EMI0001.0049
in der R einen gegebenenfalls- substituierten;

Process for the production of SS-lactams The subject of the main patent is a process for the production of SS-lactams of the formula
EMI0001.0007
in which R is a hydrocarbon radical and R1, R2 and R3 are hydrocarbon radicals and or
or hydrogen atoms, with agents having a reducing effect on the corresponding ss lactam-N-sulfochlorides, optionally in the presence of acid-binding agents.
It is known that salts of ß-amino (N - sulfonic acid) carboxylic acids are obtained when ss-lactam N sulfochloride is saponified with lye:
EMI0001.0031
It is also known that β-oxycarboxylic acid amides are obtained if the hydrolysis of the β-lactam sulfochlorides is carried out by heating in an aqueous solution without simultaneously neutralizing the mineral acid liberated:
EMI0001.0042
In an embodiment of the process of the main patent, it has now been found that one ss lactams of the formula
EMI0001.0049
in which R is an optionally substituted;
Hydrogen radicals and R1, R2 and R3 denote hydrocarbon radicals and / or hydrogen atoms, can advantageously also be produced by the corresponding ß-lactam N-sulfochloride within a pH range of 1 to 12, preferably at pH values of 2 up to 10, at temperatures from 0 to 60 C, preferably at 20 to 30 C,
using acid-binding substances in the sense of the equation
EMI0002.0003
hydrolyzed to the free fl-lactams without using reducing agents.
In. In the formulas, the radicals R, R1, R2 and R3 preferably have the following meanings: R can be a saturated alkyl radical with in particular 1 to 10, preferably 1 to 6, carbon atoms, which can also be branched, an aromatic hydrocarbon radical,. in particular a phenyl radical,
which can also be substituted by low molecular weight alkyl or alkoxy groups with preferably 1 to 3 carbon atoms, halogen atoms or a nitro group or a nitro group and a halogen atom, the remainder
EMI0002.0032
or an alkyl group substituted by an esterified carboxyl group, such as e.g.
B. the remainder -CH2 COO-Cä H2- + 1 where n can assume values from 1 to 20, be. R1, R2 and. R3 can for one. Hydrogen atom or
EMI0002.0049
to have. However, a hydrogen atom can also be used instead of the -S02C1 group.
The ss lactam N sulfochlorides used as starting materials can, for example, by conversion of olefins of the structure
EMI0002.0064
EMI0002.0065
obtainable intermediates by reaction with carbomethylene:
Medium and optionally in the presence of solubilizing a saturated alkyl radical with in particular 1 to about 10 carbon atoms, which can also be branched, are, however, in the event that R is an aromatic radical, R1, R2 and R3 must be hydrogen atoms and where, in the event that R is an aliphatic radical, R1 must also be an alkyl radical,
Furthermore, two of the radicals R, R1, R2 and R3 can be linked to form a cycloaliphatic ring with 5 to 8, in particular 6, carbon atoms in the ring. It is also possible for the grouping
EMI0002.0089
twice in the same molecule of the: ss-lactam N-sulfochloride used as the starting material. In this case, R can e.g.
B. the meaning of a radical of the constitution where R, R1, R2 and R3 have the abovementioned meanings, can be obtained with N-carbonylsulfamic acid chloride ..
ss-lactam N-sulfochlorilde of the above constitution, in which R is an optionally substituted aromatic radical or the radical
EMI0002.0108
is, for example, from aromatic aldehydes by way of the melschema according to the formula
EMI0003.0001
The compounds described in the main patent are also suitable as ss-lactam-N-sulfochlorides.
The reaction according to the invention is advantageously carried out by introducing the ß-lactam-N-sulfochloride partially or continuously into water or part of a hydrolysis mixture from a previous batch with constant stirring and the hydrochloric acid and sulfuric acid released by adding acid-binding substances
continuously binds. It is advisable to add a suitable buffer salt to the reaction mixture at the beginning, such as B. sodium acetate or citrate or disodium phosphate, advantageously in amounts of about 0.02 to.
1 mole of the salt per mole of ß-lactam-N-sulfochloride and then adds the acid-binding agent in the further course of the reaction while continuously monitoring the pH value and ensures that the temperature does not exceed 60 ° C. by suitable cooling of the reaction mixture .
In some cases, in which the hydrolysis in the aqueous medium proceeds at a rate that is too slow for practical needs, a small amount of a water-soluble inorganic iodide, in particular an alkali iodide, can advantageously be added to the reaction mixture.
The iodides generally come. in catalytic amounts of about 0.001 to 0.1, preferably 0.005 to 0.01 mol of iodide per mol of lactam N-sulfochloride for use.
