.8 g of p-toluenesulfonic acid are added. The mixture is stirred for 15 minutes at room temperature and left to stand for a further 30 minutes. Then 0.8 cm3 of pyridine and then water are added until solidification occurs. The solid obtained is collected by filtration, washed with water and air dried to give 3-ethoxy- 17 ± - (2 ', 2'-dichlorocyclopropyl) -17o- (tetrahydropyran-2' -yloxy) -östra- 3.5 diene.
A mixture of 1 g of the enol ether prepared above, 4 molar equivalents of sodium methylsulfiny1carbanion and 40 cm3 of dimethylsulfoxide is left to stand under nitrogen at room temperature for 24 hours.
The reaction mixture is then poured into water and extracted with methylene chloride. The extracts are washed with water, dried and evaporated and give 3-ethoxy-17; c-propadienyl-17ss- (tetrahydropyran- -2'-yloxy) -estra-3,5-diene.
A few drops of 36% hydrochloric acid are added to a solution of 1 g of the 3-ethoxy-17z-propadienyl compound prepared above in 10 cm3 of acetone. The mixture is heated to steam bath temperature for a few minutes, diluted with water and filtered. The solid substance collected in this way is dried and recrystallized from a mixture of acetone and hexane, 170: - -propadienyl-17ss-hydroxy-oestr-4-en-3-one being obtained.
By repeating the above procedure using 17ss- (2 ', 2'-dichlorocyclopropyl) - 17ss- (tetrahydropyran-2'- -yloxy) -1 8-ethylostr-4-en-3-one, 17, sc- (2 ', 2'-Dichloro-3' -methylcyclopropyl) -1 7) - (tetrahydropyran-2'-yloxy) -estr-4-en-3-one, 17a- (2 ', 2'-dichlorocyclopropyl ) - 17B- (tetrahydropyran-2'- -yloxy) - 1 8-methylostr-4-en-3-one, 17.a- (2 ', 2'-dichlorocyclopropyl) - 17o- (tetrahydropyran-2'- - yloxy) -18-methylandrost-4-en-3-one or 17a- (2 ', 2'-dichlorocyclopropyl) - 17p- (tetrahydropyran-2' -yloxy) -androst-sen-3-one as starting material is 17?
; Propadienyl-17ss-hydroxy-18-äthylöstr-4-en-3-one, 17, oc- (3'-methylpropadienyl) -17,8-hydroxyöstr-4-en-3-one, 17-propadienyl 17p- hydroxy- 1 8-methylöstr-4-en-3-one, 17, x-propadienyl- 17p-hydroxy- 18 -methyl-androst-4-en-3-one or
17α-propadienyl-17ss-hydroxyandrost-4-en-3-one.
Example 2
A mixture of 1 g of 17a- (2 ', 2'-dichlorocyclopropyl) - -17,3- (tetrahydropyran-2'-yloxy) -estr-4-en-3-one, 25 cm3 of dry benzene, 5 cm3 of ethylene glycol and 50 mg of p-toluenesulfonic acid monohydrate is refluxed for 4 hours using a water separator.
The reaction mixture is then washed with aqueous sodium bicarbonate solution and water, dried and evaporated to dryness, whereby 3-ethylenedioxy-17M- - (2 ', 2' - dichlorocyclopropyl) - 17p - (tetrahydropyran 2 '- yl-oxy) oestr-5 -en is obtained, which can be recrystallized from a mixture of acetone and hexane.
To a solution of 1 g of the compound prepared above in 40 cm3 of anhydrous ether, 3 molar equivalents of butyllithium in anhydrous ether are added dropwise over a period of 10 minutes with stirring, while the reaction temperature is kept at 350 ° C. The reaction mixture is left to stand for 1 hour at a temperature of -350.degree. The reaction mixture is then allowed to warm to room temperature, poured into water and extracted with methylene chloride. The extracts are washed with water, dried and evaporated to dryness and give 3-ethylenedioxy-17-propadienyl-1 78- (tetrahydro-pyran-2'-yloxy) -ostr-5-ene.
A mixture of 0.5 g of the 17x-propadienyl compound prepared above in 30 cm3 of acetone and 50 mg of p-toluenesulfonic acid is left to stand for 15 hours at room temperature. It is then poured into ice water and extracted with ethyl acetate. These extracts are washed neutral with water, dried over sodium sulfate and evaporated to dryness. The residue is triturated with ether and yields 17-propadienyl-17,3-hydroxyestr-4-en-3-one, which is recrystallized from a mixture of acetone and hexane.
Example 3
To a refluxing solution of 1 g of 17α- (2 ', 2'-dichlorocyclopropyl) -estr-4-en-17p-ol-3-one. 1.3 g of magnesium turnings and 25 cm3 of anhydrous ether are added to 5.7 g of ethyl bromide over a period of 1 hour. When the addition is complete, the reaction mixture is refluxed for a further hour and then carefully hydrolyzed by the dropwise addition of water and then 10% hydrochloric acid. The ether layer is separated off, dried and evaporated and gives 17oc-propadienyl-oestr-4-en-17p-ol-3-one.
Similarly, subjecting other 17α- (2 ', 2'-dichlorocyclopropyl) compounds to the procedure of this example, the corresponding 17, oc- -propadienyl derivatives are obtained. Example 4
A mixture of 1 g of 17a- (2 ', 2'-dichlorocyclopropyl) - -1 7P- (tetrahydropyran-2'-yloxy) - 18 -methylestr-4-en-3p-ol, 4 molar equivalents of potassium tert-butoxide and 35 cm3 of dimethyl sulfoxide is left to stand under nitrogen for 24 hours at room temperature. The reaction mixture is then poured into water and extracted with methylene chloride. The organic extracts are washed with water, dried and evaporated and yield 17% - propadienyl - 17,, 8 - (tetrahydropyran-2'-yloxy) -18-methylestr-4-en-3f3-ol.
Example 5
A mixture of 1 g of 17a- (2 ', 2'-dichlorocyclopropyl) - -17ss- (tetrahydropyran-2'-yloxy) oestr-4-en-3ss-ol, 4 molar equivalents of sodium methylsulfinylcarbanion and the like. 40 cm3 of dry dimethyl sulfoxide is left to stand under nitrogen for 24 hours at room temperature. The reaction mixture is then poured into water and extracted with methylene chloride. The organic extract is washed several times with water, dried and evaporated to give 17α-propadienyl-17ss- (tetrahydropyran-2'-yloxy) -ostr-4-en-3p-ol.
Similarly, the procedure of this example can be used to convert other 17a-dichlorocyclopropyl compounds described above to the corresponding 17, sc-propadienyl derivatives.