The implementation can especially if. the β-lactams obtained as end products are sparingly soluble in water, instead of being carried out in a purely aqueous phase, they are advantageously carried out in water-containing, low-molecular Al alcohols. As alcohols come men in particular those with 1 to 4 carbon atoms, such as.
B. methanol, ethanol, propanol, isopropanol or mixtures thereof in any ratio, into consideration. The alcohol content in the alcohol / water mixture can be up to 95 percent by volume.
Alkali metal or alkaline earth metal hydroxides, carbonates or bicarbonates, in particular the corresponding potassium and sodium compounds, can preferably be used as acid-binding agents. However, organic, especially tertiary, bases, such as trimethylamine, triethylamine or pyridine, can also be used.
When working in an aqueous reaction medium, three equivalents of the bases are required per mole of lactam sulfochloride. When working in higher percent alcoholic solutions, on the other hand, due to the more or less predominant formation of alkylsulfuric acid, even smaller amounts of acid-binding agents, namely 2 to 3 equivalents, are sufficient.
The hydrolysis in the sense of the formation of the free lactam takes place particularly easily with the 4,4-dimethylazeddin-one- (2) -N-sulfochloride obtained from isobutylene and -N-carbonylsulfamic acid chloride. But even in this case it turns out that the hydrolysis to the ß-lactam is much faster than at the beginning,
As long as almost pure water is still present as the hydrolyzing agent, then - for example, when using caustic soda or soda as an acid-binding agent - sodium chloride and sodium sulphate have formed in sufficient concentration that conditions are achieved in the aqueous solution , in which the formed 4,4-dimethylazetidin-one (2) or
part of it separates out as a second liquid phase. In this lactamp phase, the ss-lactam N-sulfochloride to be hydrolyzed is now much more readily soluble than in water.
Since the lactam phase which has separated out also contains a considerable amount of water in dissolved form, the hydrolysis of the lactam-N-sulfochloride now takes place predominantly in this layer in a homogeneous phase.
The β-lactam formed during the hydrolysis of the ß-lactam-N-sulfochloride thus accelerates the reaction due to its property of acting as a solubilizer. This effect can now be used with particular advantage when performing the method according to the invention by
that instead of pure water, part of the reaction mixture from a previous batch is submitted. It is then expedient to keep the salt concentration approximately constant by a suitable choice of the ratio of the water to be added in the course of the hydrolysis and the acid-binding agent.
This achieves a maximum hydrolysis rate right from the start, so that the hydrolysis of a given amount of lactam N-sulfochloride can be carried out in a period of a few minutes to half an hour, as long as it is possible to dissipate heat from the reaction apparatus allow:
Instead of the lactam, suitable solubilizing inert organic solvents, such as e.g. B. dimethylformamide, methylacetamide or tetrahydrofuran are added.
The indifferent organic solvents are used mixed with water, the mixture being able to consist of up to 90 parts by volume of the organic solvent or a mixture of different indifferent organic solvents.
When substituted by higher alkyl radicals ss lactam-N-sulfochlorides, such as. B. in the case of the ss lactam-N-sulfochondes obtainable from 2-methyl-pentene (1) or diisobutylene, the hydrolysis within the temperature range to be maintained is initially only so slow in water that it is better to add to the water from the start certain amount,
preferably about 5 to 10% of the amount of the β-lactam to be produced; from an earlier approach as a solubilizing component.
In those ss-lactam-N-sulfochlorides, the lactams of which are crystallized and only slightly soluble in water, such as the 8 lactam N-sulfochlorides obtainable from styrene or tetramethylethylene,
As solubilizing substances, either other liquid ß-lactams obtainable by the claimed process, such as 4,4-dimethylazetidin-one (2), and / or
or with particular advantage other substances known as solubilizers, such as dimethylformamide, N-methylacetamide or tetrahydrofuran, are added.
The isolation of the β-lactams from the reaction mixture is very simple per se. In some cases, the β-lactams crystallize out of the reaction mixture, depending on the selected solvent, by themselves, or they separate out as oils which can be separated from the solvent in a simple manner.
If appropriate, some of the salts formed in the reaction can also be separated off by cooling before the lactams are separated off from the reaction mixture. The ss lactams can also - by extraction with suitable <RTI
ID = "0004.0079"> Solvents, such as methylene chloride, diethyl ether, carbon tetrachloride, ethyl acetate or chloroform., Are removed from the reaction mixture.
Ethyl acetate and methylene chloride have proven particularly effective here. After the solvent has been distilled off, the 8 Laotams remain as oily or crystalline products that are more or less easily soluble in water, which are produced by distillation under reduced pressure or
can be purified by recrystallization.
The process products can be used as intermediate products for the manufacture of textile auxiliaries, pharmaceuticals; Pesticides and plastics are used.
A particularly advantageous use of the SS lactams of the present invention, in particular the lower alkyl radicals containing the SS lactams, is their use as selective solvents for acetylene.
When the loadability is high, the lactams mentioned are of particular industrial importance, for example for the separation of ethylene / acetylene mixtures.
For example, 1 cm3 (that is 0.976 g) of 4,4-dimethyl azetidin-one (2) at 20 C and an atmosphere 1.75 0.1 cms of ethylene (the ethylene volume was at 0 C and 760 Torr measured) and 14.0 0.1 cm3 acetylene, which corresponds to a separation factor of 8.
1 cm3 (that is 0.956 g) of the 3,4,4-trimethyl-azetidin-one- (2) dissolves at 25 C and one atmosphere 1.85 0.1 cm3 ethylene and 29.0 0.2 cm3 acetylene, which corresponds to a separation factor of 15: 1 for the acetylene / ethylene separation.
The low volatility of these ss lactams, together with the high separation factor, is of particular value for the separation of ethylene / acetylene mixtures.
Example l In. a solution of 14 g of sodium acetate in 250 cm3 of water is added with stirring at 25 to 30 ° C. in part a total of 395 g (2 times) of 4,4-dimethylazetidin-one- (2) -N-sulfochloride, and the pH is maintained the reaction mixture by simultaneous dropwise addition of 10 lye between .4 and 6.
When about half of the ß-lactam-N-sulfochloride has reacted in the course of an hour, the hydrolysis of the second half of the ß-lactam-N-sulfochloride takes place much more quickly and is completed within 15 minutes with increased cooling at the same time .
To bind the hydrochloric and sulfuric acid released, 600 cm-3 lOn sodium hydroxide solution are used. Stirring is continued for a short time at 30 ° C. until the mixture, which has been adjusted to pH 7, shows practically no change in its pH value.
When the stirring is stopped, the reaction mixture forms a two-phase liquid mixture, the ss-laetam formed being mainly contained in the lighter, usually somewhat bleached, liquid layer.
If the reaction mixture is cooled to about 10 ° C. while stirring, most of the sodium sulfate, which has a salting effect on the ß-lactam gus, separates out as decahydrate, and the separated ß-lactam dissolves.
After the excreted salt has been suctioned off and pressed off, the ss lactam is removed from the aqueous salt solution using methylene chloride.
After analogous work-up, about 180 g, that is about 90% of theory, of 4,4-dimethylazetidin-one (2) (dimethylpropiolactam) are obtained as a colorless liquid which is clear in water and has a boiling point of 65 ° C. at 0.2 Torr.
If, instead of the water, part of the two-phase liquid reaction mixture from a previous batch is presented,
the hydrolysis of the lactam N sulfochlorides takes place at high speed from the start. To keep the salt concentration constant, it is neutralized with about 7N sodium hydroxide solution and the reaction can be completed at 25 C in about half an hour or, if the cooling capacity is adequate, even in a shorter time
to lead.
Example 2 A total of 225.5 g (1 mol) of 4-methyl-4-n-propylazetidin-one- (2) -N-sulfochloride [from 2-methylpentene- (1) and N-carbonylsulfamic acid chloride] partially at 30 to 40 C with stirring in a solution of 100 cm3 of water and 14g of sodium acetate and binds the acid released by adding 5N sodium hydroxide solution,
keeping a pH of 5 to 7. After 2 hours, only about 70 ems of the 5N sodium hydroxide solution are consumed. Soon, after that, the ss-lactam formed begins to separate out as a lighter layer, and further hydrolysis now takes place much faster,
so that despite stronger cooling and a reaction temperature of 25 C, the sulfochloride can be introduced into the mixture more quickly and after a further hour the theoretical consumption of 600 cm3 5N sodium hydroxide solution is reached.
The water-soluble 4-methyl-4-n-propylazetidin o @ n- (2) is extracted from the reaction mixture with methylene chloride. After distilling off the solvent, distillation of the residue at 0.2 torr and 75 ° C. gives 110 g of the product as a colorless, glycerol-like liquid.
The hydrolysis of the ss lactam N-sulfochloride proceeds at a greater rate from the start if, instead of the water, part of the two-phase reaction mixture from a previous batch is submitted.
Example 3 A solution of 10 g of anhydrous sodium sulfate in 40 cm3 of water and 20 g of 4,4-dimethylazetidm-one- (2) (dimethylpropiolactam) is added at 30 to 35 ° C. with stirring proportionally 25 g of 4-phenylazetidin-one- (2) -N-sulfochloride and binds the released mineral acid by dropping a total of 60 cm3 of 5N sodium hydroxide solution,
A pH of 8 to 10 is maintained in the. The splitting off of the acid is complete after about 1 hour.
50 cm3 of methylene chloride are added and the two-phase mixture, which separates only poorly due to the formation of an emulsifying by-product, is filtered off over a layer of activated carbon, washed with methylene chloride, the methylene chloride layer is then separated off,
the solvent evaporates and the 4,4-dimethylazetidin-one- (2) used as a solubilizer is distilled off at 0.2 torr and a bath temperature of 100.degree.
About 5 g of 4-phenylazetidin-one- (2), which melts at 108 to 109 C after recrystallization from dilute methanol, remain as residue. The melting point of the mixture with a comparator preparation shows no depression.
The reaction takes place in the same way if, in the above example, the mixture of sodium sulfate solution and dimethylpropiolactam is replaced by a mixture of 20 cm3 of dimethylformamide and 20 cm3 of water or by a mixture of 20 cm3 of tetrahydrofuran and 20 cm3 of water and to bind the released Mineral acid used at a pH value of 7 to 9 30 cm3 10N sodium hydroxide solution.
Example 4 A mixture of 114 g (1 mol) of 2-methyl-propen- (2) -1-carboxylic acid methyl ester C H3 CH2 = C-CHZ- COOCH3 150 cm3 of diethyl ether and 142 g (1 mol) N-carbonyl sulfamic acid chloride O = C = N-SO2C1 is initially kept at 30 C for 6 hours by cooling and towards the end by gentle heating.
The homogeneous, non-crystallizing reaction mixture then contains, in addition to ether, essentially 4 "methyl-4-carbomethoxymethylazetidine-bn- (2) @ -N-sulfochloride.
To convert the sulfochloride into 4-methyl-4-carbomethoxymethylazetidinane (2), the reaction mixture described above is added dropwise to a solution of 20 g of sodium acetate in 200 cm3 of water over the course of one hour with constant stirring and with cooling at 20 ° C. and the pH is maintained -Value of the hydrolysis mixture by adding a total of about 300 cm3 10N sodium hydroxide solution at 5 to 7.
The hydrolysis of this / 3'-L; actam-N-sulfochloride is evidently due to. the somewhat hydrophilic ester group extremely smoothly and without any delay.
The hydrolysis mixture obtained, on which a layer of partially precipitated ß-lactams floats, is seeded with sodium sulfate decahydrate, cooled to about 10 ° C., from which Na2S04-10H2O is sucked off. The filter residue is washed with methylene chloride and the filtrate is repeatedly shaken out with methylene chloride.
After the solution has been distilled off, the combined methylene chloride extracts leave behind an oily residue which passes over at 0.1 torr at 115 to 116 ° C. as a colorless, crystallizing oil. The yield is 80 g.
The product dissolves in 150 cm3 of warm carbon tetrachloride and, after adding 200 cm3 of ether, provides about 70 g of coarse crystals of 4-methyl-4-carbomethoxymethylazetidinone- (2) when it cools down.

from melting point 74 to 75 C.
It is easily soluble in methylene chloride, - less easily in carbon tetrachloride, slightly soluble in cold ether, insoluble in cyclohexane.
The solution of the compound in concentrated hydrochloric acid heats up spontaneously.
Example 5 In 1500 cm3 of methanol, to which a solution of 8 g of potassium iodide in 20 liters of water was added, 1200 g of 4-phenylazetidine are added partially over the course of 2 hours at 30 ° C. (with cooling) on (2) -N-sulfocblorid entered and bound with 12N sodium hydroxide solution, the acid released.
Lye is added so that a pH of 1.5 to 2.0 is maintained in the solution. The pH range is continuously monitored with the aid of an electrical pH measuring device. The total amount of lye required is around 810 cm3. After the ss-lactam N-sulfochloride has reacted,
traces of free iodine are bound by adding a little sodium bisulfite, the reaction mixture is adjusted to pH 7 and heated to 60 C. 2.7 l of water are added to the mixture at this temperature.
On cooling, the 4-phenylazetidin-one- (2) crystallizes out of the clear solution, which is filtered off with suction and washed with water until it is free from chores. After drying, 510 g of 4-phenyl azetidin-one- (2) are obtained as a colorless crystal powder. From the remaining mother liquor, 30 to 40 g of the, B lactam can still be obtained by extraction.
The above procedure has proven to be particularly advantageous in the preparation of the f-lactams substituted by aromatic hydrocarbon radicals.
